Although their identification and documentation received a considerable attention, they remain unrecognized for many patients. Objectives: The goal of the thesis was to analyze serious drug interactions between cardiovascular drugs, to describe their management in clinical practice, which is useful during dispensation, and to evaluate occurrence of these drug interaction in a community pharmacy. Methodology: The information was gained from several databases and compared, and afterwards management of major drug interactions of betablockers and calcium channel blockers was created. Patients of a community pharmacy were screened for these drug interactions. The data were processed by frequency analysis. Results: In the theoretical part, 94 drug interactions were analysed. Title: Analysis of Didactic Interaction in Group Skiing Lessons for Adults Objectives: The aim of this thesis is to perform detailed analysis and comparison of the didactic teacher - student interaction during adult skiing lessons between two instructors with the same level of experience but different educational background. Methods: In our research we have employed a participant observation method using classification into a category system based on a standardised MADI (Modified Analysis of Didactic Interaction) technique. This includes 7 categories describing the forms of the teacher's behaviour, the didactic content of the lessons, didactic situations, and attitudinal characteristics. Results: The results of the study confirmed, among others, a higher incidence of the two most effective forms of teacher behaviour (correction and feedback information provision) in the instructor with pedagogical education, which was associated with a higher quality of the teaching process. Thesis Abstract This thesis has been performed under co-tutelle supervision between Charles University in Prague (Czech Republic) and Strasbourg University (France). It also profited from the background and cooperation of Institute of Inorganic Chemistry Academy of Science of the Czech Republic and French Commission for Atomic and Alternative energies (CEA Cadarache, France). Results of the work contribute to the OECD/NEA project Serena 2 (Program on Steam Explosion Resolution for Nuclear Applications). Presented thesis can be classed in the scientific field of nuclear safety and material science. It is aimed on the so-called "molten nuclear Fuel - Coolant Interaction" (FCI) that belongs among the recent issues of the nuclear reactor severe accident R&D. During the nuclear reactor melt down accident the melted reactor load can interact with the coolant (light water). This interaction can be located inside the vessel or outside in the case of vessel break-up. Hlavní strukturní protein myšího polyomaviru: interakce s buněčnými strukturami / Major capsid protein of mouse polyomavirus: interaction with cellular structures

Mouse polyomavirus (MPyV) is small non-enveloped DNA virus. Although this virus has been studied for almost 60 years, it still remains unclear, how can virus transport its genetic information to the cell nucleus. Also, the mechanism of virion morphogenesis is not well understood. First part of this work is focused on endocytic pathway which is used by MPyV for trafficking toward the cell nucleus. Using dominant negative mutant of caveolin-1 we showed that caveolin-1dependent endocytic pathway, described for SV40, is not used by MPyV for productive infection. MPyV is transported to early endosomes. Acidic milieu of endosomes is indispensable for productive infection. Preventing virus localisation into early endosomes (dominant negative mutant of Rab 5 GTPase) or endosomes alkalisation (by ammonium chloride or bafilomycin A1) led to dramatic decrease of virus infectivity. Alkalisation of endosomes entailed retention of MPyV in early endosomes. It indicates that virus is further transported to late endosomes. Finally, we confirmed by FRET that MPyV is in perinuclear space localized into recycling endosomes. Another poor characterized process is virion morphogenesis. Title: Didactic interaction between teacher and child in preparatory swimming lessons. Objects: The object of this thesis was to describe and analyze the interaction profiles of two kindergarten teachers. Both teachers have many years of teaching experience of preparatory swimming lessons with children 3 to 4 years old. At the same time the aim was to contribute to the objectification of ideas about the specific didactic interaction in the physical learning in the water. Methods: The method of participant observation of two kindergarten teachers with many years of teaching experience was used at this thesis. The data collection was realized by taking a video. Processing and evaluating of the video followed using a modified analysis of didactic interaction method. The method of questioning was also used for the observed teachers. Results: According to our results the most common form of behavior of two selected teachers during the lessons was instruction. The most common of one of our selected teacher was using notification. The submitted thesis is concerned with the research of individual distinctions of sociability of pupils through analysis of their social relations. The theoretical part deals with findings on general range of the concept of sociability, defines interindividual relations as a theoretical and empirical resource of the sociability research, characterizes the developmental period of early adolescence. The further part of the thesis briefly describes sociability of pupils from the view of basic psychical components. It is connected to socialization and social relations at school. The empirical part represents quantitative research, where first of all "corelation" (relationship) between individual distinctions of sociability of pupils and extraversion was found out. Futher, it describes to what extent the ability of sociability varies in relation to gender, how social characteristics between genders are different.

Mass spectrometry (MS) is a rapid, specific and very sensitive analytical method with a broad spectrum of proteomic applications such as protein identification and sequencing, 3D protein structure characterization or study of protein-protein interaction. The introduction of two ionization techniques in late 1980's that are able to ionize the large biomolecules such as proteins, oligosaccharides or nucleic acids with no or low fragmentation has started the rapidly expanding field of MS-based proteomics. The presented thesis was aimed at the application of mass spectrometric approaches to answer several proteomic questions. Firstly we have employed the chemical cross-linking in combination with MS analysis to solve the 3D structure and protein-protein interactions of three model systems: (1) homodimeric human regulatory protein 14-3-3, (2) model of 14-3-3 and regulatory domain of tyrosine hydroxylase, and (3) system of two membrane proteins, cytochrome P450 2B4 and cytochrome b5, involved in xenobiotics biotransformation. This approach works in aqueous solutions under physiological conditions and thus preserves native structure of the investigated proteins. Title: Interpersonal communication of Holmes Place company Objectives: The main objective of this work is to observe errors in interpersonal communication at Holmes Place, to find the cause of these errors and propose effective solutions on how to avoid it and improve communication within the company. Methods: At this work I used the method of interview and questionnaire survey. The goal was to determine the sample surveyed, to create questionnaires and overall, to evaluate the results. I used interview method to obtain information about the possibilities and traffic of interpersonal communication within the company. Within respondent sample, I evaluate the level of interpersonal communication at Holmes Place. Results: I have found that interpersonal communication at Holmes Place is on a bad level. Correctly answered questions are on a very low-level - only slightly above the 50%. Regardless of senior management, middle management or subordinates, communication within the company is evaluated as insufficient. This thesis deals with the relationship between family and kindergarten. It is mainly focused on communication, through which there is a mutual interaction and relationship builded between teachers and parents. In the communication both actors must strive for the best possible cooperation, since only in this case they can reach their common goal, which is the smooth development and overall development of the child's personality. The theoretical part defines the concept of communication. It analyzes current family and nursery schools as an institution and deals with the relations between them. It also mentions the conflict and some types of barriers that can lead to difficulties in the interaction. The practical part is based on research done through questionnaires which was aimed to teachers and parents. The result presents their subjective evaluation of communication.
