Vacas holandesas, n=158, produzindo 26±9 kg de leite/dia e 213±112 dias pós-parto (DPP), foram aleatoriamente distribuídas em 3 grupos: Controle (GC; n=52); GnRH (GG; n=55), 100 mcg IM. de gonadorelina (Cystorelinâ); e hCG (GH; n=51), 2500 UI IM de hCG (Vetecorâ). A temperatura retal foi verificada no momento da IA, e as amostras de sangue coletadas nos dias 5; 7 e 12 após IA para avaliar as concentrações séricas de progesterona (P4). A prenhez foi determinada entre os dias 42 e 49 após IA. Os dados foram analisados pelo procedimento MIXED do SAS, e foram incluídos no modelo efeitos do tratamento, ordem de lactação, produção de leite, temperatura retal no momento da IA e suas interações. O tratamento com GnRH ou hCG aumentou a P4 sérica (P<0,01). As concentrações séricas de P4 (ng/ml; média ± EPM) para GC, GG, e GH foram, respectivamente, no dia 5: 2,71±0,43; 2,45±0,44 e 3,23±0,45, no dia 7: 4,79±0,43; 4,15±0,44 e 5,75±0,45, e no dia 12 após IA: 5,21±0,43; 6,91±0,44 e 8,50±0,45. Devido ao aumento proporcional na concentração sérica de P4 entre os dias 5 e 7 após IA (GC: 77, GG: 69, GH: 77%; P<0,01) é provável que os tratamentos não tenham induzido o efeito luteotrópico no CL existente, porém devido ao grande aumento na P4 sérica no dia 12 nos animais tratados (GG: 112, GH: 87%; P<0,01) em relação aos animais controle (GC: 16%; P=0,31), provavelmente houve a indução de um novo CL. A temperatura retal média das vacas no momento da IA foi de 39,7°C e o tratamento com GnRH ou hCG aumentou as taxas de concepção (média ± EPM) nas vacas com temperatura retal abaixo de 39,7°C... / This study evaluated the effect of GnRH (gonadotropin releazing hormone) or hCG (human chorionic gonadotropin) injection on day 5 after artificial insemination (AI) on conception rates in lactating dairy cows during the summer. Holstein cows, 158, producing 26 ± 9 kg milk/d and 213 ± 112 days in milk, were randomly assigned to one of 3 treatments groups: control (CG: n=52); GnRH (GG: n=55), injection of 100 mcg i.m. of gonadorelin (Cystorelin®) and hCG (HG: n=51), injection of 2500 UI hCG i.m. (Vetecor®). Rectal temperature was checked at the moment of AI and blood samples collected on days 5, 7 and 12 after AI to evaluate serum progesterone (P4) concentrations. Pregnancy was determined between 42 and 49 d after AI. Data were analyzed by the MIXED procedure of SAS and were included in the model the effects of treatment, parity, milk production, rectal temperature at the moment of AI and interactions. Treatment with GnRH or hCG increased serum P4 (P<0,01). Serum P4 concentrations (ng/ml, mean±SEM) for CG, GG and HG were, respectively, on day 5 2.71±0.43, 2.45±0.44 and 3.23±0.45, on day 7, 4.79±0.43, 4.15±0.44 and 5.75±0.45 and on day 12 after AI, 5.21±0.43, 6.91±0.44, 8.50±0.45. Due to the proportional increase in serum P4 concentration from d 5 to 7 after AI (CG: 77, GG: 69, HG: 77%, P<0,01), it is likely that treatments did not have luteotropic effect on the existing CL, but due the higher increase in serum P4 on day 12 in the treated animals (GG: 112, HG: 87%; P<0,01) in relation to the control group animals (GC: 16%; P=0,31), probably induced the formation of a new CL. The average rectal temperature of cows at time of AI was 39,7°C. Treatment with GnRH and hCG increased conception rates (mean±SEM) in cows with rectal temperature below 39,7°C (CG: 10.1±0.075, n= 26, GG: 36.8±0.073, n=27, HG: 32.8±0.088%, n=21; p<.01)... Effect of roughage to concentrate ratio on ruminal fermentation and protein degradability in dairy cows

Published research suggests that it might be beneficial to increase the amount of rumen undegradable protein (RUP) that passes out of the rumen, through manipulation of rumen fermentation to establish a lower rumen pH. To test this hypothesis, a study was conducted in which three ruminally cannulated Holstein cows, 722 kg ±25.6 kg fed three different diets (treatments) were used in a Latin square design experiment to determine effects of increasing levels of dietary concentrate on some rumen parameters and ruminal crude protein (CP) degradability. The in situ method was used to determine the ruminal protein degradability of sunflower oilcake, cottonseed oilcake and roasted soya. The three treatments differed in roughage:concentrate ratio, being 60:40 (Treatment UP 60), 45:55 (Treatment UP 45) and 30:70 (Treatment UP 30). Intake of dry matter (DM) (kg/day) did not differ between treatments. The mean rumen pH in cows receiving the three experimental diets differed and was 6.00, 6.27 and 6.44 for treatments UP 30, UP 45 and UP 60 respectively. The time (hours) below pH 5.8, which is considered to be the pH where fibre degradation is substantially negatively affected, was approximately 2.5 hours, but only on treatment UP 30. Mean rumen ammonia nitrogen (N) and total volatile fatty acid (VFA) concentrations did not differ among cows receiving different treatments but, cows fed treatment UP 30 had a lower ruminal acetic acid:propionic acid (A:P) ratio compared to the other treatments. There were no differences in ruminal CP degradation within the three feedstuffs when incubated in cows fed diets with different roughage:concentrate ratios. Results suggest that roughage:concentrate ratios ranging from 60:40 to 30:70, which resulted in mean pH values ranging from 6.4 to 6.0, did not affect ruminal CP degradation of sunflower oilcake, cottonseed oilcake and roasted soya. / Dissertation (MSc(Agric))--University of Pretoria, 2009. / Animal and Wildlife Sciences / unrestricted

