Modulation of the Host Response to Tacaribe Arenavirus Infection in AG129 Mice by MY-24

MY-24 is an aristeromycin derivative previously shown to protect AG129 type I and II interferon receptor knockout mice from lethal challenge with Tacaribe virus (TCRV). TCRV is nonpathogenic to humans, but is closely related to the highly pathogenic New World arenaviruses that cause often-fatal viral hemorrhagic fever syndromes. Remarkably, MY-24 prevented mortality without reducing TCRV burden in the circulation or tissues. To investigate the mechanism by which MY-24 protects AG129 mice against TCRV infection, we first characterized the natural history of disease in the model with an emphasis on cytokine responses and vascular integrity to establish the best times to evaluate the effects of MY-24 treatment on host responses believed to contribute to pathogenesis and fatal outcome. We found that viral replication in the blood and in various tissues precedes a hyperproduction of proinflammatory mediators that may lead to the destabilization of the endothelial barrier and increased vascular leakage believed to contribute to terminal shock associated with severe cases of hemorrhagic fever. We also found slightly reduced virus titers in certain tissues from MY-24-treated mice, suggesting that there may be a weak antiviral effect; however, TCRV was not cleared from lung, spleen, brain or kidney in recovering animals out to 40 days post-infection, indicative of the establishment of chronic infection in mice that are able to survive the initial challenge. Neutralizing antibodies do not appear to play a major role in the antiviral effect of MY-24, whereas reductions in several key proinflammatory cytokines in mice treated with MY-24 may serve to reduce vascular leakage caused by TCRV infection.

Les recommandations pour la prise en charge des accès fébriles ne sont pas particulières au Cabinda ni à l’Angola. L’emploi des tests de diagnostic rapides (TDR) et/ou de la microscopie est la règle et permet de porter le diagnostic d’accès palustre. Nos observations dans des structures de santé montrent que ces examens sont très peu pris en compte. Lorsqu’un TDR est négatif, un examen de goutte épaisse sanguine est souvent demandé. Cette goutte épaisse peut dans un quart des cas être déclarée positive avec une très faible parasitémie et le diagnostic d’accès palustre retenu. Les examens microscopiques sont perçus comme plus fiables que les TDR. La plupart des enfants et des adultes fébriles reçoivent un traitement antipaludique quels que soient les résultats des examens. Ce traitement est le plus souvent une association thérapeutique à base d’artémisinine. Le choix d’utiliser la voie parentérale en traitant par la quinine ou les dérivés injectables de l’artémisinine n’est pas réservé aux cas les plus graves.Ces antipaludiques sont disponibles dans les pharmacies privées ainsi que dans le secteur informel en dehors de toute prescription. Des dérivés d’Artemesia en monothérapie y sont également accessibles.L’amélioration de la formation du personnel de santé ainsi que de leurs conditions de travail apparaissent être les mesures les plus pertinentes pour modifier la situation. Le développement d’un centre de référence, en permettant une connaissance actualisée du paludisme, est également nécessaire. Par ailleurs, la prévention de la transmission par l’usage des moustiquaires imprégnées d’insecticides de longue durée doit être développée. / The recommendations for the management of febrile attacks are similar in Angola as in other neighboring countries. The use of rapid diagnostic tests (RDTs) and / or microscopy is the rule and allows the diagnosis of access to malaria.Our observations in health facilities in Cabinda Province show that these exams are very little taken into account. When a RDT is negative, a thick blood test is often required. This thick drop can in a quarter of the cases be declared positive with a very low parasitaemia and the diagnosis of malaria access retained. Microscopic examinations are perceived as more reliable than RDTs.Most children and febrile adults receive antimalarial treatment regardless of test results. This treatment is most often a combination therapy based artemisinin. The choice to use the parenteral route by treating with quinine or derivatives of artemisinin is not reserved for the most serious cases.These antimalarials are available in private pharmacies as well as in the informal sector outside any prescription. Artemesia derivatives in monotherapy are also available.Improving the training of health personnel and their working conditions appear to be the most relevant measures to change the situation. The development of a referral center, by providing up-to-date knowledge of malaria, is also needed. In addition, the prevention of transmission through the use of long-lasting insecticidal nets must be developed.

