Influência da tetraciclina no desenvolvimento hepático e renal de filhotes de ratas /

Foram utilizadas quinze ratas sendo três controles e 12 tratadas. As ratas controle receberam injeções de solução fisiológica, enquanto as dos grupos experimentais foram tratadas com cloridrato de tetraciclina nas doses de 25, 50, 75 e 100mg/kg, via intraperitoneal, em dose única, no décimo dia da gestação. Os filhotes foram sacrificados ao nascer, com dez, vinte e trinta dias de vida. A análise histológica do fígado mostrou vacuolização, necrose, inflamação e dilatação sinusoidal principalmente em recém-nascidos. Mitoses, aumento precoce da população de células de Kupffer, hipertrofia de hepatócitos com maior síntese de glicogênio foram mais acentuadas nos filhotes com dez e vinte dias. No rim verificamos vacuolização tubular e necrose discretas, que ficaram mais evidentes nos recém-nascidos, e sinais de regeneração tubular aos dez e vinte dias. Os resultados sugerem que os órgãos estudados sofreram alterações morfológicas transitórias durante o desenvolvimento, mas revelaram indícios de regeneração ao longo do tempo / Abstract: This study aims to evaluate the possible effects of tetracycline administered to pregnant rats on kidney and liver development of their offspring. Fifteen rats were used - three controls and twelve treated ones. The control group received saline injections while the experimental groups were administered tetracycline chlorhidrate in the doses of 25, 50, 75 and 100mglkg, intraperitoneally, in a single injection, on the tenth day of pregnancy. The sucking were sacrificed at birth and after ten, twenty and thirty days of life. The liver specimes showed vacuolizations, necrosis, inflammation and sinusoidal dilatations, more evident in the newborn. Mitosis, early increase of Kupffer cells population and hypertrophy of hepatocytes with greater synthesis of glycogen were present in the 10th and twentieth days of life. The kidney specimens showed slight tubular vacuolizations and necrosis, more prominent in the newbom, as well as signs of tubular regeneration at the tenth and twentieth days. Estudo transversal, comparativo, envolvendo 180 mulheres na pósmenopausa (idade ≥45 anos e amenorréia ≥ 12 meses) acompanhada no Ambulatório de Climatério do Centro de Especialidades Médicas do Hospital Santa Casa de Belo Horizonte/Minas Gerais, de janeiro de 2011 a agosto de 2012. Os critérios de não inclusão foram: doença hepática (hepatites B e C, doença colestática, insuficiência hepática e tumor); uso de drogas que afetam o metabolismo hepático; etilistas; antecedente de HIV ou câncer; e obesidade mórbida. Foi diagnosticada SM na presença de três ou mais critérios: circunferência da cintura (CC) > 88cm; triglicerídeos (TG) ≥ 150mg/dL; HDL < 50mg/dL; pressão arterial ≥ 130/85mmHg; glicemia ≥ 100mg/dL. A DHGNA foi diagnosticada por meio da ultrassonografia abdominal. As participantes foram divididas em três grupos de acordo com a presença ou ausência de SM e/ou DHGNA: SM isolada (n=53); SM+DHGNA (n=67); sem SM ou DHGNA (controle, n=60). Foram realizadas avaliações clínicas, antropométricas e bioquímicas. O perfil inflamatório incluiu dosagens de adiponectina, interleucina-6 (IL-6) e fator de necrose tumoral alfa (TNF-α). Para análise estatística foram empregados os testes de Tukey, ANOVA, Qui-Quadrado, Correlação de Pearson e regressão logística (odds ratio-OR). As mulheres com SM+DHGNA mostraram-se obesas e com maior CC quando comparadas as mulheres com SM e controle (p<0.05). Assim como, apresentaram valores médios de glicemia e insulina superiores aos demais grupos, com resistência à insulina (RI) (p<0.05). Estudo clínico, transversal, envolvendo 188 mulheres na pós-menopausa (idade ≥ 45 anos e amenorréia ≥ 12 meses) acompanhadas em Ambulatório de Especialidades de Belo Horizonte de janeiro de 2011 a agosto de 2012. Critérios de não inclusão foram: doença hepática (hepatites B e C, doença colestática, insuficiência hepática e tumor); uso de drogas que afetam o metabolismo hepático; etilistas; antecedente de HIV ou câncer; e obesidade mórbida. A DHGNA foi diagnosticada por meio da ultrassonografia abdominal. Foram realizadas avaliações clínicas, antropométricas (índice de massa corpórea, IMC e circunferência da cintura, CC) e bioquímicas. Para análise estatística foram empregados os testes de t-student, distribuição Gama, Qui-Quadrado, regressão logística (odds ratio- OR). Das pacientes incluídas no presente estudo, 38,8% (73/188) apresentaram diagnóstico de esteatose hepática, sendo considerada leve em 45,2% (33/73) das pacientes, moderada em 42,5% (31/73) e, grave em 12,3% (9/73). As mulheres com DHGNA apresentaram valores pressóricos e CC elevados, e o IMC compatível com obesidade (31.5±4.5kg/m2) (p<0.05). Assim como, os valores médios de LDL, triglicerídeos, glicemia, insulina, ALT/TGP foram significativamente superiores as mulheres sem esteatose (controle) (p<0,05). Os valores médios do HOMA-IR indicaram resistência à insulina apenas no grupo com a DHGNA (6.1±4.6 vs 