Avaliação toxicológica e ecotoxicológica dos principais representantes de éteres de difenilas polibromadas (PBDEs) / Toxicological and Ecotoxicological evaluation of the main polybrominated diphenyl ethers (PBDEs)

Os éteres de difenilas polibromados (PBDEs) são largamente usados como retardantes de chama e têm sido detectados em diversas amostras biológicas, devido às suas características físico-químicas e bioacumuladoras, as quais fazem com que estes compostos tenham uma elevada persistência no meio ambiente. A exposição a estes compostos pode ser nociva ao homem e aos organismos vivos, uma vez que muitos estudos têm relatado que este composto possui atividade citotóxica. Desta forma, no presente estudo, investigamos o mecanismo de ação dos BDE-47, BDE-99, BDE-154 e BDE-209 sobre o processo autofágico de células HepG2, pois além de ser um processo de reciclagem de moléculas intracelular, a autofagia atua ainda na citoproteção celular. Foram realizados ensaios de imunocitoquímica para detecção de alterações no processo autofágico, no qual foi avaliado, sob microscopia de fluorescência, o padrão de distribuição das proteínas LC3-I e LC3-II; paralelamente, foi realizada a técnica de western blotting para a quantificação de três proteínas associadas ao processo de autofagia - LC3, Beclin-1 e p62. Em nossas análises de imunocitoquímica pode-se observar que os quatro PBDEs em estudo foram capazes desencadear o processo de autofagia, sendo que a magnitude do efeito tóxico foi dependente do tempo de exposição (24 e 48 horas), da concentração utilizada (0,1; 1; 5; 25 ?mol/L) e variável de acordo com o congênere em questão, sendo os BDE-47 e BDE-99 os que apresentaram maior potencial de ativação da autofagia. Além disso, esta indução de autofagia ainda pode ser detectada nos ensaios de western blotting, pois os PBDEs alteraram os níveis de LC3-II, Beclin-1 e p62, havendo um padrão de toxicidade semelhante ao visualizado nos ensaios de imunoistoquímica. Adicionalmente, devido ao fato dos PBDEs poderem se acumular no meio ambiente, principalmente em organismos aquáticos, uma análise dos efeitos destes compostos sobre um modelo ecotoxicológico foi realizada. O zebrafish (D. rerio) foi utilizado como organismoteste, a fim de avaliar a toxicidade destes contaminantes ambientais sobre seus estágios embrionários e larvais. Foram analisados endpoints de letalidade, subletalidade e teratogenicidade após a exposição aos PBDEs em estudo. Sendo que os compostos BDE-47, BDE-99 e BDE-209 (na presença de luz) apresentaram alterações em pelo menos um dos indicadores avaliados, sendo que o parâmetro mais afetado foi a inflação das bexigas natatórias, indicando uma possível teratogenicidade causada por estes compostos. Por outro lado, os BDE-154 e BDE- 209 (na ausência de luz) não causaram malformações no organismo-teste. Diante dos resultados obtidos, pode-se concluir que estes compostos podem causar alterações na homeostase celular desencadeando o processo autofágico, sendo que se absorvidos por organismos, podem afetar o desenvolvimento de espécies como o zebrafish. / Polybrominated diphenyl ethers (PBDEs) are widely used as flame retardants and they have been detected in several biological samples due to their physicochemical and bioaccumulating characteristics, which make these compounds to have a high persistence in the environment. The exposure to these compounds can be harmful to man and other living organisms, since many studies have reported that this compound has cytotoxic activity. Thus, in the present study, we investigated the mechanism of action of the BDE-47, the BDE-99, the BDE-154 and the BDE-209 on the autophagic process of HepG2 cells, because beyond being an intracellular molecule recycling process, autophagy acts also in the cell cytoprotection. Immunocytochemistry assays were performed to detect changes in the autophagic process, in which the distribution pattern of the LC3-I and LC3-II proteins was evaluated under fluorescence microscopy. In parallel, the western blotting technique was used to quantify three proteins associated with the autophagy process - LC3, Beclin and p62. In our analyzes of immunocytochemistry it can be observed that the four PBDEs in the study were able to trigger the autophagy process, and the magnitude of the toxic effect was dependent on the exposure time (24 and 48 hours), on the used concentration (0.1; 1; 5; 25 ?mol/L), and it varies according to the congener. The BDE-47 and BDE-99 are the ones with the highest potential for autophagy activation. In addition, this autophagy induction can still be detected in the western blotting assays, because the PBDEs altered the levels of LC3-II, Beclin-1 and p62, with a similar pattern of toxicity as seen in the immunohistochemical assays. In addition, due to the fact that PBDEs may accumulate in the environment, mainly in aquatic organisms, an analysis of the effects of these compounds on an ecotoxicological model was performed. The Zebrafish (D. rerio) was used as a test organism in order to evaluate the toxicity of these environmental contaminants on their embryonic and larval stages. In this study, lethality, sublethality and teratogenicity endpoints were analyzed after exposure to the PBDEs. The BDE-47, BDE-99 and BDE-209 compounds (in the presence of light) presented alterations in at least one of the indicators evaluated, and the most affected parameter was the swim bladder inflation, indicating a possible teratogenicity caused by these compounds. On the other hand, the BDE-154 and the BDE-209 (in the absence of light) did not cause malformations in the test organism. Considering the results, it can be concluded that these compounds are able to cause changes in cellular homeostasis triggering the autophagic process, and whether absorbed by organisms, they can affect the development of species such as zebrafish.

