Alternativas de controle, acúmulo de chiquimato e curva de crescimento de capim-amargoso (Digitaria insularis) suscetível e resistente ao glyphosate / Alternatives of control, shikimic acid accumulation and growth curves of sourgrass (Digitaria insularis) susceptible and resistant to glyphosate

O glyphosate vem sendo aplicado na agricultura como principal ferramenta de manejo de plantas daninhas, sendo o mesmo aplicado em alguns sistemas de forma repetitiva ao longo do ano. Esta prática contribuiu para a seleção de espécies de plantas daninhas resistentes, sendo que no Brasil mais recentemente tem sido obervado populações resistente de capim-amargoso (Digitaria insularis) ao glyphosate. Sendo assim, são necessárias pesquisas com esta planta daninha sobre alternativas de controle, caracterização biológica dos biótipos relacionadas a adaptabilidade e crescimento, e caracterização dos níveis de resistência, para que baseado nestas informações as recomendações de manejo destes biótipos sejam tomadas. Portanto, esta pesquisa foi desenvolvida com os objetivos de avaliar alternativas químicas de manejo dos biótipos resistentes, avaliar os níveis de resistência, e o crescimento diferencial entre os biótipos. Para o desenvolvimento dos experimentos foram coletadas sementes do biótipo resistente no município de Matão - SP, onde foi aplicado glyphosate de 1.440 g e.a ha-1 antes do florescimento, sendo que as sementes do biótipo suscetível foram obtidas no município de Piracicaba SP, onde não apresentava histórico de aplicação de glyphosate. Na primeira etapa da pesquisa foram realizados experimentos para determinação do fator de resistência ao glyphosate, através de curvas de dose resposta em quatro estádios de desenvolvimento do capimamargoso, além de um bioensaio de discos foliares para analisar o acúmulo de chiquimato. Na segunda etapa foi conduzido um ensaio a campo no município de Matão SP, visando encontrar herbicidas alternativos para controle do capim-amargoso resistente ao glyphosate. Por fim um estudo de curvas de crescimento foi conduzido para avaliar o crescimento comparativo dos biótipos resistente e suscetível ao glyphosate, analisando área foliar, massa seca da parte aérea e das raízes. Através do modelo de curva dose-resposta do programa estatístico R, o fator de resistência (GR 50) variou de 3 a 4 quando comparando o biótipo resistente ao suscetível. As alternativas de controle viáveis foram glyphosate em associação com clethodim e paraquat + diuron; glyphosate com clethodim e amônio-glufosinato; glyphosate e haloxyfop-methyl; glyphosate e fenoxaprop-p-ethyl + clethodim; glyphosate e tepraloxydim. O biótipo resistente teve um desenvolvimento mais rápido, todavia não foi possível inferir sobre sua capacidade competitiva. / Glyphosate has been used in agriculture as the main tool for weed management, and it has been applied in some systems repeatedly throughout the year, which contributed to the selection of resistant weed species. More recently in Brazil glyphosate resistant populations of sourgrass(Digitaria insularis) have been observed. So, researches are needed on alternative control, biological characterization of the biotypes and related growth, and characterization of the resistance levels. Based on this information it is possible to recommend management practices. Therefore, this research was developed with the objective of evaluating alternative chemical management for the resistant biotypes, assess levels of resistance, and the differential growth between biotypes. For the development of the experiments seeds from resistant plants were collected in Matao County SP - Brazil, where glyphosate was applied at 1440 g. a. e. ha-1, before flowering, and the seeds of the susceptible biotype were obtained in Piracicaba SP - Brazil, where there was no history of glyphosate application. In the first stage of the research experiments were performed to determine the resistance factor to glyphosate by dose response curves in four growth stages of sourgrass, and a leaf disc bioassay to analyze the accumulation of shikimate. The second step was to conduct a trial in Matao - SP to find alternative herbicides to control glyphosate resistant sourgrass. Finally an growth curves experiment was conducted to evaluate the comparative growth of glyphosate resistant and susceptible biotypes, analyzing leaf area, dry mass of shoots and roots. The resistance factor (GR 50) ranged from 3 to 4 when comparing the susceptible biotype with the resistant. The alternative herbicides in combination with glyphosate were clethodim and paraquat + diuron; clethodim and ammonium-glufosinate; haloxyfop-methyl; fenoxaprop-p-ethyl + clethodim; tepraloxydim. The resistant biotype had a more rapid development, however it was not possible to infer about their ability to compete.

