Etude multi-échelle de la récolte de Dunaliella salina - Développement d'un procédé d'autofloculation - flottation de microalgues / Multiscale study of Dunaliella salina harvesting - Implementation and optimization of a high-pH-induced flocculation-flotation process of microlagae

Il n’est pas constaté d’autofloculation naturelle dans les cultures hypersalines de D. salina. De plus, cette dernière ne flotte pas sans déstabilisation de la suspension. La floculation induite par augmentation du pH lors de l’ajout de soude a été choisie dans cette étude pour permettre la flottation. Des efficacités de récupération supérieures à 90% et des facteurs de concentration autour de 20 sont atteints. Un mécanisme d’autofloculation, avec précipitation de Mg(OH)2 est proposé pour expliquer la floculation par balayage de D. salina. L’influence du débit d’injection de soude ajoutée est aussi étudié pour anticiper les contraintes relatives à l’industrialisation du procédé. Ce débit n’a pas d’effet sur l’efficacité de récupération des algues et réduit le facteur de concentration seulement pour les injections abruptes.L’augmentation naturelle du pH de culture par l’intermédiaire de l’activité photosynthétique pourrait permettre de réduire le taux de base consommé. Les cellules non-récoltées restent en vie durant l’augmentation du pH et pourraient être utilisées comme inoculum d’une nouvelle culture. / Natural autoflocculation was not observed in a Dunaliella salina hypersaline culture and the microalgae did not float without destabilization of the algal suspension. High-pH-induced flocculation by sodium hydroxide addition was chosen to induce flotation. Recovery efficiencies greater than 90% and concentration factors of around 20 were reached. An autoflocculation mechanism, with precipitation of magnesium hydroxide, is proposed to explain a sweeping flotation of D. salina cells. The influence of the flow rate of sodium hydroxide addition was also studied to anticipate the constraints related to the industrialization of this process. The flow rate of sodium hydroxide addition had no effect on the recovery efficiency and reduced the concentration factor only for abrupt injections. Natural increase of culture pH by photosynthetic activity could reduce the amount of base consumed. Non-harvested cells remained viable during pH increase and could be used as inoculum for a new culture.

The objective of this diploma thesis is to assess the factors affecting the concentration of dust particles during forage pressing. For this purpose, five presses for square bales and five presses for round bales were selected. They were harvesting different kinds of straw. Thanks to measurement using an aerosol monitor, the concentration of dust particles during the harvest was detected. The measured values were evaluated and the amount of concentration of the dust particles between the square and round bale presses was compared. There were also set the methodological steps of the measurement procedure.

This thesis is focused on the evaluation of the slip wagon Strautmann Giga Vitesse in the business of primary agricultural production. Measurements were made at harvest zavadlé forage, luskovinomixed grain and cereal straw. The literary part is focused on the historical development of pick-up cars, the agro-technical requirements, the main parts from which cars of the consist, information about the manufacturer of agricultural equipment company Strautmann and wagons Giga Vitesse. The practical part contains evaluation of the quality of work from the collection of plant matter from the lines after the flush, but also the calculation of individual performance and economic evaluation. The resulting information comes from the values measured at work pick up of the car, such as working times, the amount of unpicked vegetable matter or chop length. The necessary data for the economic evaluation of the car provided by the management of the enterprise Dražovický AGROPOL s.r.o the Resulting values are given in the form of tables or graphically.

The diploma thesis on comparison of combine harvesters in terms of work quality is based on professional literary research, which describes the parts of combine harvesters from adapters to threshing devices. The methodology of this thesis describes the procedure for obtaining results and values for comparison between John Deere 9640 WTS, New Holland TC5060, Claas Lexion 450 and Sampo Comia C8 combine harvesters during work. The results and the conclusion of the work compare the calculated, measured and determined results achieved precisely according to the methodology of 2017. The combine harvesters are compared in the areas of work quality, namely in the lossability, the theoretical throughput, the quality of spreading and crushing of straw and finally in efficiency and fuel consumption. The work also includes the costs associated with opearting combine harvesters.

