The contamination of waters and groundwaters by toxic substances reduces the quality of water and can even make it inadequate for consumption. Currently, the conventional techniques for treatment of waters and wastewaters are not able to completely decontaminate them. This is attributed to the presence of partially removed/degraded andrecalcitrant substances such as chlorophenols (CPs) and endocrine-disrupting chemicals (EDCs), since they persist in water even after the conventional treatment. This emphasizes the need for the development of more complete and sustainable processes that are also economically viable. For instance, heterogeneous photocatalysis utilizing UV-activated semiconductors for the production, mainly, of hydroxyl radicals is applied in the degradation of contaminants in aqueous medium. In this aspect, titanium oxide is the most used material in virtue of its high activity, low cost, stability, availability and suitable chemical, physical and biological properties. Despite so many advantages for the application of titanium oxide in heterogeneous photocatalysis, its application as slurries (to make better use of the surface area of particles) is still restrained by drawbacks related to separation/extraction steps. In this work, titania-based photocatalist hybrids were produced to perform heterogeneous photocatalysis of organic compounds in aqueous medium. As an alternative to implement particle separation from slurries, titania and mesoporous silica were deposited on the surface of magnetite cores. The incorporation of magnetite to the hybrid aimed to enable its magnetic separation from slurries whereas the incorporation of silica had the purpose of incrementing its activity by increasing the surface area of the hybrid material. A new route has been developed to perform the titania coating, which was based on the boiling of ethanolic medium. X-ray diffraction analyses indicated that the silica layer prevented phase conversion of magnetite cores to hematite after calcination. The formation of titania anatase phase was also observed for the hybrid materials for the mass composition of 43 % or greater. Infrared spectroscopy supported the presence of titania along with silica, and also pointed to a deposition limit for the titania layer that was also verified posteriorly through x-ray fluorescence spectrophotometry. Scanning electron microscopy enabled the observation of the formation and growth of anatase phase over magnetite-silica particles for compositions of 53 % or greater. Energy-dispersive x-ray spectroscopy demonstrated that Si and Ti were well- distributed over the magnetite-silica-titania particles. The hybrid material presented mesoporous structure likewise MCM-41, but with a smaller specific surface area of approximately 248 m2 g 1.29Si nuclear magnetic resonance provided evidence that silica and titania phases were chemically bonded. Photodegradation experiments demonstrated that magnetite-silica-titania hybrids are inefficient 4-chlorophenol adsorbents. The photocatalytic activity of the hybrids demonstrated to have relation with the growth of the titania texture, being the photoactive materials those with mass compositions of 53 % or greater. The kinetic behavior of the materials was found to have zero order and was pH-independent, having similar performances at pH 3.0 and 7.2. As the efficiencies of the hybrids are compared with those of commercial materials Hombikat 100 UV and Aeroxide® TiO2 P25, the decreasing order of efficiency could be obtained as Aeroxide® TiO2 P25 > Hombikat 100 UV >silica-titania hybrid > magnetite-silica-titania hybrid. Degradation experiments allowed to conclude that the incorporation of silica and magnetite to the titania photocatalists compromise their efficiencies; on the other hand, in the case of the materials containing magnetite cores, the possibility of magnetic retrieval may consist of a compensatory property with regard to practical application. To increment the photocatalytic activity of the materials, they underwent surface modification steps to have immobilized rubeanic acid (RA) either through sinalization or through direct adsorption. The complexation of Fe3+ to the surface of the hybrids was also tested in combination with rubeanic acid or without it to perform the sensitization of the photocatalysts. The immobilization of RA via silanization demonstrated to be more efficient, whereas its desorption could be verified for the material modified with RA by direct adsorption. In the case of hybrid materials modified with Fe3+ ions, the complexation of the ions led to a decay in their performance whereas no effect was observed at pH 6 for the material Hombikat 100 UV modified with Fe3+.More experiments are necessary to determine the initial degradation rates of the studied materials. In the experiments under visible light, only Hombikat 100 UV and its modified version presented photoactivity. When Fe3+-modified Hombikat 100 UV was tested at pH 3.0, evidence of increase in activity was noted under UV illumination. / Fapesp 2013/22955-2 e 2015/05224-0

Následky výbuchu špinavé bomby / Burst effects of radiation dispersal weapon

SADÍLKOVÁ, Alexandra January 2008 (has links)
One of the relatively cheap and easily available instruments suitable for a terrorist attack is the dirty bomb. Using conventional charges (TNT, Semtex etc.) it disperses radioactive materials. Its use is based on contamination of the area where the explosion took place and on creating a radioactive cloud, which may travel rather fast depending on the wind, and which pollutes other areas with its fall-out particles. Such areas become dangerous to live or stay in for a longer period due to the danger of irradiation sickness and cancer. The polluted areas must be decontaminated, which is a very difficult task. Another problem that may occur after the explosion of such a bomb is also panic as well as burns and injuries caused by shells. This work deals with possibilities of radiological weapon construction, the results of using a dirty bomb and with the work of integrated emergency services on such an occasion.

