Effects of diet on phosphorus digestion in dairy cattle

Two studies were conducted to evaluate the effects of diet on phosphorus (P) digestion in dairy cattle. The objective of the first study was to evaluate the effects of forage and non-fiber carbohydrate (NFC) content on total P (TP) and inositol phosphates-P (IPs-P) digestion. Samples of feed, duodenal digesta and feces from a previously conducted study were analyzed for TP and IPs-P. In this study, eight lactating Holstein cows were fed diets containing either 60 or 35% forage and either 30 or 40% NFC in a 2 × 2 factorial with replicated 4 × 4 Latin square design. Dietary TP content (% DM) was 0.35, 0.36, 0.36, 0.36 and dietary IPs-P content (%DM) was 0.08, 0.13, 0.06, 0.11 for diets with forage: NFC ratio 35:30, 35:40, 60:30, and 60:40, respectively. Increasing dietary forage content decreased IPs-P and TP intake, fecal TP excretion, and total tract IPs-P digestibility (72.4 vs. 61.4%). Fecal IPs-P excretion tended to decrease as increasing forage content. Duodenal IPs-P and TP flow and apparent TP digestibility were unaffected by forage content. Increasing dietary NFC content increased IPs-P and TP intake, duodenal IPs-P flow, fecal IPs-P excretion, total tract IPs-P digestibility (61.4 vs. 72.4%), and apparent TP digestibility (32.8 vs. 41.6%). Dietary forage and NFC content affected IPs-P and TP digestion. The second study was to evaluate the effects of increasing dietary beet pulp (BP) content to replace high moisture corn (HMC) on ruminal and post-ruminal digestion of TP and IPs-P. Eight lactating Holstein cows were fed diets containing 0, 6.1, 12.1 or 24.3% BP in a replicated 4 × 4 Latin square design. Samples of rumen contents, duodenal digesta, and feces from this previously conducted study were analyzed for TP and IPs-P content. Linear and quadratic effects of BP content were analyzed using Proc Mixed of SAS. Dietary TP and IPs-P content were reduced linearly with increasing BP (0.59, 0.58, 0.57, 0.56% TP and 0.15, 0.14, 0.13, 0.11% IPs-P). Intake, ruminal content, and rumen pool size of TP decreased with increasing BP content. Digestion of TP and duodenal flow and fecal excretion of IPs-P and TP were not affected. With increasing dietary BP content, IPs-P intake was reduced, ruminal IPs-P pool size was reduced, and rumen turnover time (h) of IPs-P was increased. Apparent ruminal IPs-P digestibility (36.5, 31.8, 24.6, 13.6 %) and apparent total tract IPs-P digestibility (85.3, 82.7, 82.1, 79.1%) decreased linearly with increasing BP. Fecal excretion of IPs-P averaged 5.2 g/d. Replacing HMC with BP reduced digestion of IPs-P. The majority of IPs-P disappearance occurred post-ruminally. Magistrantūros studijų baigiamojo darbo tyrimų tikslas buvo ištirti cukrinių runkelių ligotumą, nustatyti grybinių lapų ligų paplitimą ir žalingumą cukriniuose runkeliuose bei nustatyti apsaugos priemonių, mažinančių ligų plitimą efektyvumą. Lauko eksperimentas buvo atliktas 2012 m. Lietuvos agrarinių ir miškų mokslų centro (LAMMC) Rumokų bandymų stotyje. Dirvožemis sekliai glėjiškas paprastasis išplautžemis (Haplic-Epihypogleyic Luvisol – Idg8-p). Darbo objektas - cukrinių runkelių (Beta vulgaris saccharifera) grybinės lapų ligos. Darbo metodai: Cukrinių runkelių lapų ligotumo, fungicidų biologinio efektyvumo, ligų žalingumo matavimai nustatyti tiesioginio matavimo, skaičiavimo ar svėrimo būdu pagal priimtas lauko eksperimento vykdymo metodikas (Šurkus, Gaurilčikienė, 2002). Tyrimų duomenys statistiškai įvertinti vieno veiksnio arba dviejų veiksnių kiekybinių požymių dispersinės analizės metodais, taikant kompiuterinę programą ANOVA (Tarakanovas, Raudonius, 2003). Darbo rezultatai: Tyrimų metais cukriniuose runkeliuose labiausiai plito cukrinių runkelių rudmargė ir baltuliai, kurių pažeidimo intensyvumas atitinkamai siekė 11,91 ir 1,81 proc. Didžiausias biologinis fungicido efektyvumas nuo cukrinių runkelių rudmargės nustatytas laukeliuose, kuriuose vieną ir du kartus buvo naudojamas fungicidas Maredo (vienkartinio purškimo norma 1,0 l ha-1, dukartinio - 0,75 l ha-1). [toliau žr. visą tekstą] / The main objective of Master thesis research was to investigate the intensity of sugar beet diseases, determine the fungal disease prevalence and harmfulness of sugar beets, and determine the effect of chemical plant control against sugar beet diseases.The research was made in the Rumokai Experimental Station of the Lithuanian Research Centre for Agriculture and Forestry in 2012. The soil type – Haplic-Epihypogleyic Luvisol – Idg8-p. Object of the research – sugar beet (Beta vulgaris saccharifera) fungal leaf diseases. Working methods: biological effectiveness of fungicides and disease harmfulness