Em vista disso, objetivou-se, com a realização deste experimento, avaliar a influência do parcelamento e das doses de potássio em cobertura na produção e qualidade de raízes tuberosas de batata-doce. O experimento foi conduzido na Fazenda Experimental São Manuel, de 25/03/2014 a 30/07/2014. Foram avaliados dez tratamentos em esquema fatorial 3 x 3 + 1, sendo três doses de potássio (60, 90 e 120 kg ha-1 de K2O); três parcelamentos (sendo aplicados 100% no plantio; 50% no plantio e 50% em cobertura e 25% no plantio e 75% em cobertura) e uma testemunha (sem aplicação de potássio no plantio e em cobertura), no delineamento experimental de blocos casualizados, com quatro repetições por parcela, composta por três leiras com dez plantas cada, sendo as duas leiras laterais consideradas como bordadura, assim como a primeira e a última planta da leira central de cada parcela, e oito plantas foram consideradas úteis para coleta dos dados. Foram avaliados a massa fresca e seca da parte aérea e das raízes comerciais; número, comprimento e diâmetro das raízes comerciais, produtividade, extração de macronutrientes (raízes tuberosas, parte aérea e total) e qualidade pós-colheita (acidez titulável, sólidos solúveis, pH, açúcares redutores e amido) das raízes comerciais. Foi observado ajuste linear para as doses de potássio para todas as características da parte aérea nos parcelamento 50% (plantio) + 50% (cobertura) e 25% (plantio) + 75% (cobertura). Para as características de produção das raízes foi observada interação significativa entre doses e parcelamentos, sendo que o parcelamento 50% (plantio) + 50% (cobertura) apresentou os melhores resultados. A produtividade foi crescente até a dose de 120 kg ha-1 de K2O quando parcelado 50% (plantio) + 50% (cobertura). A ordem decrescente de extração de macronutrientes pelas raízes foi K>N>Ca>P>S>Mg. Já a ordem decrescente de extração da planta foi K>N>Ca>Mg>P>S. As doses de potássio aumentaram os teores de açúcares redutores e reduziram os teores de amido. / Although there are fertilizer recommendations for comercial cultivation of sweet potato, there are few research papers that relate the effect of potassium fertilizer in top dressing and installment it production and quality. The objective of this work was to evaluate the potassium rates and installment influence in top dressing in sweet potato quality and production. The experiment was lead at São Manuel Experimental Farm and the evaluations at Horticulture Department in São Paulo State University. It was evaluated ten treatments in factorial scheme 3x3+1, with three rates of potassium (60, 90 and 120 kg ha-1 of K2O) in top dressing and three installments (100% planting; 50% (planting) + 50% (top dressing) e 25% (planting) + 75% (top dressing) and one control (without potassium and planting at top dressing). The experimental design was randomized blocks, with four replications. Fresh and dry weight of shoot and commercial roots; number, length and diameter commercials roots, yield, extraction of nutrients per plant and quality post harvest (pH, total acidity, soluble solids, reducing sugars and starch) of commercials roots were evaluated. Linear effect was observed for rates of potassium for all vegetative characteristics in installments 50% (planting) + 50% (coverage) and 25% (planting) + 75% (top dressing). For root production characteristics was observed significant interaction between rates and installment, and the installment 50% (planting) + 50% (top dressing) showed the best results. Yield increased with increasing rates of potassium till 120 kg ha-1 de K2O when installment 50% (planting) + 50% (top dressing). The decreasing order of macronutrient extraction by roots was K> N> Ca> P> S> Mg. Already the decreasing order of the plant extraction was K> N> Ca> Mg> P> S. The potassium rates increased sugars and reduced starch content.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Take into account the crescent consumers demand by quality foods and the need of constant supply of vegetables products, the aim of this study was to evaluate the conservation of Crioula and Bola Precoce onions stored in different controlled atmosphere (CA), relative humidity (RH), temperature and ethylene conditions. Were evaluated four experiments with cv. Crioula and one with cv Bola Precoce. In the Crioula experiment 1, was tested the effect of different O2 and CO2 levels in storage atmosphere, two RH and two temperatures levels. Crioula experiment 2 tested three RH levels (75, 80 and 85%), experiment 3 tested three temperature levels (0.5, -0.5 and -1.0 °C) and experiment 4 tested the effect of presence or absence of ethylene 10 μl l-1, all in cold storage (CS). Bola Precoce experiment conditions are