Application of Gis in Temporal and Spatial Analyses of Dengue Fever Outbreak : Case of Rio de Janeiro, Brazil

<p>Since Dengue fever (DF) and its related forms, Dengue Hemorrhagic fever (DHF) and Dengue Shock Syndrome (DSS) have become important health concerns worldwide, it is also imperative to develop methods which will help in the analysis of the incidences. Dengue fever cases are growing in number as it also invades widely, affecting larger number of countries and crossing climatic boundaries. Considering that the disease as of now has neither an effective vaccine nor a cure, monitoring in order to prevent or control is the resorted alternative. GIS and its related technologies offer a wealth of interesting capabilities towards achieving this goal.</p><p><strong> </strong></p><p>The intention of this study was to develop methods to describe dengue fever outbreaks taking Rio de Janeiro, Brazil as a case study. Careful study of Census data with appropriate attributes was made to find out their potential influence on dengue fever incidence in the various regions or census districts. Dengue incidence data from year 2000 to year 2008 reported by the municipal secretariat of Rio was used to extract the necessary census districts. Base map files in MapInfo format were converted to shape files.  Using ArcGIS it was possible to merge the dengue fever incidence data with the available base map file of the City of Rio according to corresponding census districts. Choropleth maps were then created using different attributes from which patterns and trends could be used to describe the characteristic of the outbreak with respect to the socio-economic conditions. Incidence data were also plotted in Excel to see temporal variations. Cluster analysis were performed with the Moran I technique on critical periods and years of dengue outbreak. Using the square root of dengue incidence from January to April 2002 and 2008, inverse distance was selected as the conceptualised spatial relationship, Euclidean distance as the distance method. More detailed analyses were then done on the selected critical years of dengue outbreak, (years 2002 and 2008), to investigate the influence of socio-economic variables on dengue incidence per census district.</p><p> </p><p>Dengue incidence rate appeared to be higher during the rainy and warmer months between December and May. Outbreaks of dengue occurred in years 2002 and 2008 over the study period of year 2000 to 2008. Some factors included in the census data were influential in the dengue prevalence according to districts. Satisfactory results can be achieved by using this strategy as a quick method for assessing potential dengue attack, spread and possible enabling conditions. The method has the advantage where there is limited access to field work, less financial means for acquisition of data and other vital resources.</p><p>A number of difficulties were encountered during the study however and leaves areas where further work can be done for improvements. More variables would be required in order to make a complete and comprehensive description of influential conditions and factors.  There is still a gap in the analytical tools required for multi-dimensional investigations as the ones encountered in this study.  It is vital to integrate ‘GPS’ and ‘Remote Sensing’ in order to obtain a variety of up-to-date data with higher resolution.</p><p> </p>

The epidemiology of Rift Valley Fever in Yemen and the risk of re-introduction from the Horn of Africa. - Lépidémiologie de la fièvre de la Vallée du Rift au Yémen et le risque de réintroduction à partir de la corne de lAfrique