Influence of 25-hydroxyvitamin D and anionic salts on the calcium status of dairy cattle

Milk fever (parturient paresis / hypocalcaemia) is a metabolic disorder that usually occurs near parturition and at the onset of lactation in high producing multiparous dairy cows. Milk fever can indirectly contribute to an increased incidence of several diseases in early lactation. This study was conducted to compare two different feeding strategies to prevent milk fever, namely (i) the established concept of feeding a diet with a negative DCAD and (ii) a feeding strategy combining a negative DCAD supplement with 25-hydroxyvitamin D3 (25-OH-D3). Thirty dairy cows were used in a randomized block design and were selected and blocked by parity (second parity and later), 305 day mature equivalent milk production in the previous lactation and expected calving date. Within each of the 15 blocks, the cows were allocated to two experimental groups named DCAD and DCAD + HyD. Fifteen animals in the DCAD + HyD group received a daily oral dosage of 3 mg of 25-OH-D3. Plasma samples were collected from day 21 prepartum to 10 days postpartum and were analysed for 25-OH-D3, 1,25-dihydroxyvitamin D3, total and ionized calcium, phosphorus and magnesium. Samples were collected on day 21, 14, 10 prepartum and every second day to calving, 4 and 6 h postpartum and every second day up to day 10 after calving. Urinary samples for determination of macro minerals (calcium and phosphorus) were collected via manual stimulation on day 21, 14, 8 and 4 prepartum and day 4 postpartum. These samples were used to ensure that mild metabolic acidosis was achieved in both treatment groups. The metabolic acidosis was demonstrated by decreased urinary pH. Milk samples were collected on day 1, 4 and 10 postpartum and used for macro mineral (calcium and phosphorus) determination. This study did not achieve all of the expected results observed in similar experiments. No treatment differences could be detected for plasma Ca2+ concentrations (P>0.05) and the mean plasma Ca2+ concentrations were [1.086a mmol/L ± 0.010 (DCAD treatment) and 1.083a mmol/L ± 0.010 (DCAD + HyD treatment)] respectively.   Furthermore 1,25-(OH)2D3 plasma concentrations did not indicate any treatment differences (P>0.05). These results could be due to the fact that the experimental animals were not sufficiently challenged and therefore the combination of a low DCAD diet and Rovimix HyD did not influence the calcium homeostatic mechanisms as expected. A clear correlation between plasma 25-OH-D3 concentration and treatment duration was however demonstrated (P<0.001), indicating effective absorption of orally supplemented 25-OH-D3. Several authors demonstrated that feeding massive doses of vitamin D2 (30 million units) for extended periods led to clinical evidence of vitamin D toxicity. When 10 million IU of vitamin D3 were however administered intramuscularly within 10 days of parturition, a reasonable measure of protection against toxicity could be provided. It can be concluded from this study that longer feeding periods (± 21 days) than the proposed 10 days prior to calving can safely be implemented when feeding 3 mg 25-OH-D3 per animal per day (=240 mg Rovimix HyD 1,25%). Associations between the gastrointestinal microbiome and nitrogen efficiency in Holstein and Jersey cows

Manure nitrogen in dairy cattle represents a substantial economic and environmental loss to the industry. Current strategies used to improve N efficiency (Neff) have shown limited progress and thus, warrant more effective approaches. Considering that the gastrointestinal bacterial community has been associated with various phenotypes of economic importance, the objective of this project was to evaluate the associations between the rumen and fecal bacteriomes and Neff phenotypes in dairy cows. Results showed similarities in the overall bacterial community composition and structure of cows differing in Neff. However, the relative abundance of specific bacterial ASV differed between low and high Neff cows. Furthermore, bacterial ASV strongly correlated with Neff could be involved in processes such as nutrient supply, performance, and feed efficiency. These suggest that the gastrointestinal bacterial community is a factor influencing Neff in dairy cows and specific bacterial members can potentially serve as markers of Neff phenotypes.

