Oxidación de proteínas y lípidos en cerebro de cobayos durante la exposición a las grandes alturas (4540 m)

Se evaluó el efecto del tiempo de exposición a las grandes alturas sobre la oxidación de proteínas y lípidos del tejido cerebral de cobayos nativos del nivel del mar trasladados a las grandes alturas (Morococha, 4540 m), y sacrificados los días 1, 3, 7 y 14 después de su arribo. Se determinó los niveles medios de cuerpos carbonílicos (CC), malondialdehído (MDA) e hidroperóxidos lipídicos (LOOH) como marcadores de la oxidación de proteínas y lípidos respectivamente; así como, las actividades de las enzimas antioxidantes superóxido dismutasa (SOD), catalasa (CAT) y glutation peroxidasa (GPx) y fosfolipasa A2 (FLA2) como mediadora de la peroxidación lipídica. Se encontró niveles de CC, LOOH y MDA incrementados al primer día; CC disminuyó por debajo del control al tercer día, LOOH mantuvo la tendencia a disminuir y MDA mantuvo sus niveles altos. Las actividades de las enzimas antioxidantes: GPx y CAT incrementaron desde el primer día; la actividad de SOD aumentó hasta el tercer día disminuyendo posteriormente; la actividad de FLA2 aumentó hasta el tercer día. Los resultados indican que la exposición por diferentes tiempos a las grandes alturas influye directamente en el proceso de oxidación de proteínas y lípidos. La disminución de los niveles de CC podría deberse a la activación del sistema proteolítico, en especial de las proteasas dependientes de Ca+2 como las calpaínas o del sistema proteasomal, las cuales degradarían las proteínas dañadas por las EROs. La exposición a la altura influye además en la actividad de las enzimas antioxidantes, especialmente en GPx, que juega un rol importante en la detoxificación de LOOH, lo que explicaría la tendencia a disminuir al final del tiempo de estudio. / -- It was determined the effect of high-altitude exposition time (Morococha - 4540 m) on protein and lipid oxidation from brain of level-sea native guinea pig for different times (1,3,7 and 14 days). It was measured the level of carbonyl groups (CC), malondialdehyde (MDA) and lipids hydroperoxydes (LOOH) as protein and lipid oxidation markers respectively. Also, the activity of antioxidants enzymes superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and phospholipase A2 (PLA2) a mediator of lipid peroxidation were evaluated. The results showed an increase of the CC, LOOH and MDA levels during the first day; CC decreased below the control levels by the third day, LOOH level had a decrease trend in time and MDA kept its higher levels. The activity of both antioxidant enzymes GPx and CAT, increased since the first day. Moreover, the activity of SOD showed an increase up to third day followed by a decrease; the activity of PLA2 increased up to the third day. The dates recorded indicated that the expositions at altitudes for different times affect directly the oxidative process of both protein and lipid. The decrease on the CC level could be caused by the activation of the proteolytic system, especially the activation of calcium-dependent proteases as calpains or the proteasomal system which could degrade damaged proteins by EROs. The exposition of altitude might affect the activity of antioxidant enzymes, especially GPx, which could play an important role in the detoxification of LOOH. / Tesis

Macroalgas lóticas do Parque Nacional Chapada dos Veadeiros e arredores (GO) : levantamento florístico e distribuição ecológica das comunidades /

Orientador: Luis Henrique Zanini Branco / Banca: Orlando Necchi Júnior / Banca: Carla Ferragut / Resumo: Vários estudos enfocaram as macroalgas de ambientes lóticos no Brasil, realizados principalmente nos estados de São Paulo e Paraná, porém ainda são escassos se considerada a extensão territorial e a diversidade de ambientes do país. Este trabalho teve como objetivo realizar o levantamento florístico e analisar a estrutura e distribuição ecológica das comunidades de macroalgas lóticas do Parque Nacional da Chapada dos Veadeiros e arredores, localizado no estado de Goiás e com vegetação predominante de cerrado. Em termos de biomas brasileiros, o cerrado é conhecido por apresentar uma das áreas mais ricas em biodiversidade e alta taxa de endemismo. Foram amostrados 16 trechos