From 1930 to 2000 Rift Valley Fever (RVF) was limited to the African continent. It is vector borne disease caused by a virus of the genus Phlebovirus, member of the Bunyaviridae family. The main vectors for transmission are Aedes and Culex. In September 2000 it was reported for the first time out of Africa, affecting Yemen and Saudi Arabia. This epidemic opened a new era in the history of RVF. It proved that the virus had the capaciity to affect new zones, different eco-systems and to spread to any area in the world. The outbreak lifted many hypothesis related to its introduction and to the factors associated with the outbreak. Although difficult to evaluate precisely its socio-economic impact was considered to be the heaviest in the modern history of Yemen animal diseases even when compared to the rinderpest outbreak that the country encountered in the seventies because of its zoonotic characteristic. To answer to these hypotheses it was of great importance to study and investigate all the factors associated with the outbreak. Thus after estimating the socio-economic impact of the disease in the world and more specifically in Yemen we studied the descriptive epidemiology of RVF in the first and most affected zone of the outbreak of 2000-2001 (Tihama Wadi Mawr) and then we analysed the socio-economic and environmental factors associated to the outbreak, to finish with the risk assessment of the re-introduction of RVFV from the Horn of Africa through legal animals trade. The descriptive study showed that at the national level 90% of the RVF cases were in the plain of Tihama coast, Hodiedah, Hajjah and Sadah governorates, the majority of the villages being located around the main canals of Wadi Mawr at an altitude < 300 m. Environmental as well as socio-economic factors likely to play a role in RVF transmission in Yemen were highlighted with the study of the period 1997-2007 in the country. As in previous RVF outbreaks in neighbouring countries in the Horn of Africa, the year 2000 presented above-normal vegetation index values, which reflected important precipitations, for both rainy seasons (the first occurring between March and May; the second between July and October). These environmental conditions favourable to the vectors populations were found concomitant with a late starting date of Eid-al Kabeer celebration (March) in 2000, related to high hosts (cattle, sheep and goats) densities. According to these criteria, 2000 was considered as an atypical year. Yemeni Veterinary Services did not declare any RVF outbreak since 2000. Thus, we assumed Yemen free of the disease when assessing the risk of introduction of RVF into Yemen via the legal trade of small ruminants from the Horn of Africa (Kenya, Somalia, Djibouti, and Ethiopia). After precisely describing the routes and volume of trade from the Horn of Africa to Yemen, the pathway and different scenarios for introduction were developed following the OIE risk assessment method. A matrix of likelihood combinations including four possible levels (very low, low, medium, high) was built and used to combine likelihood of events. The overall probability of introduction was assessed very low to medium depending on the period of the year and most likely to occur via ovine males exported during festival periods that change depending of the year considered. The uncertainty was considered to be low. The socio-economic impact although difficult to estimate was shown to be dramatic as RVF affects all the chain of life in particular of those associated to livestock and animal products trade. Despite the dramatic impact of the outbreak of RVF in 2000 but it had the advantage to draw the attention of decision makers, of international organizations and of local veterinary services on the importance of livestock diseases and their possible effects on human health and national economies. Veterinary education also improved significantly in Yemen. It enhanced the epidemiologists skills, disease surveillance in general and the cooperation between human health care people and veterinary services. A national P3 laboratory should also soon open with the help of the IAEA and the FAO and Al-Mukkah quarantine could be modernized and extended in the near future. Regional collaboration and improvement of knowledge on animal trade but also field studies related to the disease and its entomological features are seen as compulsory to hope improving the prevention and control of RVF.But the best way and strategy for prevention of Rift Valley Fever in Yemen as well as in the world is to develop more efficient surveillance and control tools to implement coordinated regional monitoring and control programmes. - Résumé - De 1930 à 2000, la Fièvre de la Vallée du Rift (RVF) était une maladie essentiellement africaine. Cest une maladie vectorielle causée par un virus du genre Phlebovirus de la famille des Bunyaviridae. Les principaux vecteurs sont les Aedes et les Culex. En Septembre 2000, des cas furent rapportés pour la