The nutritive value of faba bean silage for lactating dairy cows

Thesis (MScAgric (Animal Sciences))--University of Stellenbosch, 2009. / ENGLISH ABSTRACT: The dry matter (DM) production and chemical composition of whole crop faba beans (Vicia faba) and oats (Avena sativa) were determined according to fresh material harvested at weekly intervals. From 75 to 166 days after planting whole plants of faba beans and oats were harvested at a height of ca. 10 cm above the ground on five randomly selected areas of 0.25 m² each. The freshly harvested material was weighed “as is” and ovendried to determine the DM content of each sample. The fresh and DM forage production per hectare was then calculated. The crude protein (CP), crude fiber (CF), neutral detergent fiber (NDF), acid detergent fiber (ADF), nitrogen free extract (NFE), fat (EE), calcium (Ca) and phosphorus (P) contents were determined according to standard laboratory techniques. The change in chemical composition of fresh whole crop material was regressed on days after planting using simple linear regressions. The fresh and DM production of whole crop faba beans and oats increased with advancing growth stage. During the 2002 production year fresh yield production of both whole crop faba beans and oats peaked at 131 days (44.7±6.9 and 28.4±7.1 ton/ha respectively). DM yield peaked at 159 and 152 days for whole crop faba beans and oats being 9.4±1.3 and 8.8±0.7 tons DM/ha respectively. The chemical composition of both forage crops decreased with advancing growth stage. The CP content of whole crop faba beans decreased (P<0.05) from 25.3% at 82 days after planting to 18.4 % at 166 days after planting in the 2002 production year, while during the 2003 production year the CP content of whole crop faba beans decreased (P<0.05) from 28.2 to 19.5 % from 75 to 159 days after planting. During 2002 the CF % of faba beans increased from 20.2 to 22.6%, while during 2003 CF % similarly increased from 21.8 to 26.5%. The CF % for oats during 2003 increased from 25.6 to 36.9%. During the same time the EE, Ca and P contents also decreased (P<0.05), while the NDF, CF and ADF contents increased (P<0.05). Two milk production studies were conducted to compare the feed intake, milk yield and milk composition of Holstein cows receiving either whole plant faba bean silage or oats as a hay or silage. Faba bean (Vicia faba) silage (FBS) was compared to that of cows receiving either oat (Avena sativa) hay (OH) or oat silage (OS) and in a 50:50 combination with FBS. Faba beans (cv. Ascot) and oats (cv. Sederberg) were planted on a Glenrosa soil. Whole crop faba beans were ensiled 145 days after planting in an above ground concrete bunker using a commercial bacterial inoculant after being wilted for one day. In the first experiment, total mixed rations (TMR) containing FBS, OH or a 50:50 mixture of FBS and OH as forage, together with a concentrate, were fed to three groups of seven lactating Holstein cows each. The experiment was conducted according to a randomized block design. Cows were on average 112±44 days post calving producing 24.0±6.2 kg milk/day. Milk production parameters of cows receiving diets containing different forages were compared by analysis of variance. The DM intake and milk yield of cows receiving TMR’s containing FBS, OH or a mixture of 50:50 FBS and OH as forages did not differ (P>0.05), milk yields being 18.9±1.9, 20.6±1.3 and 20.5±1.4 kg/cow/day respectively. With the exception of MUN, milk composition parameters did not differ among treatments (P>0.10). Cows fed OH as roughage source had a higher (P=0.06) MUN level in the milk. Results from this study indicate that FBS could effectively replace OH in lactating dairy cow diets. In the second experiment, total mixed rations (TMR) containing FBS, oat silage (OS) or a 50:50 mixture of FBS and OS as forages, together with a concentrate, were fed to six Holstein cows according to a double 3 x 3 Latin Square cross-over experimental design. Each cow received 13 kg DM either FBS, OS or a 50:50 mixture of FBS and OS were fed as forages, together with three different concentrates at nine kg “as is” each, to each cow. Cows were on average 108±30 days post calving producing 22.0±2.0 kg milk/day. DM intake, body weight, milk yield and milk composition parameters of cows receiving diets containing different forages were compared statistically. The DM intake and body weight of cows receiving TMR’s containing FBS, OH or a mixture of 50:50 FBS and OS