2.4±1.4 no controle; p<0.05). Encontrou-se diferença quanto à presença de síndrome metabólica (SM), detectada em 93.1% das mulheres com DHGNA e em 46.1% no controle (p<0.05). In this cross-sectional study, 188 women (age ≥45 years and amenorrhea ≥ 12 months) followed the Belo Horizonte Specialty Clinic from January 2011 to August 2012, were included. In 45.2% (33/73) of those patients, the steatosis was considered mild, in 42.5% (31/73) moderate and severe in 12.3% (9/73). Blood pressure, WC, BMI, LDL, triglycerides, glucose, insulin, alanine aminotransferase (AST) were significantly higher in NAFLD patients when compared without NAFLD women (control group) (p <0.05). The HOMA-IR average values showed insulin resistance only in the NAFLD group (6.1±4.6 vs 2.4±1.4 in control, p<0.05). The metabolic syndrome (MetS) was detected in 93.1% of women with NAFLD, and in 46.1% of control (p<0.05). In multivariate analysis, adjusted for for age and weight, the variables considered at risk for the development of NAFLD were: high WC (OR 1.07, 95% CI 1.01-1.13); plasma insulin (OR 1.12, 95% CI 1.05-1.19), HOMA-IR (OR 3.81, 95% CI 2.01-7.13), and presence of MetS (OR 8.68, 95% CI 3.3- 24.1). Estudo da inflamação e da autoimunidade na doença hepática gordurosa não alcoólica /

Orientador: Carlos Antônio Caramori / Banca: / Resumo: A obesidade é caracterizada pelo excesso de gordura presente no tecido adiposo e em outros órgãos e está intimamente ligada à RI e outros componentes da Síndrome Metabólica. O excesso de gordura nos hepatócitos atua como um órgão endócrino que libera citocinas inflamatórias e aumenta o recrutamento de linfócitos (células de defesa). Quando a tolerância imunológica é reduzida por esse processo, podemos observar o desenvolvimento de doenças autoimunes. Segundo dados recentes, os lipídios estão especificamente envolvidos na degradação da tolerância hepática, porém mais estudos são necessários para compreender a interação entre a gordura, o sistema imunológico e a esteatose. Nosso objetivo foi estudar a inflamação e a autoimunidade na doença hepática gordurosa não alcoolica (DHGNA) e verificar se a presença do estado inflamatório poderia representar fator contributivo para a presença de marcadores de Hepatite Autoimune (HAI). Tratou-se de um estudo observacional, prospectivo, transversal e descritivo. Foram incluídos 53 sujeitos e realizadas avaliações médicas e nutricionais, exames bioquímicos e biópsia hepática. Separamos a população em dois grupos para análise: CMAI (com marcadores de HAI) e SMAI (sem marcadores de HAI). Encontramos em 58,5% dos sujeitos pelo menos um tipo de autoanticorpo sugestivo de HAI. Não ocorreram diferenças estatisticamente significantes entre os grupos nas variáveis estudadas. Ao aplicarmos o escore da International Group Autoimmune Hepatitis (IGAIH), encontramos um caso com diagnóstico definitivo de HAI. Observamos que a gordura corporal, circunferência abdominal e circunferência do pescoço, quando acima dos valores de normalidade, podem estar relacionadas com a progressão da DHGNA e com algumas características típicas da HAI. Estes resultados corroboram a importância de incentivar a mudança dos hábitos alimentares e a prática de ... / Abstract: Obesity is characterized by excess fat within adipose tissue and other organs and it is closely linked to insulin resistance and other components of Metabolical Syndrome. Excessive fat in hepatocytes acts as an endocrine organ that releases inflammatory cytokines and increases the recruitment of lymphocytes (immune cells). When the immunological tolerance is reduced by this kind of stimulus, we can observe the development of autoimmune diseases. According to recent data, the lipids are specifically involved in the degradation of liver tolerance, but more studies are required to understand the interaction between fat, the immune system and steatosis. Our objective was to study the presence of inflammation and autoimmunity in non-alcoholic fatty liver disease (NAFLD) and verify whether the inflammation could represent a contributing factor to the markers of Autoimmune Hepatitis (AIH). This was an observational cross-sectional descriptive and prospective study. Fifty three individuals were included and submeted to medical and nutritional evaluation, biochemical tests and liver biopsy. The population was splitted in two groups for analysis: with AIH markers and without AIH markers. We found that 58.5% of the individuals had at least one type of antiautobody. There were no statistically significant differences between groups in the variables studied. By applying the International Group Autoimmune Hepatitis score (IGAIH), we found one case with a definitive diagnosis of AIH. We observed that body fat, waist and neck circumferences, while above the normal range, may be related to the progression of NAFLD and some typical features of AIH. These results corroborate the importance of encouraging change in eating habits and physical exercise for the control of chronic diseases, including the AIH.