Démographie et réponses adaptatives des populations végétales aux changements environnementaux / Demography and adaptive responses of plant populations to environmental changes

La région méditerranéenne, hot-spot de biodiversité avec un fort taux d'endémisme, est classée parmi les zones les plus touchées par le changement climatique. La conservation des espèces nécessite de comprendre finement leur démographie face à ces changements, mais aussi leur capacité à s’adapter à ces nouvelles conditions. Au cours de cette thèse, deux espèces végétales rares méditerranéennes ont été étudiées, Centaurea corymbosa et Brassica insularis, en utilisant des modèles mathématiques récents pour analyser des suivis démographiques de long-terme (22 ans et 18 ans). Dans la première partie de cette thèse, des modèles de projection matriciels ont été construits afin d’analyser la variation spatio-temporelle des taux d’accroissement des populations de C. corymbosa. Cela a permis d'identifier les facteurs climatiques clefs qui impactent les taux d’accroissement des populations, et plus finement, les paramètres de survie, de floraison et de fécondité. Une analyse de viabilité des populations a été réalisée sous différents scénarios climatiques. Dans un second temps, une analyse de la capacité des populations de C. corymbosa à répondre à ces changements climatiques a été effectuée avec un modèle de projection intégral. Les changements des traits d’histoire de vie au cours du temps ont montré que la stratégie de floraison observée répond aux variations climatiques en suivant la même direction que la stratégie optimale. Ceci montre que les populations sont capables de s’adapter au changement climatique. Enfin, des modèles de capture-recapture (CR) ont été construits afin de déterminer l’influence de la faible détectabilité des individus sur l’estimation des paramètres démographiques chez B. insularis. Les probabilités de survie estimées par l’approche classique sont inférieures à celles estimées par les modèles CR. Les paramètres estimés par les deux approches sont en général très corrélés, à l'exception d'une population montrant des problèmes d'identification des individus. L'ensemble de cette thèse est replacé dans le cadre de la biologie de la conservation des espèces végétales, notamment en suggérant de prendre en compte leur capacité de réponse au changement climatique. / Mediterranean region is a biodiversity hot-spot with a high endemism rate and is classified among the areas most sensitive to climate change. Deep understanding of demography and evolution following these changes is a necessity for species conservation. During this thesis, two rare Mediterranean plant species have been studied, Centaurea corymbosa and Brassica insularis, using recent mathematical models to analyse long term demographic datasets (22 and 18 years). In the first part of this thesis, matrix projection models have been used to analyse spatio-temporal variation in population growth rate in C. corymbosa. Major climatic parameters that impact population growth rate and survival, flowering and fecundity have been identified. A population viability analysis has been performed under various climatic scenarios. In a second part, an analysis of the population ability of C. corymbosa to response to these climatic changes has been performed using integral projection models. Temporal variation of life-history traits showed that the realized flowering strategy varied with climatic conditions following the same direction than the optimal flowering strategy. It can thus be concluded that the studied populations can adapt to climatic changes. Finally, capture-recapture models have been applied on the Brassica insularis dataset to analyse the effect of plant detectability on demographic parameter estimations. Survival probabilities obtained with the classical approach are lower than the ones estimated using the CR models. However, the parameters estimated using both methods are highly correlated, except for one population where individual identification is problematic. Results of this thesis are linked to some considerations in conservation biology of plants, particularly in link to their ability to respond to climatic changes.

