Biosyntéza spodních polyketidových řetězců manumycinových antibiotik - faktory ovlivňující jejich délku / Biosynthesis of lower polyketide chains in manumycin antibiotics - the length-affecting factors

Manumycin antibiotics represent an important class of secondary metabolites produced by Streptomyces bacteria. They belong to a big class of polyketide metabolites and posses significant antimicrobial, anti-inflammatory, antitumor, and many other biological activities. They are characterized by two short polyketide chains, which are attached to a central subunit. Polyketide chains are synthesized by enzymes of the iterative type II polyketide-synthase. Mechanism of regulation of the polyketide chains length has not been known yet. Understanding mechanism can lead to biosynthesis of novel manumycin antibiotics with predetermined chain lengths what may improve their biological activities in favour of a practical use of these compounds. We prepared a mutant strain of asukamycin producer Streptomyces nodosus ssp. asukaensis with deletion of genes coding for type I/II β-ketoacylsynthase and protein AsuC14, which is a potential factor affecting lower polyketide chain length, for the identification of the chain length factor in manumycin antibiotics producers. Next, the genes for type I/II β-ketoacylsynthase and potential chain length-affecting factor C14 from strains producing manumycins with variable length of the lower polyketide chains were expressed in this mutant strain. Title: Effect of physiotherapy to reduce the length of hospital stay in patients with total hip arthroplasty: a literature review Objective: The purpose of this thesis is to research and compare available studies investigating the effect of extra physiotherapy in reducing the length of stay for patients with total hip arthroplasty. In addition, to make a compilation of available sources about issue of the hip, total hip arthroplasty in terms of anatomy, kinesiology and biomechanics and length of hospital stay. Methods: This diploma thesis has descriptively-analytical character and is structured in the form of literary review. Results: Physiotherapy decreases length of stay in patients with total hip arthroplasty. Future research could address a specific group of subjects and the issue of cost-effectiveness in reducing the length of hospital stay by physiotherapy. The theoretical part of the diploma thesis deals with the effects of sleep on human health. Lack of sleep has demonstrable effects on a person's physical and mental health. People are succumbing to strong influences of present day society, which degrade their quality of sleep. These influences do not only effect adults, but children and adolescents as well. One can improve sleep quality by following sleep hygiene rules. Nowadays these rules are often violated. The main problem is ignorance. The aim of the practical part of this diploma thesis was to compare satisfaction with quality of sleep between groups of students in elementary schools, students of secondary schools, and adult working people. It was also investigated whether respondents in schools were familiar with sleep hygiene and whether they followed it. The results of the questionnaire surveys found that the group least satisfied with sleep quality was the group with working adults. More than half of the respondents in schools were unaware of sleep hygiene. It was confirmed that more than two thirds of respondents do not follow sleep hygiene rules. In the conclusion, the author has included material on sleep problems for grade eight students of elementary schools. It could be used as a draft chaper for future school textbooks, because...

(English) The dissertation explores the vowel system of Czech L3 Spanish, focusing on three facets of its architecture: vowel quality (following SLM by Flege 1995, 1999, 2003), vowel quantity and vocalic sequences between words. Preliminary studies (Čechová 2013, 2014) suggest that there might be some evidence for the Mechanism of Equivalence in Czech L3 Spanish, since the vowel spaces of both languages consist of the same categories, with different phonetic realizations. Hence, the more similar the sounds are, the harder it is to capture the relevant difference, necessary to attain native-like pronunciation. Vowel quantity, being traditionally associated with the phonological feature of length in Czech (Palková 1994), is not present in Spanish, and compensating for that property, some Czech speakers tend to pronounce accented syllables with extraordinarily longer durations. Finally, vowel sequences in Spanish are usually subjetct to resyllabification, whereas Czech prefers glottalization to keep morphemes of words separated. These predictions were tested in 22 university Czech students with advanced level of Spanish (C1-C1). I investigated the effect of environmental factors (temperature and food availability) on the development and growth of semiaquatic bug Microvelia reticulata Burmeister, 1835. I focused on the developmental rate isomorphy. In a cross-designed experiment, M. reticulata was individually kept over whole juvenile period (eggs to reaching maturity) and subjected to treatments of three temperature (17, 21, 25 °C) and two food availability (full - daily feeding and reduced - 2 days interval) and their interaction. Development and growth was strongly influenced by temperature. Longest postembryonic development measured in individuals kept in the lowest temperature (54 days) contrasted to those kept in the highest temperature (16 days). Mortality was also significantly influenced by temperature; highest mortality rate was found in the 17 °C treatment. Effect of food availability was significant only in low temperature, resulting in the better survival of individuals on the high food level. In other temperature treatments, I found no effect of food on survival rate of M. reticulata. However, the food level influenced development significantly. In the 21 °C and 25 °C treatment, individuals on a full food developed faster than individuals on a reduced food. In 17 °C the food availability had no effect on time required to reach maturity. The best fitness (expressed as combination of low mortality rate and short development) was observed in 21 °C, both other temperature treatment were found stressing. Food availability had stronger effect on female individuals compared to males. In conclusion, I found no evidence of developmental rate isomorphy in M. reticulata. Wing polymorphism of semiaquatic bugs is affected by environmental factors as temperature, photoperiod, population density or permanency of habitat. The heritability has its own specific part, too. A lot of field studies suggest that crucial effect on wing polymorphism has temperature, but laboratory experiments often do not confirm this hypothesis. The main aim of this thesis was to describe the effect of environmental factors (photoperiod, temperature and permanency of habitat) on wing polymorphism of Microvelia reticulata, additionally of Gerris lacustris. The partial tasks included description of effect of mentioned abiotic factors on mortality and rate of development. The individuals of both species were kept at combinations of two levels of temperature, photoperiod and permanency of habitat (overall at eight treatments) from first larval instar to adult. All adults of Microvelia reticulata emerged as apterous, so there was no response of wing polymorphism to tested environmental factors. The main influence of heritability is presumable. Only ten individuals of G. lacustris developed into the adult stage, so it was impossible to describe the influence of abiotic factors on wing polymorphism, mortality and lenght of development in this species. Mortality of individuals of M. reticulata was significantly influenced by the type of substrate. Only 3.5% of individuals survived when reared on wet filter paper, while 80.5% on the water surface. Both other factors (temperature, photoperiod) had no effect on total mortality. There was no difference between mortality of males and females, so it is impossible to note sexual dependent mortality. The lenght of development did not differ between sexes either. Lenght of development was significantly affected by temperature, as low temperature reduces development of M. reticulata. The fastest development showed nymphs of second instar and slowest development nymphs of fifth instar at all treatments. Since the effect of abiotic factors on wing polymorphism was not found in M. reticulata, it would be sufficient to study the effect of heritability on development of wings in this species in the future.

This thesis focuses on unemployment of young people. Young people are among disadvantaged groups in the labor market, so they are given special attention. The main goal of this work is to analyze unemployment of young people in the Czech Republic, monitor its development and evaluate the changes. Attention is paid to the relationship between unemployment and education. The indicator of average length of education is used along with educational structure to evaluate the level of education. The work also focuses on the economic activity of young people, which has significantly reduced in recent years. At the end of this thesis is analyzed unemployment and education of young people in the regions of the Czech Republic.