Evalution of Dengue virus RNA extraction methods and the study of viral-induced apoptosis of HepG2 hepatocyte

Dengue fever is an arthropod-borne transmit disease, caused by dengue virus.The principal vectors are Aedes aegypti and Aedes albopictus. Induce Dengue fever (DF) and a more severe form of dengue hemorrhagic fever (DHF), dengue shock syndrome (DSS). Dengue virus is the most prevalent arbovirus in tropical and subtropical regions. There is no specific drug and vaccine available for treatment and prevention. Therefore, DF is an important disease among transmit diseases in humans. For the effective control and prevention of DF transmission, rapid quantitative molecular biological methods are very important for the diagnosis of dengue fever. At present, there are many methods to isolate the RNA of Dengue virus; however, the new developed magnetic method has not been used for the Dengue virus isolation yet. At first, we evaluated various methods for Dengue virus isolation. The result indicate that the best method of RNA extraction for dengue virus is the QIAamp® Viral RNA kit manual extraction. There are no apparent differences of the effect for Dengue virus RNA isolation between filting film and magnetic bead method. Furthermore, DHF caused by Dengue virus is a very serious disease and the pathologic mechanism of DHF has not been elucidated completely. Both clinical and experimental trials have confirmed that the liver cell is one of the target infected by Dengue virus. And, the mechanism of Dengue virus-induced liver cell apoptosis remains poorly understood.Furthermore, there are free radical and cytokines production in patient¡As serum in the acute phase of DF. Therefore, the role of antioxidant and p21 in the mechanisms should be elucidcited. Our preliminary data show that p21 mRNA expression increase in HepG2 after Dengue virus infection. NAC, GSH, and DPI all can attenuate Dengue-induced cell apoptosis. Howerer, the relationship between p21 expression and liver cell apoptosis should be further clarified in the near future.

TRPV Channels and Modulation by Hepatocyte Growth Factor/Scatter Factor in Human Hepatoblastoma (HepG2) Cells

Using patch clamp and Ca2+ imaging techniques, we have studied Ca2+ entry pathways in human hepatoblastoma (HepG2) cells. These cells express the mRNA of TRPV1, TRPV2, TRPV3 and TRPV4 channels, but not those of TRPV5 and TRPV6. Functional assessment showed that capsaicin (10 μM), 4α-phorbol-12,13-didecanoate (4αPDD, 1 μM), arachidonic acid (10 μM), hypotonic stress, and heat all stimulated increases in [Ca2+]i within minutes. The increase in [Ca2+]i depended on extracellular Ca2+ and on the transmembrane potential, which indicated that both driving forces affected Ca2+ entry. Capsaicin also stimulated an increase in [Ca2+]i in nominally Ca2+-free solutions, which was compatible with the receptor functioning as a Ca2+ release channel. Hepatocyte growth factor/scatter factor (HGF/SF) modulated Ca2+ entry. Ca2+ influx was greater in HepG2 cells incubated with HGF/SF (20 ng/ml for 20 h) compared with non-stimulated cells, but this occurred only in those cells with a migrating phenotype as determined by presence of a lamellipodium and trailing footplate. The effect of capsaicin on [Ca2+]i was greater in migrating HGF/SF-treated cells, and this was inhibited by capsazepine. The difference between control and HGF/SF-treated cells was not found in Ca2+-free solutions. 4αPDD also had no greater effect on HGF/SF-treated cells. We conclude that TRPV1 and TRPV4 channels provide Ca2+ entry pathways in HepG2 cells. HGF/SF increases Ca2+ entry via TRPV1, but not via TRPV4. This rise in [Ca2+]i may constitute an early response of a signalling cascade that gives rise to cell locomotion and the migratory phenotype.