Influência da terapia fotodinâmica na manutenção da crista óssea alveolar periimplantar / Influence of photodynamic therapy in the maintenance of peri-implant bone crest

João Eduardo Miranda Franco 02 July 2014 (has links)
Bacterial colonization of the peri-implant sulcus occurs very early, right after the implant placement surgery. To this purpose, the project was proposed to evaluate the effect of photodynamic therapy (PDT) in the maintenance of {the} alveolar bone crest. In a split-mouth study, 20 patients were rehabilitated with Standard Plus implants (Straumann®) and they were divided into two groups: a control group (CG) and a group submitted to photodynamic therapy (GPDT), whose protocol was: energy density of 120J/cm2 at power of 40 mW, combined with 0,005% methylene blue. The applications were performed on the day of implant placement, after 15, 30 and 45 days. Microbiological samples were collected in the initial period and after 6 weeks on both groups, using sterile paper cones. In GPDT, two other collections were made after the application of the therapy: on the day of surgery and after 45 days. The total colony forming units (UFCt) and pigmented black units (UFCpig) were counted. Clinical data (Plaque Index-IP and Sulcus bleeding index- SS modified by Mombelli) were obtained at the 6th week, 6 months and 1 year. Radiographic analysis with periapical radiographs were made using the radiographic subtraction technique. The films were digitalized with the scanner SprintScan 35 Plus- Polaroid. Using image manipulation softwares (Adobe Photoshop® CS2- California and ImageJ-National Institutes of Health) distances from the implant shoulder to the first bone-implant contact were measured in phases: initial; 6th week; installation of the prosthesis, 6 months and 1 year after prosthetic installation placement. Friedman, Wilcoxon and Mann-Whitney were the statistical tests applied. Concerning the PDT efficacy, the results presented a statistically significant reduction for both UFCt and UFCpig in the two performed moments. The number of UFCt and UFCpig of the immediate postoperative to 6 weeks indicated a statistically significant increase in both groups. Comparing both the two groups, there was no statistically significant difference between the number of UFCt and UFCpig of the initial period to the 6th week. The IP and SS were similar in both groups. Radiographic evaluation indicated that there was an increasing loss of bone crest during the initial period and the installation of the prosthesis, which remained stable after a year of function for both groups. The bone loss average from the initial period until the installation of the prosthesis was 0,73mm for the GC and 0,84mm for the GPDT. During the prosthetic function the bone loss averages were, respectively, 0,27mm and 0,19mm for the GC and for the GPDT, with no statistically significant difference for both groups. The data led to the conclusion that: PDT is effective in reducing UFCt and UFCpig present in the peri-implant groove during the osseointegration; the PDT does not avoid the re-colonization or bacterial growth over time, except for the UFCpig of GPDT, wherein the amount of bacteria during the osseointegration remained statistically stable; and regarding to the maintenance of the peri-implant bone crest, the GC and GPDT obtained had similar results, with no statistically significant difference.

Comprehensive study of the heat resistance of dried Bacillus subtilis spores / Etude de la résistance à la chaleur des spores de Bacillus subtilis déshydratées

Hauck Tiburski, Julia 17 December 2013 (has links)
In response to starvation, species from the genre Bacillus are able to form metabolicallydormant spores which are very resistant to multiple forms of stress. They are found in quitehigh concentrations in some dried foods which, upon rehydration, may lead to food deterioration or food-borne diseases. Moreover, their destruction is rather difficult and mostof the techniques commonly used to treat dry foods result in a very low spore inactivation. The aim of this work is to better understand the role spore hydration in the inactivation ofdried Bacillus subtilis spores. A fundamental study was conducted using DifferentialScanning Calorimetry pans as reactors to perform a heat treatment in dried spores andsimultaneously relate the thermograms to spore viability. Results show the persistence of arelatively high water concentration in the core of extremely dry spores. Besides, a strongrelation between this core water concentration and spore thermal sensitivity wasdemonstrated. This destruction was found to be highly related to proteindenaturation/aggregation and dipicolinic acid release through Fourier Transform InfraredSpectroscopy analysis. From this fundamental study, a procedure for the inactivation of driedspores using low pressures (2-7 bar) and high temperature was developed. The systemconsisted of a heated reactor in which gaseous nitrogen was compressed to prevent theevaporation of water from the spores and so favor spore inactivation (> 5 log10). This methodof inactivation could be an interesting new way to optimize the decontamination of driedfoods.

