La palourde japonaise, Venerupis philippinarum, a été introduite en France au début des années 70 à des fins aquacoles. Depuis 1987, d’importants épisodes de mortalité, causés par la bactérie pathogène Vibrio tapetis, touchent cette espèce le long des côtes françaises et européennes. Cette vibriose, appelée Maladie de l’Anneau Brun (MAB), est considérée comme une maladie d’eau froide. L’interaction tripartite entre V. philippinarum, V. tapetis et l’environnement a été explorée à travers le rôle du système enzymatique des phénoloxydases (POs) dans le but de mieux comprendre la réponse immunitaire de la palourde japonaise, la virulence de l’agent pathogène mais aussi l’impact de l’environnement et plus particulièrement de la température. Les POs sont des oxydoréductases impliquées dans la synthèse de la mélanine et de ses dérivés mais aussi dans les processus de reconnaissance du non-soi et d’encapsulation chez les invertébrés. Dans un premier temps, l’activité PO du sérum d’hémolymphe a été caractérisée d’un point de vue biochimique comme étant majoritairement de type laccase ; une activité minoritaire de type tyrosinase a également été identifiée. Des infections de palourdes par trois souches de V. tapetis, à différentes températures, ont mis en évidence une modulation de la réponse du système PO en fonction du temps et du compartiment étudiés. De façon générale, l’infection bactérienne s’est traduite par une augmentation de l’activité PO. Toutefois, le niveau basal d’activité PO est variable d’une population à une autre et cette variabilité semble traduire une susceptibilité différente à la MAB. L’augmentation de la température de 15 à 22°C a entraîné une augmentation des capacités immunitaires de la palourde japonaise. La températurea également eu un impact sur la pathogénicité de V. tapetis et ce, de façon différentielle selon les souches. L’inhibition de l’activité PO observée in vitro en présence de produits extracellulaires bactériens souligne la complexité de l’interaction entre V. philippinarum et V. tapetis. Ainsi, le suivi de l’activité PO constitue un biomarqueur pertinent des capacités immunitaires des invertébrés marins dans l’interaction tripartite hôte-pathogène-environnement. / The manila clam, Venerupis philippinarum, was introduced in France in the early 70’s for aquaculture purposes. Since 1987, mass mortality events, caused by the pathogenic bacterium Vibrio tapetis, have occurred in this species along French and European coasts. This vibriose, called Brown Ring Disease (BRD), is considered as a cool water disease. The tripartite interaction between V. philippinarum, V. tapetis and the environment was explored through the role of the enzymatic system of phenoloxidases (POs) in order to better understand the manila clam immune response, the pathogen virulence and also the environment impact and particularly the temperature’s one. POs are oxidoreductase enzymes involved in the synthesis of the melanin and its derivatives and also in the processes of non-self recognition and encapsulation in invertebrates. Firstly, PO activity was biochemically characterized in the hemolymph serum as a laccase-like activity; a minority tyrosinaselike activity was also identified. Clam inoculation with three V. tapetis strains, at various temperatures, highlighted a modulation of the answer of the PO system according to time post-infection and studied compartments. Overall, bacterial infection resulted in an increase in PO activity. However, the PO activity basal level varies between populations and this variability seems to reflect different susceptibility to the BRD. An increase in temperature from 15 to 22°C led to an increase in manila clam immune abilities. The temperature also had an impact on the V. tapetis pathogenicity and this impact was variable according to the strains. PO activity inhibition observed, in vitro, in the presence of bacterial extracellular products underlines the complexity of the interaction between V. philippinarum and V. tapetis. Thus, the monitoring of the PO activity constitutes a relevant biomarker of marine invertebrate immune abilities in the tripartite interaction host-pathogen-environment.

The main task of this analysis is, evaluate present progress of this region. It also describes history of brown coal mining in brown coal field of Sokolov region and negative effect for landscape. Next chapters of this thesis show: recultivation in general, project documentation, financing arrangements, company Sokolovská uhelná a.s. and its privatisation. One of the most important parts of this diploma thesis deals with recultivation in Sokolov region and description of particular locations in this region. The last part gives an evaluation of socioeconomic developmet in dependence on recultivation in Sokolov region.

