Synergistic effects of mixtures of fungicides and medicinal plant extracts against Botrytis cinerea

<p>We hypothesize that South African medicinal plants contain compounds that can act in synergism with synthetic antifungal compounds. Four fungicides - Sporekill&trade / , Rovral&trade / , Terminator&trade / and Teldor&trade / at doses 0.1, 0.2, 0.4 and 0.8 mL L-1 and plant species Galenia africana, Elytropappus rhinocerotis and Tulbaghia violacea were tested aloneand in different combinations for their potency (efficacy) on radial growth inhibition of Botrytis cinerea strains on potato dextrose plates. Four doses of plant extract for each of the respective plant species were used. A total of 48 combinations were tested for each strain. Mixtures of plant extracts were far more effective in controlling strains compared to the individual components alone, representing significant levels of in vitro synergistic interactions. Combinations of these components represent an attractive future prospect for the development of new management strategies for controlling B. cinerea. Since the in vitro tests of these mixtures showed inhibitory activity, the mixtures were tested for activity in assays on Granny Smith apples. In vitro tests can be used to screen mixtures to obtain information on their inhibitory activity on a pathogen, however, the environmental conditions of the fruit and the ability of the pathogen to grow into the fruit cannot be simulated in vivo. A series of two-fold doses of medicinal plant extracts were combined with fungicides to conduct decay inhibition studies. The incidence of gray mold was significantly reduced by mixtures of plant extracts and fungicides. Under conditions similar to those in commercial storage, a drench treatment with G. africana and Rovral&trade / significantly (p=0.05) inhibit gray mold on the apples and was more effective than the plant extract and fungicide alone. The treatments exerted synergistic effects and were markedly better than the components applied alone. The wound colonization assay was used for optimal decay control. In a drench, much higher volumes of the treatments are used to ensure that the components of the suspension are deposited evenly over the entire fruit surface. Drenching of fruit to apply other chemicals is an established practise in the pome (fleshy) fruit industry, and simplifies the commercial application of the mixtures, as no additional infrastructure at commercial packing houses will be required. This approach not only makes it possible to reduce fungicide concentrations while maintaining adequate decay control, but also ensures a reduction of the chemical residue on the fruit.</p>

Sample Preparation Methods and Pre-harvest Factors Influencing the Contents of Bioactive Compounds and Antioxidant Activity in Peppers

Peppers are a rich source of diverse bioactive compounds with potential health-promoting properties. The levels of bioactive compounds and antioxidant activity can be affected by analytical methods, pre-harvest factors, and the quality of peppers. In order to understand the nutrient composition and antioxidant activity in peppers, determination of factors influencing the contents of bioactive compounds is important. The overall objectives were to determine the efficient conditions of sample preparation and the impact of pre-harvest factors affecting bioactive compounds and antioxidant activity. Optimal extraction procedures were developed, and HPLC methods were validated for bioactive compounds in peppers. The highest flavonoids were extracted in ethanol, while myricetin was extracted using N-N-dimethylformamide. Optimized conditions for flavonoids were obtained during 3 h of extraction time and hydrolysis in M HCl for 60 min at 95 degrees C. Capsaicinoids and ascorbic acid were simultaneously separated and extracted using a solvent mixture consisting of 3% metaphosphoric acid: ethanol (2:8) after 30 min of sonication. To determine the relationship between bioactive compounds and antioxidant activities in pepper extracts from different solvent properties, bioactive compounds were analyzed, and the antioxidant activities were assayed by 2,2,-Diphenyl-1-picryl hydrazyl (DPPH), reducing power, and degradation of deoxyribose. Hexane extracts had the highest levels of capsaicinoids and carotenoids, while methanol extracts had the highest levels of flavonoids. Strong DPPH scavenging activity and reducing power were found in lipophilic extracts, while hydrophilic extracts were appropriate for inhibition of deoxyribose degradation. Variation in content of ascorbic acid, capsaicinoids, and flavonoids was evaluated at immature and mature stages of pepper cultivars in different locations over two years. Mature peppers contained the highest levels of capsaicinoids and ascorbic acid. Flavonoids were variable at different maturity stages. Interactions between pre-harvest factors and bioactive compounds were highly significant. This study demonstrated the efficient sample preparation methods and simultaneous separation of bioactive compounds, which reduces analysis time and leads to reduced cost. The antioxidant properties were strongly associated with the concentration of bioactive compounds based on selective pepper extracts. The pepper quality can be improved by using appropriate pre-harvest conditions that increase the levels of bioactive compounds in peppers.