de riachos em duas ocasiões, uma no período de chuvas (abril de 2010) e outra no período de secas (julho de 2011). Foram identificados 15 táxons específicos e seis "grupos vegetativos" (pertencentes aos gêneros Spirogyra, Oedogonium e Zygnema), além de um estágio "Chantransia pygmaea". Cyanobacteria e Chlorophyta foram os grupos mais representativos (41% dos táxons), seguidos do grupo Rhodophyta (18%). Muitas espécies tiveram ocorrência restrita a um ou dois pontos de amostragem (72,7%) e as macroalgas que se destacaram por ocorrerem em três ou mais pontos de amostragem foram Microspora floccosa (7), "Chantransia pygmaea" (4), Oedogonium sp. (4), Stigonema ocellatum (4), Sirodotia delicatula (3) e Spirogyra sp. 2 (3). Apenas uma espécie, Stigonema multipartitum, representou novo registro para o Brasil. Os dados de abundância demonstraram que o período de seca (2011) foi favorável para o crescimento das comunidades de macroalgas, porém apresentando elevada dominância de poucas espécies, o que caracteriza um padrão de pré-ocupação de nicho. It should be emphasized that most of them are restricted to São Paulo and Paraná States. This study aimed to survey the flora and analyze the structure and distribution of ecological communities of stream macroalgae of the Chapada dos Veadeiros National Park and surround area. The park is located in Goiás State and the cerrado (Brazilian savanna) is the predominant vegetational type. In terms of biomes, the cerrado biome is known for having one of the highest biodiversity and endemism rate. Sixteen stream segments were sampled in two occasions, one in the rainy season (April 2010) and one in the dry period (July 2011). Fifteen specific taxa, six "vegetative groups" (belonging to the genera Spirogyra, Oedogonium and Zygnema) and one "Chantransia pygmaea" stage were found in this study. Cyanobacteria and Chlorophyta were the most representative groups (41% of all taxa), followed by the Rhodophyta group (18%). Many species were restricted in distribution occurring in one or two sampling sites (72.7%). The macroalgae that occurred in three or more sampling sites were Microspora floccosa (7), "Chantransia pygmaea" (4), Oedogonium sp. Análisis de distribución altitudinal de mamíferos pequeños en el Parque Nacional Yanachaga Chemillén, Pasco, Perú

La Cordillera Yanachaga ubicada en la selva centro oriental del Perú, es considerada como una cadena aislada del macizo oriental de los Andes. Esta cadena montañosa y sus zonas aledañas presentan una alta diversidad de especies silvestres y gran belleza paisajística, motivos por los cuales ha sido establecida como un Área Natural Protegida en la categoría de Parque Nacional. Para conocer si la distribución de especies de mamíferos pequeños a lo largo de un gradiente altitudinal de una cadena montañosa aislada como Yanachaga, es diferente a la presentada en la Cordillera Oriental, se efectuó un análisis de la distribución vertical desde 300 m hasta los 2,800 m, tomando como estudio referencial el realizado en el Parque Nacional Manu, en Cuzco y Madre de Dios. Las especies de mamíferos pequeños consideradas en este trabajo se agruparon en cuatro conjuntos: murciélagos (67 especies), marsupiales (11 especies), roedores múridos (16 especies), y marsupiales y todos los roedores, reunidos como “mamíferos pequeños terrestres” (31 especies). Se analizó estos grupos empleando pruebas de regresión polinomial, análisis de agrupamiento, y calculando perfiles y amplitudes de rango de las distribuciones altitudinales. Para los grupos de murciélagos, marsupiales, y mamíferos pequeños terrestres, la riqueza de especies es mayor en elevaciones bajas, decreciendo conforme se asciende en altitud; en el caso de roedores múridos, la distribución de especies está más relacionada al tipo de bosque, encontrándose mayor riqueza de especies en ambos extremos de la gradiente. Resultados semejantes fueron obtenidos también en Manu. La mayor amplitud de rangos de distribución de especies se observa claramente en murciélagos y marsupiales, siguiendo la regla de Stevens, mientras en roedores múridos y mamíferos pequeños terrestres ésta solo se cumple parcialmente. Estos resultados, similares a los obtenidos en Manu, indican que los patrones de distribución en