measurements were determined while using direct measurements, calculations or weighing according field experimental techniques (Šurkus, Gaurilčikienė, 2002). Research data have been statistically evaluated with a single factor or two factor analysis of variance of quantitative methods of evidence, using a computer program ANOVA (Tarakanovas, Raudonius, 2003). Research results: The most effective fungicide against Cercospora leaf spot disease in the once and twice treatment field of sugar beet plants was Maredo (once treatment rate 1,0 l ha-1, twice treatment rate 0,75 l ha-1). Occurrence, biology, damage potential and management of Heterodera Schachtii (Nematoda: Heteroderidae) in small-scale farming in the Western Cape Province, South Africa

Thesis (PhD (Agric))--University of Stellenbosch, 2003. / ENGLISH ABSTRACT: During a survey in the greater Cape Flats Heterodera schachtii was found to be widespread on cauliflower, Brussels sprouts, beetroot and cabbage. The numbers present were above two eggs and juveniles per gram of soil, generally regarded as the economic threshhold level of infestation and requiring control. The damage potential of H schachtii on vegetables, as well as the ability of certain weeds to serve as a source of infection on subsequent crop plantings was studied under greenhouse conditions and resulted in a reduction of yield and root weight of crops. Population densities of H schachtii increased significantly under favourable hosts like cabbage where densities of 198 eggs and juveniles per gram of soil were reached. The most commonly occurring weeds maintained nematode development and increased their population densities. They can thus serve as alternative hosts in the absence of susceptible hosts and should be routinely controlled. The life cycle and biology of H schachtii was also studied. Penetration of plant tissue and subsequent development on vegetables, weeds and trap crops were observed. Penetration was successful on all crops tested reaching 37% and 52% at inoculum levels of 22 and 11 juveniles per gram of soil, respectively. Subsequent development of H schachtii on weeds and vegetables was similar, but in the case of cauliflower and black nightshade as hosts, their life cycle was shorter in comparison to other crops. The possible existence of varying susceptibility of crops to different populations of H schachtii was examined by comparing the rates of penetration in crops and reproduction of geographically isolated populations of H schachtii in the greater Cape Flats. When root penetration, virulence and juvenile emergence were examined, populations from Lynedoch and Philippi were distinct from the other populations. Subsequently, representative individuals of these populations were subjected to PCR-RFLP, but with these techniques real differences between the various populations could not be adequately detected. The environmental parameters such as soil texture, temperature and pH on H schachtii were investigated as to their influence on the root weight and yield of crops. Reductions in the yield of beetroot and cabbage were observed with soil temperatures ranging between 15 to 30°C. Migration and penetration of H schachtii juveniles declined with an increase in clay and silt content of the soil. Above a 34% silt and clay content of soil, no migration and penetration took place. Root penetration levels of 30% and higher were reached with pH varying between 4.5 and 7.4. This resulted in a significant reduction in yield of crops. Crop rotation is an essential component of non-chemical control. In the case of H schachtii, it required one host crop in four non-host rotational cycles to maintain the population of the nematode in the soilless than three eggs and juveniles per gram of soil. The inclusion of a trap crop reduced the population densities to below two eggs and juveniles per gram of soil. It therefore also forms an integral part of a control strategy. Solarization proved successful as a physical control method. Best results were obtained in summer with clear polyethylene which led to a 97% reduction of infective juveniles. This method can be applied during the late summer in the greater Cape Flats, just before the onset of winter. This may safeguard future spring plantings. The need for effective control strategies in order to reduce the numbers of H schachtii is of the utmost importance to ensure vegetable production in the future. Small-scale farmers should therefore be educated in this respect. / AFRIKAANSE OPSOMMING: Heterodera schachtii het wydverspreid in 'n opname in die groter Kaapse Vlakte voorgekom op beet, blomkool, Brusselse spruite en kopkool. Die nematode getalle by alle lokaliteite