Exogenous modulation of embryonic tissue and stem cells to form nephronal structures

Renal tissue engineering and regenerative medicine represent a significant clinical objective because of the very limited prospect of cure after classical kidney treatment. Thus, approaches to isolate, manipulate and reintegrate structures or stimulating the selfregenerative potential of renal tissue are of special interest. Such new strategies go back to knowledge and further outcome of developmental biological research. An understanding of extracellular matrix (ECM) structure and composition forms thereby a particularly significant aspect in comprehending the complex dynamics of tissue regeneration. Consequently the reconstruction of these structures offers beneficial options for advanced cell and tissue culture technology and tissue engineering. In an effort to investigate the influence of natural extracellular structures and components on embryonic stem cell and renal embryonic tissue, methodologies which allow the easy application of exogenous signals on tissue in vitro on the one hand and the straight forward evaluation of decellularization methods on the other hand, were developed. Both systems can be used to investigate and modulate behaviour of biological systems and represent novel interesting tools for tissue engineering. The novel technique for culturing tissue in vitro allows the growing of embryonic renal explants in very low volumes of medium and optimized observability, which makes it predestined for testing additives. In particular, this novel culture set up provides an ideal opportunity to investigate renal development and structure formation. Further studies indicated that the set is universally applicable on all kinds of (embryonic) tissue. Following hereon, more than 20 different ECM components were tested for their impact on kidney development under 116 different culture conditions, including different concentrations and being either bound to the substrate or dissolved in the culture medium. This allowed to study the role of ECM constituents on renal structure formation. In ongoing projects, kidney rudiments are exposed to aligned matrix fibrils and hydrogels with first promising results. The insights gained thereof gave rise to a basis for the rational application of exogenous signals in regenerative kidney therapies. Additionally new strategies for decellularization of whole murine adult kidneys were explored by applying different chemical agents. The obtained whole matrices were analysed for their degree of decellularization and their residual content and composition. In a new straight forward approach, a dependency of ECM decellularization efficiency to the different agents used for decellularization could be shown. Moreover the capability of the ECM isolated from whole adult kidneys to direct stem cell differentiation towards renal cell linage phenotypes was proved. The data obtained within this thesis give an innovative impetus to the design of biomaterial scaffolds with defined and distinct properties, offering exciting options for tissue engineering and Expression d’une variabilité génétique pour la phénologie de croissance, l’efficience d’utilisation de l’eau et la résistance à la cavitation au sein de populations naturelles de peuplier noir (Populus nigra L.) / Expression of genetic variation for bud phenology, water-use efficiency and xylem resistance to drought-induced cavitation in natural populations of black poplar (Populus nigra L.)

