The protosolar nebula heritage : the nitrogen isotopic ratio from interstellar clouds to planetary systems / Le patrimoine de la nébuleuse protosolaire : le rapport isotopique de l'azote des nuages interstellaires à des systèmes planétaires

L'existence de molécules interstellaires soulève une question, ces molécules sont-elles les mêmes molécules que nous voyons dans le système Solaire aujourd'hui ? C'est une question toujours ouverte qui implique des conséquences profondes. Il est possible d’éclaircir cette question en étant capables de retracer l'héritage d'un groupe de molécules chimiquement liées, ce que nous appelons un réservoir. Le meilleur outil pour retracer l'héritage des réservoirs sont les rapports isotopiques. L'élément qui montre les plus grandes variations du rapport isotopique dans le système Solaire est l'azote. Ces variations indiquent que le rapport isotopique de l'azote est sensible aux conditions physiques de la formation des étoiles.L'objectif principal de cette thèse est d'identifier les réservoirs d'azote à différents étapes de la formation des étoiles et des planètes. La première étape de cette entreprise était d'identifier le rapport isotopique de la masse principal d'azote du milieu interstellaire local aujourd'hui.Cela a été déterminé égale à 323 ± 30 à partir du rapport CN/C 15 N mesuré dans le disque protoplanétaire autour de TW Hya. Parallèlement à cela, nous avons également mesuré un rapport HCN/HC 15 N=128 ± 36 dans le disque protoplanétaire autour de MWC 480. Ces rapports isotopiques très distinctes mesurées sur les disques protoplanétaires sont une indication claire de la présence d'au moins deux réservoirs d'azote dans les disques protoplanétaires. La façon dont ces réservoirs se séparent est cependant inconnue. Cela pourrait peut-être se produire en raison de réactions de fractionnement chimique ayant lieu dans les cœurs prestellaires. Nous avions donc comme objectif d'obtenir une mesure précise et directe du rapport isotopique de l'azote des molécules d'HCN dans le cœur prestellaire L1498.Pour obtenir cette mesure, l'obstacle le plus important à surmonter était due aux anomalies hyperfines des molécules d'HCN. Ces anomalies hyperfines sont induites par le chevauchement des composants hyperfins. Ceci sont particulièrement sensibles à la densité de colonne d'HCN, mais aussi au champ de vitesses et aux largeurs de raies. Ainsi les anomalies hyperfines sont un outil de mesure de l'abondance d'HCN permettant aussi de sonder la cinématique des cœurs prestellaires.Pour reproduire avec précision les anomalies hyperfines, et ainsi mesurer des densités de colonne précises d'HCN, nous avions besoin d'explorer un espace de paramètres dégénéré de 15 dimensions. Pour minimiser les dégénérescences nous avons obtenu un profil de densité basé sur des cartes du continuum de L1498. Ceci permettant de réduire à 12 dimensions l'espace des paramètres. L'exploration de cet espace de paramètre a été fait grâce à l'utilisation d'un méthode de minimisation MCMC. Grâce à cette exploration, nous avons obtenu HCN/HC 15 N = 338 ± 28 et HCN/H 13 CN = 45 ± 3. Les incertitudes sur ces valeurs sont limités par les erreurs de calibration et sont dé-terminés de manière non arbitraire par le méthode MCMC. Les implications de ces résultats sont discutées dans le chapitre de conclusion,où nous présentons également quelques perspectives sur l'avenir. / The existence of interstellar molecules raises the question, are thesemolecules the same molecules we see on the Solar system today? Thisis still an open question with far reaching consequences. Some lightmay be shed on this issue if we are able to trace the heritage of agroup of chemically linked molecules, a so-called reservoir. The besttool to trace the heritage of reservoirs are isotopic ratios. The elementthat shows the largest isotopic ratio variations in the Solar system isnitrogen. For this is an indication that the isotopic ratio of nitrogen issensitive to the physical conditions during star formation.The main objective of this thesis is to identify the reservoirs of ni-trogen at different stages of star and planet formation. The first stepin this endeavour