This master's thesis deals with integrated Hall-effect current sensors. The thesis contains summary of manufacturers of current sensors suitable for low current measurement and comparison of their important parameters. Next objective of the thesis is a design of test board used for evaluation of temperature depedent accuracy of several sensors simultaneously. The device can be connected with PC through USB interface for saving measured values or automated control with other instruments.

RNDr. František Chmelík, CSc., Department of Physics of Materials Abstract: Plastic deformation of micron-scale crystalline materials differs con- siderably from bulk specimens, as it is characterized by random strain bursts. Three categories of metallic samples were investigated in this thesis: micron- scale copper micropillars with varied geometries, submillimeter-scale aluminum microwires, and aluminum and aluminum-magnesium salt-replicated foams. Very precise fabrication methods and sensitive measurement set-ups consisting of uni- axial compression and tensile tests with concurrent acoustic emission (AE) record- ing were developed. These fine methods allowed for investigations of effects re- lated to plastic deformation at micrometer scales, i.e. the dislocation dynamics associated with the stress drops. Size effects in plastic deformation, as well as clear correlations between the stress drops and the AE events, were found in mi- crosamples, confirming that dislocation avalanches are indeed responsible for the stochastic character of deformation processes also at microscales. This diploma thesis deals with the partial analysis of two teaching materials which are used while teaching German as a foreign language. These materials are Themen aktuell 1 and Direkt neu 1, which are frequently utilized at secondary schools for the teaching of German at the language level A1-A2. The analysis of these teaching materials concentrates on the presentation of the grammar curriculum. The theoretical part of this diploma thesis deals with the teaching methods, current trends and also with the role of the teaching material in teaching. In the practical part of this diploma thesis the analysis of the presentation and the practice of the selected grammatical phenomena and also the classification of the exercises, which are found in the teaching materials, is carried out. The practical part also presents the results of the test, which was carried out on the basis of selected grammatical phenomena at Akademické gymnázium, school of the capital city Prague and Biskupské gymnázium J. N. Neumanna in České Budějovice. This thesis aims to explore the effect of in situ ion gun etching on magnetic proper- ties, such as perpendicular magnetic anisotropy, of Pt/Co/Pt multilayers. Perpendicular magnetic anisotropy is of interest due to its importance in spintronic application. Calibration spectroscopic ellipsometry measurements of Pt and Co layers to obtain sputter yields of respective magnetron targets were performed as a first step. After that, ten Pt/Co/Pt multilayers with different Co and top Pt layer thicknesses were prepared. Five of these samples were etched by the ion gun, mounted inside of the magnetron chamber, during different stages of the deposition. To characterize the prepared samples, experimental and modeled Kerr rotation spectra were confronted. This provided an insight into the cross-sections of the investigated samples. Magnetic hysteresis loops measured by vibrating sample magnetometer and Kerr rotation setup demonstrated higher coercive field and change of the slope of the loop induced by ion etching. When compared to the obtained cross-sections, a correlation between the change of magnetic loops and intermixing of Pt and Co at the interface was visible. This intermixing was higher in samples treated by the ion gun. 1

This work studies various electrical properties of polymer layers of polyaniline, polythiophenes and poly(((benzothiadiazol)4,7-di(thien-2-yl)-5,6-difluoro-2,1,3-benzothiadiazol). Goal of this work is to determine electrical and photoelectrical properties as are electric conductivity, mobility of the free charge carriers, work function of the electrons and diffusion length of excitons. Conductivity was obtained through current and voltage contacts, mobility was measured by CELIV method (charge extraction by linearly increasing voltage). To determine work function a Kelvin probe was built and utilized. Diffusion length of excitons was determined from photovoltage spectrum formed by illuminating space charge region of the polymer.Space charge region on surface of the polymer can arise spontaneously or by laying suitable contact electrode. Studies of Retroviral Reverse Transcriptase and Flaviviral Protease Enzymes as Antiviral Drug Targets : Applications in Antiviral Drug Discovery & Therapy