Semen decontamination for the elimination of seminal pathogens

Fourie, Jozef Markus January 2013 (has links)
The presence of pathogens in semen can compromise the outcome of assisted reproductive treatment, together with the possibility of the female partner or offspring becoming infected. This is cause for concern, especially in South Africa with a high prevalence of HIV-1. Most of these infected individuals are in their reproductive years with the desire to have their own genetically related children. Therefore, assisted reproductive treatment with effective risk reduction procedures, such as semen processing for the elimination of these pathogens is crucial. However, during sperm preparation by standard discontinuous density gradient centrifugation, the supernatant is aspirated to allow access to the purified sperm pellet. Pathogens from the upper layers can adhere to the inside surface of the test tube and flow down to re-infect the purified sperm sample. The use of a centrifuge tube insert may prevent the re-contamination of sperm samples after discontinuous density gradient centrifugation. Furthermore, seminal pathogens can bind specifically or non-specifically to spermatozoa, rendering semen decontamination procedures ineffective. Serine proteases, such as trypsin, have been demonstrated to effectively inactivate viruses and to break pathogen-sperm bonds. However, the addition of a protease to density gradient layers during semen processing could have a negative impact on sperm parameters. This research was therefore aimed towards the determination of: i) The effect of semen processing with trypsin and trypsin inhibitor on sperm parameters. ii) The prevalence of various bacteria in semen samples from men attending the Reproductive and Endocrine Unit at Steve Biko Academic Hospital. iii) The effectiveness of semen processing by discontinuous density gradient centrifugation with a centrifuge tube insert, for the elimination of some of the most prevalent bacteria, white blood cells and in vivo derived HIV-1. Evaluation of sperm parameters after semen processing indicated that trypsin and trypsin inhibitor did not have an impact on sperm mitochondrial membrane potential, vitality, motility and zona binding potential, or acrosin activity, respectively. Seminal bacteria were highly prevalent in patients wishing to participate in the Unit’s assisted reproductive program, with 49.5% of semen samples presenting with positive bacterial cultures. Semen processing by means of discontinuous density gradient centrifugation with the tube insert, eliminated significantly more in vitro derived (spiked) bacteria and white blood cells from semen compared to processing without the insert. Furthermore, the semen decontamination procedure was effective in removing HIV-1 RNA from 100% of samples and proviral DNA from 98.1% of semen samples from HIV-1 sero-positive patients. The effectiveness of discontinuous density gradient centrifugation for the elimination of seminal pathogens could, therefore, be improved by the addition of trypsin to the upper density layer, without supplementing the bottom layer with trypsin inhibitor. Additionally, semen decontamination efficiency could also be improved by the prevention of re-contamination of processed sperm samples by the utilization of a tube insert during discontinuous density gradient centrifugation. / Thesis (PhD)--University of Pretoria, 2014. / gm2014 / Obstetrics and Gynaecology / unrestricted

Sorption et transport réactif d'ions dans des monolithes de silice fonctionnalisés aux hexacyanoferrates pour le traitement d'effluents radioactifs / Sorption and reactive transport of ions in HCF-functionalized silica monolith for radioactive effluent treatment

Cabaud, Clément 26 September 2019 (has links)
The nuclear industry produces high amounts of contaminated water from various sources that require specific treatments depending on their chemical composition. Cesium-137 and strontium-90 are among the most abundant radionuclides in those effluents, which have to be removed as efficiently as possible in order to generate the lowest amount of waste. The column process is one of the most suitable processes to achieve this solid-phase extraction. Its principle is based on the sorption capacity of the radionuclide by copper hexacyanoferrates (HCF), highly cesium-selective mineral ion-exchangers. Investigations on HCF pointed out the structural effects of the cesium insertion within the crystal, which were linked to the high affinity of HCF for this ion. The functionalization of HCF on silica monolith with hierarchical pore structure was carried out in order to benefit the remarkable properties of these supports used as a column. Sorption kinetics evaluated down to trace concentrations have shown a fast capture of the cesium, which proves the interest of this material for a column process purpose. In addition, the performances of functionalized silica monolith have been highlighted in comparison with those made of particulate fixed beds. Finally, those materials were implemented for a simultaneous decontamination of cesium and strontium by a double extraction mechanism of ion exchange and coprecipitation of barium sulfate, allowed by the high permeability of the monolith. A simplified model of reactive transport was built with the HYTEC code, based on the actual morphology of the monolith. To do so, a dispersive flow in the macroporous intraskeletal channels and a diffusive flow inside the walls of the structure and the HCF aggregates were assumed.

The fight against air pollution is a major issue in the twenty-first century. The center of Marcouleof CEA develops different waste treatment processes by incineration / vitrification that generatecombustion gases requiring treatment. To do this, the CEA uses the electrostatic precipitation, atechnical waste gas treatment employed for thin particles filtration. This thesis is dedicated to optimizing the performance of an electrofilter supplied by high pulsedpowered. One of the goals is to size and achieve a new emissive electrode adapted to thedevelopment of a new incineration process. This new electrode coupled to its High Voltage (HV)power supply, which electrical parameters were optimized, allowed to obtain maximum filtrationefficiency during operating times in line with industrial applications. The impact of thephysicochemical characteristics of dusts on the filtration efficiency was analyzed.A specific study also focused on the evolution of different discharge conditions that develop inthe electrofilter to identify the phenomena responsible for the process efficiency fall. The intake ofthe emissive electrode and a hybrid generator, combining a continuous background voltagesuperimposed with impulses, has clearly been demonstrated by their effects on back coronainitiation and therefore on the optimal efficiency operation duration.