Avaliação das alterações celulares em linhagem de hepatocarcinoma humano (HepG2) induzidas pelos principais representantes da classe de éteres difenílicos polibromados (PBDE) / Não informado

A grande evolução tecnológica tem sido responsável pela introdução de diferentes contaminantes no ambiente, dentre os quais estão os éteres difenílicos polibromados (PBDEs). Os PBDEs representam uma importante classe de retardantes de chama adicionada aos diversos bens de consumo da vida moderna com a finalidade de retardar o processo de ignição e impedir a propagação de chamas. Atualmente, a crescente presença desses compostos em compartimentos ambientais e em amostras de fluídos biológicos tem levantado preocupações relacionadas com os possíveis efeitos tóxicos à saúde, no entanto tais efeitos são incertos devido à baixa disponibilidade de dados relacionados com a ação direta sobre o organismo humano. Desta forma, esse trabalho objetivou investigar os efeitos dos principais representantes de PBDEs: congênere PBDE-209, PBDE-99 e PBDE-47 sobre células de hepatoblastoma humano (HepG2) na faixa de concentração de 0.1-25µmol/L, em períodos de 24 e 48 horas, como uma alternativa de esclarecer os efeitos tóxicos dos PBDEs sobre a saúde humana, bem como correlacionar os efeitos entre os diferentes congêneres. As células HepG2 foram escolhidas devido as semelhanças com o metabolismo das células hepáticas normais. Também foi investigado o potencial dos três congêneres em induzir a morte celular após exposição aos PBDEs, sendo demonstrado potencial citotóxico diferenciado de acordo com o congênere investigado. Pela avaliação de ensaios de proliferação celular (SRB) e viabilidade celular (MTT) foi obtido que o congênere PBDE-209 induz morte celular em altas concentrações (10µmol/L, 25µmol/L) sendo esses efeitos iniciados primeiramente, por mecanismos citostáticos em concentrações menores. Enquanto isso, os PBDE-99 e PBDE-47 demonstraram elevado potencial citotóxico de forma concentração dependente, apresentando diferenças nas intensidades das concentrações avaliadas, relacionando-as com a facilidade de absorção e com os possíveis metabólitos formados. A investigação dos mecanismos citotóxicos foi realizada pelos ensaios de avaliação do potencial de membrana mitocondrial, exposição da fosfatidilserina na membrana externa celular e fragmentação nuclear. Esses ensaios evidenciaram que os congêneres investigados agem por indução de mecanismos apoptóticos, sendo os maiores resultados relacionados com os menores congêneres investigados. A avaliação dos mecanismos por vias necróticas foi realizada pela avaliação da atividade da LDH devido ao rompimento da membrana celular característico de mecanismos necróticos, não sendo obtidos resultados significativos. A avaliação do acúmulo de EROs ocasionado pela exposição aos congêneres demonstrou que os compostos investigados apresentam elevado potencial para induzir o estresse oxidativo, sendo inclusive relacionado com os efeitos citostáticos preliminares do congênere PBDE-209. A diferença dos resultados obtidos entre os três congêneres foi relacionada com as diferenças estruturais, as quais facilitam a absorção dos menores congêneres e com os possíveis metabólitos oriundos formados. Concluindo, estes resultados mostram os efeitos tóxicos dos PBDEs em células hepáticas e sugere a presença de efeitos adversos à saúde. Além disso, embora os congêneres PBDE-99 e PBDE-47 apresentem os maiores potenciais tóxicos, o efeito do congênere PBDE-209 não deve ser ignorado uma vez que o mesmo pode se intensificado com a exposição prolongada / Technologic demand has increased the different pollutants released in the environment. Polybrominated diphenyl ether (PBDE) is an important class of the flame retardants added in several good products with a wide range of toxic effects on biotic and abiotic systems. These compounds are release into environment through the loss during manufacture and of products containing PBDEs. Their toxic mechanisms on biologic systems still have not understood due to the existence of several different congeners, with different chemical and biological characteristics in the environment, and absence of their effects on the body. Therefore, in this study we examine effects of congeners PBDE-209, PBDE-99 and PBDE-47 on HepG2 cell line in order to understand their mechanism of action and their effects on the human health. Firstly, we evaluated citotoxic mechanism by SRB assay followed by MTT assay. This way, we showed that all tested PBDEs seem to cause cell death although PBDE-209 also causes cystostatic mechanism in lower concentration. The cytotoxic mechanism was observed in higher concentrations and it was suggested be evidences of apoptosis. Apoptotic mechanism was investigated by mitochondrial membrane potential, phosphatidyl serine exposure and nuclear fragmentation. Our results showed that all tested congeners were able to decrease mitochondrial membrane potential, to cause phosphatidyl serine exposure and a nuclear fragmentation with higher results found for BDE-99 and BDE-47 due structural difference that may facilities their absorption and their toxic metabolites Exclusion of necrotic mechanism was performed by LDH release assay. We evaluated reactive oxygen species (ROS) production and it was showed that apoptotics mechanisms is followed by oxidative stress in all congeners tested and suggest cytostatic mechanism is also mediated by ROS. Investigação dos efeitos tóxicos do biossurfactante ramnolipídio e suas implicações quando usado na biorremediação de águas contaminadas por petróleo

