Role of macromolecules in coccolithophore biomineralizationWalker, Jessica Mary January 2018 (has links)
Biomineralization refers to the production of mineralized tissues by organisms. The fine control which organisms can exert over this process produces crystals with morphologies and properties contrasting to that of non-biogenic crystals and specifically altered to suit the required functional need. A key model system of biomineralization are a unicellular marine algae, coccolithophores, which produce calcium carbonate scales known as coccoliths. These coccoliths are comprised of arrangements of single crystals of calcite interlocked to form a plate-shaped structure. Coccoliths are developed intracellularly in a specialised compartment called the coccolith vesicle, before being extruded to the cell surface. In this work, two vital components of the coccolith biomineralization process are investigated - a soluble polysaccharide thought to act as a habit modifier and an insoluble organic scaffold known as a baseplate that provides the surface for nucleation and growth of the crystals. Whilst both these elements are thought to play a key part in the biomineralization process, the role of each is not fully understood. To investigate the effect of coccolith-associated polysaccharides (CAPs) on nucleation and polymorph selection, two systems that promote different polymorphs of calcium carbonate were utilised. In both systems, the intracrystalline polysaccharide fraction extracted from one species, Gephyrocapsa oceanica, was able to promote calcite nucleation in vitro, even under conditions favouring the kinetically-privileged polymorphs of calcium carbonate: vaterite and aragonite. As this property is not observed with CAPs extracted from its 'sister species', Emiliania huxleyi, the in vivo function of CAPs may differ between the two species. Both cryo-transmission electron microscopy (cryoTEM) and scanning electron microscopy (SEM) were used to determine the mechanism of calcite growth in the presence of G. oceanica CAPs, showing its impact on the forming amorphous calcium carbonate (ACC), decreasing the size of the particles and producing irregular, angular particles. Using cryo-electron tomography (cryoET), it was possible to create a 3D representation of the structure of the baseplate from the coccolithophore Pleurochrysis carterae, revealing its two-sided organisation. Examination of several stages of the coccolith growth process demonstrated the interlocking nature of the calcite crystals that make up the coccolith and the progression of the crystal morphologies over time, and the interaction of these crystals with the baseplate rim. Additionally, the effect of inhibiting carbonic anhydrase (CA), an enzyme involved in the regulation of carbonate species, revealed that inhibition of CA can affect coccolithogenesis as well as cell proliferation.

Response of Phytoplankton to Climatic Changes during the Eocene-Oligocene Transition at the North Atlantic ODP Site 612 / Fytoplanktons respons till klimatförändringar under Eocen-Oligocen övergången vid Nordatlanten ODP Site 612Rivero Cuesta, Lucía January 2015 (has links)
The development of modern glacial climates occurred during the Eocene-Oligocene transition (34 to 35.5 Ma) when a decrease of atmospheric CO2 led to a global temperature fall. The ocean was deeply affected, both in the surface and the deep-sea, suffering a strong reorganization including currents and phytoplankton distribution. Spanning that time, 35 samples from the North Atlantic Ocean Drilling Program Site 612 have been analyzed by counting coccoliths abundance in different size groups (< 4 µm, 4 to 8 µm and > 8 µm) and silica fragments abundance. Absolute coccoliths abundance were estimated with two different methods, the “drop” technique and microbeads calibration. In addition, a fragmentation index was calculated to assess the preservational state of the samples. The results obtained fit in the global picture of a decrease in phytoplankton abundance across theEocene-Oligocene boundary, although coccolith and silica fragments abundances show slight different patterns. Absolute abundances estimates showed a large difference between the “drop” and the microbeads methods. The temperature at which samples are dried seems to affect microbeads distribution, leading to an underestimation at temperatures higher than 60º C. In future work the current dataset will be updated with additional calibration and replicate counts to confirm that the “drop” estimates are the more valid results. As the fragmentation index was fairly constant in all samples, no major differences in nannofossil preservation were inferred. Coccoliths abundance drops are thought to be triggered by global temperature fall, general decrease of atmospheric CO2, changes in oceanic circulation, pulses of nutrients or a combination of those. / Under tidsspannet som täcker övergången mellan eocen och oligocen, för ungefär 35.5 till 34 miljoner år sedan, genomgick jordens klimat en stor förändring. Under eocen hade vår planet ett varmare klimat och var i ett så kallat ”greenhouse state”. Mot slutet av denna period och i början av oligocen skiftade emellertid klimatet till en kallare regim, ett så kallat ”icehouse state”. Under detta tillstånd minskade andelen koldioxid i atmosfären vilket medförde att den globala temperaturen minskade. Vidare påverkades också havet och speciellt de fytoplankton som levde där, då de påverkas av temperatur och inflödet av näringsämnen. Fytoplankton står för en betydande del av jordens pågående fotosyntes samt är basen av den organiska matkedjan. Syftet med denna undersökning är att studera förekomsten av coccoliter, små kalcitplattor som produceras av en typ av nannoplankton som kallas coccolitoforider. Coccoliter från en djuphavskärna härstammande från norra Atlanten har därför samlats in och för-ändringen av mängden fytoplankton över nämnda tidsspann mätts. Vidare har också bitar av kisel från andra växtplankton räknats. Resultatet av denna studie var att båda grupperna var rikligare under den sista delen av eocen men mängden sjönk snabbt i början av oligocen. Det finns inte tillräckligt med information för att reda ut orsakerna av detta, men det är troligt att minskningen i temperatur och CO2-tillgängligheten för fotosyntesen är viktiga faktorer.

Paléohydrologie de surface des mers nordiques à l’Holocène terminal (derniers 3000 ans) : le message du phytoplancton à squelette calcaire et organique / Late Holocene surface water changes in the eastern Nordic Seas : the message from carbonate and organic-walled phytoplankton microfossilsDylmer, Christian 17 December 2013 (has links)
La variabilité de l’intensité du flux d’eaux atlantiques et de la nature des masses d’eau de surface le long des marges occidentales de la Norvège, de la mer de Barents et du Svalbard a été reconstituée sur la base des assemblages de coccolithes et dinokystes présents dans cinq carottes sédimentaires marines représentatives de l’Holocène terminal. Les résultats sont présentés sous la forme de reconstructions qualitatives et quantitatives (fonctions de transfert MAT) à haute résolution temporelle (échelle décennale à sub-séculaire). Un travail visantà valider les traceurs micropaléontologiques utilisés a été réalisé en parallèle à l’objectif principal, et s’est en particulier nourri de la collecte et de l’examen de populations vivantes distribuées le long de plusieurs transectszonaux en mer de Norvège, mer d’Islande et à travers le détroit de Fram.Nos résultats indiquent que la partie orientale des mers Nordiques (66 à 77°N) a été sujette à une tendance globale à l’augmentation du flux d’eaux atlantiques (AW) au cours des derniers 3000 ans. La dynamique récente de ce flux méridien est supposée répondre à la modulation long-terme de la force et de la localisation de la ceinture des vents d’ouest qui est essentiellement pilotée par l’Oscillation Nord Atlantique. Ce même mécanisme atmosphérique réconcilie le déplacement zonal et contradictoire du front arctique entre le domaine ouest-norvégien, et les façades occidentales de la mer de Barents et du détroit de Fram. La variabilité rapide du flux d’AW reproduit la succession des phases climatiques historiques classiques chaudes (Période Chaude Romaine, Période Chaude Médiévale, Période Moderne : flux accentué d’AW) et froides (Période Sombre, Petit Age Glaciaire : flux réduit d’AW) des derniers 2500 ans. Un événènement rapide de renforcement du flux d’AW en mers Nordiques a été identifié pendant le Petit Age Glaciaire entre 330 et 410 ans BP (cal.). Nos résultats indiquent que les variations d’intensité du flux d’AW vers l’Océan Arctique ont eu un impact majeur sur la distribution de la glace de mer arctique au cours du dernier millier d’années, les variations reconstruites de l’extension du couvert de glace à l’echelle de l’océan arctique étant parfaitement corrélées (échelle subséculaire) avec nos reconstructions qualitatives de la dynamique de l’AW au large du Svalbard et