una cordillera aislada como Yanachaga son semejantes a los hallados en la cordillera principal. Además, en murciélagos se distinguió dos grupos: uno de especies de elevaciones bajas correspondiente a bosque lluvioso de tierras bajas, y otro de elevaciones más altas que comprenden a bosques montanos bajo y de neblina. Ambos grupos separados por una zona de reemplazo incluido en el rango altitudinal (600 – 900 m) que comprende al bosque montano bajo, aproximadamente como se encontró en Manu. De manera similar a lo encontrado en Manu, en roedores múridos se encontró tres grupos de especies, de altitudes bajas que abarcan bosques lluviosos de tierras bajas; de altitudes intermedias con bosque montano bajo y de neblina, y altas con bosque de neblina. Los tres grupos colindan con áreas de reemplazo en los rangos (600 – 900 m) y (1,800 – 2,100 m) correspondientes al bosque montano bajo, y bosque de neblina, respectivamente. Se halló tres especies nuevas, de los géneros Akodon, Thomasomys (Rodentia) y Micronycteris (Chiroptera), que sugieren su posible evolución en aislamiento geográfico en la cordillera Yanachaga. Además, se reporta una especie inédita de Monodelphis, conocida anteriormente de otras cordilleras del centro y sur de Perú. / The Cordillera Yanachaga, located in central-eastern Peru, is a mountain range east of, and isolated from the eastern chain of the Andes. This region and nearby areas is home to a large number of wild species and shows great natural beauty, and for these reasons it has been set aside as a Protected Natural Area in the category of National Park. In order to investigate if the distribution of species of small mammals along an altitudinal gradient in such an isolated mountain range as Yanachaga, is different to that present in the main Eastern Andes, an analysis of the vertical distribution of the species, from 300 up to 2,800 m, was carried out, comparing the results obtained with those found further south, at Manu National Park, in Cuzco and Madre de Dios departments. The species of small mammals studied in this work conform four groups: bats (67 species); marsupials (11 species); murid rodents (16 species); and marsupials and all the rodents, considered together as “terrestrial small mammals” (31 species). These groups were analyzed using tests of polynomial regression and cluster analysis, and range profiles and widths of altitudinal distribution were calculated. For bats, marsupials, and terrestrial small mammals, species richness is largest at low elevations, decreasing with an increase in altitude; for murid rodents, species distribution is related to forest type, being most diverse at both ends of the altitudinal gradient. Similar results were also obtained in Manu. The largest distribution of species range widths is clearly observed among bats and marsupials, supporting Stevens' rule in these two groups, whereas in murid rodents and terrestrial small mammals the rule is only partially supported. These results, similar to those obtained in Manu, indicate that the distribution patterns in an isolated mountain range like Yanachaga are similar to those of the main mountain range. Furthermore, two groups were discriminated in bats, one comprising low-elevation species found in lowland rain forest, the other including species of higher elevations, inhabiting both montane and cloud forest. The species composition of both sets changes at the vegetation belt (600 - 900 m) occupied by low montane forest, in a manner comparable to the situation found in Manu. Also, in a similar way to that seen in Manu, murid rodents are separated in three species groups, of low, intermediate and high elevations, corresponding to lowland rain forest, montane and cloud forest, and only cloud forest. These groups replace each other at the vegetation belts (600 - 900 m, and 1,800 - 2,100 m) corresponding to lower montane forest, and cloud forest, respectively. Three new species of the genera Akodon, Thomasomys (Rodentia) and Micronycteris (Chiroptera) were found, suggesting their possible evolution in geographic isolation in the Cordillera Yanachaga. In addition, an undescribed species of Monodelphis, already known from other mountain ranges in central and southern Peru, was also recorded at Yanachaga.