was bo die algemeen aanvaarbare ekonomiese drempelwaarde van twee eiers en larwes per gram grond wat beheer regverdig. Die skadepotensiaal van H schachtii op groente, sowel as die vermoë van sekere onkruide om as infeksie bronne te dien vir opvolgende gewasse, is in glashuise ondersoek en het tot 'n verlaging in opbrengs en wortelmassa by gashere gelei. Die populasie digthede van H schachtii het met die aanplant van geskikte gashere tot vlakke van 198 eiers en larvae per gram grond gestyg. Penetrasie, vyf dae na inokulasie, is met alle gashere verkry met 37% en 52% penetrasie met inokulum vlakke van 22 en 11 larwes per gram grond onderskeidelik. Wortelpenetrasie van 30% en hoër het voorgekom by pH vlakke van tussen 4.5 - 7.4 met die gepaardgaande verlaging in opbrengs van gewasse. Afwisseling van gewasse is 'n essensiële metode van nie-chemiese beheer van nematode getalle in die grond. Die mees optimale rotasie ten opsigte van H schachtii beheer is met die aanplanting van een gasheer gewas in vier gewas aanplantings verkry. Die insluiting van 'n vanggewas in die gewas rotasie siklus het die nematode populasievlakke tot onder twee per gram grond laat daal. Solarisasie is suksesvol uitgevoer met deurskynende poli-etileen in die groter Kaapse Vlakte gedurende die somer met gevolglik 'n 97% vermindering van die getalle infektiewe nematodes. Effektiewe beheermaatreëls ten opsigte van H schachtii moet in die groter Kaapse Vlakte ingestel word om groente-produksie in hierdie gebied te verseker. Kleinboere moet in hierdie tegnieke opgelei word.

Molecular mechanisms involved in the pathogenesis of beet soil-borne viruses / Mécanismes moléculaires à l'origine de la pathogenicité de phytovirus de betterave sucrière transmis par un vecteur tellurique

Le virus des nervures jaunes et nécrotiques de la betterave (Beet necrotic yellow vein virus, BNYVV) est l’agent infectieux responsable de la rhizomanie de la betterave sucrière, une maladie caractérisée par une prolifération anarchique du chevelu racinaire. Le Beet soil-borne mosaic virus (BSBMV) appartient également au genre Benyvirus mais n’est retrouvé qu’en Amérique du Nord. Ce virus, identifié pour la première fois au Texas, est morphologiquement et génétiquement semblable au BNYVV mais sérologiquement éloigné. Compte tenu des différences moléculaires existant, le BSBMV et BNYVV correspondent à deux espèces virales distinctes. Mon projet de thèse a consisté à étudier les interactions moléculaires entre le BNYVV et le BSBMV et rechercher les mécanismes impliqués dans la pathogénicité de ces deux virus. Des clones complets cDNA infectieux du BNYVV étaient disponibles, tout comme ceux de BSBMV. Compte tenu de l’aspect versatile de l’obtention de transcrits infectieux de ces différents clones, j’ai entrepris de produire des clones cDNA de chacun des ARN viraux sous contrôle d’un promoteur constitutive végétal pour initier l’infection par agroinfiltration. Les plantes hôtes Chenopodium quinoa et Nicotiana benthamiana ont été inoculées par des transcrits et agroinfiltrées pour initier l’infection virale et étudier l’interaction entre les ARN génomiques 1 et 2 des deux virus et étudier les propriétés de constructions chimères. En parallèle à ce travail, j’ai réalisé la caractérisation du suppresseur de RNA silencing du BSBMV en le comparant à celui du BNYVV. / The genus Benyvirus includes the most important and widespread sugar beet viruses transmitted through the soil by the plasmodiophorid Polymyxa betae. In particular Beet necrotic yellow vein virus (BNYVV), the leading infectious agent that affects sugar beet, causes an abnormal rootlet proliferation known as rhizomania. Beet soil-borne mosaic virus (BSBMV) is widely distributed in the United States and, up to date has not been reported in others countries. My PhD project aims to investigate molecular interactions between BNYVV and BSBMV and the mechanisms involved in the pathogenesis of these viruses.BNYVV full-length infectious cDNA clones were available as well as full-length cDNA clones of BSBMV RNA-1, -2, -3 and -4. Handling of these cDNA clones in order to produce in vitro infectious transcripts need sensitive and expensive steps, so Ideveloped agroclones of BNYVV and BSBMV RNAs, as well as viral replicons allowing the expression of different proteins.Chenopodium quinoa and Nicotiana benthamiana plants have been infected with in vitro transcripts and agroclones to investigate the interaction between BNYVV and BSBMV RNA-1 and -2 and the behavior of artificial viral chimeras. Simultaneously I characterized BSBMV p14 and demonstrated that it is a suppressor of posttranscriptional gene silencing sharing common features with BNYVV p14.

