This thesis is focused on ethical aspects of interaction between players and virtual computer game worlds, specifically on reception of situations where player faces possibility or necessity to make a moral choice that influences the future development of the game. The goal of this thesis is to find out how seriously players approach these decisions, whether they take their impact on the game world into account, what criteria they apply when deciding and what other influences affect their decisions. Another related theme is identification with the game avatar and its influence on moral decisions. This work is based on qualitative research realized with a group of five players of computer game Mass Effect 2. During the research they answered questions regarding their decisions in particular situations as well as their general approach to decisions in the game. The research is based on recent works on the related topics of game studies. Outcomes of the research are analysed by the grounded theory method. Using its procedures the answers are decomposed into basic concepts which are then put together again in mutual context. As a conclusion of the thesis the grounded theory formulated based on these analytical procedures is presented. Future thematically related research areas are also suggested.

Objectives: The aim of the thesis was to carry out a research focused on drug interactions of selected groups of cardiovascular drugs and to describe their management in clinical practice, which could be used during dispensing medicine. Mikroevoluční procesy a meziploidní interakce v sympatrických populacích více cytotypů / Microevolutionary processes and inter-cytotype interactions in mixed-ploidy populations

[Abstract] This thesis is aimed at better understanding of cytotype co-existence in mixed- ploidy populations with an emphasis on a microevolutionary processes behind it. Our past knowledge was based on a few thoroughly investigated model taxa like Chamerion angustifolium and Heuchera grossulariifolia, but some generalizations seem to be premature in the light of new findings. A detailed research of other taxa included in the thesis showed that polyploid complexes can vary dramatically in their ability to cope with the co-existence of cytotypes in mixed-ploidy popu- lations. Whereas mixed-ploidy populations are virtually lacking in some species (an example being Vicia cracca, Paper III.), ploidy-heterogeneous populations are very common in others, maintained by free mating interactions and the absence of reproductive isolation among cytotypes (e.g. Pilosella echioides, Paper II.). The strenght and cumulative effect of various breeding barriers (both pre- or post- zygotic) govern the position of a particular multi-ploidy complex between these two extremes and co-determine the type of cytotype co-existence in its mixed- ploidy populations. The paleoecology of plant - arthropod herbivory associations constitute very important source of knowledge about the phylogeny and co-evolution of both groups. The traces of herbivory interactions between plants and arthropods on the fossil leaves are preserved as so called damaged types (DTs) clustered into distinct functional feeding groups (FFGs). The diversity and frequency of these damage traces also seem to have been strongly influenced by environmental and climatic conditions. This research has been focused on rich fossil plant assemblages from the area of the Most Basin in the north-western Bohemia. The undergoing work has comprised the diagnosis of the individual damages on the basis of their specific morphological traits as their number, size, shape and distributional pattern on the leaf surface. The next issue has involved the statistical analyses concerning the differences in the frequency and diversity of the types of damage and functional feeding groups between two separate fossiliferous layers, namely those of the stratigraphically older Bílina Delta and younger Břešťany Clay. The aim of this work was to synthesize and study acid-base and coordination properties of ligands bearing a neutral or negative charge on their pendant arms. Acid-base properties of two ligands and thermodynamic stability of their complexes with gallium, copper, and zinc ions were studied using potentiometry, UV-Vis and NMR. Solid state structure of one of the ligands and its complex with copper ion was studied using RTG structural analysis. Structure of complexes in solution was studied using methods of computational chemistry.

RNA polymerase (RNAP) is a key multi-subunit enzyme of gene expression that, together with the σ factor, forms a holoenzyme and transcribes genetic information from DNA to RNA. RNAP from Bacillus subtilis and its primary factor σA were studied in this thesis. The σA factor determines the specificity for the promoters to which the holoenzyme binds. Part of its structure is domain 1.1, which is likely to prevent binding of σA to the promoter by itself (unless it is part of the holoenzyme) by binding to domains 2 and 4. The first part of the thesis verifies the hypothesis that domain 1.1 binds domains 2 and 4 and thus prevents binding of σA to the promoter. To this end, various domain constructs have been created and their interactions have been tested. Domain interaction was tested by Nitrocellulose Filter Binding Assay, EMSA, and in vitro transcription. The results did not show significant interaction between domains. The second part of the thesis deals with the creation of a tool for the study of the enzymatology of RNAP from B. subtilis - recombinant RNAP (rRNAP). First, a plasmid construct for expression of rRNAP in Escherichia coli was constructed by a series of cloning steps, followed by protein isolation and characterization. Společenstvo fytofilního bentosu v rybnících / Community of phytophilous benthos in fishponds

Community of phytophilous macrozoobenthos (PM) represents an important component of fishponds ecosystems concerning biomass as well as essential part of food webs. There are evident linkages between PM and water macrophytes. Their influence on PM is the main topic of many scientific papers which state three main water macrophyte characters with the strongest impact on PM - coverage, biomass and morphological complexity. The main aim of this diploma theses was to analyse linkages between four different water macrophyte species and PM in eight different fishponds devided into two groups according their localisation- třeboňské a rakouské. Water macrophyte species have any influence on PM abundance in both fishpond groups. Water macrophyte species have significant influence on biomass of PM in třeboňské and rakouské fishponds analysed together and also in both fishpond groups analysed separately. PM biomass was significantly higher in Glyceria maxima during first sampling period in both fishpond groups. Water macrophyte species have significant influence on PM diversity in třeboňské and rakouské fishponds analysed together and in rakouské fishponds group analysed separately. Just one variable, water macrophyte species, was analysed in my diploma thesis, but some others abiotic and biotic variables influence phytophilous macrozoobenthos community in fishponds littoral zones as well. That´s why conclusions of this thesis should be interpreted circumspectly.

The thesis concerns about the relationship between teacher and student at lower-secondary school. To be more precise, it focuses on student's self-perception and perception of a teacher; also it focuses on teacher's self-perception and perception of his/her students, all of it in the context of teacher/student interaction. The theoretical part covers student's and teacher's personality, their relationship and selected dimensions of their communication verbal correctness, sense of humour, personality and relationships and competencies. In question of dimension, there were specified so called observed spheres, which became important for determination of interactive perception of educational-process participants (e. g. "verbal correctness" dimension includes the grammatical correctness). The empirical part was realized by quantitative research represented by four questionnaires: (1) student's reflection (student evaluates behaviour of a specific teacher), (2) student's self-reflection (student evaluates his/her behaviour during classes of a specific teacher), (3) teacher's reflection (teacher evaluates the students of a specific class), (4) teacher's self-reflection (teacher evaluates his/her behaviour during specific classes). The research took place on three basic schools and two grammar schools. 16 teachers and 224 students were involved in the research. The acquired data enabled to create three outlines of results. First outline are "overall results and results obtained in a specific type of school" on basis of comparison reflections and self-reflections of teachers and students. Second outline is presented by "specific results", which came out by comparison of reflections and self-reflections of specific teachers and students of particular classes throughout all variations of questionnaire (e. g. identical and non-identical interactive perception) or just selected variations of questionnaire. Third outline is represented by "another results", these include analysis of teachers and students reflections and self-reflections according to a particular criterion (e. g. gender).
