Previous issue date: 2015-08-28 / Os primeiros fisiologistas ficaram certamente impressionados com a exist?ncia
de oscila??es peri?dicas de alta amplitude, claramente vis?veis nos tra?ados obtidos da
retina, trato ?ptico e g?nglios ?pticos. Posteriormente v?rios estudos mostraram ser a
c?lulas ganglionares os elementos respons?veis pela gera??o destes ritmos r?pidos, que
sabia-se podem propagar da retina ao geniculado lateral e ao c?rtex. Apenas
recentemente, no entanto, estas observa??es ganharam novo interesse, principalmente
a luz de teorias e conjecturas que atribuem ?s oscila??es neuronais v?rios processos
cognitivos, como a liga??o perceptual, a aten??o e a mem?ria. Segundo esta hip?tese,
oscila??es r?pidas da retina seriam importantes para a liga??o de contornos cont?guos
ou superf?cies, podendo assim constituir um mecanismo feedforward importante na
segmenta??o visual. Em acordo com estas no??es, uma s?rie de experimentos no gato
mostraram que oscila??es r?pidas da retina podem ser informativas sobre propriedades
globais do est?mulo como o seu tamanho. Uma grande limita??o nestes estudos, no entanto, foi o fato de terem sido feitos
sob anestesia e paralisia. Apenas alguns experimentos foram realizados em gatos n?oanestesiados,
mesmo assim, paralisados. Uma outra limita??o foi o uso de est?mulos
visuais limitados a breves exposi??es, que ocupavam todo o campo visual, muito longe
de condi??es naturais da vis?o. Por outro lado, muito recentemente, fizemos uma
observa??o inesperada no nosso laborat?rio: oscila??es r?pidas da retina dependem
fortemente da anestesia por halotano (e isoflurano). Tornou-se assim imperativo
investigar se as oscila??es r?pidas da retina est?o presentes ou n?o no gato n?o
anestesiado, em condi??es naturais, como por exemplo durante a observa??o-livre de
uma cena visual. Este ? o principal objetivo deste estudo. Para isto, registros simult?neos atrav?s de eletr?dios-m?ltiplos foram feitos no
geniculado lateral e na retina de gatos anestesiados (N= 3) e acordado (N= 1).
Compara??es foram feitas para respostas a filmes de cenas naturais e est?mulos
estacion?rios, como c?rculos luminosos. Para testar especificamente o papel das
oscila??es r?pidas da retina na codifica??o do tamanho do est?mulo visual aplicamos
um protocolo que consiste em apresentar sobre os campos receptores um c?rculo
luminoso de tamanho vari?vel ao longo do tempo. T?cnicas de separa??o de potenciaisde-
a??o nos permitiu estudar individualmente os componentes ON e OFF das respostas
multi-unit?rias. Nossa an?lise consistiu em obter medidas das oscila??es sincr?nicas
para c?lulas isoladas ao longo do tempo no dom?nio temporal (an?lise de correla??o
por janela deslizante) e no dom?nio espectral (an?lise espectral por afunilamento
m?ltiplo, coer?ncia por afunilamento m?ltiplo). Estes resultados estendem os nossos
achados pr?vios no gato anestesiado, que foram restritos ? an?lise de auto-correla??o
de repostas multi-unit?rias do geniculado lateral. Tanto as repostas ON como as
respostas OFF a est?mulos visuais de tamanho vari?vel mostram que oscila??es
coerentes, que aparecem apenas para est?mulos que atingem um tamanho m?nimo de
cerca de 5? (dependendo do n?vel de contraste do est?mulo). Estes resultados sugerem
que oscila??es r?pidas da retina codificam mal mudan?as sutis no tamanho do est?mulo
visual. Como nos estudos anteriores no geniculado lateral, registros obtidos diretamente
da retina mostraram que oscila??es r?pidas da retina s?o altamente dependentes dos
n?veis de anestesia por halotano. E mais importante, em uma s?rie de experimentos
pode-se registrar respostas do geniculado lateral em um gato acordado, que foi
subsequentemente anestesiado por halotano, mantendo-se o mesmo s?tio de registro. Oscila??es r?pidas da retina, ausentes durante a condi??o acordado, apareceram fortes
como usualmente na condi??o de anestesia por halotano. Estes resultados como um todo enfraquecem substancialmente a no??o de
serem as oscila??es r?pidas da retina importantes para o processamento visual. Por
outro lado, demonstram que oscila??es r?pidas da retina podem apresentar
propriedades semelhantes a oscila??es gama no cortex. Desta forma, oscila??es da
retina induzidas por halotano podem servir como uma prepara??o interessante, mesmo
se artificial, para o estudo da din?mica de oscila??es neuronais. / Early physiologists were dazzled by the occurrence of high-amplitude, periodic
oscillations, easily discernible in recording traces from the eye, optic tract and optic