Plant-Parasitic Nematodes on Sugarbeet in North Dakota and Minnesota

Field surveys were conducted in the Red River Valley (RRV) of North Dakota and Minnesota during 2016 and 2017 to determine the incidence, abundance, and distribution of plant-parasitic nematodes (PPNs) on sugarbeet. Seventy-two and 65 % of the fields surveyed were positive for PPNs in 2016 and 2017, respectively. The major genera of PPNs identified from sugarbeet production fields were Heterodera, Helicotylenchus, Tylenchorhynchus, Paratylenchus, Pratylenchus, Paratrichodorus, Hoplolaimus, and Xiphinema. Eight of PPNs were identified at the species level using species-specific PCR assays, and sequencing of the ribosomal rDNA gene. Stubby-root nematode, Paratrichodorus allius, is one of the important nematode pests for sugarbeet production worldwide. An experiment was conducted to determine the host status of sugarbeet and their rotational crops for P. allius under greenhouse conditions. Roman&quot;">Primena različitih tretmana sa ciljem redukcije sadržaja nesaharoznih jedinjenja u melasi &scaron;ećerne repe potencijalan je metod za pobolj&scaron;anje parametara kvaliteta melase, lak&scaron;e sprovođenje procesa desaharifikacije melase i sprečavanje eventualnih nepoželjnih promena pri skladi&scaron;tenju melase. Stoga je predmet istraživanja ove disertacije redukovanje sadržaja nesaharoznih jedinjenja u melasi &scaron;ećerne repe upotrebom prirodnih adsorbenata mineralnog (različiti tipovi bentonita) i celuloznog (celuloza različitog stepena čistoće i modifikovani ekstrahovani rezanci &scaron;ećerne repe) porekla. Variranjem pH vrednosti sredine, suve materije melase i koncentracije primenjenog adsorbenta ispitan je sinergistički efekat uslova tretmana na stepen redukcije sadržaja nesaharoznih jedinjenja u melasi. Therefore, the objective of this dissertation is to reduce the content of non-sugar compounds in sugar beet molasses by using natural adsorbents of mineral (different types of bentonite) and cellulosic (cellulose of different purity and modified sugar beet pulp) origin. By varying treatment conditions such as pH, molasses dry substance and the applied adsorbent concentration, the synergistic effect of the treatment conditions on the reduction efficiency of non-sugar compounds content in molasses was examined. Contrôle épigénétique de l'induction et de la tolérance à la montaison chez la betterave sucrière / Epigenetic conttrol of bolting induction and tolerance in sugar beet

La betterave sucrière est une plante bisannuelle dont le besoin de vernalisation est absolu. Ce processus correspond à l’acquisition de l’aptitude à la montaison et à la floraison et résulte d’une exposition prolongée à de basses températures. La durée de froid requise pour induire la montaison puis la floraison varie suivant les génotypes et reflète leur tolérance à la montaison, qui constitue donc un caractère agronomique essentiel. Cette thèse visait à (i) mettre en évidence un éventuel contrôle épigénétique (méthylation ADN) de l‘induction de la montaison chez des génotypes de betterave sucrière résistants ou sensibles à la montaison, (ii) identifier les séquences ciblées par des remaniements de méthylation de l’ADN et d’expression associés, et (iii) caractériser certaines séquences candidates en vue de leur utilisation comme marqueurs de la montaison. Nos travaux ont montré que l’amplitude et la cinétique des variations de méthylation de l’ADN observées au cours de la vernalisation semblent être des éléments critiques de l’induction et de la tolérance à la montaison. Par une approche ciblée, des séquences dont la méthylation de l’ADN est remaniée ont été identifiées. L’implication dans la transition florale de deux BvRNMT (RNA METHYLTRANSFERASES) et de la méthylation des ARN, tels que l’ARNm de BvFL1, un répresseur floral, a ainsi été mise en évidence chez la betterave sucrière. Enfin, grâce à une approche génomique, un réseau de gènes intégratif incluant la réponse à l’environnement, la signalisation hormonale et l’induction de la floraison a été identifié. La cinétique d’activation de ces gènes définirait le niveau de tolérance à la montaison des différents génotypes de betterave sucrière. / Sugar beet is a biennial plant with an absolute requirement of vernalization, corresponding to the acquisition of the competence to bolt and flower after a prolonged exposure to low temperatures. The cold duration needed to induce bolting and flowering varies depending on the genotypes, reflecting their bolting tolerance, which is an essential agronomic trait. This work aimed at (i) investigating a possible epigenetic control of bolting induction in bolting sensitive and bolting resistant sugar beet genotypes, (ii) identifying sequences targeted by DNA methylation and expression remodeling, and (iii) characterizing candidate sequences which could be used in marked-assisted selection for plant breeding. Our data suggest that the time course and amplitude of DNA methylation variations are critical points for the induction of sugar beet bolting and represent an epigenetic component of the genotypic bolting tolerance. In addition, we identified differentially methylated sequences exhibiting variations of gene-body DNA methylation and expression during cold exposure and/or between genotypes. Among them, two RNA METHYLCYTOSINE TRANSFERASES, in association with RNA methylation such as BvFL1 mRNA, a floral repressor, were shown to play a role in floral transition. Finally, using microarrays we identified an integrative network of genes including response to environment, phytohormone signalling and flowering induction. The activation kinetics of these genes could define the bolting tolerance level of sugar beet genotypes.