Estudos têm mostrado que, mesmo com um grande potencial de uso do ramnolipídio na indústria cosmética e na área da saúde, este composto é, principalmente, promissor para uso na indústria do petróleo. Apesar dos biossurfactantes serem considerados menos tóxicos e mais biodegradáveis que os surfactantes sintéticos, estes compostos ainda não foram investigados quanto a sua ação genotóxica direta ou indireta sob o material genético de organismos expostos. Desta maneira, o objetivo deste trabalho foi investigar os possíveis danos celulares promovidos pela ação do biossurfactante ramnolipídio e suas implicações no material genético quando utilizado como agente biorremediador de águas impactadas por petróleo, por meio dos bioindicadores Allium cepa, Oreochromis niloticus e as células humanas mantidas em cultura - HepG2. Para a realização da investigação, o biossurfactante ramnolipídio foi produzido em condições de laboratório e a partir da concentração indicada para o uso em processos de biorremediação (1g/L), foram estabelecidas as concentrações utilizadas no estudo. Para avaliar o potencial tóxico, genotóxico e mutagênico das concentrações de ramnolipídio, assim como seu feito sinérgico com o petróleo, foram realizados os testes de germinação de sementes, aberrações cromossômicas e teste do micronúcleo em A. cepa; teste do micronúcleo em eritrócitos circulantes, ensaio do cometa com sangue periférico e células de brânquias e análise ultraestruturais de eritrócitos circulantes em O. niloticus; e teste do MTT, teste do micronúcleo e ensaio do cometa em células HepG2. Nossos resultados mostraram que as concentrações 1g/L e 2g/L de ramnolipídio são tóxicas para A. cepa. No entanto, quando as concentrações 1g/L e 10g/L... / Rhamnolipids belong to the glycolipid class of biosurfactants, which are produced by the bacterium Pseudomonas aeruginosa. Studies have shown that, even being potentially used in the cosmetics industry, this compound is especially promising in the petroleum industry. Although the biosurfactants are considered less toxic and more biodegradable than the synthetic surfactants, these compounds have not yet been investigated when it comes to their either direct or indirect genotoxic action upon the genetic material of exposed organisms. Thus, the purpose of this study was to investigate the possible cell damage promoted by the action of rhamnolipid biosurfactants and their implications for the genetic material when they are used as a bioremediation agent of petroleum-impacted water, by using Allium cepa, Orechromis niloticus and human cells kept in culture – HepG2. In order to carry out this investigation, the rhamnolipid biosurfactant was produced under laboratory conditions, and the recommended concentration for use in bioremediation processes (1g/L) was also used; the concentrations used in the study were established. To assess the toxic, genotoxic and mutagenic potential of rhamnolipid concentrations, as well as their synergy with petroleum, seed germination, chromosome aberration tests and micronucleus test in A. ceppa; micronucleus test in circulating red blood cells, comet assay in the peripheral blood and gill cells, and ultrastructural analysis of circulating blood cells in O. niloticus; and MTT test, micronucleus test and comet assay in HepG2 cells. Our studies showed that the 1 g/L and 2 g/L rhamnolipid concentrations are toxic to A. cepa. However, when 1 g/L and 10 g/L concentrations were used in the bioremediation process, these concentrations did not cause toxic, genotoxic or mutagenic damage besides the ones observed for the cells exposed to soluble petroleum... Isso se deve à medicina curativa implantada principalmente no ocidente. Com frequência os NSAIDs são usados para aliviar queixas musculoesqueléticas, artrite, bursite, dor de dentes, dismenorreia, dor de estados pós-parto e na dor de metástases de câncer no osso, sendo todas essas enfermidades associadas ao aumento da síntese de prostaglandinas. Os efeitos farmacológicos dos NSAIDs são devidos à inibição da ciclooxigenase (COX) e a subsequente diminuição da síntese de prostaglandinas (PGs), levando a uma diminuição da inflamação, dor e febre. Essa diminuição de PGs leva a uma larga escala de efeitos colaterais que são associados à NSAIDs, incluindo complicações gastrointestinais (CGI), problemas cardiovasculares, toxicidade renal, hipertensão e retenção de líquidos. Devido a essas características dos NSAIDs, justificam-se estudos envolvendo a utilização de outros meios para a entrega oral de fármacos anti-inflamatórios. Uma opção muito pesquisada, que vem crescendo de maneira significativa é o uso de nanopartículas, com potencial para constituir uma nova geração de “sistemas de entrega de drogas”. Nanopartículas apresentam uma série de características, tais como: detectar, monitorar e tratar doenças; proteger o fármaco de ácidos estomacais e enzimas do trato gastrointestinal; e proteger o estômago e intestino do próprio fármaco. A indometacina é um poderoso agente NSAID, utilizado para tratamento de artrite e osteoartrite. Como inibidora não seletiva da ciclooxigenase, principalmente no caso de inibição da ciclooxigenase-2 (COX-2), que é muito relacionada às lesões da mucosa gástrica. Por esta razão, é de grande importância que a indometacina seja objeto de estudo para novos métodos e formas de distribuição desse fármaco. Considerando que não há dados na literatura sobre a análise da biossegurança do uso da Indometacina Nanoencapsulada (NI) sobre o material genético e sua toxicidade em células de mamíferos, este estudo tem por objetivo avaliar a viabilidade celular deste medicamento nanoencapsulado pelo teste de coloração com o azul de Tripan, e sua citotoxicidade pelo ensaio do MTT, e sua