Cette thèse visait à développer nos connaissances quant au contrôle génétique et environnemental et la structuration géographique de la variabilité phénotypique pour des caractères fonctionnels chez le peuplier noir (Populus nigra L.). Trois caractères fonctionnels ont été étudiés : la phénologie de croissance, WUE et la résistance à la cavitation. La variabilité phénotypique exprimée pour la phénologie de croissance et WUE a été évaluée au sein d’une collection européenne de peuplier noir regroupant 1085 génotypes échantillonnés dans 13 métapopulations et installés en test clonal en pépinière dans deux sites expérimentaux. Nos résultats ont révélé une importante variabilité génétique ainsi qu’une importante plasticité phénotypique des caractères phénologiques et de WUE au sein des métapopulations. Une différenciation génétique modérée à forte des métapopulations a été observée pour la phénologie de croissance et WUE. Les différences entre métapopulations se structuraient selon des patrons plus ou moins complexes, qui semblaient refléter, pour la phénologie de croissance, une adaptation locale des métapopulations aux conditions de photopériode et de température. Tenant compte des capacités de phénotypage limitées pour la résistance à la cavitation, la variabilité génétique exprimée pour ce caractère a été évaluée au sein d’une population naturelle de peuplier noir regroupant 33 génotypes et installée en test clonal en pépinière dans un site expérimental. Nous avons détecté une variation significative du niveau de résistance à la cavitation entre génotypes. De manière générale, aucune relation phénotypique significative n’a été détectée entre la phénologie de croissance, WUE et la résistance à la cavitation au niveau intra-population, suggérant le maintien d’une diversité de combinaisons fonctionnelles. Le développement d’une puce à 10 331 marqueurs SNPs distribués à l’échelle du génome de P. nigra, notamment dans des régions candidates pour la phénologie de croissance, devrait permettre, à court terme, de confirmer l’adaptation locale des populations de peuplier noir et d’explorer ses bases génétiques. / This work aimed at improving our understanding of the genetic and environmental control as well as the geographic structure of phenotypic variation for functional traits in black poplar (Populus nigra L.). Three functional traits were studied: bud phenology, water-use efficiency (WUE) and xylem resistance to drought-induced cavitation. The phenotypic variation expressed for bud phenology and WUE was evaluated in a European P. nigra collection composed of 1085 cloned individuals sampled in 13 natural metapopulations and established in clonal test in nursery at two experimental sites. Substantial genetic variation and substantial phenotypic plasticity for bud phenology and WUE were observed within all metapopulations. Moderate to strong genetic differentiation of metapopulations was evidenced for phenological traits and WUE. Patterns of genetic differentiation were more or less complex depending on traits and seemed to reflect, for phenological traits, local adaptation of metapopulations to photoperiod and temperature. Taking into account the limited phenotyping capacity for xylem resistance to cavitation, genetic variation expressed for this trait was evaluated within one single natural population of black poplar composed of 33 genotypes which were established in clonal test in nursery at one experimental site. Significant variations were observed between genotypes for the degree of cavitation resistance. Overall, no significant relationship was detected between bud phenology, WUE and cavitation resistance at the within-population level, indicating the maintenance of a diversity of functional combinations. The development of an array covering 10 331 SNPs distributed across the P. nigra genome, notably in candidate regions for bud phenology, will enable in the short-term to confirm local adaptation of P. nigra populations and to identify its genetic basis.

Phenology of hazelnut big bud mites in Canterbury and implications for management

Eriophyoid big bud mites are key pests of hazelnuts throughout the world, although little is known of the identity and impact of the species on New Zealand hazelnut crops. The key objectives of this study were to determine the species of mite present on New Zealand crops, explore a method of monitoring mite emergence from overwintering big buds, determine the phenology of mites in relation to tree phenology and weather, and identify the optimum timing for control measures. The presence of both Phytoptus avellanae (Nalepa 1889) (Acari: Phytoptidae) and Cecidophyopsis vermiformis (Nalepa 1889) (Acari: Eriophyidae) was confirmed, the latter species being a new record for New Zealand. Preliminary diagnostic DNA sequences were determined for both species. A sticky band technique was developed to monitor mite emergence from overwintering big buds, and mite emergence was found to occur between early and late spring. Mite emergence and movement occurred when daily temperatures were greater than 15 degrees C and when mean temperatures were greater than 9 degrees C, with mite emergence increasing with temperature. It proved difficult to relate the phenology of hazelnut to mite emergence, however, the development of new buds during mite emergence was a crucial factor in the infestation of new buds. An accumulated heat sum model (DD), started at Julian date 152 and using a lower threshold temperature of 6 degrees C, predicted the onset of emergence on two cultivars and at two sites as occurring at approximately 172 DD. A regression model based on leaf number, bud height, bud width, DD and Julian date provided a more satisfactory prediction of percent accumulated mite emergence. It is recommended both peak mite emergence and the appearance of hazelnut buds should be used to optimise the time to apply control measures. Therefore, a control should be applied before buds measure 0.5 x 0.5 mm (width x height), are enclosed within the axil, and have a rounded tip, or, when 50% accumulated mite emergence has occurred, which ever occurs first. A preliminary field experiment tested the application of sulphur (40 g/10 litres of 800 g/kg No Fungus Super Sulphur) at 2, 50 and 80% accumulated mite emergence. The greatest reduction in mite numbers was achieved with an application at approximately 50% emergence. Considerable variation in mite emergence occurred between years, therefore optimum timing of controls would need to be determined by monitoring mites, new buds and weather conditions each year. Field collection of mites also identified the presence of Typhlodromus doreenae Schicha (Acari: Phytoseiidae) which would warrant further study for inclusion in an integrated mite control programme.