was to identify the isotopic ratio of the bulk of ni-trogen in the local ISM today. This was determined to be 323 ± 30from the CN/C 15 N ratio in the protoplanetary disk around TW Hya.Along with it we also measured the HCN/HC 15 N= 128 ± 36 in theprotoplanetary disk around MWC 480. This very distinct nitrogen iso-topic ratios on protoplanetary disks are a clear indication that thereare at least two reservoirs of nitrogen in protoplanetary disks. Howthese reservoirs get separated is however unknown. This could pos-sibly happen due to chemical fractionation reactions taking place inprestellar cores. We therefore aimed to obtain an accurate direct mea-surement of the nitrogen isotopic ratio of HCN in the prestellar coreL1498.To obtain this measurement the most important hurdle to overcomewere the hyperfine anomalies of HCN. These hyperfine anomaliesarise due to the overlap of hyperfine components. They are especiallysensitive to the column density of HCN, but also to the velocity fieldand line widths. Thus hyperfine anomalies are a tool to measure theabundance of HCN and to probe the kinematics of prestellar cores.To accurately reproduce the hyperfine anomalies, and thus mea-sure accurate column densities for HCN, we needed to explore adegenerate parameter space of 15 dimensions. To minimise the de-generacies we have derived a density profile based on continuummaps of L1498. This reduced the parameter space to 12 dimensions.The exploration of this parameter space was done through the useof a MCMC minimisation method. Through this exploration we ob-tained HCN/HC 15 N = 338 ± 28 and HCN/H 13 CN = 45 ± 3. Theuncertainties on these values are calibration limited and determinednon-arbitrarily by the MCMC method. Implications of these resultsare discussed in the concluding chapter, where we also present somefuture perspectives.

Desempenho produtivo e composição isotópica de 13C e 15N no tecido muscular de tilápia do Nilo (Oreochromis niloticus) alimentada com diferentes fontes protéicas / Performance and isotopic composition of 13C and 15N in muscle tissue of Nile tilapia (Oreochromis niloticus) fed with different protein

Fontes alternativas de alimentos vem sendo estudadas em substituição à farinha de peixe na aquicultura, podendo acarretar diferença na incorporação de carbono e nitrogênio decorrentes da disponibilidade e do valor nutricional da fonte. Três experimentos tiveram o objetivo de quantificar a participação das diferentes fontes protéicas nos tecidos de juvenis e pós-larvas através da composição isotópica de C e N e sua influência no crescimento. Além disso, a digestibilidade aparente das dietas contendo mistura de plantas C3 e C4 foi estudada usando o 13C nas fezes como traçador. As dietas consistiram de quatro ingredientes protéicos. Farelo de soja FS, farelo de coco FC, farinha de vísceras de aves FVA e farinha de peixe FP que compuseram cinco combinações diferentes: FS100; FS60FC40; FS60FVA40; FS60FP40; FS19F10FP10FVA11. No primeiro experimento duzentos juvenis de tilápia com peso médio de 3,4 ± 0,20 g foram distribuídos em 20 gaiolas flutuantes e após serem alimentados com as dietas experimentais, foram coletados amostras aos 0, 22, 44, 66 e 88 dias para estimar a contribuição de C e N das dietas no tecido. Foi observada diferença significativa (P<0,05) entre peso final (PF), ganho em peso (GP), conversão alimentar (CA), rendimento de filé (RFL), taxa de eficiência protéica (TEP) e fator de condição (FC). No entanto, não observou diferença entre o consumo alimentar (CO), peso do filé (PF) e índice de perfil (IPERF). Para o experimento dois, trezentas pós-larvas de 20 dias com peso médio inicial de 0,07 ± 0,020 g, foram distribuídas aleatoriamente, perfazendo cada unidade experimental com quinze animais. Foram realizadas coletas aos 0, 6, 12, 18, 24 e 30 dias para C e N. Neste experimento foi demonstrada diferença significativa (P<0,05) entre as variáveis PF, GP, CO, CA e TEP. Para o comprimento total e padrão; IPERF e FC n(Pão foi observado diferença (P>0,05). Juvenis e pós-larvas atingiram o equilíbrio isotópico de 13C e 15N com suas respectivas dietas, permitindo assim que estes sejam usados para estimar