Proteção cruzada entre bacterinas antileptospirose produzidas com três representantes do Sorogrupo Sejroe. Foi investigada a existência de proteção cruzada entre bacterinas bivalentes formuladas com um de três representantes do Sorogrupo Sejroe: hardjo (bacterina A), wolffi (bacterina B) e guaricura (bacterina C), e uma estirpe do sorovar pomona, empregada por ser patogênica para hamsters (Mesocricetus auratus) e possibilitar a realização do teste de potência com desafio. Os ensaios foram efetuados em hamsters machos, comparando-se os níveis de anticorpos aglutinantes e neutralizantes, respectivamente obtidos nos testes de soroaglutinação microscópica (SAM) e inibição de leptospiras in vitro (ICL). Os animais receberam duas doses de vacinas via subcutânea; aos dez dias da segunda dose, foram inoculados com culturas não inativadas dos respectivos sorovares do Sorogrupo Sejroe. Aos 21 dias pós-infecção (d.p.i.), os animais foram sangrados, os soros (n=8) foram agrupados em pools e submetidos aos testes de SAM e ICL. O teste de potência com desafio para o sorovar pomona foi adaptado do protocolo preconizado pelo United States Department of Agriculture, mas as vacinas não foram diluídas e o esquema de imunização empregou duas aplicações de 1,0mL pela via subcutânea em intervalo de dez dias; o desafio foi efetuado aos dez dias da segunda aplicação; os óbitos por leptospirose foram registrados e, aos 21 d.p.i., os sobreviventes foram sacrificados e a condição de portadores renais foi investigada através de cultivos de tecido renal para isolamento de leptospiras. No teste de potência com o sorovar pomona, o número de doses infectantes empregado para desafio (100) situou-se dentro da faixa preconizada (10 a 100); respectivamente para as bacterinas A, B e C, as proporções de mortes por leptospirose entre os animais vacinados foram de 1/10, 0/10 e 3/10, e as de portadores renais de leptospiras entre os sobreviventes foram 2/9, 1/10 e 2/7. Os resultados do teste SAM revelaram que a bacterina A induziu reações para os sorovares hardjo e wolffi; a bacterina B, para hardjo, wolffi e guaricura, e a bacterina C, apenas para a guaricura, e do teste de ICL, que animais vacinados com as bacterinas B ou C apresentaram proteção para hardjo, wolffi e guaricura; entretanto, a bacterina A conferiu proteção apenas para wolffi. Apesar das variações no poder imunogênico segundo a estirpe de leptospira empregada para a produção das bacterinas, houve proteção cruzada entre os sorovares hardjo, wolffi e guaricura. / The existence of cross-protection among bivalent bacterins, produced with one of three leptospires belonging to Serogroup Sejroe: hardjo (bacterin A), wolffi (bacterin B) and guaricura (bacterin C), and a strain of serovar pomona (included because of its pathogenicity to hamsters and the possibility of performing potency assay with challenge), was investigated in male hamsters (Mesocricetus auratus) by comparison of agglutinating and neutralizing antibodies titers, respectively measured by microscopic agglutination (MAT) and in vitro growth inhibition (GIT) tests. All animals received two doses of bacterins by subcutaneous route; after ten days from the second dose, they were inoculated with non-inactivated cultures of respective serovars of Serogroup Sejroe. At 21 post-challenge day (p.c.d.), all animals were bled and their sera (n=8) were joined in pools and tested by MAT and GIT. The potency assay with challenge performed only with serovar pomona was modified from protocol of USDA, but vaccines were not diluted and the immunization schedule employed two 1.0 mL vaccine doses by subcutaneous route with 10?day interval; the challenge was performed after ten days from the second dose; the number of deaths due to leptospirosis was registered; at 21 p.c.d., the survivors were sacrificed and their renal carrier state was investigated by culture of renal tissue for leptospires isolation. In the potency assay with serovar pomona, the number of infectious doses employed for challenge (100 infective units) was in accordance with the recommended range (10-1,000 infective units); respectively to bacterins A, B and C, proportions of deaths due to leptospirosis among vaccinated animals were 1/10, 0/10 and 3/10, and proportions of leptospires renal carrier among survivors were 2/9, 1/10 and 2/7. Results of MAT showed that bacterin A induced reactions against serovars hardjo and wolffi; bacterin B, against hardjo, wolffi and guaricura, and bacterin C, against guaricura; results of GIT revealed that vaccinated animals with bacterins B or C presented protection against serovar hardjo, wolffi and guaricura; however, bacterin A induced protection only against wolffi. A cross?protection was observed among serovars hardjo, wolffi and guaricura, although variations exist in the immunogenic capacity according to the strain of leptospires used for the bacterins production.