as forages did not differ (P>0.10), although body weight changes differed significantly (P<0.10), i.e. 4.0±3.2, 8.8±3.2 and -6.0±3.2 kg respectively. The milk yield of cows receiving TMR’s containing FBS, OS or a mixture of 50:50 FBS and OS as forages did not differ (P>0.10), milk yields being 22.8±0.4, 21.4±0.4 and 21.9±0.4 kg/cow/day respectively. Of the milk composition parameters, the milk CP(%) of cows fed TMR’s containing FBS differed (P<0.05) from the cows fed the 50:50 mixture of FBS and OS, as well as cows fed the OS, being 2.82±0.02, 2.93±0.02 and 2.96±0.02% respectively. Results from this study indicate that FBS could effectively replace OS in lactating dairy cow diets. The South African database on in situ protein and fiber degradability values for whole crop faba beans and oats is limited. The chemical composition of whole crop faba beans and oats constantly change as plants mature. For optimal stage of ensiling and feed formulation it would be useful to have CP, NDF and ADF degradability values available for whole crop faba beans and oats harvested at different growth stages. The objective of this study was to determine the ruminal nutrient degradabilities of whole crop faba beans (Vicia faba) and oats (Avena sativa). DM, CP, NDF and ADF disappearance of whole crop faba beans and oats at 117 and 159 days after planting differed (P<0.05) at 4, 8, 12, 24, 48, 72 and 96 hours of incubation time. The DM, CP, NDF and ADF disappearance values were fitted to the non-linear model p = a + b (1- e-ct). The effective degradabilities (P) could be calculated using a fractional outflow rate of k = 0.05. For whole crop faba beans, parameter b (potentially degradable fraction) and parameter c (the rate at which b is degraded) all differed different forages were compared by analysis of variance. Faba beans cut as fresh crop or silage may in the foreseeable future play an increasingly larger role in the feeding of dairy cattle in the Winter Rainfall Region of South Africa. As in the case of lupin silage, though with much higher protein content, farmers will be able to produce their own quality and high protein roughage. The nutritive properties of faba bean silage holds great promise as a forage in lactating dairy cows. / AFRIKAANSE OPSOMMING: Die droëmateriaal (DM) produksie en chemiese samestelling van heelplant fababone (Vicia faba) en hawer (Avena sativa) is bepaal deur vars plantmateriaal op ‘n weeklikse basis te sny. Plantmonsters van beide fababoon en hawerplante is gesny vanaf 75 tot 166 dae na plant op ‘n hoogte van ca. 10 cm bo die grond. Plantmonsters is weekliks gesny op vyf ewekansige persele met ‘n oppervlak van 0.25 m² elk. Die vars gesnyde plantmateriaal is geweeg en daarna ge-oonddroog om die DM inhoud van elke monster te bepaal. Die varsmateriaal en DM opbrengs per hektaar is bereken. Die ruproteïen (RP), ruvesel (RV), neutraal bestande vesel (NBV), suur bestande vesel (SBV), stikstof vrye ekstrak (NVE), eterekstrak (EE), kalsium (Ca) en fosfor (P) inhoud is bepaal volgens standaardlaboratorium metodes. Lineêre regressie is gebruik om die verandering in chemiese samestelling van heelplant fababone en hawer te kwantifiseer. Vars en DM produksie van heelplant fababone en hawer het toegeneem met toename in groeistadium. Gedurende die 2002 produksiejaar het varsmateriaal opbrengs vir beide fababone en hawer gepiek op 131 dae (44.7±6.9 en 28.4±7.1 ton/ha respektiewelik). Heelplant fababoon en hawer DM opbrengs het gepiek op 159 en 152 dae na plant op 9.4±1.3 en 8.8±0.7 ton DM/ha, respektiewelik. Die chemiese samestelling van beide gewasse het afgeneem met toename in groeistadium. Die RP inhoud van heelplant fababone het verminder (P<0.05) van 25.3% op 82 dae na plant tot 18.4% op 166 dae na plant vir die 2002 produksiejaar, terwyl gedurende die 2003 produksiejaar die RP inhoud verminder (P<0.05) het vanaf 28.2 tot 19.5% vanaf 75 tot 159 dae na plant. Vir dieselfde tyd, het EE, Ca en P inhoud ook verminder (P<0.05), terwyl NBV, RV en SBV inhoud toegeneem (P<0.05) het. Twee melkproduksiestudies is uitgevoer om die effek van fababoonkuilvoer op voerinname, melkopbrengs en melksamestelling van Holsteinkoeie te bepaal. Fababoonkuilvoer (FBKV) is vergelyk met behulp van koeie wat hawerhooi (HH) of hawerkuilvoer (HKV) en in ‘n 50:50 kombinasie met FBKV as ruvoere ontvang het. Fababone (cv. Ascot) en hawer (cv. Sederberg) is gevestig op ‘n Glenrosa grond. Heelplant fababone is gesny en ingekuil op 145 dae na plant. Gesnyde materiaal is