a quantificação de nitrogênio e carbono, com excessão para a dieta FS60FC40. Em ambos os experimentos, houve aumento percentual de ganho de peso em juvenis e pós-larvas, sendo observada maior porcentagem de nitrogênio no músculo e carcaça total pelas dietas com farinha de peixe FS60FP40 e farinha de vísceras de aves FS60FVA40. Posteriormente, para o experimento de digestibilidade, 2500 juvenis com peso médio de 5,0 ± 1,0 g foram utilizados para determinação do coeficiente de digestibilidade aparente CDA,% de componentes de origem C3 e C4 contidos nas dietas experimentais. Os valores de CDA apresentaram valores inferiores para os componentes de origem C3 em relação aos de origem C4. A avaliação dos isótopos estáveis do C e do N nas dietas permitiu estimatimar a contribuição entre as diferentes fontes, o que sugeriu que a ingestão de nutrientes está relacionados com a qualidade da dieta. Verificou-se que fontes de proteína, quando combinadas, podem apresentar resultados superiores do que quando usada sozinha na composição. / Alternative sources of food have been studied in substitution for fishmeal in aquaculture, and it may result in a difference in incorporation of carbon and nitrogen, depending on the nutritional value and availability of the source. Two experiments were designed to quantify the participation of different protein sources in the muscle tissue in juvenile and post larvae through isotopic composition and its influence on growth and body composition. In addition, the apparent digestibility of feedingstuffs containing mixture of C3 and C4 plants has been studied using 13C as tracer. The experiments were conducted in a random design with five treatments and four repetitions. Diets consisted of by four sources of protein ingredients. soybean meal SBM, coconut meal CM, poultry product meal PPM, meal and fish meal FM, composed five different combinations of diets: SBM100 SBM60CM40 SBM60PPM40 SBM60FM40 SBM19CM10FM10PPM11. In the first experiment, it used 200 juveniles with an average weight of 3.4 ± 0.20 g, distributed in 20 vinyl cages. Fish collections were held after being fed the experimental diets for 0, 22, 28, 38, 44, 66 and 88 days. In this assay, it was observed significant difference (P<0.05) for final weight, weight gain, feed conversion, fillet yield, protein efficiency ratio and condition factorHowever, there was no difference between food consumption, weight and content of the fillet profile. For the experiment two, 300 post-larvae, 20 days old and with average weight starting at 0.07 ± 0.020g, were randomly distributed in the five treatments, making each experimental unit with twenty animals. Six fish collections were held at 0, 6, 12, 18, 24 and 30 days. This assay also showed significant difference (P<0.05) for final weight, weight gain, food intake, feed conversion and protein efficiency ratio. It was observed that juvenile and post larvae did reach the isotopic equilibrium of 13C and 15N with their own diets, thus allowing it to be used for estimating the quantification of nitrogen and carbon incorporated into fish tissue, with exception for the SBM60CM40 diet. The higher percentage of nitrogen in the muscle and carcass were obtained by total diets with fish meal SBM60FM40 and poultry product meal SBM60PPM40To test the digestibility of the diets, 2,500 juvenile with average weight 5.0 ± 1.0 g were used for determination of apparent digestibility coefficient ADC, % of source components C3 and C4 contained in the experimental diets. The values of CDA constituents of experimental diets of fingerling showed lower values for source components C3 in respect of C4 origin. The evaluation of stable isotopes of carbon and nitrogen contained in protein ingredients permitted the estimation of contributions among the different sources, which suggests that the intakes of nutrients are related to the higher quality of the diet. It was noted that protein sources when combined can present superior results when used alone in the composition. The contribution of different protein sources in feed of Nile Tilapia, through the natural variation of stable isotopes of carbon and nitrogen, proved promising.