Carbon metabolism in clear-water and brown-water lakesAsk, Jenny January 2010 (has links)
The trophic state of lakes is commonly defined by the concentration of nutrients in the water column. High nutrient concentrations generate high phytoplankton production, and lakes with low nutrient concentrations are considered low-productive. This simplified view of lake productivity ignores the fact that benthic primary producers and heterotrophic bacteria can be important basal producers in lake ecosystems. In this thesis I have studied clear-water and brown-water lakes with respect to primary production, respiration and bacterial production based on allochthonous organic carbon. These processes were quantified in pelagic and benthic habitats on temporal and spatial scales. I also calculated the net ecosystem production of the lakes, defined as the difference between gross primary production (GPP) and respiration (R). The net ecosystem production indicates whether a lake is net heterotrophic (GPP < R), net autotrophic (GPP > R) or in metabolic balance (GPP = R). Net heterotrophic lakes are sources of carbon dioxide (CO2) to the atmosphere since respiration in these lakes, by definition, is subsidized by an external organic carbon source. External organic carbon is transported to lakes from the terrestrial environment via inlets, and can serve as a carbon source for bacteria but it also limits light availability for primary producers by absorbing light. On a seasonal scale, four of the clear-water lakes studied in this thesis were dominated by primary production in the soft-bottom benthic habitat and by respiration in the pelagic habitat. Concentrations of dissolved organic carbon (DOC) were low in the lakes, but still high enough to cause the lakes to be net heterotrophic. However, the lakes were not low-productive due to the high production in the benthic habitat. One of the clear-water lakes was studied also during the winter and much of the respiration under ice was supported by the benthic primary production from the previous summer. This is in contrast to brown-water lakes where winter respiration is suggested to be supported by allochthonous organic carbon. By studying lakes in a DOC gradient (i.e. from clear-water to brown-water lakes) I could draw two major conclusions. The lakes became less productive since benthic primary production decreased with increasing light extinction, and the lakes became larger sources of CO2 to the atmosphere since pelagic respiration was subsidized by allochthonous organic carbon. Thus, lake carbon metabolism can have an important role in the global carbon cycle due to their processing of terrestrial organic carbon and to their possible feedback effects on the climate system.

Seaweeds are organisms which live in oceans and seas. They have ability to perform photosynthesis and feed many species of aquatic organisms. Macroalgae, which can be divided by characteristic colors in: Red algae (Phylum Rhodopyta), Green algae (Phylum Chlorophyta) and Brown algae (Class Phaeophyceae). Seaweeds are used for centuries by Asians as an important part of their diet, and also as source of compounds to Pharmaceutical and Cosmetic industries. The Brazilian Northeastern has a large coastal area; it possesses several forms of marine organisms especially for being in a Tropical hot water zone. Based on the importance of macroalgae, a chemical study was conducted using five species of algae (Bryothamnion seaforthii; Colpomenia sinuosa; Dictyosphaeria versluysii, Digenea simplex and Galaxaura rugosa) to prepare methanolic extracts and sequential fractions. 72 substances
were identified using the GC-MS technique under the following operation conditions: helium as the carrier gas; 1.52 mL/min flow rate; oven temperature 60 º (25 ºC/min 300 ºC, heating rate); Splitless; injected volume 1 μL; solvent cutting 3 minutes. The mass spectra were obtained in the same equipment by electron impact ionization (EI) at 70 eV; and the ion source was maintained at 300 ° C. Hexadecanoic acid, in all fractions; 1-isopropyl-1,3,4-trimethyl-cyclohexane, 1,4-diisopropyl-cyclohexane and isopentyl decanoate, first identified in seaweeds; several esters, hydrocarbons and fatty acids can be mentioned as identified substances. / Conselho Nacional de Desenvolvimento Científico e Tecnológico / Alga é uma forma de vida que vive em oceanos e mares e possui a capacidade de realizar fotossíntese e servir de alimento e/ou abrigo para muitas espécies de organismos aquáticos. As macroalgas, que podem ser divididas pela cor característica que possuem, em algas vermelhas (Filo Rhodophyta), verdes (Filo Chlorophyta) e pardas (Classe Phaeophyceae), são utilizadas pelos orientais como parte importante de sua alimentação há vários séculos, além de utilizá-las como fonte de produtos farmacêuticos e da indústria de cosméticos. O nordeste brasileiro possui uma vasta área litorânea e costeira sendo rico em diversas formas de vida marinha, principalmente por estar em uma zona tropical de águas quentes. Com base na importância destas macroalgas, foi realizado um estudo químico de cinco espécies de algas, Bryothamnion seaforthii, Colpomenia sinuosa, Dictyospharia versluysii, Digenea simplex e Galaxaura rugosa, através de extratos metanólicos e frações. A técnica de cromatografia gasosa acoplada à espectroscopia de massas foi utilizada com as seguintes condições de operação: hélio como gás de arraste; velocidade do fluxo, 1,52 mL/min; a temperatura inicial
do forno foi 60 ºC, com uma taxa de aquecimento de 25 ºC/min até 300 ºC; “Splitless”, como modo de injeção; 1 μL de volume injetado; e corte do solvente em 3 minutos. O tempo total de corrida foi 25 minutos. Os espectros de massas foram obtidos no mesmo equipamento, através de ionização por impacto de elétrons (EI) de 70 eV; e a fonte de íons foi mantida a 300 ºC. Desse modo foram identificadas 72 substâncias, dentre as quais podem ser citadas: ácido hexadecanoico, presente em todas as frações; os hidrocarbonetos 1-isopropil-1,3,4-trimetilcicloexano, 1,4-di-isopropilcicloexano e decanoato de isopentila, identificados pela primeira vez em algas; além de diversos ésteres, hidrocarbonetos e ácidos graxos.