ganglia. Numerous studies thereafter pointed to retinal ganglion cell as the
elements responsible for the generation of these fast rhythms, which were known to
propagate to the lateral geniculate and to the cortex. Only recently, however, these early
observations gained renewed interest, mainly in the light of recent theories linking
neuronal oscillations to various cognitive processes, such as perceptual binding, attention
and memory. In this context, fast retinal oscillations have been associated to the binding
of contiguous contours or surfaces, which in principle could support a fast feedforward
segmentation process. In addition, a series of experiments in the cat have shown that fast
oscillations in the retina may convey global stimulus properties, such as size. A limitation in these previous studies, however, was that most of them where were made in the anesthetized and paralyzed cat. Only a few early studies have been performed in the non-anesthetized but still paralyzed cat. Another concern was that, in these latter experiments, visual stimuli were often limited to ganzfeld flashes, far from
natural vision conditions. Moreover, very recently we made the surprising observation
that fast retinal oscillations depend strongly on halothane (and isoflurane) anesthesia. It
was therefore imperative to verify whether oscillatory activity is also present in the
awake cat, under naturalistic conditions, such as during free-viewing of a visual scene.
This is the main goal of the present study. Simultaneous multiple-electrode recordings were made from the lateral geniculate nucleus (LGN) and the retina of anesthetized cats (N= 3) and from the LGN
of an awake cat (N= 1). Comparisons were made for responses to natural movies and
flashed stationary light stimuli. To test specifically the role of retinal oscillations in
encoding stimulus size we designed a protocol made of a light circle of varying size along
the trial. Spike sorting techniques allowed us to study separately the ON- and OFFcomponents
of the responses. Analysis consisted in measuring synchronous oscillations
for single cell spiking activity in the time (sliding correlation analysis) and spectral
domains (multitaper spectral analysis, multitaper coherence). Our present results based
on single-cells extend our previous findings in the anesthetized cat, which were
restricted to an autocorrelation analysis of LGN mutiunitary responses. Both ON- and
OFF-responses to varying size stimuli show that coherent oscillations appear only after
the stimulus attained a minimum size of about 5? (depending on the contrast level),
suggesting that oscillations in the retina are rather limited in encoding subtle changes in
stimulus size. Recordings obtained directly from eye showed that oscillations in the
retina, as in the LGN, are highly correlated with the concentrations level of halothane.
Notably, in a series of sessions we were able to record LGN responses in an awake cat,
which was subsequently anesthetized with halothane, keeping the same recording site.
Oscillations were completely absent in the awake condition and appeared strong as usual
during the halothane anesthesia. Overall these results weaken substantially the notion that fast retinal oscillations are meaningful for vision. Nevertheless, as shown from our single cell analysis, retinal
oscillations share many of the properties of cortical gamma oscillations. In this respect,
oscillations in the retina induced by halothane serve as a valuable preparation, even
though artificial, for studying oscillatory neuronal dynamics.

intergeniculate leaflet (IGL) of the thalamus are considered the central components of the
circadian timing system (CTS) of mammals. This system is responsible for the generation and
regulation of circadian rhythms by establishing a temporal organization of physiological
processes and behaviors. The neuronal specific nuclear protein (NeuN) has been widely used
as a neuronal marker in several studies. Since glial fibrillary acidic protein (GFAP) is a
component of intermediate filaments found in the cytoplasm of astrocytes and is commonly
used as a specific marker for these cells. This study aims to identify, in the marmoset, the
NeuN immunoreactive neurons and glial cells immunoreactive to GFAP, as well as map the
major route of photic synchronization of the STC, retinohypothalamic tract (RHT), and
identify the indirect pathway to the SCN and pregeniculate nucleus (PGN) - structure
homologous to IGL rodents, using immunohistochemical and cytoarchitectonic techniques.