Desenvolvimento de metodologias utilizadas nas áreas de salvaguardas e forense nuclear baseadas na técnica LA-HR-ICP-MS / Development of methodologies used in Nuclear Safeguards and Nuclear Forensic based on LA-HR-ICP-MS technique

A amostragem ambiental por esfregaço ou swipe samples é uma metodologia que vem sendo empregada rotineiramente pela Agência Internacional de Energia Atômica (AIEA) para verificar se os Estados signatários do Acordo de Salvaguardas estão realizando atividades nucleares não declaradas. A análise dessas amostras ambientais é complementar aos procedimentos de salvaguardas convencionais para a verificação das informações cedidas pelos Estados. Neste trabalho, foi descrita uma metodologia que visa a fortalecer os processos de salvaguarda nuclear e da ciência forense nuclear no país. A proposta é estudar e avaliar a técnica de ablação à laser acoplada a um espectrômetro de massa de alta resolução com fonte de plasma indutivamente acoplado (LA-HR-ICP-MS Laser Ablation High Resolution Inductively Coupled Plasma Mass Spectrometry), como técnica alternativa para a análise das amostras de esfregaço. Adicionalmente, foram empregadas outras técnicas complementares (atividade radioativa, microscopia eletrônica de varredura e espectroscopia de energia dispersiva) com o intuito de garantir maior segurança ao processo de inspeção de salvaguardas e investigações forenses nucleares. A precisão, medida através do desvio padrão relativo (DPR),dos resultados obtidos para as razões isotópicas 234U/238U, 235U/238U e 236U/238U do padrão analisado (CRM -125A) para a medida do fator de discriminação de massa foram, respectivamente, 1,3 %, 0,2 % e 7,6 %. Já as incertezas percentuais (u %), que também contemplam o DPR das medições, variaram de 3,5 % a 29,8 % para as medições da razão isotópica 235U/238U e de 16,6 % a 42,9 % para a razão isotópica 234U/238U. Esses valores mostraram-se compatíveis com outros estudos que utilizaram a mesma técnica para análise de amostras reais coletadas em uma instalação nuclear. As amostras coletadas apresentaram nível de enriquecimento que variou de (2,3 ± 0,7) % (amostra 3 corredor de acesso à instalação) a (17,3 ± 2,8) % (amostra 18 bancada de recuperação de UF4). A partir da coleta de amostras nesses diversos pontos, desde a entrada da instalação até a bancada em que se manuseia o urânio enriquecido, foi possível detectar diversos níveis de enriquecimento no isótopo 235U. / Environmental sampling performed by means of swipe samples is a methodology frequently employed by International Atomic Energy Agency (IAEA) to verify if the signatory States of the Safeguards Agreements are conducing unauthorized activities. Swipe samples analysis is complementary to the Safeguards ordinary procedures used to verify the information given by the States. In this work it was described a methodology intending to strengthen the nuclear safeguards and nuclear forensics procedures. The proposal is to study and evaluate the laser ablation high resolution inductively coupled plasma mass spectrometry (LA-HR-ICP-MS) technique as an alternative to analyze the real-life swipe samples. The precision achieved through the standard(CRM -125A)measurements, represented by the relative standard deviation (RSD), was respectively 1.3 %, 0.2 % e 7.6 %for the 234U/238U, 235U/238U e 236U/238Uisotopes ratios. The percent uncertainties (u %), which covers the RSD, ranged from 3.5 % to 29.8 % to the 235U/238U measurements and from 16.6 % to 42.9 % to the 234U/238U isotope ratio. These results were compatible with former studies performed by the LA-HR-ICP-MS that analyzed real-life swipe samples collected at a nuclear facility. Swipe samples collected from several points of the nuclear facility presented enrichment level ranging from (2.3 ± 0.7) % (sample 3) to (17.3 ± 2.8) % (sample18). They also allowed detecting different enrichment levels within the facility.