toegelaat om vir ‘n dag te verlep, waarna dit in ‘n bogrondse kuilvoersloot ingekuil is met behulp van ‘n kommersiële bakteriële entstof. In die eerste eksperiment is volvoere met FBKV, HH en ‘n 50:50 mengsel van FBKV en HH as ruvoer, saam met ‘n konsentraat, gevoer aan drie groepe koeie wat bestaan het uit sewe Holsteinkoeie elk. Die eksperiment is uitgevoer volgens ‘n ewekansige blokontwerp. Koeie was gemiddeld 112±44 dae in melk en het 24.0±6.2 kg melk/dag geproduseer. Melkproduksie-veranderlikes van koeie wat diëte ontvang het met verskillende ruvoere is met ‘n variansie-analise vergelyk. Die DM inname en melkopbrengs van koeie op volvoere bevattende vii FBKV, HH of ‘n 50:50 mengsel van FBKV en HH, het nie betekenisvol verskil (P>0.05) nie. Melkopbrengs was 18.9±1.9, 20.6±1.3 en 20.5±1.4 kg/koei/dag, respektiewelik. Met die uitsondering van melkureumstikstof (MUN), het melksamestelling-veranderlikes nie betekenisvol (P>0.10) verskil tussen behandelings nie. Koeie wat HH as ruvoer ontvang het, het ‘n hoër (P=0.06) MUN vlak in die melk gehad. Resultate van hierdie studie dui daarop dat FBKV effektief HH in lakterende melkkoeidiëte kan vervang. In die tweede eksperiment is volvoere, betsaande uit FBKV, hawerkuilvoer (HKV) of ‘n 50:50 mengsel van FBKV en HKV as ruvoer, saam met ‘n konsentraat aan ses Holsteinkoeie gevoer volgens ‘n dubbel 3 x 3 Latynse Vierkant omskakel proefontwerp. Elke koei het 13 kg DM van FBKV, HKV of ‘n 50:50 mengsel van FBKV en HKV ontvang as ruvoere, saam met drie verskillende konsentrate van 9 kg op ‘n natuurlike vogbasis elk. Koeie was gemiddeld 108±30 dae in melk en het 22.0±2.0 kg melk/dag geproduseer. DM inname, liggaamsmassa, melkopbrengs- en melksamestelling- veranderlikes van koeie op verskillende diëte wat verskillende ruvoere ingesluit het, is statisties vergelyk. Die DM inname en liggaamsmassa van koeie op volvoere bevattende FBKV, HKV of ‘n 50:50 mengsel van FBKV en HKV, het nie betekenisvol (P>0.10) verskil nie. Verandering in liggaamsmassa het betekenisvol (P<0.10) verskil, te wete 4.0±3.2, 8.8±3.2 en -6.0±3.2 kg, respektiewelik. Die melkopbrengs van koeie op volvoere bevattende FBKV, HKV of ‘n 50:50 mengsel van FBKV en HKV as ruvoer, het nie verskil (P>0.10) nie. Melkopbrengs was 22.8±0.4, 21.4±0.4 en 21.9±0.4 kg/koei/dag, respektiewelik. Ten opsigte van die melksamestelling-veranderlikes van die koeie op die onderskeie volvoere, was dit slegs melk RP(%) van koeie wat volvoere met FBKV ontvang het, wat betekenisvol (P<0.05) verskil het van die koeie wat HKV en die 50:50 mengsel van FBKV en HKV as ruvoer ontvang het. Melk RP(%) was 2.82±0.02, 2.93±0.02 en 2.96±0.02%, respektiewelik. Resultate van hierdie studie dui daarop dat FBKV effektief HKV in lakterende melkkoei diëte kan vervang. Die Suid-Afrikaanse databasis van in situ proteïen- en veseldegradeerbaarheidswaardes vir heelplant fababone en hawer is beperk. Die chemiese samestelling van heelplant fababone en hawer verander gedurig soos plante toeneem in ouderdom en groeistadium. Vir optimale stadium van inkuiling en voerformulering sou dit belangrik wees om DM, RP, NBV en SBV degradeerbaarheidswaardes van heelplant fababone en hawer ge-oes op verskillende groeistadiums te hê. Die doel van die studie was om die ruminale degradeerbaarheidswaardes vir heelplant fababone en hawer te bepaal. Effektiewe DM, RP, NBV en SBV degradeerbaarheidswaardes is vir fababone en hawer bepaal vir groeistadiums 117, 131, 145 en 159 dae na plant deur gebruik te maak van die in situ nylon sakkie tegniek. Drie nie-lakterende Holsteinkoeie elk toegerus met ‘n rumen- kannula is gebruik om ruminale degradeerbaarheidswaardes te bepaal. Plantmateriaal wat in Dacron sakkies afgeweeg is, is in die rumen geplaas vir 4, 8, 12, 24, 48, 72 en 96 ure. Die degradeerbaarheid van DM, RP, NBV en SBV fraksies van heelplant fababone en hawer vir vier verskillende groeistadiums (117, 131, 145 en 159 dae na plant) het nie betekenisvol (P<0.05) tussen koeie verskil nie. Die degradeerbaarheid van verskillende fraksies van beide ruvoere het verskil (P<0.05) tussen groeistadiums en inkubasie-ure. DM, RP, NBV en SBV verdwyning van heelplant fababone en hawer op 117 en 159 dae na plant het betekenisvol (P<0.05) verskil by 4, 8, 12, 24, 48, 72 en 96 inkubasie-ure. Die DM, RP, NBV en SBV verdwyningswaardes is gepas op ‘n nie-lineêre model p = a + b (1- e-ct). Die effektiewe degradeerbaarheid (P) kon bereken word met ‘n fraksionele uitvloeitempo van k = viii 0.05 vanuit die rumen. Vir heelplant