première fois hors du continent africain touchant le Yémen et lArabie Saoudite. Cette épidémie ouvrit une nouvelle ére dans lhistoire de la RVF. Elle prouva la capacité du virus à infecter des zones nouvelles, des écosystèmes différents et sa capacité à se propager à nimporte quelle région du monde. Plusieurs hypothèses furent avancées concernant son introduction et les facteurs socio-économiques associés aux foyers. Sil est difficile dévaluer précisement son impact socio-economique, il fut considéré comme le plus lourd de lhistoire moderne des maladies animales au Yémen même comparé à celui de la peste bovine dans les années 70 à cause de son caractère zoonotique. Pour répondre à ses hypothèses, il était important détudier et de rechercher tous les facteurs associés aux foyers. Ainsi après avoir estimé limpact soci-èconomique de la maladie dans le monde et plus particulièrement au Yemen, lon sintéréssa à lépidémiologie descriptive de la RVF dans les premières zones touchées mais aussi les plus affectées lors des foyers de 2000-2001 (Tihama Wadi Mawr) Puis lon analysa les facteurs socio-économiques et environmentaux associés aux foyers pour finir avec lévaluation de risque de la réintroduction du RVFV à partir du commerce légal danimaux de la Corne dAfrique. Létude descriptive montre quau niveau national, en 2000-2001 environ 90 % des cas étaient dans la plaine de la côte de Tihama, Hodiedah, Hajjah et Sadah, la plupart des villages étant localisées autour des canaux principaux de Wadi Mawar à une altitude infereure à 300 Mètres (m). Les facteurs environmentaux et socioèconomiques susceptibles davoir un rôle dans la transmission de la RVF au Yémen furent soulignés avec létude de la période 1997-2007 dans le pays. Comme dans les foyers précédents de RVF dans les pays voisins de la Corne de lAfrique, lannée 2000 présentait des valeurs anormalement élevées dindex normal de végétation refletant dimportantes précipitations lors des deux saisons des pluies (la première entre mars et mai, la seconde entre juillet et octobre). Ces conditions environmentales favorables aux populations de vecteurs apparurent concommitantes avec une date tardive de début des festivités de lEid-al Kabeer en (mars) 2000 entraînant à une densité importante de populations hôtes (bovins, moutons, chèvres). Considérant ces facteurs lannée 2000 fut considérée comme une année atypique. Les services vétérinaires yéménites ne déclarèrent pas de foyers de RVF depuis 2000. Ainsi on fit lhypothèse que le Yémen nétait plus infecté pusqon lon sintéressa au risque de ré-introduction de la RVF au Yémen via le commerce légal de petits ruminants depuis la Corne de lAfrique (Kenya, Somalie, Djibouti et Ethiopie). Après avoir précisemment décrit les routes et volumes déchanges depuis la Corne de lAfrique jusquau Yémen, les chemins événementiels et différents scénarios dintroduction furent développé en suivant la méthode de lOIE. Une matrice de combinaison des probabilités incluant quatre niveaux (très bas, bas, modéré, élevé) fut construite et utilisée pour combiner les probabilité de réalisation des évènements. La probabilité globale dintroduction fut trouvée très basse à modérée en fonction de la période de survenue dans lannée et plus probablement via lintroduction dovins males exportés pendant les périodes de fêtes dont loccurrence change en fonction de lannée considérée. Lincertitude fut considérée basse. Si l'impact socio-économique de la RVF est difficile à évaluer, il est pourtant dramatique en atteignant tous les maillons du réseau de ceux dont la vie sorganise autour du bétail et de son commerce ou du commerce de ses produits. Malgré limpact dramatique des foyers de RVF en 2000, ceci a eu lavantage de dattirer l'attention de décideurs, dorganisations internationales et des services vétérinaires locaux sur l'importance des maladies de bétail et leurs effets possibles sur la santé humaine et les économies nationales. L'éducation vétérinaire s'est aussi améliorée de façon significative au Yémen. La qualification des épidémiologistes et la surveillance des maladies en général sen sont trouvées meilleures, ainsi que la coopération entre les services médicaux humains et les services vétérinaires. Un laboratoire national P3 devrait voir le jour prochainement avec laide de lIAEA (International Atomic Energy Agency) et de la FAO (Food and Agriculture Organization). La modernisation et l'extension de la quarantaine Al-Mukkah, est aussi prévue dans un avenir proche. La collaboration régionale, lamélioration de la connaissance du commerce du bétail mais aussi des études de terrain relatives à la maladie et à ses caractéristiques épidémiologiques sont indispensables pour espérer améloirer la prévention et le contrôle de la RVF. Mais la meilleures stragtègie pour la prevèntion de la fièvre de la vallée de Rift au Yemen comme dans le monde est de developper une surveillance plus efficace et controler des outils pour mettre en place un suivi regional coordonnée et des programmes de contrôle.