ESTABLISHMENT OF SEROLOGICAL REFERENCE RANGES FOR ELK IN KENTUCKY AND EFFECTS OF QUARANTINE AND TRANSLOCATION ON ELKHildreth, Aaron M. 01 January 2017 (has links)
Establishing blood serum profiles is important for understanding animal disease ecology and nutrition, the effects of capture and immobilization, and general physiological variation among individuals and populations. Elk (Cervus elaphus nelsoni) have been successfully translocated to several states for reintroduction or population augmentation, including most recently in Kentucky, where over the course of three years (2011-13) wild elk were captured, temporarily quarantined, and subsequently moved to Missouri and Virginia to establish populations in those states. I used this opportunity to collect a variety of biological samples, including blood from which I present and compare serological profiles for a variety of elk age and sex classes. Additionally, I took the opportunity to investigate the effects of the translocation process on some morphological and physiological parameters of elk. Quarantine and processing of elk can be stressful for animals and lead to injury or death. I characterized how elk respond to this process by measuring biochemical serum values along with various physiological parameters at 5 handling events from the time of capture until final transport to recipient states. Changes observed in parameters between paired workups were indicative of the physical exertion and stress associated with handling activities.

The feasibility of Fourier transform infrared imaging spectroscopy in discriminating benign prostatic hyperplasia from prostate cancer in blood serum samplesMonjardez, Geraldine January 2013 (has links)
The feasibility of Fourier transform infrared (FTIR)-imaging spectroscopy as a tool to discriminate samples from patients suffering from benign prostatic hyperplasia (BPH) and prostate cancer (CaP) samples in blood serum was investigated. Prostate cancer is known to be an age related disease, with the risk of developing the disease dramatically increasing in men past forty years old. Currently the PSA blood test is notoriously unreliable and is non specific for CaP thus leading to overtreatment of the disease. It is important therefore to develop diagnostic method that is non-invasive, reliable, and specific for CaP.In order to achieve the objective of establishing a robust protocol, which could be applied to a clinical study, obtaining optimal sample preparation for the FTIR analysis of serum smears, had to be achieved. A protocol was developed to prepare the serum samples prior to their FTIR analysis. First, the samples were centrifuged with ultrafiltration devices of different sizes to obtain several fractions which were then smeared to obtain thin films of serum. The spectra from the larger (>100 kDa components) and medium (containing the 10–100 kDa components) fractions were utilised for both a pilot and a clinical study, while the spectra from the smaller fractions (containing the 3–10 and <3 kDa components) were affected by fringing and could therefore not be used. A major novelty of this project involved the application of FTIR-imaging to the analysis of serum smears. The use of the Focal Plane Array detector system enabled the collection of a spectral image containing 16,384 spectra, on which a Quality Testing and pre-processing techniques were applied to select the “good spectra” and reject the spectra that failed the Quality Test. Several types of substrates were assessed to determine the most appropriate for the analysis of the smears and it was established that the spectra obtained from the serum smeared on CaF2 windows gave the most reproducible results. 5 BPH and 5 CaP samples were analysed for the pilot study following the developed protocol. While no clear separation was observed in the Principal Component Analysis (PCA) plots between the BPH and the cancerous samples, a trend emerged throughout the results, with the CaP samples clustering together and the BPH samples scattered around them. A larger clinical study was conducted with 60 BPH samples and 60 CaP samples. PCA was applied on the “good spectra” and while the over 100 kDa fraction did not show a clear separation between the two types of samples, the 10–100 kDa fraction showed a distinct classification between the BPH and CaP samples. An artificial neural network was then applied to create a model using patients from the database used for the PCA analysis to determine whether the discrimination between the two types of samples could be increased or highlight different classification trends. For the >100 kDa fraction, the sensitivity value was calculated to be 97.8% and the specificity value was calculated to be 44.3% while the sensitivity and the specificity value for the 10 to 100 kDa fraction were calculated to be 78.9% and 60% respectively. A complementary study using mass spectrometry was carried out on healthy and diseased samples to identify the components contained within the different fractions and determine whether they could be correlated with the components identified from the spectral features of the FTIR data. While no quantitative information was obtained from this study, the components found in the different fractions were identified, confirming the results of the FTIR studies.