genotoxicidade por intermédio do ensaio cometa e teste do micronúcleo com bloqueio na citocinese. Todos os ensaios foram feitos com o uso de culturas de linfócitos humanos de sangue periférico (células não metabolizadoras) e de células do hepatoma humano (HepG2) (células metabolizadoras) (exceto o MTT). Os ensaios dos testes de viabilidade foram realizados com tempo de exposição à substância teste por 24 horas, com as seguintes concentrações: 5, 10, 50, 125, 250, 500 e 1250 μg/ml. Os resultados obtidos permitiram a seleção de seis concentrações (5, 10, 50, 125, 250 e 500 μg/ml) 11 onde foi observado uma viabilidade celular ≥ 80%. Também foi possível realizar o ensaio de citotoxicidade utilizando as células HepG2, com as mesmas concentrações e pelo mesmo período de tempo usado no teste de exclusão azul de tripan. Os resultados foram bastante semelhantes resultados com o azul de tripan, onde apenas a concentração de 1250 μg/ml obteve uma viabilidade menor que 80%. Para o teste de genotoxicidade as células foram tratadas com as concentrações que obtiveram uma viabilidade maior que 80%, por um período de 24 horas para o teste do micronúcleo e por 4 horas para o teste do cometa. Os resultados obtidos mostraram que apenas a concentração máxima nas células metabolizadoras (HepG2), no teste do micronúcleo, foi observado um aumento estatisticamente significativo de células micronucleadas quando comparado ao controle negativo. Nas outras concentrações testadas, a NI não causou efeito genotóxico e/ou mutagênico significativo em ambas as linhagens celulares. Os resultados obtidos indicam que o nanoencapsulamento da indometacina pode ser um processo bastante promissor no sentido de desenvolver uma forma de administração deste medicamento, menos nociva a mucosa gástrica dos pacientes. Por outro lado, os resultados também apontam para a necessidade de realização de mais estudos relacionados com o potencial genotóxico de concentrações mais altas da NI, bem como de sua metabolização. / Non-steroidal anti-inflammatory drugs (NSAIDs) are among the drugs most widely used worldwide, mainly by curative medicine that is implanted. Commonly NSAIDs are used to relieve muscle-skeletal complaints, arthritis, bursitis, toothache, dysmenorrhea, postpartum states of pain and pain of cancer metastases in the bone. Such diseases and complications are associated with increased prostaglandin synthesis. Pharmacological effects of NSAIDs are due to inhibition of cyclooxygenase (COX) and subsequent decrease of prostaglandin synthesis (PGs), causing a decrease in inflammation, pain and fever. PGs decrease leads to a wide range of side effects that are associated with NSAIDs, including gastrointestinal complications (CGI), cardiovascular disorders, renal toxicity, hypertension and water retention. Because of these characteristics of NSAIDs are justified studies involving the use of other means for oral delivery. A solution that is very researched, and has been growing significantly is the use of nanoparticles with potential to constitute a new generation of “drug delivery systems”. Nanoparticles have many characteristics, such as: detect, monitor and treat diseases; protecting the drug from stomach acids and enzymes in the gastrointestinal tract; and protect the stomach and intestines of the drug itself. Indomethacin is a potent NSAIDs agent used to treat arthritis and osteoarthritis. It has the ability to inhibit not selectively cyclooxygenase, particularly cyclooxygenase – 2 (COX-2) that is closely associated with gastric mucosal injury. For this reason, it is important that the Nanocoated Indomethacin (NI) is the subject of this study, to ensure the safety of new methods and forms of distribution of this type of drugs. There are no data in the literature on the analysis of biosafety using Nanocoated Indomethacin of the genetic material and its toxicity in mammalian cells. This study aims to assess cell viability of this medicine nanocoated by Trypan blue exclusion test, and cytotoxicity by MTT assay, and genotoxicity through the comet assay and micronucleus test with block in cytokinesis. All assays were performed using cultures of human lymphocytes from peripheral blood (not metabolizing cells) and human hepatoma cells (HepG2) (metabolizing cells) (except MTT assay). Tests of viability were performed by treatment of 24 hours with the following concentrations: 5, 10, 50, 125, 250, 500 and 1250 μg/ml. Results obtained enabled the selection of six concentrations (5, 10, 50, 125, 250 and 500 μg/ml) which was observed cell viability ≥ 80%. Was possible to perform cytotoxicity assay using the HepG2 cells with same concentrations and same period of treatment as trypan blue exclusion test. Results were quite similar with trypan blue, only the concentration of 1250 μg/ml achieved < 80% viability. In genotoxicity assay cells were 13 treated with the following concentrations: 5, 10, 50, 125, 250 and 500 μg/ml, for a period of 24 hours for micronucleus test and 4 hours for the comet assay. Results showed that only the maximum concentration in the metabolizing cells (HepG2), on micronucleus test, there was a statistically significant increase in micronucleated cells compared to the negative control. Other tested concentrations, NI did not cause any genotoxic and/or mutagenic effect, significantly, in both cell lines. The results indicate that Nanocoated Indomethacin can be a very promising method to develop an administration form of the drug, less harmful to the gastric mucosa of patients. However, the results also point to the need for further studies related to the genotoxic potential of higher concentrations of NI as well as your metabolism.

