Transkriptionelle Interaktionen morphogener Signalwege in der adulten Leber und im Hepatozellulären Karzinom

Die evolutionär konservierten morphogenen Signalwege Wnt/β-Catenin und Hedgehog (Hh) spielen vor allem in der Embryogenese, Zelldifferenzierung und Kanzerogenese eine große Rolle. Es ist bekannt, dass es zwischen Komponenten der beiden Signalkaskaden zu verschiedensten Wechselwirkungen und Hintergrundreaktionen in unterschiedlichsten Organismen und Geweben kommt. Ziel dieser Arbeit war die Aufklärung einzelner mole- kularer, transkriptioneller Prozesse hinter diesen Kreuzreaktionen in primären Hepatozyten und dem Hepatozellulären Karzinom. Dafür sind die beiden Signalwege durch verschiedenste Einflüsse, wie dem Einsatz von siRNAs, transgenen Mausmodellen und rekombinanten Proteinen gegen einzelne Faktoren der Hedgehog, aber auch der Wnt/β-Catenin Kaskade in ihrer Genexpression verändert und die Reaktionen der Signalwegskomponenten mittels der quantitativen Real-Time-PCR untersucht worden. Neben dem Organismus Maus haben einzelne vergleichende Experimente auch auf der humanen Ebene zum Erkenntnisgewinn beigetragen. Durch die Einflussnahme auf den Hedgehog Signalweg in murinen Hepatozyten wird deutlich, dass die Antworten auf die einzelnen Veränderungen in den Signalkaskaden sehr vielschichtig und umfangreich sind. Abhängig vom induzierten bzw. reprimierten Gen, aber auch vom gelenkten Signalweg variieren die Genexpressionen auf unterschiedlichste, und zum Teil gegenläufige Weise. Ferner wird deutlich, dass es zu Unterschieden in der Genantwort bezüglich der verschiedenen Organismen Maus und Mensch, aber auch zu Variationen der Interaktionen in den diversen Gewebe bzw. Zelltypen kommt.

Regulation of Leptin by Sexual Maturation and Energy Status in Male Atlantic Salmon (Salmo salar L.) Parr

Leptin is a peripheral adiposity signal and a key hormone in energy balance regulation in mammals, acting as a link between nutritional status and the endocrine reproductive axis. If this is also the role of leptin in fish is not fully understood. This thesis investigates how different components of the leptin system are affected by sexual maturation and seasonal changes in energy balance in male Atlantic salmon (Salmo salar L.) parr under fully fed and feed-restricted conditions. Moreover, the role of sex steroids as being one of the possible mechanisms by which sexual maturation interacts with leptin is explored. The salmon leptin-a genes, lepa1 and lepa2, were expressed mainly in liver and the leptin receptor (lepr) in brain and ubiquitously in peripheral tissues. Seasonal characterization of the lepa genes and lepr during the growth and reproductive season in one-year old males showed that hepatic lepa1 and lepa2 mRNA levels and plasma leptin levels were down-regulated concomitantly with an increase in weight and body fat. Feed restriction up-regulated hepatic leptin, and pituitary lepr expression as well as plasma leptin levels. Correlation between leptin levels and body lipid stores were either lacking or negative. These findings show that leptin and lepr are sensitive to changes in energy balance, but that leptin might not reflect adiposity in juvenile salmon. Hepatic lepa1 and lepa2, and testicular lepr expression increased during mid- to late spermatogenesis in early maturing males. This up-regulation was preceded by rapid gonadal growth and elevated pituitary follicle-stimulating hormone gene expression levels, whereas peak leptin levels coincided with peak pituitary luteinizing hormone expression and the presence of running milt in the testes. The sex steroids testosterone (T), 11-ketotestosterone and 17-β estradiol stimulated lepa1 and lepa2 gene expression in Atlantic salmon hepatocytes in vitro differentially depending on developmental stage. T was also able to stimulate hepatic lepa1 and pituitary lepa1 and lepr gene expression in immature male salmon in vivo. These results suggest that leptin plays a role in male fish reproduction during later stages of the maturational process and that the elevation of leptin expression during spermatogenesis could be caused by androgen stimulation.