Efecto del ketoprofeno sobre la presión arterial pulmonar en terneros Jersey sometidos a hipoxia de la altura

Se ha estudiado el efecto del ketoprofeno, un antiprostaglandínico bloqueador de la enzima ciclooxigenasa, sobre la presión arterial pulmonar media (PAPm) en 10 terneros Jersey, machos, de 1 a 2 meses de edad, nacidos a nivel del mar y expuestos durante 3 días a una hipoxia ambiental de 3 320m. de altitud; los cuales fueron divididos en 2 grupos de 5 animales cada uno: Grupo Control que recibió placebo y Grupo Tratamiento al que se le suministró ketoprofeno, en dosis de 3mg/kg de peso vivo, por 5 días consecutivos, durante su estadía a nivel del mar. La PAPm se determinó mediante la técnica de cateterismo cardiaco a nivel del mar y al tercer día de su arribo a dicha altitud. Los valores promedio de la PAPm (mmHg) a nivel del mar fueron de 19,00 ± 0,94 para el Grupo Control y 21,00 ± 3,39 para el Grupo Tratamiento, y en la altura de 39,50 ± 6,00 para el Grupo Control y 31,25 ± 5,70 para el Grupo tratamiento. Al no existir diferencia por efecto del ketoprofeno, se analizaron ambos grupos en conjunto obteniéndose los siguientes resultados de la PAPm: A nivel del mar 20,00 ± 2,68 y al tercer día de exposición a la altura 35,28 ± 7,16. Estos resultados nos inducen a sugerir que el ketoprofeno no tuvo efecto significativo (p>0.05) sobre la PAPm en los terneros Jersey sometidos durante 3 días a la hipoxia de la altura; sin embargo sólo la exposición a dicha hipoxia incrementó significativamente (p≤0.05) la PAPm. / The effect of ketoprofen, an antiprostaglandin sustance that inhibits the enzyme ciclooxigenase, had been studied on the mean pulmonary arterial pressure (mPAP) in 10 Jersey, male calves, from 1 to 2 months old. They were born at sea level and exposed during 3 days to a environmental hypoxia of 3 320m. of altitude and were divided in 2 groups of 5 animals each: Control Group which received destiled water as placebo, and Treatment Group which received ketoprofen in dose of 3mg/kg body weight for 5 consecutive days during its permanence at sea level. The mPAP was determinated by cardiac cateterism at sea level and 3 days after arriving to altitude. The mean values of mPAP (mmHg) at sea level were: Control Group 19,00 ± 0,94 and Treatment Group 21,00 ± 3,39; whereas at the altitude were: Control Group 39,50 ± 6,00 and Treatment Group 31,25 ± 5,70. Since there was not significative effect due to ketoprofen, the two groups were joined and analysed together, obtaining the following values for mPAP: At sea level 20,00 ± 2,68 and at day 3 of exposition to altitude 35,28 ± 7,16. These results induce us to sugest that ketoprofen have not significative effect (p>0.05) on the mPAP in Jersey calves exposed during 3 days at altitude hypoxia, however just the effect of this hypoxia increased significatively (p≤0.05) the mPAP.

Evidencia de la participación del oxido nítrico en la adaptación a la altura en ratas albinas

El NO participa en una amplia gama de procesos biológicos, entre ellos se presume la adaptación a la hipoxia medioambiental. Por tal motivo se determinaron los niveles de nitritos en plasma en un total de 270 ratas albinas machos, divididas en dos ensayos: la mitad sometidos a hipoxia a la altura de 3,320 m.s.n.m confrontados a un grupo control a nivel del mar. La determinación del óxido nítrico (NO) fue indirecta mediante la cuantificación de los nitritos (NO2) y nitratos (NO3) por la reacción de Griess, (1879). Todos los animales fueron muestreados en grupos de 15 los días 1, 2, 5, 7, 14, 21, 28, 35 y 42 respectivamente. La mayor concentración de óxido nítrico en el grupo hipóxia medioambiental se encontró en el día 2 de muestreo ((α = 0.05). Los niveles plasmáticos de óxido nítrico en el grupo hipóxia medioambiental presentaron diferencias significativas con el grupo control (p – valor = 0.001) encontrándose incrementados con respecto al grupo control (α = 0.05). Estos resultados proponen que la hipoxia medioambiental produce un aumento de la síntesis de ON, como consecuencia de procesos adaptativos a la altura, corroborando los resultados de otros trabajos e investigaciones. / NO participate in an ample rate of biological processes, among them presumes the adaptation to the environmental hypoxia. All the animals were tested in groups of 15, on the days 1, 2, 5, 7, 14, 21, 28, 35 y 42 respectively. The greater concentration of NO in the group submissive environmental hypoxia was in day 2 of sampling (α = 0.05). The NO levels in the hypoxia environmental group presented significant differences with the group control (p – value = 0,001) being increased with respect to the group control (α = 0.05). These results propose that environmental hypoxia produces an increase of the synthesis of nitric oxide, consequence of adaptive processes to the height, corroborating the results of other works and investigations.