Avaliação das alterações celulares em linhagem de hepatocarcinoma humano (HepG2) induzidas pelos principais representantes da classe de éteres difenílicos polibromados (PBDE) / Não informado

A grande evolução tecnológica tem sido responsável pela introdução de diferentes contaminantes no ambiente, dentre os quais estão os éteres difenílicos polibromados (PBDEs). Os PBDEs representam uma importante classe de retardantes de chama adicionada aos diversos bens de consumo da vida moderna com a finalidade de retardar o processo de ignição e impedir a propagação de chamas. Atualmente, a crescente presença desses compostos em compartimentos ambientais e em amostras de fluídos biológicos tem levantado preocupações relacionadas com os possíveis efeitos tóxicos à saúde, no entanto tais efeitos são incertos devido à baixa disponibilidade de dados relacionados com a ação direta sobre o organismo humano. Desta forma, esse trabalho objetivou investigar os efeitos dos principais representantes de PBDEs: congênere PBDE-209, PBDE-99 e PBDE-47 sobre células de hepatoblastoma humano (HepG2) na faixa de concentração de 0.1-25µmol/L, em períodos de 24 e 48 horas, como uma alternativa de esclarecer os efeitos tóxicos dos PBDEs sobre a saúde humana, bem como correlacionar os efeitos entre os diferentes congêneres. As células HepG2 foram escolhidas devido as semelhanças com o metabolismo das células hepáticas normais. Também foi investigado o potencial dos três congêneres em induzir a morte celular após exposição aos PBDEs, sendo demonstrado potencial citotóxico diferenciado de acordo com o congênere investigado. Pela avaliação de ensaios de proliferação celular (SRB) e viabilidade celular (MTT) foi obtido que o congênere PBDE-209 induz morte celular em altas concentrações (10µmol/L, 25µmol/L) sendo esses efeitos iniciados primeiramente, por mecanismos citostáticos em concentrações menores. Enquanto isso, os PBDE-99 e PBDE-47 demonstraram elevado potencial citotóxico de forma concentração dependente, apresentando diferenças nas intensidades das concentrações avaliadas, relacionando-as com a facilidade de absorção e com os possíveis metabólitos formados. A investigação dos mecanismos citotóxicos foi realizada pelos ensaios de avaliação do potencial de membrana mitocondrial, exposição da fosfatidilserina na membrana externa celular e fragmentação nuclear. Esses ensaios evidenciaram que os congêneres investigados agem por indução de mecanismos apoptóticos, sendo os maiores resultados relacionados com os menores congêneres investigados. A avaliação dos mecanismos por vias necróticas foi realizada pela avaliação da atividade da LDH devido ao rompimento da membrana celular característico de mecanismos necróticos, não sendo obtidos resultados significativos. A avaliação do acúmulo de EROs ocasionado pela exposição aos congêneres demonstrou que os compostos investigados apresentam elevado potencial para induzir o estresse oxidativo, sendo inclusive relacionado com os efeitos citostáticos preliminares do congênere PBDE-209. A diferença dos resultados obtidos entre os três congêneres foi relacionada com as diferenças estruturais, as quais facilitam a absorção dos menores congêneres e com os possíveis metabólitos oriundos formados. Concluindo, estes resultados mostram os efeitos tóxicos dos PBDEs em células hepáticas e sugere a presença de efeitos adversos à saúde. Além disso, embora os congêneres PBDE-99 e PBDE-47 apresentem os maiores potenciais tóxicos, o efeito do congênere PBDE-209 não deve ser ignorado uma vez que o mesmo pode se intensificado com a exposição prolongada / Technologic demand has increased the different pollutants released in the environment. Polybrominated diphenyl ether (PBDE) is an important class of the flame retardants added in several good products with a wide range of toxic effects on biotic and abiotic systems. These compounds are release into environment through the loss during manufacture and of products containing PBDEs. Their toxic mechanisms on biologic systems still have not understood due to the existence of several different congeners, with different chemical and biological characteristics in the environment, and absence of their effects on the body. Therefore, in this study we examine effects of congeners PBDE-209, PBDE-99 and PBDE-47 on HepG2 cell line in order to understand their mechanism of action and their effects on the human health. Firstly, we evaluated citotoxic mechanism by SRB assay followed by MTT assay. This way, we showed that all tested PBDEs seem to cause cell death although PBDE-209 also causes cystostatic mechanism in lower concentration. The cytotoxic mechanism was observed in higher concentrations and it was suggested be evidences of apoptosis. Apoptotic mechanism was investigated by mitochondrial membrane potential, phosphatidyl serine exposure and nuclear fragmentation. Our results showed that all tested congeners were able to decrease mitochondrial membrane potential, to cause phosphatidyl serine exposure and a nuclear fragmentation with higher results found for BDE-99 and BDE-47 due structural difference that may facilities their absorption and their toxic metabolites Exclusion of necrotic mechanism was performed by LDH release assay. We evaluated reactive oxygen species (ROS) production and it was showed that apoptotics mechanisms is followed by oxidative stress in all congeners tested and suggest cytostatic mechanism is also mediated by ROS. These results are evidences of toxic effects of PBDEs on the cell line and health

Avaliação da citotoxicidade, genotoxicidade e mutagenicidade da mandioca (Manihot esculenta Crantz) em célula tumoral HepG2 / Assessment of Cytotoxicity, genotoxicity and mutagenicity of cassava (Manihot esculenta Crantz) in HepG2 cells

