Underexploited (ipso, ortho) microbial arene dihydroxylation : uses in synthesis & catalysisGriffen, Julia Anne January 2013 (has links)
This thesis sought to expand upon the synthetic application of the underexploited ipso, ortho diene cis-diol microbial arene oxidation product from benzoic acid. The microbial oxidation of benzoic acid by mutant strains of bacteria to give the ipso, otho diene cis-diol may be considered to be a green and clean method. This biocatalytic route yields large quantities of an enantiopure chiral building block, which is not assessable via traditional synthetic methods. The fermentation product has seen application towards the synthesis of aminocylitols, which have been tested for their biological activity. Attempts to synthesise the fully oxygenated counterparts, cyclitols, were investigated. Expansion of previous work using a bromine substituted derivative led to a range of cross-coupled and iron co-ordinated products. Finally, a range of novel chiral acids and ketones were synthesised and evaluated for their catalytic activity towards asymmetric epoxidation.

Inositols belong to an important class of biologically active compounds, named as
cyclitols (cyclic polyols), which have attributed high interest in the past decade due
to the glycosidase inhbition property that affects many biological processes.
Initially, 1,3,3a,7a-tetrahydro-2-benzofuran (69) was synthesized as a key compound
to synthesize bishomo-inositol derivatives. Then photooxygenation and
manganese(III) acetate oxidation reactions of this key compound were studied in
order to obtain isomeric inositol derivatives. Moreover, dihydroxylation and acid
catalyzed ring opening reactions were investigated. Finally, new synthetic
approaches for the synthesis of bishomo-inositol derivatives have been developed. In
addition to that whole products were conscientiously purified and characterized and
mechanism for the formation of the products has been discussed.

Le diabète de type 2 est aujourd'hui une maladie de plus en plus répandu. En ce sens il est nécessaire de mettre au point de nouveau composé permettant d'inhiber la gluconéogenèse. C'est pourquoi nous nous sommes intéressé à la synthèse du FR225654 1, décaline présentant une activité hypoglycémiante in vivo après administration par voie orale et inhibiteur de la néoglucogenèse in vitro (IC50 = 1,1.10-7 M). Ce composé, isolé en 2005 du champignon Phoma sp N°00144 et jamais encore synthétisé à ce jour, possède un mécanisme d'action inconnu. La nécessité d'une synthèse par voie chimique convergente et flexible, permettant l'accès à des analogues, est donc évidente. La stratégie consistait à préparer une trans-décaline par le biais d'une réaction de Diels-Alder intramoléculaire à partir d'un triène précurseur. La combinaison de ces travaux a constitué une avancée importante dans le cadre de la synthèse du FR225654, un hypoglycémiant potentiel. La mise au point d'une synthèse convergente du précurseur de la réaction de Diels-Alder permettra notamment d'effectuer par la suite des modifications aisées en vue de la préparation d'une vaste gamme d'analogues simplifiés. A ce jour, le produit de cyclo-addition a été isolé et caractérisé, validant ainsi l'étape clé de la stratégie de synthèse. Ainsi, l'accès rapide au FR225654 est rendu possible et la synthèse d'analogues est maintenant envisageable. Les produits synthétisés feront l'objet d'une évaluation biologique, l'objectif ultime étant d'accéder à de nouveaux médicaments pour le traitement du diabète de type 2.