Adaptation and acclimation of red alder (Alnus rubra) in two common gardens of contrasting climate

Red alder (Alnus rubra Bong.) is the only tree in British Columbia and the Northwest US to engage in actinorhizal symbiosis to fix atmospheric nitrogen. This study was conducted to explore the plasticity in growth and physiology among 58 17-year-old red alder families in response to variation in climate in two common garden plots, one at Bowser, BC and one at Terrace, BC. Physiological assessments included height and diameter growth, bud flush, water use efficiency as measured by δ13C, cold hardiness as measured by controlled freezing and electrolyte leakage, autumn leaf senescence, and instantaneous and seasonally integrated rates of nitrogen fixation as measured by acetylene reduction and natural abundance δ15N isotope analysis, respectively. Significant differences were identified among families for growth (height and diameter), bud burst stage, leaf senescence, cold hardiness, and bud nitrogen content. No significant differences among families were identified for water use efficiency as measured by δ13C, or for rates of nitrogen fixation as measured by either acetylene reduction or natural abundance δ15N. This study identified possible adaptive differences among red alder genotypes, especially in traits such as bud flush timing, cold hardiness, or nitrogen fixation and their respective contributions to growth. These differences often reflected a tradeoff between growth and the ability to tolerate an extreme environment. Cold hardiness results indicate that red alder families are well adapted to their climate of origin, and may not be able to acclimate sufficiently to a northward assisted migration of genotypes. Nitrogen fixation results demonstrated gaps in our current knowledge of Frankia distribution and impact on the actinorhizal symbiosis in British Columbia. / Graduate

Adventivní odnožování krátkověkých rostlin v přírodních populacích / Adventitious sprouting of short-lived plants in natural populations

Disturbance is one of most important selective factor causing removal of plant biomass. Man-made habitats are characterized by strong and unpredictable disturbances, providing bare soil surface colonized by plants with short life cycle. Populations of the short-lived plants are, however, vulnerable to the strong disturbance removing all stem parts with reserve axillary meristems in the case it occurs before plant flowering and fruiting. Nevertheless, 2 % of annual and 14 % of biennial plants are able to overcome meristem limitation by adventititous sprouting from hypocotyle or/and roots. This thesis is composed of four original studies describing the occurrence of adventitious sprouting in natural populations of 22 monocarpic weeds of Central Europe and one species in Indonesia. The studied phenomenon was analyzed in relation to various environmental factors and plant traits in the field and in experimental conditions.