Rift Valley fever : development of diagnostics and vaccines

Rift Valley Fever virus (RVFV) causes an infection with severe impact on animal and human health. The disease is endemic throughout almost the entire African continent and large regions of the Arabian Peninsula. During epidemics, high mortality is observed in animals, especially among cattle, goats, and sheep. In humans, the symptoms vary from a benign influenza-like disease to a life-threatening hemorrhagic fever. Due to the devastating effect on communities in endemic regions and the possibility of further spread of this virus, there is an imperative need to improve and develop control measurements against this emerging disease. Therefore, this thesis focuses on diagnostics and vaccines against RVFV. RVFV infection kinetics was studied in a mouse model system by detection and quantification of viral genomes, using a developed quantitative real-time PCR (QRT-PCR) method. This novel QRT-PCR method proved to be reliable and serves as a supplement to standard diagnostics, direct virus isolation and serological methods. High levels of viral RNA were found in blood and liver samples from experimentally infected mice during the first days post infection. Thereafter the levels declined rapidly and dropped below detection limit approximately seven days post infection. The QRT-PCR technique was also used in a study aimed to improve diagnosis of RVFV from field samples collected on filter strips. Today, the available RVFV vaccines are only approved for animal use and these vaccines have several shortcomings. Since RVFV is a highly pathogenic organism requiring bio-safety level 3 laboratories, two different none-replicating vaccine approaches have been applied and evaluated using a mouse model. A DNA based vaccine, administered via gene-gun, and the use of virus-like particles (VLP), by the intra-peritoneal route. RVFV specific and neutralising antibodies were raised with both vaccine approaches. However, VLP vaccination against Rift valley Fever proved to be more promising as a future vaccine, since higher titres of neutralising antibodies and improved survival rate were found upon a lethal RVFV challenge in mice. In conclusion, a sensitive and specific method for quantifying RVFV infection and a promising vaccine candidate against RVFV were developed.

Immunopathogenesis of relapsing fever borreliosis

Relapsing fever (RF) is caused by different species of Borrelia transmitted by soft ticks or by the human body louse. Illness is characterized by reappearing peaks of high concentrations of spirochetes in blood, concordant with fever peaks separated by asymptomatic periods. Neuroborreliosis is one of the most severe manifestations of RF borreliosis. To understand the immune response during early RF, we analyzed immune cells in brain and kidney of mice infected with B. crocidurae during the acute infection. Our results indicate that brain defense is comprised primarily of innate immune cells. Despite the infiltration of innate immune cells, Borrelia was not completely eradicated. A failure of the host brain to clear the bacteria may give the pathogen a niche where it can persist. Using our mouse model, we revealed that Borrelia duttonii could persist in the mouse brain for up to 270 days, without being present in the circulation. The infection was silent with no change in host gene expression, and the spirochetes could re-enter the circulation after immunosuppression. We propose that the brain is used by the pathogen to evade host immunity and serves as a possible natural reservoir for B. duttonii, a spirochete that has rarely been found in any mammalian host other than man. Borrelia-induced complications during pregnancy have been reported, and are especially common in RF. In our established mouse model of gestational RF, we could show that the fetuses suffered from severe pathology and growth retardation, probably as a consequence of placental destruction. We could also show trans-placental transmission of the bacteria leading to neonatal RF. Surprisingly, pregnant dams had a lower bacterial load and less severe disease, showing that pregnancy has a protective effect during RF. We have used the gestational RF model to investigate host factors favoring disease resolution. Because the spleen is the primary organ responsible for trapping and removing blood-borne pathogens, we have compared temporal changes in spleen immune cell populations and cytokine/chemokine induction during the infection. Spleens of pregnant mice had earlier neutrophil infiltration, as well as faster and higher production of pro-inflammatory mediators. This rapid, robust response suggests a more effective host defense. Thus, an enhanced pro-inflammatory response during pregnancy imparts a distinct advantage in controlling the severity of relapsing fever infection.