HARVEST FREQUENCY AND CULTIVAR EFFECTS ON YIELD, QUALITY, AND REGROWTH RATE AMONG NEW ALFALFA CULTIVARS

Alfalfa (Medicago sativa L.) is the most important forage crop in the United States and consistently produces high yields and quality, but harvest frequency is the most significant factor for maximizing forage yield and quality. The objective of this research was to determine forage yield, quality, and regrowth rate among new alfalfa cultivars under four different harvest frequencies. Some of these cultivars have been marketed as having rapid rates of regrowth after cutting to maximize the number of harvests per year. Five cultivars were placed under four harvest frequencies of 25, 30, 35, and 40 days in a split-plot design. There was a significant yield and regrowth rate effect across cultivars and harvest frequencies, but little forage quality effect during the two years of this research. These results confirm previous findings that a 35-d harvest frequency is optimal for forage yield, quality, and stand persistence.

FOREST HARVEST EQUIPMENT MOVEMENT AND SEDIMENT DELIVERY TO STREAMS

Streamside management zones (SMZs) have become important management techniques to prevent the introduction of sediment to stream networks. This study examined the current Kentucky best management practice (BMP) guidelines for SMZs by outfitting mobile forest harvest equipment with global positioning system (GPS) receivers, enabling modeling of equipment traffic and spatial analysis of stream sediment delivery. Three SMZ configurations were implemented during commercial timber harvest, along with four different techniques of crossing ephemeral channels, in order to determine where and why sediment was introduced to the stream network. Results indicate that increasing the SMZ buffer width leads to decreased sediment delivery, and that requiring an SMZ buffer with some canopy retention on ephemeral channels will lead to improvements in stream water quality. Care should be taken in the placement and construction of water control measures for skid trail retirement, and improved stream channel crossings such as bridges and pipe culverts should be required to improve water quality over unimproved fords. A northeasterly aspect of harvested areas was shown to be related to increased sediment delivery to streams, while surface roughness downslope from the skid trail system was shown to decrease sediment delivery.

EFFECTS OF STREAMSIDE MANAGEMENT ZONE TIMBER HARVEST ON SALAMANDER COMMUNITIES IN ROBINSON FOREST

Salamanders are critical components of forest ecosystems, in terms of total biomass, as well as for their value as indicators of ecosystem stress. Considering the worldwide decline in amphibian populations, the known effects of timber harvest on salamander populations, and the importance of the forest products industry in Kentucky and elsewhere, the impacts of silvicultural operations on salamander communities cannot be overlooked. The objective was to investigate the effects of three different silvicultural treatments, each involving different streamside management zone (SMZ) characteristics, on salamander communities in ephemeral streams. Data were collected by regular checks of pitfall traps, coverboards, and transect searches. Using both pre- and post-harvest data, abundance estimates were acquired using binomial mixture models. Declines in some species of terrestrial and stream-breeding salamanders were detected, and were shown to be likely related to characteristics of the corresponding silvicultural treatment. Applying modest SMZ regulations to ephemeral streams would likely alleviate these declines significantly.

Variable Retention Harvesting: Mortality of Residual Trees and Natural Regeneration of White Spruce

In this thesis I examined the impacts of variable retention harvesting on residual tree mortality and natural regeneration of white spruce [Picea glauca (Moench (Voss)] in northern Alberta. The VR was done in four overstory canopy compositions (ranging from deciduous dominated to conifer dominated) and at six rates of canopy retention (2%, 10%, 20%, 50%, 75% and 100%). After 10 years there was 32.9 % mortality of aspen (Populus tremuloides Michx.) and 16.9 % mortality of spruce in the VR cuts. Mortality of individual trees was greater with low density of trees, in the conifer stands and for trees with short live crowns, which are large and trees near machine corridors. Natural regeneration of spruce was greatest with higher availability of seed trees (>30 ha-1) and on machine corridors, where stocking reached 74%. By contrast, stocking was 14% on retention strips, when seed tree density was 11 seed trees ha-1. / Forest Biology and Management