The search is proposed to develop a beet flour using simple technology and procedures available for the majority of small producers. It was compared different types of longitudinal and transverse cuts of beet (long 2mm; long 4.5mm; long 9mm, and, trans 2mm; trans 4.5mm; trans 9mm) as the time for air-drying, drying rate, time in the grounding process, income in flour product and its size. The conditions for the material drying, the cooked slices (2 mm) of beet, were similar, so it was recommended among the treatments the use of 100° C for 2 hours and 70° C to the end, for this condition finishes the process of air-drying in temperature more lenient. The treatments showed no significant difference in income for the full production of flours and brans, but the flours were significantly different as to size. Minors were drying times of the treatment of cuts 2 mm (7.3 h = longitudinal, 7.3 h = transverse) and sticks 4.5 mm (7.3 h = longitudinal, 7.7 h = transverse). The treatments in sticks 9mm remained, longitudinal = 9.7 h and transverse = 9.3 h, in the air-dryer. The beet flour showed percentage posted for nutrients, carbohydrate (50.32 to 5.39%), dietary fiber (23.30 to 26.90%), protein (11.32 to 13.40%) and total of minerals (4.70 to 6.82%). Already, the level of lipids, as expected, was less than 0.75%. Considering the ease of slices preparation, the dry time, the production of low size flour, and the flour income, the treatment that produced cooked slices of beet in sticks of 4.5 mm in thickness was recommended to the flour processing. / A produção de hortaliças desidratadas tem sofrido significativo aumento, devido ao maior tempo de conservação comparado a in natura, a praticidade de uso e a possibilidade de aproveitamento da produção excedente, reduzindo efeitos de sazonalidade. A pesquisa se propôs a desenvolver um produto farináceo de beterraba aplicando procedimentos tecnológicos simples e disponíveis para uma grande parte dos pequenos produtores. Foram comparados diferentes tipos de cortes longitudinais e transversais de beterraba (long 2mm; long 4,5mm; long 9mm; trans 2mm; trans 4,5mm; trans 9mm) quanto ao tempo de secagem, taxa de secagem, tempo no processo de trituração, rendimento em produto farináceo e sua granulometria. As condições de temperaturas testadas para a secagem das fatias de beterraba foram semelhantes, por isso foi recomendado o uso de 100° C por 2 horas seguido de 70º C, por esta condição terminar o processo de secagem em temperatura mais branda. Os tratamentos de fatias de beterraba não apresentaram diferença significativa para o rendimento na produção de farinhas e farelos, mas, as farinhas foram significativamente diferentes quanto à granulometria. Os menores tempos de secagem foram dos tratamentos de cortes laminares 2mm (long = 7,3 h; trans = 7,3 h) e em palitos 4,5mm (long = 7,3 h; trans = 7,7 h), sendo que os tratamentos em palitos 9mm permaneceram, long = 9,7 h e trans = 9,3 h, no secador. As farinhas de beterraba apresentaram valores porcentuais destacados para os nutrientes, carboidrato (50,32-55,39 %), fibra alimentar (23,30-26,90 %), proteína (11,32-13,40 %) e total de minerais (4,70- 6,82 %). Já, o teor de lipídeos, como esperado, foi mínimo, 0,75 %. Considerando a facilidade de preparo das fatias, o tempo de secagem, a produção de farinhas de baixa granulometria, bem como o rendimento apresentado, o tratamento que produziu fatias cozidas de beterraba em palito de 4,5mm de espessura foi o recomendado para o processo de obtenção de farinha de beterraba.
