Zpracování signálu UHF RFID čtečky / Signal Processing for UHF RFID Reader

The master’s thesis is focused on the UHF RFID reader EXIN-1 signal processing. The first part describes the concept of the EXIN-1 front end, its basic testing and possible communication interfaces for reader control and for receiving and transmitting baseband signals. The second part of this work is aimed to a simple description of EPCglobal Class-1 Generation-2 UHF RFID Protocol, especially to used modulations and codings. In the last part, a block connection between the front end and an ARM Cortex-M4 microcontroller discovery board is designed. The microcontroller is used for generating of all required signals and also for receiving incoming signals and processing them for identification numbers of RFID cards (tags), which are in the reading range of the reader. A decoding algorithm is designed in MATLAB software and implemented to the selected microcontroller. Obtained identification data are displayed on an LCD display and also sent to a PC through a serial communication.

This master’s thesis deals with the issue of scanning human fingerprints, which is currently very topical and represents the most widespread biometric technology. The theoretical part of the work acquaints the reader with basics of dactyloscopy and biometrics and concerns technologies used for fingerprinting, image preprocessing methods and commercially available contactless optical scanners. The practical part is a realisation of a contactless optical scanner based on a Raspberry Pi minicomputer, implementation of preprocessing algorithms in Python and testing of the device from the perspective of image quality.

Based on these results it is possible to propose an enhancement of the current state of the firm and its prospective development in the future.

Designed algorithms are implemented in a control unit of the experimental vehicle (filters signal used in the closed-loop controller). The improvement of the position estimation using Kalman Filter is demonstrated on the experimental vehicle. In the next part the design process of algorithm developing for dsPIC microcontroller using Matlab is described.

Návrh vestavěného systému pro řízení výukového modelu rotačního kyvadla / Design of embedded system for control of educational model of rotary pendulum

The basic aim of this work is to improve existing model of rotational inverted pendulum by adding new mechanical features, implement the control algorithm to dsPIC microcontroller and develop related control electronics thus extending the functionality of current model while making it more compact. The work contains derivation of dynamic equations both by means of analytical methods and multi-body formalism of SimMechanics. These are used to design a state controller stabilizing the pendulum in inverse position. In addition, parameters of the system are being estimated experimentally. Swing-up controller is developed to drive the pendulum to unstable position. Various state estimators are added to controller to improve the control process while comparing their overall performance. The last point is devoted to development of superior state-automaton designed to switch between different regulating modes including fail-detection algorithms providing smooth operation of the model.

This diploma thesis elaborated on the subject „Contemporary Health and Safety at Work Problems and their Influence on the Functioning of an Organization“ focuses on the description of contemporary state of Health and safety at work problems in work activities, where the source of risk is represented by chemical substances, from the point of view of a company. The thesis sets out a view of legislative definition for the mentioned topic and also the possible application of safety at work management programs. This part also includes a summary description of risk analysis methods. In the practical part of the thesis, there is an organizational directive for a fictive company engaged in chemical production made. This directive will identify possible occupational risks resulting from the manufacturing process, and it will also evaluate the amount of risk for particular work activities in conjunction with hazard factors. The next part of the thesis contains a proposal of measures for risk reduction, based upon the previously mentioned evaluation. In the final part of the thesis, the author contemplates the possibilities of health and safety at work system improvement.

Izolace, identifikace a charakterizace mikroflóry vína a vybraných potravin / Isolation, Identification and Characterisation of Microbial Communities of Wine and Selected Foods

Proposed dissertation thesis deals with wine and artisanal cheeses microbiology. The first part is focused on identification of yeasts isolated from grapes and musts during production of white and red wines. The grape varieties were grown under the integrated and organic farming on Moravian vineyard. Yeasts were identified by ITS-PCR-RFLP method (amplifying internal transcribed spacer ITS: ITS1, ITS2 and 5.8S rDNA) and unknown species were subjected to partial sequencing of ITS rDNA region. In total, 524 isolates were divided into 14 different species belonging to six genus were identified from. The first stages of fermentation process were characterised by predominance of non-saccharomyces species especially H. uvarum. Due to increased ethanol concentration strains of S. cerevisiae prevailed in the later phases of the process. Further, partial aim of this study was to isolate and to apply selected autochthonous S. cerevisiae strains as starter culture during controlled industrial wine fermentation process. Genus Saccharomyces was distinguished from other non-saccharomyces species by ITS-PCR-RFLP. Further, in order to distinguish Saccharomyces genus at the species and the strain level, several molecular methods were applied including PCR-fingerprinting (rep- and RAPD-PCR), species-specific primers (multiplex and touchdown PCR), LSU-DGGE and interdelta PCR. Species-specific primers enabled us to distinguish some species of the Saccharomyces sensu stricto complex. Furthermore, interdelta PCR seems to be useful tool for S. cerevisiae strains identification. Among 120 isolated autochthonous strains belonging to Saccharomyces genus, 45 different strains were identified. Based on its sufficient technological properties (osmo- and ethanol tolerance, low H2S production etc.), S. cerevisiae 1-09 strain isolated from grape berries coming from moravian vineyard was chosen. Strain S. cerevisiae 1-09 was tested in small amount of must and after that also during industrial fermentation of red and white wine production. Based on the results of chemical and sensorial analysis, the strain seems to be suitable for application as the starter culture for winemaking process. The final part of this thesis is focused on quantification and identification of the yeasts isolated from artisanal cheeses and their by-products coming from Western Balkan Countries. Isolated species were identified by ITS-PCR-RFLP, partial sequencing and by physiological tests. Among the 20 yeast species found, D. hansenii, C. zeylanoides and Y. lipolytica were found to be predominant. Moreover, we developed culture-independent, semi-quantitative technique based on construction of ITS-clone library from metagenomic DNA to investigate complex fungal communities associated with artisanal cheeses and their by-products. Novel technique is based on direct extraction of total DNA from the sample. This was compared with culture-dependent ITS-PCR-RFLP and culture-independent LSU-DGGE methods. The results highlighted the discrepancies among these methods. Finally, the divergences among applied methods were confirmed by correlation analysis and by indices of general biodiversity and dominance of species. ITS-clone library approach combines the advantages of cultivation-based analysis and LSU-DGGE with semi-quantification of fungal species without the requirement of their cultivation. This study might open new perspectives in direct and complex analysis of yeasts and moulds in food matrices.
