Violence, sexualité et double : les représentations féminines dans Perfect Blue et Paprika de Kon Satoshi

Le présent mémoire consiste en une analyse thématique des représentations féminines dans l’œuvre de Satoshi Kon, de Perfect Blue à Paprika. L’objectif de ce travail est de démontrer que ces images de la femme reflètent la place des femmes dans la société japonaise contemporaine. À cet effet, nous avons examiné les films du réalisateur selon l’approche des études féministes du cinéma. Nous avons divisé notre analyse en trois thèmes : la violence, la sexualité et le double. Il apparaît que les représentations féminines des longs-métrages de Kon possèdent effectivement des parallèles au sein la société nippone actuelle. Le réalisateur emploie des figures et des motifs narratifs communs au Japon et l’anime afin de produire et reproduire les stéréotypes de genre. Par ailleurs, il utilise les éléments filmiques et les particularités du médium de l’anime pour appuyer ces définitions des rôles sexuels. Cette étude est originale par son angle d’approche féministe et psychanalytique qui est rarement adopté par les théoriciens de l’anime. Les études portant sur ce médium sont d’ailleurs récentes et s’intéressent généralement à l’esthétique de l’anime ou à la formation d’une identité nationale japonaise plutôt qu’à la construction du genre dans un média de culture populaire. / The present thesis consists of a thematic analysis of the feminine representations in Satoshi Kon’s work, from Perfect Blue to Paprika. Our objective is to demonstrate that these female depictions reflect the status of women in contemporary japanese society. To this end, we examined the director’s movies according to feminist film theory. Also, we separated our analysis in three themes : violence, sexuality and the double. It seems that Kon’s feminine representations possess parallels to the present Japanese society. Indeed, the director uses figures and narrative motifs common to Japan and anime in order to produce and reproduce gender stereotypes. In addition, he utilizes filmic elements and the particularities of the anime medium to support these definitions of sexual roles. This study is original in its feminist and psychoanalytic approach which is rarely employed by anime theorists. Furthermore, the studies regarding this medium are fairly new and usually focus on the anime easthetic and the establishment of a Japanese national identity rather than the construction of gender in a popular culture media.

Regulation of mitochondrial ATPase by its inhibitor protein IF1 in Saccharomyces cerevisiae

ATP synthase is an essential protein complex located in the mitochondrial inner membrane, which synthesize ATP by coupling to a rotary proton transport across the membrane at the expense of the electrochemical proton gradient created by the electron transport chain. This reaction guarantees the supply of energy to biological processes in a cell. When mitochondria get deenergized, i.e. the protomotive force across the mitochondrial inner membrane collapses, the ATP synthase switches from ATP synthesis to hydrolysis. This hydrolytic activity is then immediately prevented by a natural soluble mitochondrial ATPase inhibitor, IF1. This efficient reversible inhibition system protects cells from wasting energy. In yeast, IF1 is a small protein consisting of 63 amino acids. It binds to one of the three (αβ) catalytic interfaces of ATP synthase and thereby blocks the rotary catalysis. Although the crystal structure of the dead-end IF1 inhibited F1-ATPase complex has been resolved, IF1 initial binding and locking to ATPase still remain unclear events at the molecular level.During my thesis, we have been interested in the dynamic mechanism of ATPase inhibition by IF1. By means of analyses of published structures and protein sequence alignment, we selected numerous residues located in different regions of Saccharomyces cerevisiae ATP synthase α, β subunits, which might potentially paticipate in IF1 binding process. Using site-directed mutagenesis combined with kinetic experiments, we studied the effect of mutations of the selected candidates on the rate and extent of ATPase inhibition by IF1. In this way we identified residues or motifs in ATP synthase α, β subunits involved in IF1 recognition and/or locking steps, which allows complementing structural studies and drawing an outline of IF1 binding.