<p>&nbsp;Acorus calamus L. је вишегодишња зељаста биљка влажних подручја, чија су лековита својства одавно позната. Дрогу иђирота чини ризом који се употребљава као чај, прах, сок, гел, уље или крема. Због прекомерне експлоатације и високог степена угрожености ове дивље лековите биљне врсте у Србији је уведена забрана његовог сакупљања из природе. Да би се задовољила повећана потражња од стране индустрија које га користе, гајење иђирота се намеће као једно од најпрагматичнијих решења. За потребе утврђивање начина гајења, прво се приступило истраживању услова успевања и варирање својстава иђирота са пет локалитета природних станишта: Обедска бара, Делиблатска пешчара, Засавица, Раковац и Дубовац. Потом је заснован двогодишњи пољски оглед на ком је испитиван утицај примене основних агротехничких мера, односно густине садње и ђубрења азотом, на својства биљака. Оглед је постављен 2013. године, на подручју засеока Ћумуране у насељу Рипањ. Код биљка са природних станишта и са огледног поља праћена су следећа својства: висина биљака, дужина ризома, број и дужина бочних грана на ризому, број пупољака на ризому и бочним гранама, број нодуса на ризому и бочним гранама, проценат суве материје ризома, маса свежег и сувог ризома, однос масе свежег и сувог ризома. Код гајених биљака праћен је и принос свежег и сувог ризома. Код свих узорака иђирота испитиван је садржај и састав етарског уља из ризома. Идентификоване су, такође, најзаступљеније компоненте и утврђен је садржаја &beta;-азарона. За утврђивање нивоа плоидије популација из Србије рађено је пребројавање хромозома. Испитивањем биљака са природних станишта, констатовано је да на већину њихових морфолошких одлика, локалитет није имао утицаја. Утицај локалитета испољен је само на: број нодуса на ризому (највећи је био у Дубовцу - 58 а најмањи на Обедској бари - 15), проценат суве материје (највећи је био у Раковцу - 50,2%, а најмањи на Делиблатској пешчари - 37,9%) и однос масе свежег и сувог ризома (највећи је на Делиблатској пешчари - 2,64, а најмањи у Раковцу - 1,90). У пољском огледу добијени су следећи резултати: висина биљака је била највећа при најмањој густини усева (35000 биљака/ha) и најмањој дози азота (60 kg/ha) дужина ризома, као и број и дужина њихових бочних грана су највећи при средњој густини садње (48000 биљака/ha) и најмањој дози азота; број пупољака на ризому има највећу вредност при најмањој густини садње и најмањој дози азота. Број пупољака прве бочне гране је био највећи на највећој густини садње (62000 биљака/ha) и при најмањој дози азота, а број нодуса на ризому није зависио од густине садње, као ни од доза ђубрења. Највећа маса свежег и сувог ризома остварена је при средњој густини садње (48000 биљака/ha) и најмањој (62000 биљака/ha) дози азота, док је проценат суве материје био највећи при највећој густини и најмањој дози азота (60 kg/ha). На однос масе свежег и сувог ризома утицај нису имали ни ђубрење ни густина</p><p>садње усева, док су приноси свежег и сувог ризома били највећи на највећој густини садње и при најмањој дози азота. Садржај етарског уља иђирота из природе није показао значајна одступања између локалитета. Садржај етарског уља ризома гајеног иђирота опадао је са порастом густине садње, док различите дозе азота нису утицале на ову особину. Максимална концентрација &beta;-азарона у етарском уљу иђирота са природног станишта била је 17,07 % (локалитет Раковац), док је највећи садржај &beta;-азарона код гајеног иђирота био 21,41 %. Повећана концентрација &beta;- азарона се објашњава интензивнијим метаболизмом азота, због повећане количине из ђубрива. обијени резултати потврђују да је концентрација &beta;-азарона у етарском уљу ризома иђирота слична резултатима из других земаља Европе. Бројањем хромозома утврђено је да иђирот припада триплоидном, европском варијетету Аcоrus cаlаmus vаr. cаlаmus.</p> / <p>&nbsp;Acorus calamus L. je višegodišnja zeljasta biljka vlažnih područja, čija su lekovita svojstva odavno poznata. Drogu iđirota čini rizom koji se upotrebljava kao čaj, prah, sok, gel, ulje ili krema. Zbog prekomerne eksploatacije i visokog stepena ugroženosti ove divlje lekovite biljne vrste u Srbiji je uvedena zabrana njegovog sakupljanja iz prirode. Da bi se zadovoljila povećana potražnja od strane industrija koje ga koriste, gajenje iđirota se nameće kao jedno od najpragmatičnijih rešenja. Za potrebe utvrđivanje načina gajenja, prvo se pristupilo istraživanju uslova uspevanja i variranje svojstava iđirota sa pet lokaliteta prirodnih staništa: Obedska bara, Deliblatska peščara, Zasavica, Rakovac i Dubovac. Potom je zasnovan dvogodišnji poljski ogled na kom je ispitivan uticaj primene osnovnih agrotehničkih mera, odnosno gustine sadnje i đubrenja azotom, na svojstva biljaka. Ogled je postavljen 2013. godine, na području zaseoka Ćumurane u naselju Ripanj. Kod biljka sa prirodnih staništa i sa oglednog polja praćena su sledeća svojstva: visina biljaka, dužina rizoma, broj i dužina bočnih grana na rizomu, broj pupoljaka na rizomu i bočnim granama, broj nodusa na rizomu i bočnim granama, procenat suve materije rizoma, masa svežeg i suvog rizoma, odnos mase svežeg i suvog rizoma. Kod gajenih biljaka praćen je i prinos svežeg i suvog rizoma. Kod svih uzoraka iđirota ispitivan je sadržaj i sastav etarskog ulja iz rizoma. Identifikovane su, takođe, najzastupljenije komponente i utvrđen je sadržaja &beta;-azarona. Za utvrđivanje nivoa ploidije populacija iz Srbije rađeno je prebrojavanje hromozoma. Ispitivanjem biljaka sa prirodnih staništa, konstatovano je da na većinu njihovih morfoloških odlika, lokalitet nije imao uticaja. Uticaj lokaliteta ispoljen je samo na: broj nodusa na rizomu (najveći je bio u Dubovcu - 58 a najmanji na Obedskoj bari - 15), procenat suve materije (najveći je bio u Rakovcu - 50,2%, a najmanji na Deliblatskoj peščari - 37,9%) i odnos mase svežeg i suvog rizoma (najveći je na Deliblatskoj peščari - 2,64, a najmanji u Rakovcu - 1,90). Brojanjem hromozoma utvrđeno je da iđirot pripada triploidnom, evropskom varijetetu Acorus calamus var. calamus.