Stanovení tepelné ztráty novostavby domu z přírodních materiálů

Aim of the practical part is to evaluate a real project of a family house assigned by investors in terms of heat loss properties. The ecological trace connected with the heat insulation quality is being determined. There are three options: The first option consists of a house with no outer heat insulation system. The second house is insulated with straw bales. For the third option, a system with mineral wool is used. The main goal is to show that straw bale insulation is not any worse than broadly used materials, and it is also a much better option from the ecological perspective. In theoretical part, there is an explanation of common rules to follow when using straw bales for construction. Also some common prejudices connected with this topic are explained.

In the text part of this thesis is described procedure of designing the house, materials, which are used in this house, composition and construction of the building. The practical part consist of the draft of the strawbale house, drawings, documentation and other parts regarding about the design of the house.

Lucie Cahlíková, Ph.D. Title of Doctoral Thesis: Alkaloids of Zephyranthes citrina (Amaryllidaceae): isolation, structure identification, biological activity. Key words: Amaryllidaceae alkaloids, Zephyranthes, Alzheimer's disease, AChE, BuChE, POP, oncological diseases Zephyranthes citrina Baker was chosen, based on result of previous screening study and literatury research, for detailed phytochemical work for the purpose of isolation of the widest range of AAs. From 35 kg of fresh bulbs was obtained 151 g of purified alkaloidal extract, which was processed using column chromatography to almost 700 fractions. These fractions were connected into 27 subfractions, which were processed by liquid-liquid extraction, flash chromatography, preparative TLC and crystallization. Finally, 27 pure alkaloids were isolated. All compounds were identified by MS (EI, ESI), HRMS, 1D- and 2D-NMR, CD, optical rotation and by comparison with literature data. Lucie Cahlíková, Ph.D. Title of Doctoral Thesis: Alkaloids of the genus Hippeastrum (Amaryllidaceae): isolation, identification, biological activity. Hippeastrum x hybridum cv. Ferrari was chosen based on results of previous screening studies for detailed phytochemical study for the purpose of isolation of the widest range of AAs. From 25 kg of fresh bulbs was obtained 29,46 g of purified alkaloidal extract, which was processed using column chromatography. Finally, 18 pure alkaloids were isolated including one new homolycorine-type alkaloid (9-O-demethyllycorenine). All compounds were identified by spectrometric techniques (GC-MS, ESI-MS, NMR, optical rotation) and by comparison with literature data. All alkaloids, isolated in sufficient amount, were tested for their biological activities associated with Alzheimer's disease (inhibition of hAChE, hBuChE, and POP), and cytotoxic activity. The inhibitory activity against human cholinesterases was determined in vitro by a modified Ellmanʼs spectrophotometric method. Pancreatic cancer (PC) is extremely severe malignant disease with a five-year survival of less than 5%. Currently there is no reliable tool for the diagnosis of PC in its early stages. At the time of clinical symptoms most patients are in an advanced stage of the disease and the treatment does not usually have a significant effect. For these reasons emphasis is gradually shifting to the search for the suitable molecular markers for improvement of the diagnosis and assessment of the survival prognosis with respect to a possibility of surgical treatment. MiRNA represent one of the most promising markers, although, their examination in pancreatic tissue is a complicated process. One of the reasons is the very small amount of the source material coming from a fine needle biopsy. A second cause of problems is the subtle character of the pancreatic tissue resulting in significantly lower yields of molecular genetic analysis when compared to other epithelial tissues. An additional negative factor is heterogeneity of the tissue resulting in disproportionate representation of tumor cells within the sample. A suitable choice of procedures for isolation of nucleic acids (NA) and subsequent analysis including quantification of tumor cells is critical for accurate evaluation of the miRNA levels. This theses deals with a structural design comparison of heat insulations implementation in flat roofs in light of heat engineering. It also adverts to possible structural design in terms of heat insulations distribution in a flat roof. There are many materials options. I show the possibilities of material solution and also advantages and disadvantages of the single sealing materials at these layers. In the aplication part of this thesis I deal with possible concepts of insulation and solution for a specific roof of an older apartment house in Brno, where I conducted a constructive exploration. There was developed and evaluated a few options of solutions considering the heat insulator used. The following computational programs were used for the evaluations: TEPLO, AREA, CUBE 3D.

The increasing quantity of waste represents serious environmental, social and economic problem. Wastes produced from industry and households. A large part of the economy transforms a certain amount of raw materials to waste in their production, because it is currently emphasis is placed on finding sustainable sources of raw materials. One of the suitable secondary raw materials can be discarded textiles or waste from the textile industry. These kinds of textile waste often ends up in landfills or incinerators, so it is from an ecological and environmentally advantageous to their further use. The diploma thesis deals with the study and development of thermal and acoustic insulation materials based on textile fibers. It focuses on the legislative requirements for waste management for the sorting of textile wastes and their subsequent modification before reuse. Further it contains kinds of textile fibers and bonding technologies thermal and acoustic insulation materials. Finally, an evaluation of thermal insulation, acoustic and mechanical properties of fibrous insulation materials.

The thesis solves the project of a kindergarten in the field of inclination 7%. The kindergarten is located on the plots number 570, st. 373, st. 374 in the village Vrané nad Vltavou. The building has two floors and one basement. The kindergarten is for 60 children and for 11 employees. The building serves as a preschool institution for raising children. The supporting structure consists of a precast concrete frame supplemented in the basement by a cast-in-place wall and a foundation slap. External walls and partitions form sand-lime bricks Sendwich. The external walls are completed with Orsilt TF Profi mineral insulation th. 280 mm. The floors are made of prestressed concrete floor slabs Spiroll PPD 256 th. 250 mm supported by T and L primary beams. The building ends with a could extensive green roof. The slope and space for the blow-in insulation Climatizer Plus uses Steico Joist 400 I-beams. Windows are wooden frame Optiwin Purista and doors are Optiwin Entrada.
