Received Signal Strength-Based Localization of Non-Collaborative Emitters in the Presence of Correlated Shadowing

RSS-based localization is a promising solution for estimating the position of a non-collaborative emitter using a network of collaborative sensors. This paper examines RSS-based localization and differential RSS (DRSS) localization in the presence of correlated shadowing with no knowledge of the emitter's reference power.  A new non-linear least squares (NLS) DRSS location estimator that uses correlated shadowing information to improve performance is introduced. The existing maximum likelihood (ML) estimator and Cram\' er Rao lower bound (CRLB) for RSS-based localization given do not account for correlated shadowing. This paper presents a new ML estimator and CRLB for RSS-based localization that account for spatially correlated shadowing and imperfect knowledge of the emitter's reference power. The performance of the ML estimator is compared to the CRLB under different simulation conditions. The ML estimator is shown to be biased when the number of sensors is small or the shadowing variance is large. The effects of correlated shadowing on an RSS-based location estimator are thoroughly examined. It is proven that an increase in correlated shadowing will improve the accuracy of an RSS-based location estimator. It is also demonstrated that the ideal sensor geometry which minimizes the average error becomes more compact as correlation is increased. A geometric dilution of precision (GDOP) formulation is derived that provides a metric for the effect of the position of the sensors and emitter on the location estimator performance. A measurement campaign is conducted that characterizes the path loss at 3.4 GHz. The measurements are compared to the log-distance model. The errors between the model and the measurements, which should theoretically be Gaussian, have a Kurtosis value of 1.31. The errors were determined to be spatially correlated with an average correlation coefficient of 0.5 at a distance of 160 meters. The performance of the location estimators in simulation is compared to the performance using measurements from the measurement campaign. The performance is very similar, with the largest difference between the simulated and actual results in the ML estimator. HIV-1 Tat Affects Interorganelle Communication in HIV-Associated Neurocognitive Disorders (HAND)

Among Human Immunodeficiency viruses (HIV), type-1 (HIV-1) is the most common worldwide and has the highest virulence and infectivity. Though the virus only infects a few cell types, the infection affects almost every organ system causing multiple comorbidities. One of the comorbidities is HIV-associated Neurocognitive Disorders (HAND). Interorganelle communication regulates many cellular functions including calcium exchange, lipid exchange, intracellular trafficking, and mitochondrial biogenesis. Interestingly, all these processes have been implicated in HAND suggesting that dysregulation of interorganelle communication plays a role in the progression of HAND. Using neuronal cell cultures, I show that mitochondrial-associated ER membranes (MAM)-associated protein and MAM-tethering protein expression and interactions are altered in the presence of the HIV-1 protein Tat. I also show, PTPIP51 and VAPB, two MAM-tethering proteins, expression is altered in the MAM fraction but not the whole cell fraction, indicating a localization problem. Phosphorylation of PTPIP51 has been shown to regulate the subcellular localization and I show that tyrosine phosphorylation is upregulated with Tat. In addition, I show that PTPIP51 binding with VAPB can be rescued with the addition of kinase inhibitors blocking PTPIP51 phosphorylation suggesting that Tat is altering the phosphorylation of PTPIP51 affecting its subcellular localization and binding to VAPB. Furthermore, I show that ER-Golgi communication is altered in the presence of Tat where there is an increase in the interactions between YIF1A and VAPB, two ER-Golgi tethering proteins. The altered iv interactions between MAM and ER-Golgi tethering proteins in the presence of Tat lead to the disruption of cellular pathways associated with dysfunctional interorganelle communication that can lead to neuronal dysfunction and can contribute to HAND. / Biomedical Sciences

The Role of ER-Alpha and the Ovaries in the Enduring Altered Behavioral Response to Pubertal Immune Stress

Peripubertal immune stress alters adult responsiveness to estradiol (E2) and progesterone (P). When female mice are injected with the bacterial endotoxin lipopolysaccharide (LPS) at six weeks of age, or during pubertal development, they display a decrease in response to ovarian hormones. In contrast, females ovariectomized prior to peripubertal immune stress display typical levels of sexual behavior following sequential injections of E2 and P in adulthood. Additionally, intact females exposed to peripubertal immune stress display a decrease in estrogen receptor alpha (ER-α)-immunoreactive (ir) cells in the medial preoptic area (MPOA) and ventromedial nucleus of the hypothalamus (VMH) in adulthood. However, ER-α has not been studied in mice that have been ovariectomized prior to receiving LPS. The objective of the present study is two-fold: to replicate the finding that ovariectomy prior to pubertal development prevents the deleterious effects of LPS administration, and to examine the status of ER-α in areas of the brain important to sex behavior. We predicted that mice ovariectomized after LPS injection would display fewer ER-α-ir cells and a decreased responsiveness to ovarian hormones than saline controls and those mice ovariectomized prior to LPS injection. To test this, female mice were ovariectomized or sham-operated prior to LPS treatment. Then, at six weeks of age, all mice were injected with saline or LPS. Following that, sham-operated mice were ovariectomized and ovariectomized mice were sham-operated. Mice were primed weekly with E2 and P, and sex behavior testing occurred once a week for 5 weeks. After the final behavior test, all mice were euthanized, their brains removed, and stained for ER-α via immunocytochemistry. Results revealed a large variability in hormone responsiveness. However, animals that received peripubertal LPS, but still had their ovaries, had significantly lower sexual receptivity when compared to animals that were ovariectomized prior to the pubertal period and given LPS. Further, there were no differences between groups in ER-α-ir numbers. External environmental stressors, such as animal housing and vibrations and noise from nearby construction, may have caused some of the results found here, which are inconsistent with previous findings.

The aim of this study is to address how “skevt” [skewed] girlhood is constructed in relation to age, time, and sexuality in three literary works of the Swedish author Sara Stridsberg: the novel Darling River (2010), the play Konsten att falla (2017) and the short story Tre systrar (2021). In the study, I pose the following questions: How is the skewed girlhood portrayed in relation to constructions of time and age? How does the construction of skewed femininity interact with the construction of sexual practice and sexual desires?     I use the term “skev” – a Scandinavian variation and hybrid of queer – to address how the girlhood is subverted in the texts, as well as to analyze how this not only occurs in relation to heteronormativity, but also within it. To explore this, I work with “skeva” [skewed] readings of the texts. My analysis is based on “skev” [skew] theory and queer temporality, through which I illustrate how the characters of the three literary works are challenging normative assumptions of girlhood and femininity, by performing gender and age as well as sexuality in non-normative ways. I discuss the girl’s bedroom as a form of heterotopia, in which both time and ageing are constructed in a skewed way. Furthermore, I locate a “skev” [skewed] temporality in the texts, connected to an absence of futurity.