Viruses are a major threat to humans due to their unique adaptability, evolvability and  capability to control their hosts as parasites and genetic elements. HIV/AIDS is the third largest cause of death by infectious diseases in the world, and drug resistance due to the viral mutations is still the leading cause of treatment failure. The flaviviruses, such as Dengue virus (DEN) and Japanese encephalitis virus (JEV), represent other major cause of morbidity and mortality, and the areas where these viruses are endemic are spreading rapidly. No curative therapy for any flavivirus could be made available as yet. The first part of this thesis focuses on the HIV-1 drug resistance caused by mutations in a major HIV drug target, the HIV-1 reverse transcriptase (RT) as a response to the largest class of clinically used anti-retrovirals, the NRTIs. A robust proteochemometric model was created to analyse the complex mutation patterns in RT drug resistance. The model identified more than ten frequently-occurring mutations, each conferring at least two-fold decrease in susceptibility for one or several NRTIs. Using our prediction server (hivdrc.org), the model can be applied to propose optimum combination therapy for patients harbouring mutated HIV variants. The second part of the thesis encompasses studies on a promising drug target, the NS2B(H)-NS3pro, in two flaviviruses, namely the dengue virus (DEN) and Japanese encephalitis virus (JEV). Functional determinants of DEN NS2B(H)-NS3pro were identified by site-directed mutagenesis. Further, peptide inhibitors were designed using proteochemometrics (PCM) and statistical molecular design (SMD), synthesized and assayed on DEN proteases, which resulted in some novel peptides with low micromolar or sub-micromolar inhibitor activity. The very poorly characterised JEV NS2B(H)-NS3pro  was cloned, purified and the kinetic parameters of this attractive drug target were determined for a series of model substrates and inhibitor. The results identified the role in target-ligand interaction of different residues on specific positions in the target (NS2B(H)-NS3pro) and ligands (substrates/inhibitors). Overall, the findings in this thesis contribute to rational antiviral drug discovery and therapy.

Human Immune Response To Japanese Encephalitis Virus Guides Development Of Vaccines With Long Lasting Immunity

Chapter 1: Role of JEV NS1 in protective immunity and in immunopathology. Previous studies from our laboratory revealed T cell immunodominance of non structural proteins NS3 and NS1 during natural JEV infections in humans where as the structural protein E, which is a good target for neutralizing antibody response is a poor inducer of T cells. Flavivirus NS1 is also known to induce humoral immune response. Several studies in different flaviviruses have indicated a role for NS1-specific immune responses in protection against flaviviruses. Paradoxically, studies also pointed to the contribution of NS1 in pathology and immune modulation. We screened serum samples from 72 convalescent JE patients for the presence of anti-NS1 antibodies by ELISA and radioimmunoprecipitation and found NS1 reactivity in 45 samples. These antibodies to NS1 are capable of inducing complement mediated cytolysis of cells expressing NS1 on the surface. Additionally, we demonstrated twenty two fold reduction in the infectious virus produced at 48h in SW-13 cells in the presence of human complement and NS1 antiserum compared to control serum, suggesting that complement mediated cytolytic activity of anti NS1 antibody helps the host in controlling the virus propagation. Chapter 2: Comparison of immune responses to JEV structural proteins Capsid and Envelope in human volunteers vaccinated with inactivated JE vaccine and naturally exposed to live JEV. We compared the CMI responses to structural proteins E and C in human volunteers vaccinated with commercially available killed JE vaccine and in humans naturally exposed to live JEV. The results revealed that structural proteins E and C are inherently poor inducers of T cells even in killed vaccine preparation, where there is no competition from immunodominant non structural proteins. Therefore inclusion of nonstructural proteins NS1 and NS3 along with neutralizing antibody inducing envelope should improve memory and efficacy of a JE vaccine. Chapter 3: Construction and testing in the mouse model of experimental recombinant poxvirus vaccines expressing prM, E, NS1, and NS3 of JEV. Guided by the information on immune responses to JEV in the JE endemic human cohort and volunteers vaccinated with killed JE vaccine, we designed experimental vaccines as recombinant vaccinia viruses expressing NS1, NS3, prM, and E proteins of JEV (vNS1NS3prME) or NS1, NS3, prM, and C-terminally truncated E (vNS1NS3prMΔE) and studied the immune responses elicited by these vaccines in mice. Our data showed that a recombinant vaccinia virus expressing prM, ΔE, NS1, and NS3 of JEV is superior to killed JE vaccine in eliciting long lived neutralizing antibodies as well as NS1 and NS3-specific cytotoxic T lymphocytes (CTL) in addition to NS1-specific cytolytic antibodies, resulting in long lasting and enhanced protection from lethal JEV infection in mice. Our results thus identified all B and T cell antigens whose inclusion in a live-vectored vaccine would provide a vaccine with far superior efficacy over the inactivated JE vaccine.

The thesis aims to specify the place of this work in the context of medieval preaching, to critically review the previous research and to find new paths of research in evaluating this work. It is divided into three chapters, their common topic being attribution of both the postils on the grounds of codicological, content, and especially language features. Special emphasis is put on description of non-standard language phenomena, their interpretation being a key factor for transcription rules (one of the main results achieved apart from precising probable dating and localisation is a complete transcription of the Old Czech postil).