O objetivo do presente estudo foi avaliar, através da análise de n-alcanos e de suas razões isotópicas, a influência da abertura do canal artificial do Valo Grande (VG) nas fontes de matéria orgânica (OM) do Sistema Estuarino-Lagunar de Cananéia-Iguape (CIELS). Os compostos foram analisados em quatro testemunhos, 22 sedimentos superficiais e em folhas de nove espécies de plantas dominantes na região de estudo. Os parâmetros avaliados nas folhas amostradas permitiram separar as espécies estudadas em monocotiledôneas e dicotiledôneas, e em vegetação lenhosa de mangue e de restinga. Esta separação foi útil na identificação das fontes de OM do CIELS. Os dados dos sedimentos superficiais mostraram que a principal fonte de OM no sistema é a vegetação de restinga. Esta se dá principalmente na região norte, onde está presente o VG. A influência fitoplanctônica, das macrófitas emersas e da vegetação de mangue é maior na região sul. Os dados dos testemunhos mostraram que a abertura do VG aumentou a contribuição da vegetação de restinga e das macrófitas aquáticas na OM presente ao longo do CIELS, diminuindo a influência fitoplanctônica e das macrófitas emersas. Assim, a qualidade e quantidade de OM do CIELS foram influenciadas pela presença do VG. / This study aimed to verify, through the use of n-alkanes and their isotopic composition, the influence of the Valo Grande artificial channel (VG) on organic matter (OM) sources of the Cananéia-Iguape Estuarine-Lagoon System (CIELS). The compounds were analyzed in four cores, 22 surface samples and nine dominants plants species leaves sampled on the studied area. The parameters evaluated on the leaves permitted to group the studied species in mono and dicotyledonous, and in mangrove and resting vegetation. This was useful to identify the OM sources on the CIELS. The surface sediment data showed that the CIELS OM is mainly composed by restinga vegetation. This occurs mainly on the northern region, where the VG is present. The influence of phytoplankton, of emergent macrophytes and of mangrove trees is higher on the southern region. The cores data showed that the VG opening increased the influences of Atlantic Forest and aquatic macrophytes on the CIELS OM, decreasing the emergent macrophytes and the phytoplankton influence. So, the type and the amount of OM on the CIELS are influenced by the VG presence.

Développement d'une plateforme analytique jetable basée sur l'isochophorèse pour la séparation et la caractérisation isotopique des lanthanides / Development of a micro total analytic system based on isotachophoresis for the separation and characterization of lanthanides

La caractérisation juste et reproductible en isotopie et en concentration des radioéléments est l’une des thématiques essentielles des laboratoires d’analyse dans le domaine du nucléaire. Afin de minimiser les temps de manipulation en boite à gants des personnels et la production de déchets radioactifs liés à l’analyse de combustibles nucléaires, il est nécessaire de proposer des solutions efficaces et innovantes. Depuis quelques années, la miniaturisation des systèmes séparatifs constitue l’un des axes de développement majeurs de la chimie analytique et ces microsystèmes constituent certainement une des solutions pour répondre aux exigences de l’analyse nucléaire. Ce travail a pour objectif la conception d’une plateforme analytique miniaturisée et à usage unique, dédiée a la séparation des lanthanides, issus des combustibles usés, en amont de leur analyse par spectrométrie de masse. Destiné à remplacer une étape de séparation chromatographique au centre d’un processus analytique de trois étapes, le nouveau protocole basé sur l’isotachophorèse (ITP) doit satisfaire un cahier des charges précis. Les propriétés de complexation des lanthanides ont d’abord été exploitées afin d’obtenir avec un agent chélatant unique et rigoureusement sélectionné, l’acide 2-hydroxy-2-methylbutyrique (HMBA), la sélectivité intra période nécessaire à leur séparation complète par ITP. Basées sur des modèles théoriques existants, des études complémentaires, notamment des paramètres influençant la résolution, ont permis l’amélioration des performances globales du système ainsi que son dimensionnement. Pour réduire drastiquement le volume de déchets liquides secondaires (solutions de rinçage) et la manipulation de matériaux et de matériels radioactifs, le protocole a été implanté sur un microsystème polymérique jetable en COC, spécialement développé pour cette application. Ce microsystème a ensuite été couplé à un spectromètre de masse a multi collection et source à plasma à couplage inductif pour mesurer les rapports isotopiques / The accurate and reproducible characterization of radioactive solutions in isotope composition and concentration is an essential topic for analytical laboratories in the nuclear field. In order to reduce manipulation time in glove box and production of contaminated wastes, it is necessary to propose innovative and efficient solutions for these analyses. Since few years, microchips are a major field of development in analytical chemistry and those devices could provide a solution which fits the needs of nuclear industry. The aim of this work is to design a disposable analytical micro-device devoted to lanthanide separation from spent nuclear fuel before their analysis in mass spectrometry. Designed to be used in place of a separation process by liquid chromatography which is involved in a three step protocol, the new protocol based on isotachophoresis (ITP) keeps compatible with the other two steps. The complete separation of lanthanides by ITP was obtained by the use of only one chelating compound rigorously selected: the 2-hydroxy 2-methyl butyric acid (HMBA). The main parameters involved in solute resolution were defined from the theoretical models of ITP and experimental studies of the influence of these parameters allowed to optimize the geometry of the system and to improve its performances.