O arroz (Oryza sativa L.), principal alimento para cerca de metade da população mundial, é consumido principalmente na forma polida. Contudo, o arroz integral vem se destacando, devido principalmente aos compostos bioativos presentes nas camadas mais externas do grão. Os benefícios à saúde são atribuídos, em parte, à sua capacidade de combater radicais livres e exercer atividades biológicas, tais como a inibição de determinadas enzimas. Neste trabalho foram analisados os teores de compostos fenólicos totais (FT), solúveis (FS) e insolúveis (FI) e avaliado o efeito do cozimento de 17 genótipos de arroz integral, sete com pericarpo pigmentado e dez genótipos não-pigmentados. Ainda foi avaliada a inibição da enzima conversora de angiotensina I (ECA) e da -amilase por esses compostos, no arroz cru e cozido. O arroz pigmentado se mostrou rico em compostos fenólicos, em média da ordem de 4200 µg eq. ácido ferúlico/g, devido aos seus altos teores de FS, constituídos principalmente por antocianinas e proantocianidinas. Os FI, representados principalmente pelos ácidos fenólicos contribuíram com apenas 20% dos compostos fenólicos totais. O arroz não-pigmentado contém, em média, ao redor de 1000 µg eq. ácido ferúlico/g, distribuídos quase equitativamente entre a fração solúvel e insolúvel. O cozimento do arroz provocou redução nos teores de FT e alteração na proporção entre FS e FI. Essas alterações foram mais pronunciadas no arroz com pericarpo vermelho, afetando principalmente a fração solúvel. O arroz preto, contudo, manteve a proporção entre FS e FI após o cozimento. A -amilase não foi inibida de forma significativa pelos fenólicos das amostras de arroz cozido. O arroz pigmentado inibiu mais fortemente a ECA do que o arroz não-pigmentado, levando a crer que a pigmentação seja um fator importante. No entanto, entre os diferentes genótipos pigmentados, o perfil de fenólicos parece ser o fator determinante para a maior ou menor atividade inibitória. O cozimento do arroz reduziu significativamente a inibição da ECA pelos fenólicos, fato observado principalmente nos genótipos vermelhos, devido à diminuição dos teores de FS e da capacidade inibitória dos fenólicos presentes. O arroz preto se destacou por ter o maior teor de fenólicos solúveis e a maior ação inibidora da ECA, após o cozimento. / Rice (Oryza sativa L.) sustains at present about half of the world´s population. Consumption is mainly in its milled form, but brown rice has prompted further research due to bioactive compounds present in the pericarp of the grain. Some of the positive health effects have been attributed to radical scavenging activity and other biological effects such as inhibition of certain enzymes. In this study it was analyzed the contents of total, soluble and insoluble phenolic compounds of 17 different genotypes of brown rice as well as the effect of cooking. Seven genotypes had pigmented pericarp and ten were non-pigmented. In addition, the extracts from crude and cooked rice were tested for their capacity to inhibit the angiotensin I (ACE) converting enzyme and -amylase activities. Pigmented rice genotypes were highest in phenolic compounds, with an average of about 4200 µg ferulic acid eq./g, due to their high contents of soluble phenolics, mostly represented by anthocyanins and proanthocyanidins. Insoluble phenolics, represented mainly by phenolic acids, contributed with only 20% of total phenolics. Non-pigmented rice showed overall lower levels of phenolics. The mean content was about 1000 µg ferulic acid eq./g, almost equally distributed between the soluble and insoluble fractions. Levels of total phenolics were significantly reduced by rice cooking and proportions between soluble and insoluble fractions were altered. These alterations were more pronounced for pigmented rice, and soluble phenolics were the most affected. However, after cooking, black rice was the only that maintained the original proportion between soluble and insoluble phenolics. Alpha-amylase was not significantly inhibited by phenolics after cooking. Pigmented rice showed a potent inhibition of ACE, much higher than of non-pigmented rice, which seems to indicate that color of the pericarp is an important factor. Nevertheless, different profiles of phenolic compounds may explain why individual pigmented genotypes with similar phenolic levels can have different ACE inhibiting capacities. Rice cooking reduced significantly the inhibition of ACE by phenolics, which feature was more pronounced in pigmented rice due to the reduction of soluble phenolics and the activity of individual phenolics. Among pigmented rice, the highest soluble phenolic content and the most potent ACE inhibition after cooking was observed for black rice turning it the most distinguished notable.

It is calculated heat balance of the boiler, the boiler losses and the thermal efficiency of the boiler is determined. After designing the combustion chamber thermal calculation is made. Then, individual heating surfaces are proposed and at the end of the calculations are controlling the overall energy balance of the boiler. For calculation was design documentation added.

First chapters describe technical characteristics of the boiler and possible measurements which would lead to lowering NOx emissions. In order for boiler to meet emission standards valid from 1.1.2016 flue gas treatment by selective catalytic reduction is proposed. Main part of the thesis is thermal calculation of the boiler with modifications necessary in order to apply SCR. The aim of calculation is to determine flue gas exit temperature and temperature at catalyst area in order to assess the suitability of proposed modifications.

First, stoichiometric calculation and the calculations enthalpy of flue gases and air are performed. After calculating the heat balance and loss of the boiler was counted boiler efficiency. The design of individual heating surfaces is proposed and at the end are controlled the overall heat balance of the boiler. The calculation is supplemented by drawings.