identical to Crioula experiment 1. The analyses were realized after six months of storage. CA decreased the breaking of dormancy and the bulb sprout onset in relation to CS. Low O2 and high CO2 increased the response from CA on sprout development process. Respiratory rate of bulbs gradually decreased during shelf-life, being less in CA stored bulbs. The mainly form of mass loss was due to respiration in Crioula and due to water loss in Bola Precoce . CA, mainly with 0.5+2.0 (kPa O2+kPa CO2), reduced aliinase activity and the generation of precursor compounds of onion flavor, beyond decrease the soluble solids consume and polyphenols level. Therefore, CA decrease the bulb metabolism during storage, reflecting in retard in breaking of dormancy and sprout onset, beyond maintain better post-harvest bulb quality. Better results were obtained in AC 0.5+2.0 (kPa O2+kPa CO2). / Tendo em vista a crescente demanda do mercado consumidor por alimentos com qualidade e a necessidade de suprimento constante de produtos de origem vegetal, o objetivo deste trabalho foi avaliar a conservação de cebolas Crioula e Bola Precoce armazenadas em diferentes condições de atmosfera controlada (AC), umidade relativa (UR), temperatura e etileno. Foram realizados quatro experimentos com a cv. Crioula e um com a cv. Bola Precoce. No experimento 1, com Crioula , testou-se o efeito de diferentes níveis de O2 e CO2 na atmosfera de armazenagem, dois níveis de UR e duas temperaturas. O experimento 2 testou três níveis de UR (75, 80 e 85%), o experimento 3 testou três níveis de temperatura (0,5, -0,5 e -1,0 °C) e o experimento 4 testou a presença constante de 10 μl l-1 de etileno durante o armazenamento, todos mantidos em armazenamento refrigerado (AR). Os tratamentos testados em Bola Precoce foram iguais aos do experimento 1 de Crioula . As análises foram realizadas após seis meses de armazenamento. A AC reduziu a superação da dormência e início do processo de brotamento dos bulbos em relação ao AR. Condições com baixo nível de O2 e alto de CO2 aumentaram a resposta da AC no controle do processo de desenvolvimento dos brotos. A taxa respiratória dos bulbos reduziu gradativamente durante a vida de prateleira, sendo menor nos bulbos armazenados em AC. A principal forma de perda de massa foi através da respiração na Crioula e pela perda de água na Bola Precoce . A AC, principalmente com 0,5+2,0 (kpa O2+kpa CO2), reduziu a atividade da enzima aliinase e a geração de compostos precursores do flavor de cebolas, além de reduzir o consumo de sólidos solúveis e o teor de polifenois. Portanto, a AC reduz o metabolismo dos bulbos durante o armazenamento, refletindo em retardo na superação da dormência e início do processo de brotamento, além de manter melhor qualidade pós-colheita dos bulbos. Melhores resultados são obtidos em AC 0,5+2,0 (kPa O2+kPa CO2). .

Esta premissa e a dúvida sobre a adequação do programa dos trabalhadores motivaram esta proposta. Certamente o programa deve ter sido concebido para contemplar não só a produtividade para a empresa, mas também, melhores condições de vida no trabalho para os cortadores. Mas que opinião tem os mesmos? Assim, na busca de resposta a esta questão o objetivo desse trabalho é o de avaliar o Programa Cana Limpa, desde o ponto de vista do cortador verificando seus pontos fortes, fracos e melhorias necessárias ao Programa. Embora a participação do homem se dê em várias das fases dos processos produtivos do setor sucro-alcooleiro, a presente proposta enfoca apenas o trabalho durante a colheita de cana de açúcar. Esta pesquisa quanto ao método tem duas fases. A primeira é qualitativa e exploratória. Nesta fase busca levantar as variáveis para avaliar o programa. Após a identificação das variáveis tem-se uma segunda fase, quantitativa descritiva, onde se avalia o grau de existência das variáveis encontradas, ou seja, estas variáveis da primeira fase não utilizadas na elaboração de questões do instrumento de coleta de dados. Nesta fase é construído um instrumento de coleta de dados com questões fechadas utilizando escala ordinal... / Due to the high consumption of sugar cane, industries have been searching to improve environmental management in the sugar and alcohol production processes, thus resulting in improved production of sugar cane and its by products as well as improved harvesting techniques. The Cana Limpa (clean sugar cane) is one of the programs that arose as a result of this search for improvement in sugar cane production and harvesting. To say that a program is sustainable depends not only on the sponsor or the entrepreneur, but also on the people that are directly affected by such a program. This and the question whether or not the program will be adequate to the workers, motivated