The aim of my master thesis was to reconstruct the pedigree of wild cherry (Prunus avium L.) half sib progeny using microsatellite markers. The cherries (Prunus avium L.) are grown for the purpose of establishing plantations half-sib progeny, to verify the registration of mother and traced his father between the clones represented in the seed orchards, or reveal paternal gamete contamination. Technically there is a transfer of individuals half-sib offspring to full-sib offspring. Literary analysis was performed focusing on the issue of pedigree reconstruction using molecular markers and the theory of the breeding cycle focusing on offspring testing was incorporated. The study was conducted on material from the seed orchard Obrovice, owner VLS s. p. (Parental population) and seedlings grown from these individuals for the purpose of establishing half-sib offspring testing in the forest nursery Lhota. As part of their research were collected samples, all clones of parental populations and 268 individuals from the offspring recording evenly respective parent clones.
Subsequently DNA was isolated. Microsatellite analysis was performed on 16 polymorphic loci and realized the pedigree reconstruction using software GeneMarker and software Cervus. Results are recorded at the end and progeny structure was described in detail. Pedigree knowledge will continue to be used in the future in genetic evaluation of individuals based offspring and enables significant acceleration of the whole of the breeding cycle.

This thesis deals with mikrobiology of curd and selected curd products. It describes the microbiological quality and curd products, their unwanted contamination by micro-organisms. Contamination occurs most often during production. The most common con-taminating microorganisms are gram – negative bacteria, yeats and fungi. It defines their negative impact on the quality of curd and curd products. It further characterizes other possible contaminants tnat can cause foodborne disease in humans. The thesis also describes technologically important lactic acid bacteria. Part of the thesis is an experi-mental microbiological analysis of curd and curd products during storage. Relatively low numbers of contaminating microorganisms were found. Higher numbers of microor-ganisms showed samples after expiration date and samples of Budapest spread contai-ning flavoring components.

Roadside soils are affected by traffic. Topsoil samples were collected at 35 Moravian localities with varying traffic intensity. They were analyzed by XRF spectrometry to obtain total element composition. The evaluation of results was carried out with an emphasis on risk elements (Cr, Ni, Cu, Zn, As, Mo, Cd, Pb). Positive correlation between traffic intensity and Cu and Pb content in the soil was discovered. The evaluation of the level of contamination was carried out on the basis of the limits announced by the Czech Ministry of Environment. The whole diploma thesis is supplemented by a review in order to provide a comprehensive view on this extensive issue.

This diploma thesis deals with the effect of compost on heavy metal (Cd, Pb, Zn) contaminated soil. There are theoretically discussed contaminations, their sources and their impact as well as remediation possibilities. Work validates the assumption that compost has a positive effect on mobility reduction of heavy metals and their bioavailability. Verification is performed by the pot experiment, where the indicator plant lettuce (Lactuca sativa L.) has been grown on differently contaminated soils. When the experiment was finished, the lettuce biomass and soils were analysed. Detected concentrations of heavy metals have confirmed a positive effect on Cd and Pb immobilization. The immobilization of zinc was not affected by compost.