Background: The method of serum bactericidal assay represents an alternative possibility of optimization of anti-infectious therapy and administration of antibiotics. It mirrors the real activity of one or more administered antibiotics in the complex system of the antibacterial effect of patient's serum. The paper aimed to confirm non-inferiority of bactericidal testing using the broth dilution method according to CLSI M21-A Guidelines (time to results 48, 72 hours) in comparison with modified methods of testing on the basis of turbidimetry (time to result 6, 8, 24 hours) and resazurin color (time to results 8, 24 hours). Methods: Four antibiotics were tested: gentamicin, amikacin, piperacillin/tazobactam and meropenem with 30 Escherichia coli strains isolated from blood cultures of 29 pacients hospitalised in different wards, University Hospital in Hradec Kralove. Human blood sera (n = 76) from ten hematological patients (4th Department of Clinical Medicine, University Hospital, Hradec Kralove) were tested to establish bactericidal titer. Patients' blood was withdrawn prior to and in the course of the first and third day of antibiotic therapy of febrile neutropenia. Testing employed the reference strain Escherichia coli ATCC 25922. Previous issue date: 2013-02-25 / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES / This study aims to evaluate the energy, protein and mineral metabolism in Santa Inês ewes, both healthy and with subclinical mastitis, followed up during late gestation and lactation periods. Ewes subjected to the same semi-intensive nursing system were followed up. The animals were evaluated according to the following stages: 10 days before parturition (dbp) and 15 days postpartum (dpp), 30 (dpp), 60 (dpp), and 90 (dpp). Blood metabolites were evaluated starting from the stage previous to parturition and whey metabolites were evaluated in the subsequent stages. A screening of the ewes followed up in this study (12 healthy and 18 with subclinical mastitis) was performed after a clinical and bacteriological examination. During lactation, maintaining the same screening criteria, 11 healthy and 20 infected mammary glands were selected; the milk for whey extraction was collected from these glands. Energy profile metabolites (non-esterified fatty acids [NEFAs], ß-hydroxybutyrate [BHB], fructosamine, cholesterol and triglycerides), protein profile (total protein, albumin, urea and creatinine) and mineral profile (iron, copper, zinc, magnesium, total calcium, ionized calcium, sodium, and potassium) were measured in the blood serum. Calcium, sodium and potassium ions, as well as NEFAs and BHB were measured in the whey. Blood biochemistry revealed an influence (P<0,05) of the peripartum and lactation periods on the blood concentrations of NEFAs, BHB, cholesterol, albumin, urea, ionized calcium. An analysis of the whey also revealed an influence on the potassium ion. Ewes with subclinical mastitis showed higher (P<0,05) blood levels of cholesterol, albumin and copper; higher sodium ion concentrations and NEFAs, and lower potassium ion in whey. Good physical score of ewes observed during this study, combined with the biochemical findings, allowed us to conclude that there was a larger energy requirement in the first month of lactation; however, this requirement was not enough to trigger any metabolic disorder or the emergence of ketonemia, and these discrete changes were more apparent in ewes with subclinical mastitis. / Este estudo teve por objetivo avaliar o metabolismo energético, proteico e mineral de ovelhas Santa Inês hígidas e com mastite subclínica acompanhadas durante o final da gestação e na lactação. Foram acompanhadas ovelhas submetidas ao mesmo sistema de criação semi-intensivo. Os animais foram avaliados conforme os momentos a seguir: 10 dias que precedeu o parto (dap) e 15 dias após o parto (dpp), 30 (dpp), 60 (dpp) e 90 (dpp). Os metabólitos sanguíneos foram avaliados a