Regulation of Cholesterol Biosynthesis in Hepatocytes

Hypercholesterolemia, a condition of high cholesterol levels in the circulation, poses a major risk for developing cardiovascular disease, such as atherosclerosis. A common method of reducing plasma cholesterol levels relies on the administration of drugs that limit cholesterol synthesis or uptake, many of which have undesirable side effects. Thus, some patients are turning to an alternative treatment, namely natural health products. Natural health products are often equally or even more effective at treating illness than synthetic drugs and may produce fewer side effects. The goal of this study was to identify a natural health product that regulates hepatic cholesterol synthesis by inhibiting HMG-CoA reductase, the enzyme which catalyzes the rate-limiting step of the cholesterol synthesis pathway. Several natural compounds were screened using the human hepatoma cell line HepG2. One compound, berberine, showed great potential as a regulator of cholesterol synthesis and so became the subject of this investigation. Berberine inhibited HMG-CoA reductase activity and decreased cellular accumulation of cholesterol. Berberine was shown to regulate HMG-CoA reductase through activation of metabolic regulator AMP-activated protein kinase, which modifies HMG-CoA reductase post-translationally and thereby decreases its activity. In conclusion, this study demonstrates that the natural health product berberine decreases cholesterol synthesis by activating a cellular signalling pathway to bring about post-translational modification of HMG-CoA reductase, and in doing so, inhibits this enzyme. This novel mechanism supports berberine’s potential for a cholesterol-lowering therapy and its role in reducing the risk for cardiovascular disease.

Partial hepatectomy and liver regeneration in PCSK9 knockout mice

The proprotein convertase subtilisin/kexin type 9, PCSK9, belongs to the proprotein convertase (PC) family. Human mutations in the gene encoding PCSK9 lead to either familial hyper- or hypocholesterolemia, resulting from a gain or loss of function, respectively. Mice lacking PCSK9 are viable and show a 42% decrease in plasma cholesterol levels. The enzyme triggers the degradation of the low density lipoprotein receptor (LDLR) through a partially unknown mechanism. / PCSK9 is very abundant in the liver and intestine during development and adulthood. Hepatocytes have a capacity to reproduce themselves and, upon injury, can repopulate the liver. For a better understanding of the role of PCSK9 in the liver, partial hepatectomy was performed on Pcsk9 +/+, Pcsk9+/- and Pcsk9-/- mice. The absence of PCSK9 resulted in defective liver regeneration, while wild type (WT) and heterozygous mice had no phenotype. Regeneration defects could be prevented by a high cholesterol diet. PCSK9 deficiency, by contributing to maintaining low circulating cholesterol levels may thus hamper liver regeneration. This knowledge is critical for the analysis of future PCSK9 inhibitors expected to be developed in the near future. / Key words. Energetic Costs of AhR Activation in Rainbow Trout (Oncorhynchus mykiss) Hepatocytes

Aquatic organisms in response to toxic insults from environmental pollutants activate defence systems including the aryl hydrocarbon receptor (AhR) in an attempt to metabolize and excrete these toxicants and their metabolites. These detoxification mechanisms however may come with certain energetic costs. I hypothesize that the activation of the AhR by β-Naphthoflavone (β-NF), a model AhR agonist, results in increased energetic costs requiring metabolic reorganization in rainbow trout hepatocytes. While the results obtained suggest that there are no significant energetic costs of AhR activation, analysis of enzyme activities suggests possible metabolic reorganization. This study also showed significant changes in cellular processes in hepatocytes over the incubation periods which previously were not reported. Furthermore, for the first time in fish hepatocytes, metabolic flux analysis (MFA) was used to examine intra-cellular metabolism, the applicability of which is discussed.