Em serpentes, como em outros ectotérmicos, o controle da temperatura corpórea (Tc) é dependente da utilização de fontes externas de calor e principalmente de ajustes comportamentais, embora ajustes fisiológicos e morfológicos também ocorram. Em geral, esse controle é alcançado por meio da escolha de ambientes quentes ou frios, cuja disponibilidade pode variar ao longo do dia e das estações do ano. A termorregulação, portanto, constitui aspecto central da biologia das serpentes, as quais dedicam porção considerável de seu tempo e energia a essa atividade. Os objetivos do presente estudo foram documentar a variação sazonal e circadiana da temperatura corpórea da jararaca-ilhoa, Bothrops insularis, em seu ambiente natural, a Ilha da Queimada Grande (IQG), a fim de prover análise descritiva da Tc e do comportamento termorregulatório destes animais à luz dos custos e benefícios da regulação da Tc na região tropical; e entender como estes parâmetros são ditados/influenciados por fatores ambientais (temperatura local e de microhábitats, umidade relativa, precipitação, radiação solar, vento, características do substrato) e bióticos (tamanho dos indivíduos, sexo, estágio reprodutivo, estado digestivo). (1) As jararacas-ilhoa são usualmente termoconformadoras, embora consigam atingir a temperatura corpórea preferida em todas as estações do ano; (2) as temperaturas ambientais indicam que a IQG é um ambiente termicamente favorável para essas serpentes; (3) a Tc destes animais sofre forte influência da variação sazonal e circadiana da temperatura ambientel... / Given the pervasive effects of temperature on most biological processes, studies focusing on thermoregulation are pivotal to the understanding of ectotherm’s vertebrate ecology. In snakes, like other ectotherms, body temperature (Tb) control is dependent of external heat sources and based mainly on behavioral adjustments, although physiological and/or morphological adjustments can also occur. In general, such control is achieved through the choice of warm or cold environments, whose availability may vary along the days and/or seasons. Thus, thermoregulation can be regarded as a central aspect of snake biology, and these animals invest considerable time and energy into this activity. The aims of the present study were to document the seasonal and circadian variation in the Tb’s of the golden lancehead, Bothrops insularis, in its natural habitat, the Queimada Grande Island (QGI). We then used such database to provide a descriptive analysis of Tb variation and thermoregulatory behavior for this species in the light of the costs-benefits theory applied to a tropical region. Our second goal was to understand how body temperature is dictated/influenced by environmental (local and microhabitat temperatures, relative humidity, precipitation, solar radiation, wind, substrate characteristics) and biotic factors (individual size, sex, reproductive stage, digestive state). The main findings of the present study were: (1) Golden lanceheads can be regarded as being thermoconformers, although they do reach their preferred body temperature in all seasons; (2) environmental temperatures indicate that QGI has a high thermal quality, i.e., it is thermically suitable for the snakes; (3) Tb is strongly influenced by seasonal and diel variation of the environmental temperature; (4) B. insularis selected... The utility of standardized DNA markers in species delineation and inference of the evolutionary history of symbiotic relationships in the Malagasy ant Melissotarsus insularis Santschi, 1911 and its scale associate (Diaspididae)

A subset of 199 Melissotarsus insularis and 130 Diaspididae specimens were analyzed to 1) determine the species status of M. insularis and 2) to explore the relative intimacy of the relationship between M. insularis and Diaspididae. An analysis of molecular variance and the observed lack of association between clades and distinct habitats on the M. insularis phylogeny suggested that while M. insularis exhibits isolation by distance, it does not apparently diversify by habitat. When cryptic COI pseudogenes were accounted for, the majority of the genetic diversity exhibited by M. insularis was limited to a divergence of 3% or less suggesting that M. insularis represents a single, albeit broadly distributed, species. A cophylogenetic reconstruction of the relationship between M. insularis and Diaspididae yielded 14 “cospeciation” events but was not significant unlike reconstructions of host-parasite relationships.