de la mer de Barents. La diminution importante de l’extension de la banquise durant le 20ème siècle est synchrone d’un flux record d’AW à travers le détroit de Fram, flux qui, d’après nos données, est sans précédent pour les derniers 3000 ans. / Five marine sediment cores distributed along the Norwegian, western Barents Sea, and Svalbard continental margins have been investigated in order to reconstruct late Holocene changes in the poleward flow of the Norwegian Atlantic Current (NwAC) and West Spitsbergen Current (WSC) and the nature of the upper surface water masses within the eastern Nordic Seas. This research project is based on the use of dinocyst and coccolith assemblages for qualitative and quantitative reconstructions of surface water conditions from high resolution sediment cores, and involve upstream investigations on proxy reliabilities. The investigated area (66 to 77°N) was affected by an overall increase in the strength of the AW flow from 3000 cal. yrs BP to the Present. The long-term modulation of westerlies strength and location which are essentially driven by the dominant mode of the North Atlantic Oscillation (NAO), is thought to explain the observed dynamics of the AW flow. The same mechanism also reconciles the recorded opposite zonal shifts in the location of the Arctic Front between the area off western Norway and the western Barents Sea-eastern Fram Strait region. Submillenial changes in AW flow are organised according to known pre-Anthropocene warm (RWP, MCA and the Modern period: strong poleward flow) and cold (LIA, DA: weak poleward flow) climatic spells. A sudden short pulse of resumed high WSC flow interrupted the LIA in the eastern Nordic Seas from 330 to 410 cal. yrs BP. Our results are indicative of a major impact of AW flow dynamics on the Arctic sea ice distribution during the last millenium, when changes in reconstructed sea-ice extent are negatively correlated with the strength of the WSC flow off western Barents Sea and western Svalbard. The extensive decrease in sea ice extent during the last century is synchronous with an exceptional increase in AW flow. The previously reconstructed high amplitude warming of surface waters in eastern Fram Strait at the turn of the 19th century was therefore primarily induced by an excess flow of AW which stands as unprecedented over the last 3000 years.

Size and Abundance of Late Pleistocene Reticulofenestrid Coccoliths from the Eastern Indian Ocean in Relation to Temperature and Aridity / Storlek och abundans av Pleistocen coccoliter från östra Indiska oceanen i förhållande till temperatur och torrhetvan Dijk, Jeroen January 2017 (has links)
Measurements on coccolith abundance and mass can be used as a signal of primary productivity and pelagic calcification in response to environmental change. The Leeuwin Current (LC) is known to transport warm and low-salinity waters from the Indo-Pacific Warm Pool (IPWP) southwards along the coast of West Australia. Along with the onset of continental aridity during late Neogene, increased strength of the LC may have played a role in reef expansion on the Northwest Shelf. In this study the morphological variation in size and mass of reticulofenestrid coccoliths was assessed in material from IODP Site U1461 in the eastern Indian Ocean spanning the past 500 ka. Both the absolute abundance of all reticulofenstrid coccoliths (Emiliania huxleyi, Reticulofenestra spp., Gephyrocapsa spp. and Pseudoemiliania spp.) was determined, as well as the relative abundance of large versus small coccoliths. Coccolith size and mass were measured quantitatively under circularly polarized light. The data was compared to variations in sea surface temperatures (SST) of the LC, and to continental aridity of Australia. SST fluctuations could influence coccolithophore productivity by affecting their metabolic rate, whereas continental aridity may influence the influx of terrestrial matter by wind. The investigated interval is dominated by small species of Gephyrocapsa. Peak values of absolute abundance and mass were observed during Marine Isotope Stage (MIS) 11, an interglacial period of extended warmth and humidity. These results coupled with high densities of aragonite needles in the same samples indicate the sediments were diluted by material overflowing from the adjacent shallow- water carbonate platform, analogous to the whiting events observed in the modern-day Bahamas. A decrease in abundance of Gephyrocapsa caribbeanica at 240 ka can be linked to the timing of