Observed in SCN the presence of neurons immunoreactive to NeuN and terminals
immunoreactive subunit b of cholera toxin (CTb), neuropeptide Y (NPY) and serotonin (5-
HT). In the PGN noted the presence of the NeuN and NPY immunoreactive neurons and the
immunoreactive terminals CTb and 5-HT. Astrocytes are present throughout the extent of the
SCN and the PGN this New World primate / O n?cleo supraquiasm?tico (NSQ) do hipot?lamo anterior, juntamente com o folheto
intergeniculado (FIG) do t?lamo s?o considerados os componentes centrais do sistema de

neuronal having a cap shaped located dorsomedially to the main relay visual
information to the cerebral cortex, the dorsal lateral geniculate nucleus (GLD). Several
cytoarchitectonic, neurochemical and retinal projections studies have pointed PGN as
a structure homologous to intergeniculate leaflet (IGL) of rodents. The IGL receives
retinal terminals and appears to be involved in the integration of photic and non-photic
information relaying them, through geniculo-hypothalamic tract (TGH), to the main
circadian oscillator in mammals, the suprachiasmatic nucleus (SCN) of the
hypothalamus. Thus, the IGL participates in the control of the biological rhythm by
modulating the activity of the SCN. Pharmacological and IGL injury studies conclude
that it is critical in the processing of non-photic information which is transmitted to the
SCN. Other studies have found that especially neurons immunoreactive to
neuropeptide Y (NPY) respond to this type of stimulation, determined by its colocation
with the FOS protein.
Has not been determined if the PGN responds, expressing the FOS protein, to
the non-photic stimulus nor the neurochemical nature of these cells. Thus, we apply a
dark pulse in the specifics circadian phases and analyze the pattern of expression of
FOS protein in PGN of the marmoset (Callithrix jacchus). We found that in all
animals analyzed the FOS expression was higher in the experimental than in the
control group. There was a higher expression of FOS when the dark pulse was applied
during the subjective day between the groups. Still, a subregion of the PGN, known by
immunoreactive to NPY, had a greater number of FOS-positive cells in relation to his
other just close dorsal region.
Our data corroborate the theory that the PGN and IGL are homologous
structures that were anatomically modified during the evolutionary process, but kept
its main neurochemical and functional characteristics. However, injury and
hodological studies are still needed for a more accurate conclusion / O n?cleo pr?-geniculado (NPG) do t?lamo de primatas ? um aglomerado

aplicamos um pulso de escuro em fases circadianas espec?ficas e analisamos o padr?o
de express?o da prote?na FOS no NPG do sagui (Callithrix jacchus). Verificamos que
em todos os animais analisados a express?o de FOS foi maior em rela??o ao grupo
controle. Houve uma maior express?o de FOS quando o pulso de escuro foi aplicado
durante o dia subjetivo entre os grupos estudados. Ainda, uma sub-regi?o do NPG,

The SCN is considerate the site of an endogenous biological clock because can to generate rhythm and to synchronize to the environmental cues (zeitgebers) and IGL has been related as one of the main areas that modulate the action of SCN. Both receive projections of ganglion cells of retina and this projection to SCN is called retinohypothalamic tract (RHT). Moreover, the IGL is connected with SCN through of geniculohypothalamic tract
(GHT). In primates (include humans) was not still demonstrated the presence of a homologous structure to the IGL. It is believed that the pregeniculate nucleus (PGN)
can be the answer, but nothing it was still proven. Trying to answer that question, the objective of our study is to do a comparative analysis among PGN and IGL through of
techniques immunohystochemicals, neural tracers and FOS expression after dark pulses. For this, we used as experimental model a primate of the new world, the
common marmoset (Callithrix jacchus). Ours results may contribute to the elucidation of this lacuna in the circadian system once that the IGL is responsible for the
transmission of nonphotic information to SCN and participate in the integration between photic and nonphotic stimulus to adjust the function of the SCN. In this way to find a same structure in primates represent an important achieve in the