partir do momento que antecedeu ao parto e os metabólitos no soro lácteo nos momentos subsequentes. Após exame clínico e bacteriológico foi realizada a triagem das ovelhas acompanhadas neste estudo, sendo 12 hígidas e 18 com mastite subclínica. Durante a lactação, mantendo os mesmos critérios de triagem, foram selecionadas 11 glândulas mamárias sadias e 20 infectadas, das quais foi colhido o leite para obtenção do soro lácteo. Foram mensurados no soro sanguíneo os metabólitos do perfil energético (ácidos graxos não esterificados (AGNEs), ß-hidroxibutirato (BHB), frutosamina, colesterol e triglicérides), ao perfil proteico (proteína total, albumina, uréia e creatinina) e ao perfil mineral (ferro, cobre, zinco, magnésio, cálcio total, cálcio ionizado, sódio e potássio). No soro lácteo foram mensurados os íons cálcio, sódio e potássio, bem como dos AGNEs e BHB. A bioquímica sanguínea revelou haver influência (P<0,05) do período do periparto e da lactação sobre as concentrações sanguíneas dos AGNEs, BHB, colesterol, albumina, uréia, cálcio ionizado e no soro lácteo sobre o íon potássio. As ovelhas portadoras de mastite subclínica apresentaram valores sanguíneos superiores (P<0,05) de colesterol, albumina e cobre e no soro lácteo teores superiores do íon sódio e dos AGNEs e inferiores do íon potássio. O bom escore corporal das ovelhas observado durante o estudo aliado aos achados bioquímicos permitiu concluir ter ocorrido maior requerimento energético no primeiro mês da lactação, porém não o suficiente para desencadear qualquer transtorno metabólico e o aparecimento de um quadro de cetonemia, sendo estas discretas alterações mais expressivas nas ovelhas com mastite subclínica.

No. of bitstreams: 0 / A nanociência e a nanotecnologia vêm abrindo inúmeros desenvolvimentos de dispositivos e sistemas em escala nanométrica, com novas organizações moleculares, propriedades e funções distintas. Nesse contexto, as nanopartículas magnéticas poliméricas são compósitos formados por materiais magnéticos com tamanhos de partículas entre 1 e 100 nm combinados com polímeros funcionais. São materiais bem conhecidos e têm sido amplamente estudados devido às suas aplicações em diversas áreas tecnológicas. Nas áreas biológica e médica, as aplicações incluem separação e imobilização de enzimas e proteínas, melhoria nas técnicas de imagem de ressonância magnética para diagnóstico e sistemas de liberação controlada de fármacos. Neste trabalho, proteínas foram imobilizadas na superfície de um biopolímero combinado com partículas superparamagnéticas de magnetita para formar o compósito magnético. Utilizou-se o biopolímero quitosana, reticulada e funcionalizada com glutaraldeído, aplicável em ensaios biológicos. Obtiveram-se 3 tipos de compósitos magnéticos, os quais foram nomeados QM1Glu, QM2NaGlu e QM3Glu. Foram caracterizados por difratometria de raios X, microscopia eletrônica de varredura, magnetometria de amostra vibrante, calorimetria exploratória diferencial, termogravimetria e espectroscopia por infravermelho. Foram avaliados quanto à imobilização das proteínas albumina de soro bovino (SAB), colágeno e tripsina. A imobilização das proteínas no biopolímero ocorreu em 30 min de incubação. O compósito magnético de quitosana não funcionalizada (QM3) também foi avaliado. Para a tripsina verificou-se que QM3 apresentou maior potencial de imobilização do que QM3Glu. Após 30 dias, QM3-Trip e QM3Glu-Trip ainda apresentavam a tripsina ativada. <p> In serum, there are two manganese-binding proteins. The first is a(beta)1 globulin with a molecular weight of 70,000. This forms a relatively labile manganese complex both in vitro and in vivo, and is remarkably similar in both its chromatographic and electrophoretic behaviour to the iron-binding protein, transferrin. The second protein is a higher molecular weight (beta) globulin. It is found to incorporate radiomanganese in vivo only, thereupon forming a very stable entity. </p>
