Three dimensional evaluation of the TMJ condyle position in different types of skeletal patterns

Objective: To evaluate three-dimensional position of the TMJ condyle within the glenoid fossa in different types of skeletal patterns. Materials and methods: Ninety CBCT images were consecutively selected and divided into skeletal class I, class II and class III. The images were analyzed locating landmarks in the different areas of the condyle and glenoid fossa. All landmarks presented acceptable reliability. The mean results were compared using ANOVA and Bonferroni post-hoc test (p < 0.05). Results: There was a tendency for the anterior joint space to be smaller than the posterior joint space. Statistical analysis, however, evidenced no significant differences between the anterior, superior and posterior joint spaces and the different skeletal patterns or between sides. Conclusion: There was non-concentricity of the condyle for all the groups studied, and no particular direction was statistically significantly favored. It is unclear whether the differences found would be clinically significant, considering anatomical individual variations.

Circular source and detector trajectories in cone-beam computed tomography (CT) are known to collect insufficient data for accurate object reconstruction. One model predicts that the lacking information corresponds to a shift-variant cone of missing spatial frequency components in the local Fourier domain. These predictions were experimentally verified by imaging small, localized objects and observing their Fourier transforms. Measurements indicated that the internal angle of the ‘missing cone’ varies as the angle of locally intersecting x rays with respect to the horizontal plane, as expected. Object recovery was also found to depend greatly on the distribution of the object’s frequency spectrum relative to the missing cone, as predicted. Findings agreed with more anatomically relevant phantoms, which showed preferential intensity discrepancies at gradients oriented within or near the missing cone. Methods for artefact correction are in general limited to approximation unless a priori information is incorporated.

The role of Tm5NM1/2 on early neuritogenesis

Master of Philosophy (Medicine) / The actin cytoskeleton is important in many cellular processes such as motility, and establishing and maintaining cell morphology. Members of the tropomyosin protein family associate with the actin cytoskeleton along the major groove of actin filaments (F-actin), stabilising them and regulating actin-filament dynamics. To date over 40 non-muscle tropomyosin isoforms have been identified, which are encoded by 4 different genes (α, β, γ, δ). Individual tropomyosin isoforms define functionally distinct F-actin populations. Previous studies have shown that tropomyosins sort to distinct subcellular compartments at different stages of development in polarised cells. Neuronal growth cones are highly dynamic polarised structures, dependent on a constant reorganisation of the actin cytoskeleton. By eliminating tropomyosins in a knockout (KO) mouse model, we investigated the role of two tropomyosin isoforms, Tm5NM1 and Tm5NM2 (γTm gene products) in growth cone dynamics and neurite outgrowth. Growth cone protrusion rates were significantly increased in one day old Tm5NM1/2 KO hippocampal neurons compared to WT controls. Neuritogenesis was significantly affected by the elimination of Tm5NM1/2, with a slight decrease in neurite length and an increase in neuronal branching in neurons cultured for four days. At the molecular level, the depletion of Tm5NM1/2 had no impact on the protein levels and activity of ADF/cofilin in hippocampal neurons while in cortical neurons a subtle but significant increase in ADF/cofilin activity was observed. The subtle phenotype in the early stages of neuritogenesis observed from eliminating Tm5NM1/2 may be explained with functional compensation by other tropomyosin isoforms. Functional compensation for the loss of Tm5NM1/2 may be provided by isoforms Tm5a/5b, TmBr2 and Tm4 as they localise to the growth cones, structures where Tm5NM1/2 are normally found. These results suggest that Tm5NM1/2 may not be required for early stages of neuritogenesis but may still play a fine-tuning role for this process.