O fator alimentar pode ser considerado um promotor tumorigênico responsável pela etiologia do câncer gástrico no estado do Pará. A mandioca (Manihot esculenta Crantz) é uma das muitas espécies da Amazônia consumida de modo indiscriminado pelos habitantes da região norte. O cianeto, seu principal componente tóxico, pode ser liberado durante o processamento da mandioca por meio da hidrólise do glicosídeo cianogênico linamarina. No organismo, o cianeto bloqueia a cadeia de transporte de elétrons inibindo a respiração celular e, provocando, entre outras coisas, a produção de radicais livres que podem agir no DNA, através da formação de adutos exocíclicos. O objetivo deste trabalho foi a avaliação da atividade citotóxica, genotóxica e mutagênica de folhas e tucupi, crus e cozidos, de mandioca mansa e brava, usando os ensaios do MTT, cometa e citoma em células HepG2. Os resultados obtidos demonstraram que a viabilidade celular decai à medida que a concentração aumenta na maioria dos grupos de tratamento. No ensaio do cometa, a análise visual demonstrou que o cianeto de potássio, usado como padrão, foi genotóxico em todas as concentrações testadas (5,0; 15,0 e 25,0 ?g/mL). As amostras de folhas de mandioca brava foram genotóxicas somente nas concentrações 15,0 e 25,0 ?g/mL (cruas) e 5,0; 15,0 e 25,0 ?g/mL (cozidas). As amostras de mandioca mansa foram genotóxicas apenas quando cozidas (5,0 e 25,0 ?g/mL). Já as amostras de tucupi, tanto cruas quanto cozidas, demonstraram dano ao DNA de células HepG2 em todas as concentrações testadas (20,0; 40,0 e 60.0 ?g/mL). Utilizando análise de fragmentação de DNA observamos que o cianeto de potássio foi genotóxico apenas na avaliação da porcentagem de DNA na cauda. Já amostras de mandioca brava e mansa, de maneira geral, demonstraram valores de porcentagem de DNA na cauda, tail moment e olive moment maiores em relação ao grupo controle negativo, mas, somente as folhas de mandioca, em algumas concentrações, demonstraram valores estatisticamente significativos. Observando o ensaio do citoma, as concentrações de folhas e tucupi, crus e cozidos, tanto de mandioca brava quanto de mandioca mansa mostraram um discreto aumento no número de micronúcleos em células HepG2 binucleadas, estatisticamente não significativo, em relação ao grupo controle negativo. Já o número de pontes nucleoplasmáticas e brotos nucleares foi menor que no grupo controle. Os danos aqui observados pelo ensaio do cometa podem estar transitoriamente presentes ii como intermediários formados durante o reparo de lesões no DNA. A ausência de brotos nucleares, pontes nucleoplasmáticas e resultados estatísticos significativos em relação ao grupo controle negativo, não nos permitem afirmar presença de mutagenicidade em células HepG2 tratadas com mandioca tanto brava quanto mansa. De maneira geral, o cozimento das amostras não foi um fator determinante na diminuição do dano observado em células HepG2. Nossos resultados confirmam apenas citotoxicidade e genotoxicidade das variedades da mandioca nas concentrações e sistema celular utilizados. Os mecanismos moleculares que envolvem a genotoxicidade da mandioca requerem estudos futuros. Para o consumo da mandioca devem ser levados em consideração tipo de processamento realizado para extração de compostos cianogênicos, níveis de glicosídeos cianogênicos nos produtos consumidos, quantidade de mandioca consumida e estado nutricional do consumidor / The food factor can be considered a tumor causing agent reponsasible for the etiology of gastric cancer in the state of Pará Cassava ( Manihot esculenta Crants is one of the many food species of the Amazon indiscriminately consumed by the inhabitants of the northern region Cyanide, its main toxic component, can be released during cassava processing through the hydrolysis of the cyanogenic glycoside linamarin. In the body, cyanide blocks the electron transport chain by inhibiting cell respiration which brings about, among other things, the production of free radicals which can act upon DNA through the formation of exocyclical adducts. The main goal of this study was the assessment of the citotoxic, genotoxic and mutagenic role of the leaves and tucupi juice of wild and sweet cassava, both raw and cooked, using the MTT, comet and cytome assays in HepG2 cells. The results gathered have shown that cell viability decreases as the concentration increases in most treatment groups. In the comet assay, a visual analysis has shown that potassium cyanide, used as a standard, was genotoxic in all concentrations tested (5.0, 15.0 and 25.0 ?g/mL). Samples of wild cassava leaves were genotoxic only in concentrations of 15.0 and 25.0 ?g/mL for raw leaves, and 5.0, 15.0 and 25.0 ?g/mL for cooked leaves. Samples of sweet cassava leaves were genotoxic only when cooked (5.0 and 25.0 ?g/mL). Yet, samples of tucupi juice, both raw and cooked, have shown damage to DNA in HepG2 cells at all concentrations tested (20.0, 40.0 and 60.0 ?g/mL). In performing DNA fragmentation analysis, it was observed that potassium cyanide was genotoxic only in the assessment of percentage DNA in tail. Whereas the wild and sweet cassava samples, in a general fashion, have shown greater values of percentage DNA in tail, tail moment and olive moment than the negative control group, but only the cassava leaves revealed statistically significant values, in some concentrations. For the cytome assay, concentrations of leaves and of tucupi juice, raw and cooked, of both wild and sweet cassava, have shown a slight increase in the number of micronuclei in binucleated HepG2 cells, not statistically significant as compared to the negative control group. On the other hand, the number of nucleoplasmic bridges and nuclear buds in binucleated cells was lower than the value found in the negative control group. The damages verified herein by the comet assay may be transiently present as intermediates formed during repair of DNA lesions. The absence of iv nucleoplasmic bridges, nuclear buds and of statistically significant results vis-à-vis the negative control group do not warrant the assertion for the presence of mutagenicity in HepG2 cells treated with either wild or sweet cassava. In general, the cooking of the samples was not determining factor of the decrease in damage observed in HepG2 cells. Our results only confirm cytotoxicity and genotoxicity of wild and sweet cassava species in the concentrations and cell system used. The molecular mechanisms involving genotoxicity of cassava require further studies. For the consumption of cassava, the way of processing performed for extracting cyanogen contents, the levels of cyanogenic glycosides in the products consumed, amount of cassava consumed as well as the nutritional condition of the consumer must be taken into account.

