Assessment of the anti-proliferative and anti-inflammatory pollen extract Cernitin™ in prostatic cells and isolated human peripheral blood mononuclear cells

Benign prostatic hyperplasia (BPH) and chronic prostatitis (CP) are common diseases in aging men. Though medications are available to alleviate these conditions, problems of possible side-effects of first-line synthetic drugs for prostatic conditions have allowed patients to switch to a safer plant-based medication. CernitinTM, a pollen extract, is used to alleviate these conditions. A recent in vitro study showed that CernitinTM inhibits cell proliferation and induce a regulatory effect on inflammatory parameters. To validate those results, the inter-batch variability of CernitinTM was assessed using the active ingredients CernitinTM T60 and CernitinTM GBX on the human prostatic cell lines BPH‐1 and WPMY‐ 1 and on human peripheral blood mononuclear cells (hPBMCs) in vitro. Cell proliferation assay was performed in prostatic cell lines, while inflammatory parameters were analyzed in hPBMCs. Results revealed that both CernitinTM active ingredients, regardless of batch production, significantly inhibited the proliferation of both prostatic cell lines after 48 and 72 hours, respectively (p < 0.05 to p < 0.001). Among the batches, there were no significant differences observed. Notably, the GBX batches 14164, 14548 and 14160 had a more pronounced effect on cell proliferation right after 48 hours on both cell lines. Whilst, T60 batches 11539 and 14144 had a pronounced effect right after 48 hours on BPH cells. In hPBMC, the production of the anti-inflammatory cytokine interleukin (IL)- 10 and its receptor IL-10 receptor subunit beta (RB), as well as pro-inflammatory cytokine IL-6 was significantly increased after treatment with the T60 formulation regardless of the batch, but not after treatment with the GBX batch. Moreover, IL-10 receptor subunit alpha (RA) and tumour necrosis factor‐related apoptosis‐inducing ligand (TRAIL) expression increased after the use of both formulations (p < 0.05 to p < 0.001). The pro-inflammatory cytokine IL-8 and chemokine CXCL-10 was significantly decreased using both batches of T60 (p < 0.05 to p < 0.001). Collectively, these results support the claim of the role of CernitinTM as an anti-proliferative agent and as a cytokine regulator.

Congenital adrenal hyperplasia (CAH) due to 21-hydroxylase deficiency is the most common inherited endocrinopathy. This disorder is associated with many complications caused either by illness itself or inadequate treatment. The late consequence of this disorder is a reduced final height. Absences of cortisol, androgen excess, with or without salt wasting, virilization of external genitalia in new-born girls are main findings. Androgen excess causes virilization in girls, accelerated bone maturation and early epiphyseal fusion in both sexes. CAH patients are dependent on lifelong treatment with oral glucocorticoids. Growth retardation and obesity are common side effects resulting from glucocorticoid use. The aim of this thesis was to analyse the success of obtaining a final height that is within a genetic potential regarding to treatment compliance. The other objective was the analysis of auxological profile of CAH patients. This study was mainly focused on the analysis of body height, linear proportionality, the weight parameters, body composition, body fat distribution, skeletal robusticity and body structure. (ENGLISH) Background: The investigation of the immunosuppression usage in cardiovascular surgery and interventional cardiology is, at present, concentrated on three main topics: 1) influence on intimal hyperplasia of coronary and peripheral vascular reconstructions 2) influence on rejection of allogeneic vascular grafts and 3) influence on intimal hyperplasia of coronary arteries after endovascular interventions. Modern immunosuppressive drug FK506 (Tacrolimus) could have a positive effect for these indications. In experimental study, FK506 inhibited rejection of arterial allografts and also inhibited intimal hyperplasia in percutaneous coronary interventions. Aims: The purpose of this study was to evaluate the effect of systemic tacrolimus treatment on the process of arterialisation of allogeneic and syngeneic venous grafts in a rat vein-to-artery implantation model. Material and Methods: Lewis (LEW) rats were used as recipients of syngeneic (Lewis) or allogeneic (Brown-Norway; BN) iliolumbar veins which were implanted into abdominal aorta. Aloe striata Plant Defense Compounds Produced in Response to Jasmonic Acid, Salicylic Acid, and Eriophyid Mite Salivary Extract Elicitor Compounds

Aloe mites are herbivores of the genus Aloe (ALOACEAE) and are associated with hyperplastic growth in various aloe species, but the biochemistry of this interaction is poorly understood. In an effort to characterize plant defense responses to herbivory in the genus Aloe, a salivary extract was isolated from aloe mites (Aceria aloinis Keifer) and its bioactivity was tested using a hypocotyl elongation assay. Subsequently, Aloe striata plants were treated with jasmonic acid (JA), salicylic acid (SA), and the mite salivary extract. Using water and methanol, compounds of different polarity were extracted from aloe tissues that had been frozen and crushed at 4, 12, and 24h after treatment. Extracts were analyzed by HPLC and three compounds were found. One of these compounds was SA (mean concentration of 4µg/mL), and this is the first time that this aloe species has been found to produce SA. Two additional peaks of unknown identity were observed in JA- and SA-treated plants. These results suggest that A. striata may in fact undergo a JA-mediated change in secondary metabolism as part of a plant defense response.