Remanescentes contínuos da Mata Atlântica apresentam um ambiente altamente heterogêneo, resultado de fatores ambientais e impactos antrópicos. Os resultados apresentados nos 4 capítulos dessa tese destacam que, embora a Mata Atlântica seja o bioma mais intensamente estudado para mamíferos no Brasil, ainda não se atingiu o conhecimento necessário para a conservação e gestão eficaz desta classe na Mata Atlântica. Estudos adicionais são necessários para permitir a definição de zonas dentro das áreas protegidas, como definido pela legislação brasileira (Lei: 9.985/2000 (SNUC)). Este zoneamento é necessário para a realização dos objetivos múltiplos das áreas protegidas (incluindo a manutenção da biodiversidade e o fornecimento dos serviços dos ecossistemas) dentro do contexto sócio-econômico em um escala regional e nacional / There is an urgent need to develop ecological knowledge to support conservation and management of the protected area network within the Serra do Mar biogeographical region. Results of previous research showed a high variation in the abundance of large mammals within large protected areas of Atlantic Forest within this region that is comparable if not greater than that found in fragmented Atlantic Forest ecosystems. This thesis shows how environmental variables and anthropogenic impacts influence large mammals within the remaining continuous Atlantic Forest remnants of the Serra do Mar. Continuous Atlantic Forest remnants are highly heterogeneous and this heterogeneity results from environmental factors and human impacts. The results presented in the four chapters of this thesis highlight that, although the Atlantic Forest is the most intensely studied biome for mammals in Brazil, the knowledge necessary for the conservation and effective management of this class in the Atlantic Forest is still lacking. Additional studies are needed to allow the establishment of zones within protected areas as defined by Brazilian legislation (Law: 9.985/2000 (SNUC)). Gradientes ambientais e sua influência na variação da estrutura da vegetação no Parque Botânico do Morro do Baú - SC

Defesa: Curitiba, 23/02/2017 / Inclui referências : f. 91-109 / Resumo: O presente estudo teve como objetivo analisar a variação estrutural das sinúsias epifítica e arbórea ao longo de um gradiente altitudinal no Morro do Baú, Ilhota, SC, buscando correlações com as variações pedológicas (árvores) e atributos dos forófitos (epífitos). Instalamos 150 parcelas (100 m2) distribuídas equitativamente nas altitudes de 340 m (Área 1), 540 m (Área 2) e em 810 m s.n.m. (Área 3). Coletamos dados para cálculos fitossociológicos dos espécimes arbóreos com PAP ? 15 cm. Coletamos dados de solos em 28 parcelas (10 na Área 1, 10 na Área 2 e oito na Área 3). Abrimos dois perfis por patamar altimétrico, um em cada parcela, e coletamos amostras complementares de solos nas demais parcelas. Selecionamos 30 forófitos distribuídos equitativamente nos três pisos altitudinais para amostrar a sinúsia epifítica. Registramos, em cada forófito, todas as espécies epifíticas, bem como algumas características dos forófitos. Construímos modelos de regressão múltipla utilizando a estrutura das duas sinúsias (eixos da NMDS) e, no caso da epifítica, também a riqueza, como variáveis resposta. Utilizamos os atributos pedológicos como variáveis preditoras da estrutura da sinúsia fanerofítica e os dados dos forófitos para a sinúsia epifítica. Adicionamos MEMs (Moran Eigenvector Maps) aos modelos a fim de isolar o efeito da dependência espacial e testamos as suas premissas. Dentre as árvores, amostramos 3.470 indivíduos, pertencentes a 183 espécies. A riqueza decresceu conforme o aumento da altitude, tendo sido registradas 125, 103 e 47 espécies, respectivamente, nas Áreas 1, 2 e 3. Apenas nove espécies foram comuns às três Áreas. Detectamos uma forte diferença na composição da flora entre os extremos do gradiente explicados pela DCA e pela NMDS. Identificamos forte contraste também nos solos, havendo nas Áreas 1 e 2 Cambissolo Háplico e na Área 3 Espodossolo Humilúvico. Os melhores preditores da estrutura da sinúsia fanerofítica dentre os descritores pedológicos