Eddy-covariance carbon balance, photosynthetic capacity and vegetation indices in a harvested boreal jack pine stand

Eddy-covariance (EC) CO₂ flux data were analysed and annual carbon (C) balances estimated for a four-year period (2004-2007) following clearcut harvesting of a boreal jack pine stand in northern Saskatchewan. The site was a source of C to the atmosphere for all years, with annual net ecosystem productivity (NEP) increasing from -153 g C m⁻² yr⁻¹ in 2004 to -63 g C m⁻² yr⁻¹ in 2007. This increase was mainly due to gross primary productivity (GPP) increasing significantly from 78 to 200 g C m⁻² yr⁻¹ , while ecosystem respiration (R) increased only slightly from 231 to 263 g C m⁻² yr⁻¹ over the same period. In the 2006 growing season (GS), a field campaign was conducted to investigate the relationships between monthly destructive measurements of leaf area index (LAI) and daily measurements of the normalized difference vegetation index (NDVI) and photosynthetic capacity (Amax). The latter was derived from 5-day, 16-day, 30-day and annual Michaelis-Menten light response analyses using daytime measurements of NEP and incident photosynthetically active radiation. Digital-camera data were used to evaluate the potential of using the rectilinear-lens vegetation index (RLVI) as a surrogate for NDVI of a young forest stand. Results showed that LAI was linearly related to NDVI and RLVI, which was largely the result of changes in the deciduous vegetation component across the GS. These results indicate that RLVI could be used as a surrogate for NDVI up to a GS maximum LAI of 0.91 m2 m⁻² observed in 2006. Measured mean (± 1 S.D.) GS LAI was 0.67 (± 0.24) m² m⁻² in 2006. LAI accounted for the majority of the variability in Amax at the 30-day time scale, while at shorter time scales air temperature was the dominant control. For 2004 to 2007, mean spring estimates of LAI were 0.25, 0.29, 0.38 (compared to 0.40 m² m⁻² from measurements) and 0.41 m² m⁻², respectively. Results suggest that a steady increase in the jack pine LAI component accounted for the annual increases in GPP and hence NEP over the four years.

Non-linear analysis of jack-up structures subjected to random waves

There is a steadily increasing demand for the use of jack-up units in deeper water and harsher environments. Confidence in their use in these environments requires jack-up analysis techniques to reflect accurately the physical processes occurring. This thesis is concerned with the models appropriate for the dynamic assessment of jack-ups, an important issue in long-term reliability considerations. The motivation is to achieve a balanced approach in considering the non-linearities in the structure, foundations and wave loading. A work hardening plasticity model is outlined for the combined vertical, moment and horizontal loading of spudcan footings on dense sand. Empirical expressions for the yield surface in combined load space and a flow rule for prediction of footing displacements during yield are given. Theoretical lower bound bearing capacity factors for conical footings in sand have been derived and are used in a strain-hardening law to define the variation in size of the yield surface with the plastic component of vertical penetration. The complete incremental numerical model has been implemented into a plane frame analysis program named JAKUP. The spectral content of wave loading is considered using NewWave theory, and the importance of random wave histories shown by constraining the deterministic NewWave into a completely random surface elevation. Using this technique, a method for determining short-term extreme response statistics for a sea-state is demonstrated. A numerical experiment on an example jack-up and central North Sea location is shown to emphasise the difference in long-term extreme response according to various footing assumptions. The role of sea-state severity in the variation of short-term extreme response statistics is also highlighted. Finally, probabilistic methods are used to develop further understanding of the response behaviour of jack-ups. A sensitivity study of influential variables (with probabilistic formulations as opposed to deterministic values) has been conducted using the response surface methodology.

Development of non-linear numerical models appropriate for the analysis of jack-up units

Jack-up units have considerable economic significance because they are used to carry out a large proportion of the world's oil and gas exploration in water depths less than 90.0m. Due to the increase in use of jack-ups in harsher environments, analysis techniques assuming quasi-static and linear structural behaviour have had to be reassessed. This thesis is concerned with non-linear dynamic analysis methods appropriate for a jack-up assessment. Jack-up modelling requires realistic representation of the structure, the foundations and the environmental loading, together with the implementation of appropriate dynamic analysis algorithms. Techniques for each of these aspects of jack-up analysis are reviewed and the implementation of several of the methods in an advanced plane frame analysis program called JAKUP is described. Geometric non-linearity in the structure and work hardening plasticity at the foundations are accounted for in the program. Test cases are presented to verify the implementation of the methods and then some illustrative plane frame quasi-static and dynamic analyses are described. These simple models highlight the importance of accounting for dynamic motions in a jack-up analysis. For the quasi-static analyses, the assumption of pinned footing behaviour is seen to always result in the most conservative displacement and moment predictions. However, the analyses show that this is not always the case when dynamic amplification is accounted for.

Response of Pinus banksiana (Lamb.) families to a global change environment

We examined how fast- and slow-growing families (based on height at 10 years) of Pinus banksiana Lamb. are affected by a climate altered by CO$ sb2$ during their first growing season. Our primary objective was to evaluate the possibility that genotypes performing best under present conditions may not necessarily do best under projected warmer climate. Seedlings were grown for six months in two climatic environments (350 $ mu$L/L CO$ sb2$ x present temperatures and 700 $ mu$L/L CO$ sb2$ x 4$ sp circ$C warmer temperatures) and with 100 ppm and 5 ppm nitrogen. / The CO$ sb2$T$ sp circ$ environment had a significant effect on most biomass components of seedlings and water-use efficiency but not on height and other growth variables. The nitrogen fertilization was generally the most significant effect of the treatments for most growth variables. / All the families responded in a similar way to variations in the growing environments except for WUE. Family differences were more important for measurements of height and growth variables than for biomass components. The architecture of seedlings was also highly variable between families. Norm of reaction graphs were built for several growth variables to outline which families were overall most successful in an enriched CO$ sb2$T$ sp circ$ environment. Of the 15 families studied, four of them were classified as most successful in a projected high CO$ sb2$T$ sp circ$ climate.