</p> / <p>&nbsp;&nbsp;Acorus calamus L. is a perennial herbaceous plant found in wet areas, whose medicinal properties have been long known. The drug of sweet flag is made from the rhizome which is used as tea, powder, juice, gel, oil or cream. Because of overexploitation and the high degree of endangerment of this wild medicinal plant species in Serbia collecting was banned. In order to meet the increased demand for this plant by industries that use it, cultivation of sweet flag is emerging as one of the most pragmatic solutions. For the purpose of determining the method for plant growth, the first approach was studying the growing conditions and the variation of properties of sweet flag from five natural habitats: Obedska pond, Deliblato Sands, Zasavica, Rakovac and Dubovac.After that a two-year field experiment was designed in which the effect of application of basic agrotechnical measures, i.e., different planting density and doses of nitrogen fertilization on the plants, was studied. The experiment was set up in 2013, in the area of the hamlet Ćumurana in the settlement Ripanj. In plants from natural habitats and the experiment, the following properties were measured: the height of the plants, the length of the rhizome, the number and length of lateral branches on the rhizome, the number of buds on the rhizome and lateral branches, the number of leaf scars on the rhizome and lateral branches, the percentage of dry matter of the rhizome, the mass of the fresh and the dry rhizome, the ratio of mass between the fresh and the dry rhizome. In the experiment with cultivated plants, the yield of the fresh and the dry rhizome was also measured. For all samples of sweet flag, the content and composition of the essential oil from the rhizome was measured. Also, the main components were identified and the content of the &beta;-asarone was determined. For the purposes of identifying the ploidy, i.e., which varieties the sweet flag from Serbia belongs to, counting of chromosomes was done. By examining plants from natural habitats, it was noted that the site had no influence on the majority of morphological features of plants. The impact of the sites was manifested only in the number of leaf scars (the largest number was in Dubovac-58,0 and the smallest number was in Obedska pond-15,0), percentage of dry matter (the largest percentage was in Rakovac-50.2% and the smallest percentage was in Deliblato Sands-37,9 %) and the ratio of mass between a fresh and a dry rhizome (the largest ratio was in Deliblato Sands-2,6 and the smallest ratio was in Rakovac-2,2). In the field experiment, the following results were obtained: the height of the plants is the largest at the lowest planting density (35000 plants/ha) and lowest nitrogen dosage (60 kg/ha), the length of the rhizome, as well as number and length of lateral branches of the rhizome are largest at medium planting density (48000 plants/ha) and the lowest dosage of nitrogen (60 kg/ha), the number of buds on the rhizome has the highest value at the lowest planting density (35000 plants/ha) and the lowest dosage of nitrogen (60 kg/ha). The number of buds on the first lateral branch was largest at the largest planting density (62000 plants/ha) and the lowest dosage of nitrog (60 kg/ha) , and the number of leaf scars on the rhizome does</p><p>not depend on the planting dosage or the dosage of fertilizing. The largest mass of the fresh and the dry rhizome is accomplished at medium planting density and the lowest dosage of nitrogen, while the percentage of dry matter was largest at the largest density and the lowest dosage of nitrogen. The fertilization and the crop density had no effect on the ratio of mass between the fresh and the dry rhizome, while the yields of the fresh and the dry rhizome were largest at the largest planting density and the lowest dosage of nitrogen. The content of essential oils of natural sweet flag showed no significant discrepancies between the sites. The content of essential oil of cultivated sweet flag rhizomes declined with the increase of planting density, while different nitrogen doses had no effect on this property. The maximum concentration of &beta;-asarones in the essential oil of natural sweet flag was 17,07 % (Rakovac), while the largest content of &beta;-asarones in cultivated sweet flag was 21,41 %. The larger concentration of &beta;-asarones is explained through increased nitrogen metabolism, because of increased amounts of fertilizer. The results confirm that the concentration of &beta;-asarone in the essential oil of sweet flag rhizomes is similar to the concentration of essential oil of sweet flag rhizomes from Europe. By counting the chromosomes it is established that sweet flag belongs to the triploid, European variety of Аcоrus cаlаmus vаr. cаlаmus.</p>