our proposal. It is certainly true that the program was conceived to embrace not only productivity, but also better working conditions to the harvesters. But what do they think about it? The objective of this study is to evaluate the Cana Limpa Program from the point of view of the harvester, and analyze its strengths and weaknesses as well as the need for improvements. Even though man participates in various stages of the sugar and alcohol production process, this study focus only on the harvest of sugar cane. The method used in this research is twofold: the first part is qualitative and exploratory. We aimed at obtaining variables to evaluate the program; the second part is quantitative and descriptive, that is, we measurehow many times the variables occur. During this part of the research, we built an instrument to collect data, with closed questions and ordinal scale. The questionnaires were distributed to the harvesters in their place of work. Os frutos foram colhidos na Fazenda Sacramento Agropastoril Ltda e transportados para o Laboratório de Frutas e Hortaliças – UNESP/FCA onde foram selecionados, higienizados e submetidos aos tratamentos, no caso da aplicação do cloreto de cálcio, os frutos foram imersos por 10 minutos nas concentrações de 0% (controle), 0,5, 1,0, 2,0 e 3,0% CaCl2 e embalados com filme de policloreto de vinila 0,020mm, enquanto que os submetidos a diferentes concentrações de gases: 0,03% CO2 e 21% O2 (controle); 5% CO2 e 4% O2; 6% CO2 e 4% O2; 7% CO2 e 4% O2; 8% CO2 e 4% O2 foram embalados em sacos nylon+polietileno. A seguir, os frutos foram armazenados à temperatura de 0±0,5ºC e 85±5% UR por 35 dias e analisados a cada 7 dias quanto à perda de massa, atividade respiratória, coloração, acidez titulável, sólidos solúveis, “Ratio”, pH, firmeza, pectinametilesterase, poligalacturonase, açúcar redutor, ácido ascórbico e índice de adstringência. O delineamento estatístico empregado foi inteiramente casualizado, com cinco repetições para perda de massa e atividade respiratória e com três repetições para as demais análises, utilizando-se o Teste de Tukey a 5% de probabilidade. Nas condições em que os experimentos foram realizados, pode-se concluir que os frutos destanizados e não destanizados submetidos à aplicação do cloreto de cálcio apresentaram menor produção de CO2 e pico climatérico tardio, porém, para as demais análises o cálcio não apresentou incremento positivo na manutenção da qualidade e da firmeza dos frutos... / This study aimed to evaluate the effect of application of calcium chloride and use of modified atmosphere in cold storage of persimmon 'Giombo‟ destanizado and not destanizado order to maintain the quality and delay ripening. The fruits were harvested in the Sacramento Agropastoril Farm Ltda and transported to the Laboratory of Fruit and Vegetables - FCA / UNESP where they were selected, cleaned and subjected to treatment in the case of application of calcium chloride, the fruits were immersed for 10 minutes at concentrations 0% (control), 0.5, 1.0, 2.0 and 3.0% CaCl2 and packed with polyvinyl chloride film of 0.020 mm, while subjected to different concentrations of gases: 0, 03% CO2 and 21% O2 (control), 5% CO2 and 4% O2; 6% CO2 and 4% O2; 7% CO2 and 4% O2; 8% CO2 and 4% O2 were packed in polyethylene + nylon bags. Then the fruits were stored at 0±0,5ºC with 85±5% RH for 35 days and analyzed every 7 days for loss of mass, respiratory activity, color, titratable acidity, soluble solids, Ratio, pH, firmness, activities of pectinmethylesterase and polygalacturonase, reducing sugar, ascorbic acid and astringency index. The employed statistical design was thoroughly randomized, with five repetitions for weight loss and breathing and three replicates for further analysis, using the Tukey‟s Test to 5% of probability. In conditions where the experiments were performed, one can conclude that the fruits destanizados and not destanizados of subjected to the application of calcium chloride had lower CO2 production and the climacteric peak later, but for the other calcium analysis did not show a positive increase in maintenance of quality and fruit firmness. Para tanto foram realizados três experimentos durante o período de 2006 a 2008. Experimento 1: espaçamento de plantas (0,5 x 0,5; 0,75 x 0,5; 0,5 x 1,0 metros) x idade de colheita (6 meses e 12 meses) em esquema fatorial com parcelas sub-divididas no tempo com plantas procendente de Jundiaí/SP; Experimento 2: população de plantas coletadas no Estado de São Paulo (Piquete, Iporanga, Barra do Turvo e Jundiaí) x sazonalidade (inverno/07, primavera/07, verão/08 e outono/08), com o mesmo delineamento estatístico anterior; Experimento 3: horário de colheitas (6:00; 9:00; 12:00; 15:00, 18:00 