Bayesian model of axon guidance

An important mechanism during nervous system development is the guidance of axons by chemical gradients. The structure responsible for responding to chemical cues in the embryonic environment is the axonal growth cone -- a structure combining sensory and motor functions to direct axon growth. In this thesis, we develop a series of mathematical models for the gradient-based guidance of axonal growth cones, based on the idea that growth cones might be optimised for such a task. In particular, we study axon guidance from the framework of Bayesian decision theory, an approach that has recently proved to be very successful in understanding higher level sensory processing problems. We build our models in complexity, beginning with a one-dimensional array of chemoreceptors simply trying to decide whether an external gradient points to the right or the left. Even with this highly simplified model, we can obtain a good fit of theory to experiment. Furthermore, we find that the information a growth cone can obtain about the locations of its receptors has a strong influence on the functional dependence of gradient sensing performance on average concentration. We find that the shape of the sensitivity curve is robust to changes in the precise inference strategy used to determine gradient detection, and depends only on the information the growth cone can obtain about the locations of its receptors. We then consider the optimal distribution of guidance cues for guidance over long range, and find that the same upper limit on guidance distance is reached regardless of whether only bound, or only unbound receptors signal. We also discuss how information from multiple cues ought to be combined for optimal guidance. In chapters 5 and 6, we extend our model to two-dimensions, and to explicitly include temporal dynamics. The two-dimensional case yields results which are essentially equivalent to the one dimensional model. In contrast, explicitly including temporal dynamics in our leads to some significant departures from the one-dimensional and two-dimensional models, depending on the timescales over which various processes operate. Overall, we suggest that decision theory, in addition to providing a useful normative approach to studying growth cone chemotaxis, might provide a framework for understanding some of the biochemical pathways involved in growth cone chemotaxis, and in the chemotaxis of other eukaryotic cells.

The mandibular canal at the region of the molar teeth: an evaluation of cone beam volumetric tomography

Master of Science in Dentistry / Objectives: The aims of this study were: • to evaluate the exact level of the mandibular canal using Cone Beam Volumetric Tomography (CBVT) using measurements taken on images from the NewTom3G and i-CAT machines and manually • to determine the course of the mandibular canal in the regions of the first, second , and third molars • to compare the course of the mandibular canals bilaterally • to compare variables measured between the CBVT and panoramic units • to determine appropriate positions for the implant placement at the region of the mandibular molars in relation to the mandibular canal. Methods: Ten mandibles were selected, including seven edentulous and three dentate ones. They were marked at four positions from the distal border of the mental foramina in the posterior direction at intervals of 10.00 mm. On each dry mandible, at four sites namely M0, M1, M2, and M3, Gutta Percha (GP) points, known as markers, were attached to the mandible so that they were parallel to the midline of the mandible on both buccal and lingual sides. On the NewTom 3G and i-CAT, variables of cross-sectional images were measured from the alveolar crest of the mandible to the superior border of the mandibular canal (AC); the lingual rim of the canal to the lingual margin of the mandible (LC); the buccal rim of the canal to the buccal margin of the mandible (BC); the inferior rim of the canal to the lower border of the mandible (IC), and from the lingual margin to the buccal margin of the mandible (BW: Bone Width). Dry mandibles were subsequently sacrificed by cutting at the four marked sites. On each cross-section of mandibles, distances AC, BC, LC, IC, and BW were measured using a caliper as the manual measurement. IC distances on a conventional OrthophosIII panoramic machine were also measured to compare with the CBVT. Data were managed by Microsoft Office Excel 2003 and transferred to the software of Statistics Package for Social Sciences (SPSS) version 15.0 for Windows for analysis. Data were presented as Mean, Standard Deviation (SD), and Mean Difference, and Standard Error of Mean (SEM) with decimal at 0.00. T-test and One-way ANOVA were used to analyse variables measured in which T-test was used to analyse variables with paired samples and One-way ANOVA was used with adjustment for multiple comparisons of Bonferroni. Statistical significance has an assumed P- value of 0.05 or less. Results: The findings showed that there was no significant difference among measured variables from the NewTom 3G, i-CAT and manual measurement (P>0.05). There was significant statistical difference between the Orthophos OPG machine and CBVT system (P=0.00<0.05). There was no significant difference in the course of the mandibular canals bilaterally (P>0.05). On average, Distances AC, BC, LC, and IC were obtained for reference purposes. The bone width of the mandible on the right side was slightly different from that on the left side. Conclusions: The findings implied CBVT was an accurate diagnostic tool for locating the course of the mandibular canal and for placing dental implants in the region of the mandibular molars. The course of the mandibular canal on the left and right sides was variable. The distances measured at the region of the first, second, and third mandibular molars should be considered as a valuable reference. The bone width of the mandible on the right and left sides was slightly different. The accuracy of the NewTom3G and i-CAT was superior to the panoramic Orthophos machine. However, a panoramic radiograph is still valuable in the daily dental clinic.