Es handelt sich um die Forschungen zu der Auswanderungsbewegung der sog. Altlutheraner unter Führung des Dresdner Pfarrers Martin Stephan nach Missouri/USA. Ein zufällig gefundener Vermerk des Radeberger Superintendenten Martini 1839 wurde zum Ausgangspunkt der Recherchen zu Martin Stephan, der sich mit seinen Vertrauten im Radeberger Bad /Augustusbad aufgehalten hatte. Ein Großteil der Vorbereitungsarbeiten für die Auswanderung, einschließlich der konkreten und abschließenden Auswanderungspläne entstanden hier im Augustusbad. Der Spuk fand ab Frühjahr 1838 rund um das „Radeberger Bad“, Lotzdorf und Liegau statt, argwöhnisch beobachtet von der Dresdner Polizei. Unter Führung Stephans erfolgte 1838 die sächsische Lutherische Auswanderung mit über 700 Anhängern aus Sachsen und Thüringen in die Vereinigten Staaten. Damit wurde der Grundstein für die heutige „Lutheran Church - Missouri Synod“ gelegt, die heute zweitgrößte lutherische Kirche der USA, die sein Nachfolger, Pfarrer C.F.W. Walther, gründete und die heute über 2,3 Millionen Mitglieder in den USA verzeichnen kann.

Mit den revolutionären Dresdner Unruhen 1830 änderte sich die staatliche Situation im Königreich Sachsen und der Kirche grundlegend. Es kam zum Sturz und zur Amtsenthebung des bisherigen Förderers pietistischer Ideen, des Grafen Detlev von Einsiedel als Kabinettsminister im Sächsischen Ministerium. Mit der Einsetzung des aus Altenburg stammenden liberalen Bernhard August von Lindenau (1779-1854) in dieses Amt veränderte sich die Gesamtsituation durch Re-formen und neue Gesetzgebungen grundlegend. Für konservative Theologen und Anhänger der Erweckten erschien es durch die Einflussnahme des Staates auf das Schulsystem und die kirchliche Agende immer unerträglicher, in der Heimat zu leben, zu arbeiten und ihre Auslegungen ihres lutherischen Glaubens in alter Form zu praktizieren. Unter Führung Martin Stephans erfolgte 1838 die sächsische Lutherische Auswanderung mit über 700 Anhängern aus Sachsen und Thüringen in die Vereinigten Staaten.