fababone het parameter b (potensieel degradeerbare fraksie) en parameter c (die tempo waarteen b degradeer word) betekenisvol verskil (P<0.05) vir alle groeistadiums (117, 131, 145 en 159 dae na plant) vir degradeerbaarheidswaardes van RP, NBV en SBV. Vir hawer het parameter b en parameter c nie betekenisvol (P>0.05) tussen die vier verskillende groeistadiums (117, 131, 145 and 159 dae na plant) verskil nie. Die resultate van hierdie studie kan ‘n belangrike bydrae maak tot die Suid-Afrikaanse databasis van fababoon en hawer voedingswaardes, en kan aangewend word in dinamiese voerformulering. Die gebruik van varsgesnyde of ingekuilde heeplant fababone kan in die toekoms ‘n al groter rol speel in melkkoeivoeding in die Winterreënstreek van Suid-Afrika. Soos in die geval met lupiene, maar met ‘n hoër proteïen inhoud, sal produsente hul eie kwaliteit en hoë proteïen ruvoer kan verbou. Die voedingswaarde van fababoonkuilvoer hou groot belofte in as ruvoer vir lakterende melkkoeie.

The influence of teat wash failure on milk yield in dairy cows

Data for the period 2015-04 to 2015-09 was analyzed in order to examine the possible relationship between teat wash failure and the result on milk yield for dairy cows. Data provided by Sveriges Lantrbruksuniversitet over 49 093 specific milking events were used. Two linear mixed-effects models and one basic OLS-model were estimated. In order to perform the analysis a lot of data manipulation also had to be performed. The data analysis was divided into to two parts. First the variable of interest (teatwash) was examined by constructing two versions of the different models; an unrestricted- and a restricted version were teatwash had been excluded. Because of the large sample and linear mixed-effect models an out-of-sample forecasting method was used as the primary evaluation criteria. The prediction errors were evaluated on the basis of root mean squared error (RMSE) and mean squared error (MSE). The difference between the unrestricted- and restricted models was very small and no indication of a relationship between teat wash failure and milk yield was found. The second part involved the comparison of prediction errors between the two mixed-effect models and the OLS-model. Surprisingly, the basic OLS-model resulted in the lowest prediction error although obvious breach of assumptions.

Evaluation of Dietary Niacin and Supplemental Cooling for Alleviation of Heat Stress in Lactating Dairy Cows

Four studies were conducted to evaluate the effects of supplemental cooling and niacin on heat stress (HS) responses in vivo and in vitro in lactating dairy cows. For experiment one, lactating dairy cows were fed four levels of dietary niacin, NIASHURE® (0,4,8,12 g/d) for 21 d. Heat stress decreased niacin levels in whole blood, red blood cells and plasma as compared to thermoneutral. Water intake, plasma and milk niacin concentrations increased linearly with increasing dietary niacin in HS cattle. In thermoneutral, but not HS cows, niacin increased skin temperature compared to controls suggesting niacin increased skin blood flow and sensible heat loss. In experiment 2, lactating cows were used to evaluate the impact of feed-line soaking (FLS) combined with niacin supplementation. In evaporative cooled barns, FLS reduced body temperatures; however the addition of niacin did not improve heat status of these cows. For experiment 3, 200 lactating dairy cows were used to determine the effects of conductively cooled bedding (CC) compared to feed-line soaking with fans (FLSF). Conductively cooled bedding can reduce skin and vaginal temperatures in cows after nighttime rest. However, FLSF were more effective in decreasing body temperature, as cows had lower heat parameter indices, higher milk yield and longer rest time. For experiment 4, three cell types were used to evaluate niacin in vitro. Niacin induced heat shock proteins (HSP) that protected cells during HS in transformed bovine mammary epithelial cells but not in primary bovine mammary epithelial cells (BMEC) or bovine endometrial cells (BEND). Effect of niacin on HSP may depend on enzymes involved in DNA-binding activity of heat shock factor 1. These results suggest that niacin may be involved in whole body metabolism during heat stress and is cell dependent. We did not find dietary niacin to be commercially efficacious in treating HS in cows. Further research is warranted to improve efficacy of CC and FLSF under high temperature humidity index conditions.