A indústria de software é muito dinâmica e novas ideias surgem a todo instante em todas as partes do mundo. Nem sempre é fácil fazer com que essas ideias sejam adotadas, pois, para isso, é preciso fazer as pessoas mudarem sua forma de pensar. Deve-se sempre considerar o fato de que o ser humano, diferente do computador, é inusitado e imprevisível. Apesar disso, podemos encontrar determinados padrões de comportamento, que não resolvem todas as questões, mas ajudam a lidar com situações e continuar caminhando para atingir um determinado objetivo. Trazemos nesta dissertação uma pequena introdução sobre o conceito de padrões e, em seguida, apresentamos 48 Padrões para Introduzir Novas Ideias, propostos por Linda Rising e Mary Lynn Manns. Esses Padrões têm o objetivo de ajudar na difícil tarefa de introduzir uma nova ideia dentro de alguma organização, pois se essa ideia pressupõe mudanças culturais, o trabalho é ainda mais complicado. Propomos também quatro novos padrões, que podem ser incorporados ao catálogo original. Num desses novos padrões, mostramos a importância de se usar atividades artísticas no dia-a-dia de pessoas que trabalham com desenvolvimento de software; mostramos também como a Arte pode nos ajudar a introduzir novas ideias. Pesquisamos algumas práticas como teatro, pintura, poesia, música e meditação. Pudemos encontrar elementos de ligação entre o lado puramente matemático e bem definido do ser humano e o seu lado abstrato, analógico e artístico. Desenvolver software deve ser encarado como uma atividade humana, acima da questão técnica e puramente lógica. Existem pessoas envolvidas no processo: as que usam e as que criam o software. Existe uma barreira que separa os programadores das pessoas que usam o software. Essa barreira pode ser quebrada se pessoas da Computação começarem a desenvolver, além das habilidades lógicas que já lhes são óbvias, habilidades artísticas e de relações humanas. / The software industry is very dynamic and new ideas arise all the time from virtually any part of the world. It is not guaranteed that these ideas will be adopted, mainly because, among other obstacles, the solution may imply in having people change their way of thinking. Different from people, computers receive well defined commands and execute them precisely. We should take into account that human beings are independent and unpredictable. Despite of this unpredictability, we can find some behavioral patterns to help us deal with several situations, allowing us to achieve our objectives. This work brings a small introduction into the pattern concept and a presentation of the 48 Patterns for Introducing New Ideas proposed by Linda Rising and Mary Lynn Manns. These patterns provide some insight into the difficult task of introducing new ideas into any organization. We show that if the idea brings cultural changes of any kind, then the task is even more complicated. We also propose four new patterns that can be added to the original catalog. In one of these new patterns, we show the great importance of combining artistic activities with the day-to-day activities of people who work in software development and how Arts can help us to introduce new ideas. The study of some practices such as theater, painting, poetry, music and meditation allowed us to find some connective elements between the purely mathematical side of the human mind and its creative, artistic one. Software development must be approached as a ``human activity\'\', rather than a solely technical or logical one. Above all, there are two groups of people involved in the process: software creators and users. Although a barrier appears to separate these two groups, in this thesis we will show evidences that this division or gap can be bridged if IT professionals nurture their artistic and interpersonal relationship skills in addition to their obvious technical ones.

Detekce dopravních značek a semaforů / Detection of Traffic Signs and Lights

The thesis focuses on modern methods of traffic sign detection and traffic lights detection directly in traffic and with use of back analysis. The main subject is convolutional neural networks (CNN). The solution is using convolutional neural networks of YOLO type. The main goal of this thesis is to achieve the greatest possible optimization of speed and accuracy of models. Examines suitable datasets. A number of datasets are used for training and testing. These are composed of real and synthetic data sets. For training and testing, the data were preprocessed using the Yolo mark tool. The training of the model was carried out at a computer center belonging to the virtual organization MetaCentrum VO. Due to the quantifiable evaluation of the detector quality, a program was created statistically and graphically showing its success with use of ROC curve and evaluation protocol COCO. In this thesis I created a model that achieved a success average rate of up to 81 %. The thesis shows the best choice of threshold across versions, sizes and IoU. Extension for mobile phones in TensorFlow Lite and Flutter have also been created.

The diploma thesis examines new phenomena of electronic democracy which is becoming more popular with the internet expansion. The thesis summarizes existing research in the field of electronic democracy from a view of forms that could be or are taken. The aim of this summarization is to show that new transformational changes affecting our society do not automatically implicate a shift towards direct democracy. Using theoretical summarization a state of the art of electronic democracy in the Czech Republic is described. Importance is given to an analysis of contemporary evolving tools and initiatives which were developed from two directions - top-down and a bottom up. A part of this analysis is an assessment of important factors that are stimulating a development of electronic participation - freedom of information right, internet penetration and computer literacy.

Attachment of this master´s thesis is drawings of site equipment, selected construction details, working safety, instructions for using the building, budget by items and a time plan. The aim of the project is to design appropriate and economically effective procedure for realization of the building.

The context, purpose, and dissemination of legendary genealogies in northern England and Iceland, c.1120-c.1241

The thesis is a comparative and multidisciplinary study of legendary genealogies in the historical writing of northern England and Iceland c. 1120 – c. 1241. Historical writing was produced in abundance over this period in both areas and the frequent contact between England and Scandinavia, as well as shared use of early medieval insular sources make them especially suitable for comparison. The Viking invasions and settlement in England had a significant impact on English culture, language and literature and changed attitudes to their own legendary past. The Danish conquest of England in the early eleventh-century also brought the insular and Scandinavian worlds closer together, and even after the Norman Conquest in 1066, England and Scandinavia engaged in scholarly and textual exchange The theoretical framework for the thesis combines approaches from religious history, art history, political history, literature history and gender history. The main research questions of the thesis consider the dissemination, development, and purpose of legendary genealogies. The sources are a collection of Durham related manuscripts with illuminations of the pagan god Woden (c. 1120–88) in two historical works De Primo Saxonum Aduentu and De Gestis Regum; Genealogia Regum Anglorum (Rievaulx, 1153x54) by Aelred of Rievaulx; two works attributed to Snorri Sturluson’s Prose Edda (Iceland, 1220s) and Heimskringla (Iceland, 1225x35). Common to the sources is the inclusion of genealogies that stretch from legendary generations to living individuals at the time of writing. Thus, genealogies connected dynasties and civilisations in mutual descent from pagan, Trojan and biblical ancestors. By analysing textual dissemination as well as political contexts, literary patronage and mechanisms in legitimisation of power, the thesis address amalgamations of origin myths, the use and significance euhemerised pagan gods, and female generations in genealogies.
