The purpose of this study is to investigate an animal model of distress (conditioned suppression) to assess the effects of magnesium (MAG) and nicotinamide (B3) administration on recovery of anxiety-like behavior following traumatic brain injury (TBI). Post-traumatic stress disorder (PTSD) is comorbid with TBI and both affects a victim’s ability to maintain daily activities and have a good quality of life. Administration of MAG decreases swelling of the brain considerably and lessens cell death. B3 is a neuroprotective precursor to NAD+ and enhances energy levels as well has help reduce free radicals after TBI. A conditioned suppression procedure is an established method for generating disruptive fearful anxiety-like responses in animals and these treatments may help to reduce anxiety responses.

Celecoxib enhances sorafenib/sildenafil lethality in cancer cells and reverts platinum chemotherapy resistanceWebb, Timothy A 01 January 2016 (has links)
The present studies sought to determine whether the lethality of the drug combination [sorafenib + sildenafil] could be enhanced by the anti-inflammatory agent celecoxib, using ovarian cancer and other tumor cell lines as models. Also, in a dose dependent fashion celecoxib enhanced [sorafenib + sildenafil] lethality in multiple ovarian cancer cell lines. In a dose dependent fashion celecoxib enhanced the ability of [sorafenib + sildenafil] to reduce expression of multiple chaperone proteins in parallel with lower levels of the drug efflux pumps ABCB1 and ABCG2. Over-expression of GRP78 and HSP27 maintained pump expression in the presence of drugs. Cell killing by the 3 drug combination was mediated by mitochondrial / caspase 9 -dependent apoptotic signaling and by RIP-1 / caspases 2 and 4 / AIF -dependent necroptotic signaling. Pre-treatment of intrinsically resistant primary ovarian cancer cells with [celecoxib + sorafenib + sildenafil] significantly enhanced tumor cell killing by a subsequent cisplatin exposure. Similar data were obtained in some cancer cell lines, but not all, using the related platinum containing drugs, oxaliplatin and carboplatin. As our prior publications have also validated in vivo the combinations of [celecoxib + sildenafil] and [sorafenib + sildenafil] as cytotoxic to multiple tumor cell types, combined with the present findings, we would argue that the combination of celecoxib/sorafenib/sildenafil should be explored in a new phase I trial in ovarian cancer.

The combinational logic circuits, which are performing some kind of logic function, can have several realizations. Realizations differ from each other, because of the elements of the database used in circuit. The test of one scheme realization do not necessarily fully verify mistakes of the other realization. The number of pathways in different realizations may also differ. The determination of dependence between test's propriety and the number of pathways for different circuits is the main task in this paper. After finding pathways in different realizations of circuit, and comparing these pathways, the number of fictional pathways in every realization is detected. Special software was developed for calculating pathways in VHDL combinational logic circuits. The software was used for testing pathway calculation operations of circuit realization and for comparing pathways of realizations. The main purpose of this paper was to develop software for pathways in VHDL circuit calculation, to perform experiments using this software, and to estimate the dependence for the number of pathways. The practise of developed software is wide. This system may be implemented for optimising the algorithm of test generator. Usually the test generation program generates more than minimum of possible tests. If the number of pathways in circuit is known, the developed software will help to optimise the algorithm of test generation so that the minimum number of tests would be generated.

Drug repositioning of protease inhibitors in combinational therapy as a treatment forCOVID-19Malmberg, Max January 2021 (has links)
There is currently no standardized treatment of COVID-19. Despite several vaccines being deployed, the need for an effective treatment in hospitalized patients is heavily sought after. In this systematic review, six clinical trials were analyzed for their findings on the effect of commercially available anti-viral protease inhibitors in treatment of COVID-19. The aim of the study was to evaluate the effect on the hospitalization period and mortality of patients suffering from COVID-19 and if HIVprotease inhibitors could be repositioned to treat COVID-19 in combinational therapy. The findings suggest that protease inhibitors targeting the hepatitis C virus (HCV) proteases could potentially beeffective at reducing the hospitalization period and that the effect can be further enhanced by using them in combination with drugs targeting the RNA polymerases. Further studies are needed confirm these findings. Lopinavir/ritonavir which was the most common protease inhibitor included in the study did not reduce the hospitalization period or the mortality significantly. It is unclear whether the HCV protease inhibitors could reduce mortality. Further studies regarding this outcome are warranted.