Além do risco de doenças clínicas e subclínicas, favorecidas pela imunossupressão e alterações hormonais, as vacas podem sofrer distocias, que são partos marcados por dificuldade, tempo prolongado e necessidade de assistência para remoção do feto. Por ser multifatorial, além das causas maternas e fetais relacionadas à distocia, pode haver interação com variáveis comportamentais, ambientais e de manejo. O impacto da distocia no sistema produtivo está relacionado à ocorrência de doenças, o que aumenta o risco de descarte e morte, tanto para as vacas quanto para suas crias, impactando nos custos de produção e afetando a composição e longevidade do rebanho. Portanto, a presente dissertação teve como objetivos: identificar os fatores de risco associados à distocia e seu impacto sobre as doenças do período de transição pós-parto e sobre a produção de leite em vacas leiteiras; e avaliar se bezerras leiteiras nascidas de partos distócicos tiveram seu desenvolvimento, saúde, reprodução e produção de leite prejudicados. Para responder aos objetivos, realizou-se um estudo observacional retrospectivo utilizando registros de uma propriedade leiteira no Rio Grande do Sul, obtidos do software de gerenciamento leiteiro Dairy Plan C-21(GEA). O estudo foi dividido em dois experimentos, cujos resultados estão em um artigo científico. Para responder a um dos objetivos do primeiro experimento, foram analisados dados de 1.001 partos de vacas Holandesas, primíparas e multíparas, ocorridos entre 2013 e 2017, agrupadas em partos eutócicos e distócicos. As variáveis incluídas como preditoras de risco de distocia foram idade da vaca, categoria da vaca (primípara vs multípara), distocia ao nascimento, sexo do bezerro, tipo de parto, viabilidade do bezerro, estação climática no parto e ano. Para determinar o impacto da distocia sobre as doenças do pós-parto e produção de leite, foram analisados separadamente os dados das vacas primíparas (n= 540) e multíparas (n=461). Foram avaliadas as taxas de doenças clínicas como mastite, metrite, retenção de placenta e outras, bem como o número de episódios ocorridos, além das taxas de descarte e morte. A incidência de partos distócicos foi de 10,4% e os fatores de risco significativos para ocorrência de distocias foi o nascimento de bezerros machos, gêmeos, natimortos e partos ocorridos no inverno. Observou-se uma alta taxa de doenças clínicas no pós-parto e um risco aumentado para desenvolvimento de metrite em vacas multíparas distócicas. Para responder ao objetivo do segundo experimento, foram analisados registros de 447 bezerras nascidas na propriedade entre 2012 e 2015 agrupadas como nascidas de partos eutócicos ou distócicos. O impacto da distocia sobre a saúde, desenvolvimento e produção das bezerras foi analisado mediante avaliação dos dados de peso ao nascimento, 30 e 60 dias de idade, ocorrência de doenças até os 60 dias e até os 365 dias de idade, bem como o número de eventos ocorridos nestes períodos e as taxas de mortalidade e descarte. Também se avaliou o número de inseminações por concepção, a idade ao primeiro parto, taxas de distocia e natimortos, as doenças clínicas mastite, metrite, retenção de placenta e outras ocorridas após o primeiro parto e a produção de leite na primeira lactação. Observou-se que bezerras crias de partos distócicos apresentaram maiores taxas de metrite após o primeiro parto. Outros parâmetros não foram afetados pela distocia. / Parturition is an important process of the dairy production system and imposes a great challenge to the cow. In addition to the risk of clinical and subclinical diseases favored by immunosuppression and hormonal changes, cows may suffer from dystocia, which are difficulty deliveries with prolonged time and sometimes the need for assist in fetal removal. Since it is multifactorial, in addition to the maternal and fetal causes related to dystocia, it could have interaction with behavioral, environmental and management variables. The impact of dystocia on the productive system is related to the occurrence of diseases, which increases the risk of discarding the cows from the dairy and even death of the cows and their offspring, impacting production costs and affecting the composition and longevity of the herd. Therefore, the present dissertation had as objectives: identify the risk factors associated with dystocia and its impact on the diseases of the postpartum transition period and on milk production in dairy cows; and evaluate if dairy heifers born from a dystocic birth had their development, health, reproduction or milk production impaired. In order to respond to the objectives, a retrospective observational study was carried out using records from a dairy farm in Rio Grande do Sul, using Dairy Plan C-21 dairy management software (GEA). The study was divided into two experiments, which the results were published in a scientific paper. In order to achieve one of the objectives of the first experiment, data collected between 2013 and 2017, from 1,001 primiparous Holstein’s heifers and multiparous Holstein’s cows, grouped into normal or difficult birth was analyzed. The variables included as predictors of risk of dystocia were age, category (primiparous vs. multiparous), dystocic birth, calf sex, type of delivery, calf viability, climatic season