Comportamento do peptídeo entomotóxico jaburetox-2Ec em solução e a sua interação com lipossomas miméticos de plaquetas humanas

No presente trabalho, o peptídeo inseticida recombinante Jaburetox-2Ec, derivado da urease de Canavalia ensiformis (Jack bean) foi estudado, principalmente através de técnicas de espectroscopia de espalhamento de luz e raios-X. Foi analisada a tendência do peptídeo a se agregar em diferentes condições físico-químicas quando em solução aquosa, e a sua interação com membranas de lipossomas miméticos de plaquetas humanas. A partir dos dados de raio hidrodinâmico, raio de giro, e massa molar ponderal média obtidos do peptídeo em solução, foi possível observar uma tendência para maior agregação numa faixa em torno de pH 5,5, bem como ausência de agregação em pH inferior a 4,0 e superior a 7,0. Não se verificou nenhum efeito visível no nível de agregação com a adição de agente redutor ou íons, sendo que a exposição ao oxigênio do ar levou a um aumento da mesma. Os estudos de interação do Jaburetox-2Ec com lipossomas revelaram uma significante mudança estrutural na membrana, que se mostrou mais intensa com uma maior concentração do peptídeo. Foram, através de software, feitos ajustes teóricos das curvas de espalhamento de raios-X à baixos ângulos dos lipossomas, a fim de quantificar as mudanças nos parâmetros físicos das membranas causados pela ação do peptídeo. A formação de poros na membrana pelo peptídeo foi proposta como explicação para os resultados encontrados. / In the present work, the insecticide recombinant peptide Jaburetox-2Ec, derived from Canavalia ensiformis (Jack bean) urease, was studied, primarily by light and X-ray scattering techniques. Its tendency to aggregate in aqueous solution under different physical-chemical conditions, and its interaction with human platelet mimetic liposome membranes were analyzed. From the data relative to hydrodynamic radii, radii of gyration and mean molecular weight obtained from the peptide in aqueous solution, it was possible to observe a tendency for greater aggregation around pH 5.5, as well as an absence of aggregation at values of pH below 4.0 and above 7.0. No visible effect on the level of aggregation was verified with the addition of reducing agent or ions, whereas the exposure to oxygen in the air resulted in an increase in this level. The studies of Jaburetox-2Ec interaction with liposomes revealed a significant structural change in the membrane, which was shown to be more intense in higher peptide concentrations. Software-assisted theoretical fits were made for the experimental small-angle X-ray scattering curves aiming to quantify the changes in the physical parameters of the membranes caused by peptide action. The formation of pores in the membrane was proposed as an explanation to the results found.

The goal of the present study was to evaluate the effect of inoculation with the fungus Rhizophagus clarus and the addition of grape bagasse vermicompost on phytoremediation by Canavalia ensiformis of a sandy soil with high Cu concentration. Methods: Soil was contaminated with 100 mg Cu kg-1, fertilized with five levels of vermicompost, and cultivated with C. ensiformis with and without inoculation with Rhizophagus clarus. Availability of Cu and other nutrients in the soil and in the soil solution, shoot and root accumulation of Cu and other nutrients, plant growth, and Cu phytotoxicity using photochemical efficiency, concentration of photosynthetic pigments, and oxidative stress enzyme activities as indicators of Cu phytotoxicity were evaluated. Results: Phytostabilization showed better performance with the addition of the vermicompost level equivalent to 20 mg P kg-1 and in the presence of R. clarus. Phytoextraction was higher with the addition of the vermicompost level equivalent to 40 mg P kg-1 and without R. clarus inoculation. However, C. ensiformis was not a good phytoextractor. Conclusions: The system C. ensiformis vermicompost R. clarus exhibited potential for Cu phytostabilization in sandy soils. / Introdução e objetivos: Fungo micorrízico arbuscular e vermicomposto podem reduzir os efeitos deletéricos do cobre às plantas. O objetivo do trabalho foi avaliar o efeito da inoculação do fungo Rhizophagus clarus e da adição do vermicomposto de bagaço de uva na fitorremediação por Canavalia ensiformis de um solo arenoso com alto teor de Cu. Métodos: Solo foi contaminado com 100 mg Cu kg-1 e adubado com cinco doses de vermicomposto para o cultivo de C. ensiformis, com e sem a inoculação com Rhizophagus clarus. Foram avaliados a disponibilidade de Cu e outros nutrientes no solo e em solução, o acúmulo de Cu e dos outros nutrientes na parte aérea e nas raízes, o crescimento vegetal e a fitotoxicidade do Cu, através da eficiência fotoquímica, da concentração de pigmentos fotossintéticos e da atividade de enzimas do estresse oxidativo. Resultados: A fitoestabilização ocorre melhor na dose de vermicomposto equivalente a 20 mg P kg-1 e na presença do R. clarus. A fitoextração é maior na dose de vermicomposto equivalente a 40 mg P kg-1 e sem a inoculação do R. clarus, mas C. ensiformis não se apresenta como uma boa planta fitoextratora. Conclusões: O sistema C. ensiformis- vermicomposto- R.clarus tem potencial de aplicação na fitoestabilização de Cu em solos arenosos.
