Izolace DNA ze sýrů pro použití v polymerázové řetězové reakci / DNA extraction from cheeses for polymerase chain reaction analysis

This work was focused on DNA isolation from cheeses for the use in polymerase chain reaction. First, there was optimised the procedure of homogenisation of different types of cheeses from commercial sources, cell lysis and DNA isolation. DNA was isolated using magnetic microspheres and phenol extraction. It was shown that the DNA was amplified in PCR for domain Bacteria after dilution. Next, there was optimised the procedure of DNA isolation from fresh cheeses and from contaminated fresh cheeses and their pickles. DNA from all samples was amplified in PCR. The presence of DNA of domain Bacteria and yeast DNA was demonstrated. In the last part of the work, there were optimised the preparation of PCR mixtures and bacterial DNA amplification in PCR with primers with clamp (F357-GC and R518). Synthetized PCR products were analysed using DGGE. It was shown that amplicons of DNA isolated from cheeses and pickles differ in positions and numbers. Larger number of bands of different intensities was detected after amplification of DNA isolated from contaminated pickles.

The master's thesis is dedicated to the development of thermally insulating fittings for brick construction with integrated thermal insulation. In the theoretical part of the thesis are summarized legislative regulations for building construction, the technical requirements for insulation and ceramic products. Furthermore is here described the production of ceramics and technology of filling the ceramic fittings with interated insulation. The practical part of the thesis deals with new possibilities of filling the bulk ceramic fittings with thermal insulating materials based on polyurethane foams in order to improve their thermal insulation and mechanical properties. In the conclusion of this master thesis is an overall evaluation of taken measurements, the filling technology and comparison of new fittings with the current market and their potential use.

Probiotics products are an integral part of the current market. Products containing probiotics cultures are also cosmetic products. The first part of the study focuses on testing of bacterial survival abilities in the environment of preservatives presented in cosmetic products. Collection strains of genus Lactobacillus were used for these tests. Another part of the study focuses on isolation of bacterial DNA from probiotic cosmetic products Ryor, Yoghurt of Bulgaria, FeminaMed and Lactovit Activit in PCR-ready quality. DNA was isolated by fenol extraction and with magnetic particles. Presence of bacteria was proved by genus and species specific PCRs Lactobacillus. Species specific PCR for identification of Lactobacillus pentosus was optimalized. Species identification was in accord with data declared by producers.

With a development of molecular biology methods it is an increasing interest in new procedures of DNA isolation of high quality. DNA isolation is performed on crude cell lysates by many techniques e.g. phenol extraction, salting out or adsorption on solid phase. Classical DNA isolation, such as phenol extraction is quite complicated and time consuming. New alternative methods of DNA isolation was development using reverse immobilizing DNA to a solid phase. Widespread is the use of the magnetic particles as carriers, which allow the isolation of DNA in high quality directly from crude cells lysates of complex samples. The current method of DNA adsorption onto the surface of magnetic particles does not provided sufficiently pure DNA for analysis of some comlex samples (e.g. food). Some inhibitors of the polymerase chain reaction (PCR) are apparently adsorbed onto the tube wall and the next step of DNA elution leads to their release into the solution and cpnsequent negative effect on quality of DNA (e.g. decreasing of PCR amplification). The principle of the developed procedure is design a device, which utilizes transfer of magnetic particles by paramagnetic newddle from one to another Eppendorf tube, in which further processing of the sample extends. Transfer of magnetic particles with DNA using needle prevents transmission of contaminating impurities. The proposed device allows to realize above-mentioned procedure. The functionality of the device being tested in the isolation of plasmid pUC19 DNA from crude lysates of E. Coli JM 109 (pUC19).

This thesis deals with biting midges of the genus Culicoides, which are tiny nematoceran insects belonging to the Diptera, and their parasites. Biting midges partake in transmissions of several parasitical organisms of various groups. Most notably, they are the vectors of several pathogenic viruses which might have a serious impact on livestock. However, the thesis deals with detection of parasites belonging to Trypanosomatids and Filarioids related to two independent biting midges collections - from the Czech Republic and from the Central African region. Apart from testing biting midges, there were carried out the tests on the occurrence of the same group of parasites within ruminant hoofed games in the Czech Republic. Our goal was to find out whether the same parasites occur with biting midges and their hosts (hoofed games) and whether biting midges could play a role as vectors. Biting midges are relatively overlooked group of haematophagous insects. Until recently, they had not been paid much attention, which changed with the spread of Bluetongue virus over Europe. This stimulated a widespread monitoring of biting midges in several European countries, including the Czech Republic. Based on the selected criteria, a suitable contractor will be selected and the production and assembly procedures of the selected company will be thoroughly described. In the practical part the project documentation will be prepared according to the assignment using the knowledge from the previous part of the final thesis. The scope and content of the project documentation will be made in the construction phase and will meet all required requirements.

Cytochrome b5 is a small heme protein. There are three isoforms present in human organism, one is located in the membrane of endoplasmic reticulum (microsomal cyt b5), second in the outer mitochondrial membrane and the third (soluble form) was found in cytoplasm of matured erythrocytes. The main role of cytochrome b5 is to transport single electron in various reactions including cytochrome P450-dependent reactions. First aim of the thesis was to prepare and to isolate the soluble form of rabbit microsomal cytochrome b5, using heterologous expression in Escherichia coli strain BL-21 Gold. The plasmid pET22b containing synthetic gene for rabbit microsomal cytochrome b5, lacking the sequence encoding the membrane associated C-terminal domain, was used as an expression vector. The second aim was to synthesize the expression vectors carrying genes for human microsomal and erythrocytic cytochromes b5. These genes were prepared by gene synthesis, ligated to cloning vector pUC19, amplified in E. coli DH5α competent cells and their sequences were verified by DNA sequencing. Consequently the pET22b expression vectors containing genes for human microsomal and erythrocytic cytochrome b5 were constructed and finally their suitability for heterologous expression was evaluated. The thesis topic is The needs of women suffering from feelings of loneliness. The author focuses on the work of women who suffer from feelings of loneliness. It can be both a woman living alone, and also women who live with her husband or children, but feels lonely. My work is aimed to map their needs from the perspective of social work. The thesis is normally divided into theoretical and research, practical. The theoretical part is based on basic human needs such as the need of belonging, the need for integration, the need for association, the need for integration and the need for interpersonal relationships. An important chapter of the theoretical part are sections covering old age and aging, as an important factor that can increase the risk of feeling lonely. The practical part of the thesis focuses on mapping the current supply and state entities and facilities, services, self-help groups that focus on providing assistance and support to single women. The main goal of this work is to analyze and define the needs of single women in terms of social work. Outcome of this work should be to design specific programs that are aimed directly at women. It should be a support or self-help groups where women could share their feelings and to share them with someone who can understand them.