understanding of the biological rhythms in those animals / O sistema de temporiza??o circadiana (STC) ? respons?vel pela gera??o e modula??o dos ritmos circadianos que s?o oscila??es end?genas manifestadas pelos seres vivos para a maioria das fun??es e comportamentos, com per?odo em torno de 24 horas. Estes ritmos s?o sincronizados principalmente ao ciclo claro-escuro di?rio. O STC ? constitu?do por um conjunto de estruturas neurais interligadas, incluindo na sua
composi??o um marca-passo encarregado da gera??o do ritmo, vias sincronizadoras e de sa?da aos efetores comportamentais. O n?cleo supraquiasm?tico do hipot?lamo (NSQ) ? tido como principal marcapasso circadiano. A les?o desse conjunto de c?lulas deixa o animal arr?tmico para
algumas fun??es circadianas. A principal via direta de sincroniza??o ? o tracto retinohipotal?mico (TRH), que leva informa??o f?tica ambiental da retina ao NSQ. Uma

contra NPY em diversos primatas estudados. Sabe-se que o sistema FIG-TGH al?m de estar relacionado ? modula??o f?tica
do NSQ, parece tamb?m estar fortemente envolvido na sincroniza??o n?o-f?tica desse sistema. Ainda, as c?lulas imunorreativas a NPY est?o claramente mais envolvidas na

proje??o retiniana e a express?o da prote?na do gene c-fos ap?s pulso de escuro, para melhor definir o papel do NPG dentro do STC. Para isso, usamos como modelo experimental um primata do novo mundo, o Callitrhix jacchus, conhecido
popularmente como sag?i. Nossos dados confirmaram a hip?tese inicial de que o NPG, ou parte dele, seria

Previous issue date: 2009-10-29 / Conselho Nacional de Desenvolvimento Cient?fico e Tecnol?gico / The thalamus plays an important role in the sensorial processing information, in this particular case, the visual information. Several neuronal groups have been characterized as conductors and processors of important sensorial information to the cerebral cortex. The lateral geniculate complex is one to them, and appears as a group very studied once it is responsible, in almost all totality, for the processing of visual information.
Among the nuclei that constitute the lateral geniculate complex we highlight the dorsal lateral geniculate nucleus of the thalamus (DLG), the main thalamic relay for the visual information. This nucleus is located rostral and lateral to medial geniculate nucleus and ventral to thalamic pulvinar nucleus in most of the mammals. In the primates humans and non-humans, it presents as a laminate structure, arranged in layers, when observed in coronal sections. The objective of this work was to do a mapping of the retinal projections and a citoarchictetonic and neurochemical characterization of DLG in the marmoset (Callithrix jacchus), a New World primate. The retinal projections were traced by anterograde transport of subunit b of cholera toxin (CTb), the citoarchicteture was described by Nissl method, and to neurochemical characterization immunohistochemicals technical were used to examine the main neurotransmitters and neuroatives substances present in this neural center. In DGL of marmoset thalamus, in coronal sections labeled by Nissl method, was possible to visualize the division of this nucleus in four layers divided in two portions: magnocellular and parvocellular. The retinal projections were present being visualized fibers and terminals immunorreactives to CTb (IR-CTb) in the DLG ipsilateral and contralateral. And through the immunohistochemicals techniques was observed that DLG contain cells, fibers and/or terminals immunoreactives against neuronal nuclear protein, subunits of AMPA 15 glutamate receptors (GluR1, GluR2/3, GluR4), choline acetyltransferase, serotonin, glutamic acid decarboxylase, binding calcium proteins (calbindin, parvalbumin and calretinin), vasopressin, vasoactive intestinal polypeptide, and an astrocyte protein, glial fibrillary acidic protein. / O t?lamo exerce um importante papel no processamento de informa??es sensoriais, em particular, a informa??o visual. V?rios grupos neuronais j? foram caracterizados como condutores e processadores de informa??es sensoriais importantes para o c?rtex cerebral. O complexo geniculado lateral ? um deles e aparece como um grupo muito estudado uma vez que ? respons?vel, em quase toda sua totalidade, pelo processamento de informa??o visual. Entre os n?cleos que constituem o complexo geniculado lateral destacamos o n?cleo geniculado lateral dorsal do t?lamo (GLD), o principal rel? tal?mico para as informa??es visuais. Este n?cleo se localiza rostral e lateral ao n?cleo geniculado medial e ventral ao n?cleo pulvinar do t?lamo na maioria dos mam?feros. Nos primatas humanos e n?o humanos, apresenta-se como uma estrutura