at calving and year. To determine the impact of dystocia on postpartum diseases and milk production, data from primiparous cows (n = 540) and multiparous cows (n = 461) were analyzed separately. Rates of clinical diseases such as mastitis, metritis, retained placenta and others, as well as the number of episodes occurred and rates of discard or death were evaluated. The incidence of dystocia was 10.4% and the significant risk factors to the occurrence of dystocia were the birth of male calves, twins, stillbirths and births during winter time. There was a high rate of postpartum clinical disease and an increased risk of developing metritis in distocic multiparous cows. In order to achieve the second experiment, we analyzed the records from 447 calves born at the farm between 2012 and 2015 and grouped as born from normal or difficult births. The impact of dystocia on the health, development and production of heifers was analyzed by weight at birth, 30 and 60 days of age, occurrence of diseases up to 60 days and up to 365 days of age, as well as the number of events occurred in these periods and mortality and discharge rates. The number of inseminations per conception, age at first calving, dystocia and stillbirth rates, clinical mastitis, metritis, retained placenta and others after first calving and milk production at the first lactation were also evaluated. It was observed that heifers born from a dystocic parturition had higher rates of metritis on their first calving. Other parameters were not affected by dystocia.

Genetics of health and lameness in dairy cattle

For the modern dairy cow, advances in genetics and breeding for productivity has resulted in an increasing incidence of health disorders and reduced longevity. One of the most important health problems is lameness, which has led to significant economic, production and welfare consequences. A reduction in lameness will improve the economic future of the dairy industry through increased profitability and decreased welfare-related problems. Although positive attempts have been made by researchers and the industry towards improving lameness, it has remained a persistent ailment for dairy farmers. Further analysis of the genetic and environmental factors influencing lameness is warranted so that selection indices and management practices can be modified leading to improved health and welfare of the dairy cow. Several factors that cause dairy cow lameness have been implicated. I reviewed previous studies on these causative factors as well as the association between lameness, longevity and fertility. It has also been suggested that lameness affects milk production of dairy cows, but reports on the association between lameness and daily milk yield of cows have varied among researchers. Using locomotion score data on 248 cows from the Langhill herd, I investigated the relationship between locomotion score which has a high genetic correlation with lameness and various explanatory variables and also the association between daily milk yield and lameness. The study revealed that the most significant factors affecting locomotion are management regime (high concentrate feed and all year indoor housing; low concentrate feed and outdoors in summer) and time of year when cows are locomotion scored. It also showed that lameness adversely affects the milk yield of later lactation cows, and that high yielding cows are more susceptible to lameness. Housing environment plays a significant role in the health and welfare of dairy cows. With national type evaluation records, I estimated the association between housing systems and lameness-related type traits as well as genetic parameters for the locomotion traits. The analysis indicated that cows kept at pasture had favourable linear and composite type trait scores compared with cows in cubicles, straw yards and slatted floors or loafing yards. Locomotion score had strong genetic and phenotypic correlations with the leg and feet composite. Bone quality, which is a new trait in the UK type classification scheme, was moderately heritable (0.23) and had a moderate and positive genetic association with locomotion and leg and feet composite. This suggests that breeding for flatter, more refined bones could reduce locomotion disorders and help improve the longevity of the dairy cow. Analysis of national data again showed reduced incidence of digital dermatitis (DD) for cows at pasture and those with flatter, more refined bones, higher locomotion score and better leg and feet composite. Estimates of genetic parameters indicated heritable variation of DD among cows and moderate genetic associations between DD and production traits and longevity. Incorporating DD in future selection indices will be useful for increased productive life. Using random regression, I analysed changes in type traits associated with lameness (locomotion, rear legs, side view, foot angle and leg and feet composite) in relation to time (months) that cows had spent in cubicles before being classified. The general trend supported the fact that cubicle housing is unfavourable to these traits. There was significant evidence of a genotype x environment interaction, suggesting variation between bulls in the sensitivity of their daughters to cubicle housing with time.