Borrelia channel-forming proteins : structure and function

Borrelia is a Gram-negative, corkscrew-shaped bacterium transmitted by infected ticks or lice. Borreliae are subdivided into pathogens of two diseases: Lyme disease, caused mainly by B. burgdorferi, B. afzelii and B. garinii; and relapsing fever caused primarily by B. duttonii, B. hermsii, B. recurrentis or B. crocidurae. Both diseases differ in their manifestations, duration times and dissemination patterns. Antibiotics are the major therapeutics, although unfortunately antibiotic treatment is not always beneficial. To date, drug resistance mechanisms in B. burgdorferi are unknown. Transporters of the resistance-nodulation-division (RND) family appear to be involved in drug resistance, especially in Gram-negative bacteria. They consist of three components: a cytoplasmic membrane export system, a membrane fusion protein (MFP), and an outer membrane factor (OMP). The major antibiotic efflux activity of this type in Escherichia coli is mediated by the tripartite multidrug resistance pump AcrAB-TolC. Based on the sequence homology we conclude that the besA (bb0140), besB (bb0141) and besC (bb0142) genes code for a similar efflux system in B. burgdorferi. We created a deletion mutant of besC. The minimal inhibitory concentration (MIC) values of B. burgdorferi carrying an inactive besC gene were 4- to 8-fold lower than in the wild type strain. Animal experiments showed that the besC mutant was unable to infect mice. Black lipid bilayer experiments were carried out to determine the biophysical properties of purified BesC. This study showed the importance of BesC protein for B. burgdorferi pathogenicity and resistance to antibiotics, although its importance in clinical isolates is not known. Due to its small genome, Borrelia is metabolically and biosynthetically deficient, thereby making it highly dependent on nutrients provided by their hosts. The uptake of nutrients by Borrelia is not yet completely understood. We describe the purification and characterization of a 36-kDa protein that functions as a putative dicarboxylate-specific porin in the outer membrane of Borrelia. The protein was designated as DipA, for dicarboxylate-specific porin A. DipA was biophysically characterized using the black lipid bilayer assay. The permeation of KCl through the channel could be partly blocked by titrating the DipA-mediated membrane conductance with increasing concentrations of different organic dicarboxylic anions. The obtained results imply that DipA does not form a general diffusion pore, but a porin with a binding site specific for dicarboxylates which play important key roles in the deficient metabolic and biosynthetic pathways of Borrelia species. The presence of porin P66 has been shown in both Lyme disease and relapsing fever spirochetes. In our study, purified P66 homologues from Lyme disease species B. burgdorferi, B. afzelii and B. garinii and relapsing fever species B. duttonii, B. recurrentis and B. hermsii were compared and their biophysical properties were further characterized in black lipid bilayer assay. Subsequently, the channel diameter of B. burgdorferi P66 was investigated in more detail. For this study, different nonelectrolytes with known hydrodynamic radii were used. This allowed us to determine the effective diameter of the P66 channel lumen. Furthermore, the blockage of the channel after addition of nonelectrolytes revealed seven subconducting states and indicated a heptameric structure of the P66 channel. These results may give more insight into the functional properties of this important porin.