foram pH e carbono, além de dois MEMs. Dentre os epífitos, registramos 168 espécies, sendo 126 na Área 1, 120 na Área 2 e 57 na Área 3, sendo esta última Área significativamente distinta das demais. Os modelos de regressão apontaram altura do fuste e altura da copa relacionados com a riqueza e estrutura epifítica, embora também a idade da árvore figure como preditor no modelo de estrutura. Quando analisadas apenas as Áreas 1 e 2, a riqueza se relacionou com a idade dos forófitos. A seleção dos descritores pH e carbono não devem ter relação causal com a variação da vegetação arbórea e a natureza abrupta da variação deve estar relacionada com a atuação conjunta de fatores como altitude, clima, geologia e geomorfologia que afetam simultaneamente a vegetação e a formação dos solos. Dentre os preditores da riqueza e estrutura da sinúsia epifítica, além da seleção daqueles associados ao tamanho do forófito, a idade do forófito parece ser um atributo muito importante, mas pouco discutido na literatura. Idade do forófito. / Abstract: Environmental gradients and their influence on the variation of the vegetation structure in "Morro do Baú" Botanical Park - SC. This study aims to analyze the structural variation from epiphytic and arboreal sinusiae along an altitudinal gradient in Morro do Baú, Ilhota municipality (Santa Catarina state, Southern Brazil), looking for correlations with the pedological variations (trees) and phorophytes attributes (epiphytes). We installed 150 plots (100 m2) evenly distributed in altitudes of 340 m (Area 1), 540 m (Area 2) and 810 m (Area 3). We collected data for phytosociological description from arboreal species with DBH ? 15 cm. We collected soil samples only in 28 plots (10 in Area 1, 10 in Area 2 and eight in Area 3). We digged two trenches for soil profiles description per altimetric level and collected additional soil samples in remaining 22 plots. We selected 30 phorophytes evenly distributed in the three altitudinal levels to sample the epiphytic sinusiae. We registered, in each phorophyte, all the epiphytic species, as well as some characteristics from the phorophytes. We constructed multiple regression models using the structure from the two sinusiae (NMDS axes) and also the richness (in the case of the epiphytes) as response variables. We used the pedological attributes as predictors variables of arboreal sinusiae structure and the phorophytes descriptors for the epiphytic sinusiae. We added MEMs (Moran's Eigenvector Maps) to the models to isolate the effect of spatial dependence and, finally, tested the premises of the model. Among the trees, we sampled 3.470 individuals, belonging to 183 species. The richness decreased as the altitude increased, registering 125, 103 and 47 species in Areas 1, 2 and 3, respectively. Only nine species are shared between the three Areas. We detected a strong difference in flora composition between the two extremes of the gradients as defined by the DCA and NMDS. We also identified a strong contrast in soils characteristics predominating Inceptisols in Area 1 and 2, and Histosols in Area 3. The best structure's predictors from fanerophytic sinusiae were pH and carbon, plus two MEMs. Among the epiphytes, we registered 168 species: 126 in Area 1, 120 in Area 2 and, 57 in Area 3, being the last one significantly different from others. The regression models showed that trunk height and crown height are related to richness and epiphytic structure, although age of the tree also figures as predictor in the structure model. When we analyze only Areas 1 and 2, richness was related with forophytes' age. The selected soil descriptors (pH and carbon) should have no causal relation with the arboreal vegetation's variation and the abrupt nature of the variation should be related to the joint action of factors as altitude, climate, geology and geomorphology which affect simultaneously vegetation and soils formation. Among the predictors of richness and epiphytic sinusiae structure, besides the selection of the ones associated to the phorophyte's size, phorophyte's age seems to be a very important attribute, but little discussed in literature.