Quantificação de lesões em DNA e RNA em cultura de células expostas a tetraidrofurano / Quantitation of DNA and RNA lesions in cell culture exposed to tetrahydrofuran

Tetraidrofurano (THF) é um solvente muito utilizado industrialmente, com demonstrada ação carcinogênica em animais experimentais, mas pouco se sabe sobre sua toxicidade celular, danos a biomoléculas e genotoxicidade. Em trabalho anterior realizado no laboratório, foi verificado que adutos de DNA são formados a partir da reação de THF oxidado com 2\'-desoxiguanosina (dGuo), 2\'-desoxiadenosina (dAdo) e 2\'-desoxicitidina (dCyd) in vitro. A ocorrência dessas lesões em sistemas biológicos expostos a THF nunca foi demonstrada e pode indicar uma via de ação genotóxica desse solvente, até o momento não considerada nos estudos de carcinogênese e toxicidade. Neste trabalho foi investigada a indução de lesões em DNA e RNA de sistemas biológicos expostos ao THF, assim como alterações epigenéticas. Como sistemas biológicos foram utilizados homogenato de fígado de camundongos, cultura de células HepG2 e fígado e rim de camundongos que foram, em trabalho anterior, expostos a vapores do solvente. Métodos de HPLC-ESI-MS/MS foram validados para quantificação de todas as lesões em DNA. Foi verificado que THF é um solvente de baixa citoxicidade para células HepG2, sendo necessárias concentrações acima de 50 mM por período de incubação superior a 24 h para ser observada perda de viabilidade. Houve indução de dano oxidativo em DNA de células HepG2 e de fígado de camundongos expostos. Uma vez que o solvente esteja oxidado, a espécie reativa THF-OH presente no meio de cultura é capaz de reagir com RNA e DNA das células e DNA de homogenato de fígado de camundongos, gerando adutos com dAdo, dGuo e Ado. A espécie reativa possui maior afinidade por dGuo que por dAdo no DNA. Entretanto, as células HepG2 não biotransformaram THF para o intermediário reativo. Além disso, THF induziu hipermetilação no DNA das células HepG2 expostas a 50 e 100 mM do solvente e em DNA de fígado de camundongos fêmeas C57BL/6J expostos a vapores de THF (vapor de 1 mL/h, 6h/dia, 5 dias). Este estudo mostra pela primeira vez a reatividade do solvente THF oxidado (contendo THF-OH) com DNA e RNA em sistemas biológicos, assim como alteração epigenética induzida pelo solvente, e servirá para uma melhor compreensão dos mecanismos envolvidos na indução de câncer por THF. / Tetrahydrofuran (THF) is a widely used industrial solvent with demonstrated carcinogenic action in experimental animals, but little is known about its cellular toxicity and genotoxic damage to biomolecules. Previous work of this group showed that DNA adducts are formed from the reaction of oxidized THF with 2\'-deoxyguanosine (dGuo), 2\'-deoxyadenosine (dAdo), and 2\'-deoxycytidine (dCyd) in vitro. The occurrence of these lesions in biological systems exposed to THF has never been demonstrated and may indicate a genotoxic pathway of this solvent, so far not considered in carcinogenesis and toxicity studies. It was investigated here the induction of DNA and RNA lesions in biological systems exposed to THF, as well as epigenetic changes. The biological systems used were mice liver homogenate, HepG2 cell culture, and liver and kidney of mice that were exposed to solvent vapors in a previous work. HPLC-ESI-MS/MS methods were validated for quantitation of all lesions in DNA. It was found that THF is of low cytotoxicity to HepG2 cells, requiring concentrations above 50 mM for incubation period over 24 h to induce loss of viability. DNA oxidative damage was induced in HepG2 cells and liver of exposed mice. Once the solvent is oxidized, the reactive species THF-OH present in the culture medium is able to react with DNA and RNA of HepG2 cells and DNA from mice liver homogenate, generating adducts with dAdo, dGuo and Ado. The reactive species has higher affinity for dGuo than for dAdo in DNA. However, HepG2 cells were not able to activate THF to the reactive intermediate. In addition, THF induced DNA hypermethylation in HepG2 cells exposed to 50 and 100 mM of the solvent and in liver of C57BL/6J female mice exposed to THF vapors (1 mL vapor / h 6h/dia, 5 days). This study shows for the first time the reactivity of oxidized THF (containing THF-OH) with DNA and RNA in biological systems, as well as epigenetic change induced by the solvent, and may contribute for a better understanding of the mechanisms involved in the induction of cancer by THF.