Research of Affects of Anti-Mosquito Volunteer Work and Environmental Literacy on Dengue Fever Vector Prevention Effectiveness The Case of Kaohsiung City¡¦s Gushan District

This thesis undertakes analysis of members of the neighborhood anti-mosquito volunteer team which was co-established by the Kaohsiung City Gushan District Office and the Department of Health and finds members of the team have, for the most part, not sufficiently encouraged others to join or participate in dengue fever vector mosquito prevention. In addition, these team members lack proper concepts concerning individual prevention of dengue fever carrying mosquitoes. This study therefore advises anti-mosquito volunteers that anti-mosquito teams must emphasize education concerning environmental behavior, as well as increase public awareness of vector mosquito prevention and measures for individual prevention. In order to make dengue fever vector prevention more effective, it is necessary to start with environmental behavior. In addition to advocating for proper environmental behavior, education and information dissemination concerning environmental behavior must be strengthened so as to increase the effectiveness of the anti-mosquito volunteer workers in dengue fever vector prevention and allow them to attain better results. The possibility of a dengue fever epidemic is an annual threat to the health of the citizens in Southern Taiwan¡¦s Kaohsiung City. The most important tasks in creating a new precedent with respect to mosquito control are to determine how to utilize community and neighborhood anti-mosquito volunteer team resources, to increase efforts to instruct neighborhood citizens to rid their home environments of objects which can retain water, to rid communities of breading sites, to improve environmental sanitary conditions, and to continue advocating neighborhood environmental health works in order to lower vector density for dengue fever and reach the goal of having cleaner and healthier home environments. Needless to say, this type of policy requires the selfless contributions and enthusiastic participation of the community¡¦s citizens. With this in mind, we must ask whether assistance the government provides in can facilitate the effectiveness of anti-mosquito volunteer teams. In addition, we should inquire into what impressions anti-mosquito volunteers have concerning dengue fever vector prevention. This thesis examines the extent to which anti-mosquito volunteer team members¡¦ cognition of environmental literacy influences the effectiveness of dengue fever vector prevention through an analysis which examines the interaction between anti-mosquito volunteer team attributes, members¡¦ environmental literacy, and dengue fever vector prevention results. This study uses the questionnaire survey method and various statistics in compiling data and utilizes various statistical techniques to understand the relationship between these factors, find a clear policy direction, and make this work less arduous and more productive.

The Molecular Characterization of a Diuretic Hormone Receptor (GPRdih1) From Females of the Yellow Fever Mosquito, Aedes aegypti (L.)

In the yellow fever mosquito, Aedes aegypti (L.), hemolymph-circulating diuretic hormones act upon the renal organs (Malpighian tubules) to regulate primary urine composition and secretion rate; however, the molecular endocrine mechanisms underlying rapid water elimination upon adult eclosion and blood feeding are not fully understood. Bioinformatic analysis of the current Aedes aegypti genome assembly reveals only a single predicted corticotropin releasing factor (CRF)-like diuretic hormone 44 (DH44) gene, but two DH44 receptor genes. The tissue expression profiles of the DH44 receptor(s), and specifically the identity of the DH44 receptor(s) in the Malpighian tubule, are undetermined in any mosquito species. This dissertation shows that Vectorbase gene ID AAEL008292 encodes a DH44 receptor (AaegGPRdih1) transcribed in Malpighian tubules. Sequence analysis and transcript localization indicate that AaegGPRdih1 is the co-ortholog of the Drosophila melanogaster DH44 receptor (CG12370-PA). The presence of conserved amino acid residues between AaegGPRdih1 and vertebrate CRF receptors suggests this mosquito receptor modulates multiple G protein-dependent intracellular signaling pathways. Quantitative PCR analysis of a time course of Malpighian tubule cDNA reveals AaegGPRdih1 abundance increases paralleling periods of observed urination. This suggests that target tissue receptor biology is linked to the known periods of release of diuretic hormones from the nervous system, pointing to a common up-stream regulatory mechanism. Higher relative abundance of AaegGPRdih1 transcript in female Malpighian tubules 24 hours after blood feeding suggests a role for AaegGPRdih1 in the excretion of nitrogen waste. RNA-mediated silencing to establish the significance of AaegGPRdih1 to mosquito Malpighian tubule physiology was inconclusive.
