Kognitivní prediktory zdravého stárnutí / Cognitive Predictors of Healthy Ageing

Worldwide we are becoming witnesses to trend leading to rising number of people living to an older age. Focus of the research changes compared to previous studies in last few decades which studied the pathological aging and shifts to identification of factors leading to healthy (also "successful" aging). The focus is thus on finding the definition of successful and unsuccessful aging, so we would be able to timely identify the risks connected with aging and their influence on health or the level of quality of life. In connection with this, new concepts are becoming known, meaning "SuperAging" and "SuperAger" (SA). SuperAging is a phenomenon presenting successful (also as healthy) aging. SuperAger is the given individual, who is successfully (also as healthy) ages. In the theoretical part of the submitted thesis is presented the summary of findings in the field of neuropsychological changes connected with aging. The concept of successful (or healthy) aging is presented and the methodology of cognitive deficit detection in old age is described. Following part deals with requirements specification for design of diagnostic system and the system design itself. The aim of this thesis is to design and implement two methods of predictive analysis for company Škoda Auto a.s. In the first part I have conducted a literature search on methods of predictive diagnostics. In the next part, with help from thesis consultant in Škoda Auto a.s., the analysis of the assembly line and data blocks from machinery and measuring has been made. Then I designed and programmed data generator based on real data. I created two methods of predictive diagnostics, capable of analyzing input data and deciding about their condition. In the end I tested these two methods and evaluated accuracy of their prediction. Main output of my thesis is two methods of predictive diagnostics, feasible in real world.

Influenza viruses cause respiratory illnesses which can vary in severity depending on the strain of the virus, as well as the age and health condition of the host. Influenza remains a major threat to public health due to its nature prone to suffer mutations. As a result, vaccines have to be reformulated annually and new strains may cause sporadic global pandemics. Furthermore, the recent emergence of resistant strains of the virus against the current standard of care (oseltamivir and zanamivir) underlines the need of novel anti-influenza therapeutics. The aim of this dissertation work is to contribute to the discovery of new anti-influenza inhibitors either by rational drug-design and optimization of oseltamivir structure, or by developing screening assays suitable for the discovery of novel inhibitors of the enzymes neuraminidase or RNA-polymerase. Diagnostika karcinomu pankreatu / Pancreatic Cancer Diagnosis

Pancreatic cancer is a malignant disease with an unfavorable prognosis. Currently, the diagnosis of pancreatic cancer is based mainly on CT, MR and EUS imaging methods, because a reliable biomarker of pancreatic cancer is not available. It is considered a success in the current clinical practice if patients suitable for surgical resection can be selected, because in this group the overall survival is slightly better than in the group where surgery is not technically possible. In the first part, the presented dissertation focuses on testing of already established imaging diagnostic methods used in common practice, i.e. EUS and EUS FNA, as well as modern examination possibilities, such as contrast endosonography, and the comparison of the accuracy of the methods used. The dominant role of the EUS FNA was confirmed. In the next phase of the work, the methodological procedure of EUS FNA collection and the preprocessing of pancreatic tissue samples was tested with regard to the quantity and quality for epigenetic examination and further testing using the prognostic role of KRAS and miR-21. It has been suggested that cytological smears are the most suitable source of biological material for DNA and miRNA analysis, where non-tumor tissue contamination is low. This diploma thesis deals with methods of determining and treatment of coeliac disease. In the theoretical part of the work sums up informations about coeliac disease - history, prevalence, etiopathogenesis, immunopathogenesis, possibilities of diagnosis, discusses about gluten-free diet, mentions legislation of the Czech Republic and the European Union and compares expensiveness of gluten-free diet and normal diet. The experimental part of this thesis compares PCR, ELISA and indirect immunofluorescence methods for assesment of determination of genetic predisposition and antibodies in coeliac disease (greatest emphasis was placed on the comparison of antibodies against gliadin and deamidated gliadin antibodies in IgA and IgG).

Diploma thesis displaying black and white image in false color includes a study of methods and creation of computer software. The main function of the program is to convert the monochrome image into the pseudo colors. This procedure is achieved increasing the diagnostic yield of image. The program may find application in medical field. The pseudocoloring is used several different methods for obtaining the best results, combined with modifications increasing brightness scale image contrast images. The program is created in the programming environment of Visual Studio 2008 and written in C # programming language. Entry program is monochrome images in various formats (JPEG, BMP). The output of the program can be printed in a false-color image or stored image again in several different formats.