The influence of management on runoff and water quality in a coastal lowland PINUS plantation, Southeast Queensland

The exotic Pinus plantations of southeast Queensland occupy approximately 130 000 ha and are prominent in catchments which drain to estuarine and marine waters that are economically, socially, and environmentally important. Recently, the deterioration of estuarine and marine water quality has raised concerns about the possible off-site impacts from the intensive management of the Pinus estate in southeast Queensland. Additionally, forest managers have raised questions over the effects of the currently adopted management practices on soil, water, and nutrient resources within plantations. A paucity of information regarding the impacts of these plantations in the humid sub-tropics of southeast Queensland initiated the research presented here. The objectives of this study were to: (i) determine the influence specific Pinus management techniques (harvesting, site-preparation, prescribed burning and forest roads) have on runoff generation; (ii) quantify fluxes of some nutrients (nitrogen (N), phosphorus (P), dissolved organic carbon (DOC) and iron (Fe)) and suspended solids (SS) in runoff from these management treatments; and, (iii) assess the overall effectiveness of the currently adopted Best Management Practices (BMP's) in protecting on-site soil, water and nutrient resources, as well as protecting off-site waters from pollution. The study site was located in an intensively managed sub-catchment within the Beerburrum State Forest Pinus plantation on the coastal plain of the Pumicestone region, southeast Queensland. This study was established in October 2001 and consisted of a 141 ha catchment based investigation into water quality and hydrology, which received a 50 ha harvest treatment in February and March 2002. Water was monitored for two water years (October 2001 - September 2003), and incorporated site-preparation and the establishment of the subsequent rotation. The influence of a forest road stream crossing was also monitored in this component of the investigation. Two discrete forest road plots were monitored for the same period to measure the response of runoff, nutrient and sediment fluxes to different road surface materials (gravelled and ungravelled), road maintenance and traffic intensity. Rainfall simulation was used on small plots covering specific management treatments (clearfall harvest, cultivation, fertilised cultivation, prescribed fire and established trees) within the general plantation area to determine their influence on water, sediment and nutrient fluxes. The investigation in the catchment receiving inter-rotation management revealed that that there was very little difference in water quality indices up and downstream of the forest road stream crossing, which suggests that road borne runoff contributed only minor amounts of N, P, Fe and SS to the stream. Perched groundwater quality within the general plantation area was similar to that observed in the adjacent stream. Water quality monitoring within the Coochin-Mellum and Coonowrin Creek catchments showed that the mean annual concentrations of N and P in surface waters were highest from catchments hosting agriculture and residential areas, respectively. Mean annual DOC and Fe concentrations were highest from the catchment hosting native Wallum vegetation. The mean annual concentration of SS was highest from an unmanaged native forest catchment. The rainfall simulation on specific management treatments revealed that mean losses of N and P from unfertilised and unburnt treatments were comparable to loads reported from catchment scale studies in the Pinus plantations of south-east Queensland. Mean SS loads from all treatments were considerably higher than stream loads reported in the literature from catchment scale investigations, and suggest that the currently adopted mitigation practices between the general plantation area and streams are effective in promoting the deposition of entrained solids. The investigation into gravelled and ungravelled forest roads revealed that the mean runoff coefficient (runoff depth / rainfall depth) was consistently higher from the gravelled road plot with 0.57, as compared to the ungravelled road with 0.38. Total sediment loss over the two year period was greatest from the gravelled road plot. Suspended solids contributed 86% of the total sediment loss from the gravelled road and 72% from the ungravelled road over the two years. When road and drain maintenance (grading) was performed runoff and sediment loss was increased from both road types. It should be noted that the results presented herein were based on only two water years, and both years experienced below average rainfall. As such it is important that future research in a catchment prone to waterlogging be conducted over a longer term so as to increase the chance of quantifying water, nutrient and sediment fluxes in response to average and above average rainfall years. It is likely that in above average rainfall years the results for nutrient and sediment fluxes from the general plantation area would be significantly different as runoff would be more readily generated and sustained for longer periods. Overall, the research presented suggests that the management of an exotic Pinus plantation during the inter-rotation period results in relatively low fluxes of N, P, SS Fe and DOC in stream water and vindicates the use of the current practices in protecting on-site water, soil and nutrient resources.
