This thesis deals with the determination and evaluation of antibacterial properties of coatings and surface finishing. The thesis verified antibacterial, physico-mechanical and chemical properties of water-based coatings themselves with added antibacterial particles. For better orientation in issue of this thesis are generally described coatings, antibacterial substances and tested bacteria. The aim is to determine the effect on the properties of coatings with added antibacterial substances. How effective is the resistance of the tested coatings with added ingredients to the action of bacteria and fungi as well as to assess whether they are suitable for use in surface finishing furniture parts and whether they have the expected properties.

During the evolution some groups of organisms have become coevolutionary associated with other groups, as is the case of host symbiont systems. To explore coevolutionary history of hosts and their associated symbionts, phylogenetic reconstruction of symbionts and phylogenetic reconstruction of hosts are usually compared. Coevolution is described by coevolutionary events (cospeciation, host switch, duplication, failure to diverge events and linage sorting events). The aim of this work was to test the suitability of MLST method for the complex of entomopathogenic nematodes from the genus Steinernema (with detailed analysis of Steinernema feltiae) and their symbiotic bacteria Xenorhabdus bovienii and subsequently use cophylogenetic comparative analysis to determine their level of cospeciation.

The diploma thesis deals with the antibiotics policy and is focused on resistance and mechanism of action of the antibiotics. A great attention is also paid to novel substances testing in terms of their impact on the selected bacteria. Theoretical part of thesis describes the system of antibiotic policy including its structure and organization in the Czech Republic. It also briefly summarizes this issue globally as well as individual organizations involved in the organization of the antibiotic policy. The theoretical part is also concerned with the history, classification, effects and significance of the antibiotics. A particular attention is paid to the chapter describing the macromolecular mechanism of action of 2 selected groups of substances (antibiotics) on bacteria, including physiological functions of some bacterial structures. This thesis primarily focuses on one segment of freshwater Betaproteobacteria, the Limnohabitans genus (including the RBT lineage). As opposed to other recent research directions, the major aim was to recover the members of the previously uncultured RBT lineage through their isolation from various freshwater habitats. However, the results presented in this thesis have also ambitions to go far beyond the taxonomic descriptions only; the dissertation intends to contribute significantly to unveiling of important ecophysiological characteristics of the studied lineage in a set of both laboratory and field research. Therefore, understanding of growth characteristics, mortality, diversity and life strategies of aquatic microbes is of highest importance regarding profound human impact on water quality and increasing need of drinking water supplies.

Since the introduction of advanced molecular methods the research on insect bacterial symbioses underwent a major focus shift towards large scale phylogenetics and comparative genomics. These new fields provided answers to several fundamental questions of symbiont evolution, functional capabilities of the host-associated bacteria, and the role of symbionts in the host?s biology. However, the vast diversity and complexity of symbiotic relationships still leaves gaps in our understanding to a rich mosaic of various symbiont types, effects and transitions from facultative association to obligate mutualism. The presented study focuses on distribution, diversity, phylogenetic patterns, evolutionary transitions and genome evolution of two less known but ecologically diverse bacterial genera, Arsenophonus and Sodalis. The thesis also takes advantage of the knowledge on a well established symbiotic model between aphids and Buchnera and reveals several evolutionary patterns in the host and symbiont.

The object of the dissertation was the monitoring of the quantity of the yogurt culture microbes Lactobacillus delbrueckii subsp. bulgaricus and Streptococcus salivarius subsp. thermophilus in selected yogurts after the end of the mature process and the consideration of the observed values with the requirements of the legislation. The literary search mentions the fermented dairy products and their classification, the characterization of individual types of lactic fermentation microbes and also the pure milk cultures used for the production of soured dairy products are not left out. The experimental part deals with the evaluation of microbiological analysis and the acidity of selected yogurts according to the influence of the producer and the storage life. The observed results showed, that the legislative requirement for the quantity of live microbes has been kept not only during the prescribed best-before period, but also during the complete experiment, it means also on the 56th day of the storage.

In diploma thesis I dealt with American foulbrood infection. At first I was focusing on disease hazards of foulbrood and I was investigating the pathogenesis of the disease bacteria Paenibacillus larvae. Then I carried out analysis of slumgum by culture test in the laboratory. I watched the bacteria P. larvae under a microscope. Next, I followed outbreaks announced by Regional Veterinary Administration. I investigated procedures of the Regional Veterinary Administration in accordance with legislation and decree during detecting the disease of AFB on the habitats. At the end, I developed a list of proclaimed outbreaks in South Bohemia into graphs and chart for the period from 2006 to 2013. The only treatment against this disease so far is burning hives, including all equipment of apiary. That is why I pointed out the importance of reporting suspected outbreaks of American foulbrood, respecting the rules and precautions by beekeepers, and beekeepers should also breed only healthy and strong beehives. Only then the spread of American foulbrood can be prevented and the originator of American foulbrood, the P. larvae, which is very durable, can be suppressed.

This diploma thesis deals with resistance of microorganisms to antibiotics and public awareness of this topic. The aim of the thesis was to find out what the public awareness of the given topic is and what knowledge the public has about the possibilities of prevention of creation and spreading of resistant microorganisms. Three main hypotheses have been set: Hypothesis 1: The public has no deeper knowledge of antibiotic treatment. Hypothesis 2: The public will in most cases get information on antibiotic treatment from a doctor. Hypothesis 3: Most people are unaware that they can contribute by rational use of antibiotics to prevent antibiotic resistance. In order to achieve the stated goals of the diploma thesis, a quantitative method of research was used. Data collection was conducted through anonymous questionnaire survey of the public population using the Click4Survey.cz web server and distribution of paper questionnaires in FN Plzeň. Results of the questionnaire survey were evaluated based on statistical data processing in Microsoft Office Excel 2007, and a Chi-squared test of independence in the pivot table and a relative frequency test were used to verify the hypotheses. Based on the evaluation of the results, it was found that among the groups of respondents, depending on whether they filled in a questionnaire on the Internet or a printed questionnaire at the hospital, there are fundamental statistical differences in knowledge and awareness of the problem. That is why it was necessary to further evaluate these two groups separately. Antibiotic resistance nowadays is a very important medical issue. The main cause is inconsistent use or overuse of antibiotics in both human and veterinary medicine, as well as insufficient awareness of the extent of the problem and the consequences of antibiotic resistance. For this reason, I have chosen this issue as a topic for my diploma thesis to highlight the risks of antibiotic resistance and to provide information primarily to the general public.

Společenstvo metanotrofních bakterií v půdách zimoviště skotu / The methanotrophic community in soils of cattle overwintering area

The long-term cattle impact on soil methanotrophs was investigated at cattle overwintering area located at the ecofarm in the South Bohemia. Four sets of soil samples were sampled in spring and fall of 2009 and 2010. Differences in methanotrophic community structure among experimental sites and were investigated by DGGE and MISA. Real-Time PCR was used for estimation of quantity of Type I methanotrophs. The CARD-FISH method was used to estimate the percentage shared and cell counts of Type I and Type II methanotrophs among all prokaryotic cells. Substantial part of the thesis was focused on optimization of methods used.