e 21:00 horas) x temperatura de secagem (folha fresca, 40ºC; 60ºC e 80ºC) em esquema fatorial, utilizando plantas procedentes de Piquete/SP. The objective of this work was to accomplish agronomical and phytochemistry studies, to obtain a protocol with information initial spacing of plants, harvest age, interpopulation variation, seasonal variation, time harvest and drying temperature, to a culture that ensures a systematic and regular production standardized, and thus contribute to species conservation. For this three experiments were accomplished during the period 2006 to 2008. Experiment 1: plant spacing (0.5 x 0.5, 0.75 x 0.5, 0.5 x 1.0 meters) x age harvest (6 months and 12 months) in a factorial scheme with “split-splot in the time” with plants at Jundiaí/SP, Experiment 2: Plants Population (Piquete- SP, Iporanga-SP, Barra do Turvo-SP and Jundiaí-SP) x seasonality (winter/07, spring/07, summer/08 and fall/08) with the same previous statistical design; Experiment 3: Time harvest (6:00, 9:00, 12:00, 15:00, 18:00 and 21:00 hours) x drying temperature (fresh leaf, 40C, 60C and 80C) in factorial scheme, with plants at Piquete/SP. They were evaluated biometric variables (plant height, number of branches and leaves, leaf area, fresh and dry leaves biomass) and phytochemistry (yield and chemical composition of essential oil and yield crude leaf extract and the compound 4-nerolidilcatecol in extract). Data were submitted to variance analysis and Scott- Nott test. Adubação orgânica da bananeira prata-anã e experiências com outras cultivares nas ilhas canárias

A fonte de adubo orgânico foi mantida a mesma desde o primeiro ciclo, aplicando-se composto produzido a partir de serragem de madeira e esterco bovino, que constituíram os tratamentos (doses de composto orgânico): 0, 43, 86, 129 e 172 kg de composto por planta, o que correspondeu a 0; 98,5; 197,0; 290,5 e 394,0 g de K2O por planta, sendo estas doses calculadas de acordo com o teor de potássio presente no mesmo. No solo, a eficiência dos tratamentos foi avaliada mediante análises químicas de amostras de solo, enquanto que nas plantas avaliaram-se os teores de nutrientes presentes nas folhas, circunferência do pseudocaule, altura de inserção da inflorescência, número de folhas por planta, massa do cacho; número de frutos por cacho; número de pencas por cacho; peso da 2ª penca e número de frutos na 2ª penca. Também foram avaliados teores de nutrientes presentes em partes da plantas e a decomposição e liberação de nutrientes dos restos culturais. Para os frutos produzidos no 4º e 5º ciclos avaliaram-se a qualidade dos frutos por meio de análises físicas e químicas como textura, pH, acidez titulável, sólidos solúveis e amido da polpa. O delineamento experimental adotado foi em blocos casualizados, com cinco tratamentos, cinco repetições e duas plantas por parcela, sendo os dados submetidos à análise de variância e regressão. Diante das melhorias observadas no solo (manutenção do pH dentro de uma faixa adequada, elevação nos teores da matéria orgânica, do fósforo... / Aiming to evaluate the effects of organic compost rates in the production of banana plants `Prata-Anã´ in a five production cycle, as well as the nutrient liberation of this compost in the soil, the decomposition and nutrient liberation of the cultural residue, this present work was carried out at FCA/UNESP dependences, in Botucatu-SP from November of 2002 to July of 2007. The organic compost was the same produced since de first cycle, where it was applied compost produced by wood residue and bovine manure, which constituted the treatments (organic compost rates): 0, 43, 86, 129 and 172 kg of compost per plant, which corresponds to 0; 98,5; 197,0; 290,5 and 394,0 g of K2O per plant. These compost rates were calculated based on the quantity of potassium contained in the compost. The effects of organic fertilization in the soil were evaluated by chemical soil analyses. The efficiency of the treatment in plant development and production were evaluated by the quantity of nutrient present in the leaves, pseudostem circumference, plant height, number of leaves per plant, bunch weight, number of fruits in the bunch, number of hands per bunch, 2nd hand weight and number of fruits in the 2nd hand. It was also evaluated the nutrient levels in parts of the banana plants, decomposition and nutrient liberation of the cultural residues. Fruit quality produced in the 4th and 5th cycles was evaluated by physical and chemical analyses such as firmness, pH, acidity, soluble solids and starch. The experiment was arranged in randomized blocks design, with 5 treatments, 5 replications and 2 plants per plot. The obtained data were submitted to variance analyses and to regression analyses.