their last common occurrence (LCO), within MIS 7. The subsequent shift to Gephyrocapsa oceanica as the dominant large species may indicate an ecological replacement of G. caribbeanica, or signify warm and low-salinity waters. / Mätningar av abundans och massa hos coccoliter kan användas som en signal för primärproduktion och pelagisk förkalkning som resultat av miljöförändringar. Développement du thermomètre Δ47 appliqué sur coccolithes : de la calibration en laboratoire à l’applicabilité au registre sédimentaire / Development of a coccolith-based Δ47 thermometer : from laboratory cultures to the applicability to the sedimentary archiveKatz, Amandine 05 July 2017 (has links)
Le géothermomètre Δ47 est basé sur la relation entre l’abondance des liaisons 13C–18O des carbonates et la température de calcification. Ce proxy contourne potentiellement les limites des autres thermomètres (δ18O, Mg/Ca) pour reconstruire les paléo-températures des océans, expliquant son développement exponentiel depuis dix ans. Cette thèse teste pour la première fois le potentiel et les limites de la thermométrie Δ47 sur les coccolithes, des nannofossiles calcaires produits par des organismes calcifiants dans la zone photique. Ces biominéraux calcitiques et ubiquistes constituent une part importante de l’archive sédimentaire. Des cultures in vitro nous ont permis d’établir que trois espèces de coccolithes actuelles enregistrent la même relation Δ47 – T que la calcite inorganique, alors qu'elles présentent de très larges effets vitaux en δ18O (±5‰). Nous concluons que ces espèces de coccolithes d'importance géologique ne présentent pas d’effets vitaux en Δ47. Nous avons ensuite appliqué le Δ47 à l’étude des sédiments enregistrant l’événement d’anoxie océanique du Toarcien (–183 Ma) au cours duquel les reconstructions de températures restent encore ambigües, notamment du fait de la méconnaissance de la composition isotopique en oxygène de l’eau de mer. Sur la base des données Δ47 acquises, nous proposons des températures élevées (de l’ordre de 36°C), mais restant relativement stables sur l'intervalle d'étude. En couplant ces températures aux données de δ18O des carbonates, nous suggérons une variation importante du δ18O de l'eau de mer dans le Bassin de Paris lors de la mise en place des faciès black shales. Enfin, sur des sédiments pélagiques subactuels, l’une des espèces étudiées présente des déséquilibres isotopiques en Δ47 non observés en culture et explicables par d’autres paramètres environnementaux comme l’intensité lumineuse. Cette thèse illustre le potentiel du thermomètre Δ47 des coccolithes en différents contextes, ouvrant un vaste champ d’application de reconstruction des paléo-environnements sur le Méso-Cénozoïque / The Δ47 geothermometer relies on the relationship between the 13C–18O abundance in carbonateand temperature of calcification. This proxy has the potential to overcome limitations of other thermometers(δ18O, Mg/Ca) to reconstruct oceanic paleotemperatures. This thesis evaluates for the first time the potentialand the limitations of the Δ47 thermometry of the coccoliths, the calcareous nannofossils produced byorganisms calcifying in the photic zone. These calcitic and ubiquitous biominerals represent an importantpart of the sedimentary archive. In vitro cultures allow us to establish that three modern coccoliths speciesrecord the same Δ47–T relation than inorganic calcite, although exhibiting substantial δ18O vital effects(±5‰). We conclude that these coccoliths species do not present any Δ47 vital effect. We subsequentlyapplied the Δ47 proxy to sediments from the Toarcian oceanic anoxic events (–183 Ma) during which thetemperatures reconstructions are still elusive, mainly due to the unknown oxygen isotopic composition ofseawater. On the basis of our Δ47 data, we infer relatively high, yet steady temperatures (around 36°C) duringthe examined interval. By combining these Δ47-derived temperatures to carbonate δ18O data, we can suggestdrastic change in the seawater δ18O composition in the Paris Basin at the onset of black shale deposition. Acore top calibration of Δ47 of coccoliths revealed that one of the studied cultured species exhibits Δ47disequilibrium that is accountable by other environmental parameters, such as light irradiance in the naturalenvironment. Thus, this thesis illustrates the potential of the coccolith Δ47 thermometer in different settings,opening a wide range of application to reconstruct the palaeoenvironments over the Meso-Cenozoic Eras