laminar, disposto em camadas, quando observada em sec??es coronais. O objetivo neste trabalho foi fazer um mapeamento da proje??o retiniana e uma caracteriza??o citoarquitet?nica e neuroqu?mica do GLD no Callithrix jacchus (sag?i), um primata do Novo Mundo. As proje??es retinianas foram tra?adas por transporte anter?grado da subunidade B da toxina col?rica (CTb), a citoarquitetura foi descrita atrav?s do m?todo de Nissl, e para a caracteriza??o neuroqu?mica t?cnicas imunoistoqu?micas foram utilizadas para examinar os principais neurotransmissores e subst?ncias neuroativas presentes neste centro neural. No GLD do t?lamo do sag?i, nas sec??es coronais coradas pelo m?todo de Nissl, foi poss?vel visualizar a divis?o desse n?cleo em quatro camadas dividas em duas por??es: magnocelular e parvocelular. As proje??es retinianas estavam presentes visualizando-se fibras e terminais imunorreativos a CTb (CTb- IR) no GLD ipsolateral e contralateral. E atrav?s das t?cnicas imunoistoqu?micas observou-se que o GLD cont?m c?lulas, fibras e/ou terminais 13 imunorreativos a prote?na nuclear neuronal, subunidades dos receptores AMPA de glutamato (GluR1, GluR2/3, GluR4), colina acetiltransferase, serotonina, descarboxilase do ?cido glut?mico, prote?nas ligantes de c?lcio (calbindina, calretinina e parvalbumina), vasopressina, polipept?deo intestinal vasoativo, e uma prote?na astrocit?ria, prote?na ac?dica fibrilar glial.

In this study, these visual related centers in the rock cavy were evaluated by using the retinal innervations pattern and Nissl staining cytoarchigtecture. After unilateral intraocular injection of cholera toxin B subunit and immunohistochemical reaction of coronal and sagittal sections from the diencephalon and midbrain region of rock cavy. Three subcortical centres of primary visual system were identified, superior colliculus, lateral geniculate complex and pretectal complex. The lateral geniculate complex is formed by a series of nuclei receiving direct visual information from the retina, dorsal lateral geniculate nucleus, intergeniculate leaflet and ventral lateral geniculate nucleus. The pretectal complex is formed by series of pretectal nuclei, medial pretectal nucleus, olivary pretectal nucleus, posterior pretectal nucleus, nucleus of the optic tract and anterior pretectal nucleus. In the accessory optic system, retinal terminals were observed in the dorsal terminal, lateral terminal and medial terminal nuclei as well as in the interstitial nucleus of the superior fasciculus, posterior fibres. All retinorecipient nuclei received bilateral input, with a contralateral predominance. This is the first study of this
nature in the rock cavy and the results are compared with the data obtained for other species. The investigation represents a contribution to the knowledge regarding the organization of visual optic systems in relation to the biology of species. / Os sistemas visual prim?rio e ?ptico acess?rio compreendem dois conjuntos de grupamentos neurais, que recebem proje??o direta da retina. Nesse estudo, estes dois sistemas foram avaliados com rela??o a citoarquitetura e padr?o de inerva??o retiniana, usando como tra?ador neural anter?grado, a subunidade B da toxina col?rica revelada por imunoistoqu?mica para detectar este tra?ador em terminais sobre grupamentos neuronais (alvos) no enc?falo do moc? (Kerodon rupestris), um roedor nativo do Nordeste Brasileiro. Os resultados permitiram identificar os componentes do sistema visual prim?rio o complexo geniculado lateral, o complexo pr?-tectal e o col?culo superior. O complexo geniculado lateral cont?m o n?cleo geniculado lateral dorsal, o n?cleo geniculado lateral ventral e o folheto intergeniculado. Todos recebem fibras da retina com predomin?ncia contralateral, menos intensa para o folheto intergeniculado. O complexo pr?-tectal cont?m os n?cleos pr?-tectal anterior, pr?-tectal medial, pr?-tectal posterior, olivar pr?-tectal e n?cleo do trato ?ptico, os quais recebem proje??o retiniana predominantemente contralateral. Do mesmo modo, o col?culo superior recebe fibras da retina contralateral nas camadas superficiais. Tamb?m foi identificado o sistema ?ptico acess?rio completo no moc?, constitu?do pelos n?cleos terminal medial, terminal lateral, terminal dorsal e intersticial do fasc?culo superior posterior. Esses n?cleos recebem inerva??o retiniana com forte predomin?ncia contralateral, sendo que o n?cleo terminal medial, embora preserve a predomin?ncia contralateral, exibe uma evidente inerva??o ipsolateral.
