Life strategies for substrate assimilation by freshwater bacterioplankton

The availability of substrates is one of the most important environmental constraints on the diversity and functioning of microorganisms. Substrate quantity and quality as well as the metabolic features of heterotrophic microorganisms determine the efficiency, speed and type of transformation that can occur in nature. As such their interplay with the environment regulates how much carbon and energy is incorporated by bacteria and subsequently reaches higher trophic levels. In lakes the bulk substrate that is available for bacteria is composed of a complex mixture of compounds, varying in lability and distribution in the environment. This thesis addresses the coupling of organic substrates, their metabolic use and the composition and ecology of the microbial community. Controlled laboratory experiments with mixed bacterial communities in either batch cultures or chemostats were designed to shed further light on bacterial use of labile and quantitatively significant carbon compounds. I show that different amino acid substrates only exert a minor influence on bacterioplankton community composition and growth. Hence the ability to use a wide range of such abundantly produced protein monomers seems to be widespread among freshwater bacteria. In contrast, when acetate was provided as the only carbon substrate, in either pulsed or continuous amendments, this very different substrate input mode had a strong effect on bacterial community composition. Biomass yield, for example, was twice as high when acetate was given in the form of pulses rather than provided continuously. In another set of experiments, I show that the oxidation of the globally significant greenhouse gas methane is a process that can potentially take place at the water-ice interface of seasonally ice-covered lakes and was not constrained by temperature as suggested in previous studies. This work also suggests that methane oxidation in ice-covered lakes can be constrained by competition for nutrients between specialized methanotrophs and heterotrophic bacteria. Combined these studies suggest that some labile substrates cause minor selection on bacterial community structure and functioning. This probably reflects the competitive advantage of using a broad range of low molecular weight substrates. However, as in the case of methanotrophs there is specialization for a specific low molecular weight substrate such as methane. In which case, competition with other community members i.e. for nutrients can constrain methane oxidation. In both cases it might however not depend just on the availability of substrate, but also on how substrates are distributed in time and space.

Comparison of the benthic fauna within the littoral in two affected lakes situated in central Sweden

The benthic animals have important roles in benthic communities. Indicator organisms can tell things about a lakes status. Many of them are sensitive to acidification but there are also tolerant taxa. Some prefer eutrophic environments. The benthic fauna can therefore decide a lakes status since some species prefer different environments. The focus of this study was to compare benthic fauna within the littoral in the lakes Sidsjön and Vintertjärnen. They are situated a little south of Sundsvall, central Sweden and belong to the water system of Selångersån. The comparison was done at eight random locals in each lake at two occasions, during early and late summer to analyze if the benthic fauna differed between the two lakes and if it changed through the season. Five of the locals in each lake were protected and had soft bottoms and the remaining were exposed locals with hard bottom. Sidsjön is bigger than Vintertjärnen and has more variated bottom types so the biodiversity should be higher in Sidsjön. I also expect that the number of taxa should decrease during the later occasion due to hatchings. Three statistical analyses were done to see if these hypotheses were right or if they should be rejected. The two first analyses were ANOVA analyses, two-way with replication. The first ANOVA was for number of taxa and the second was for number of animals. The third analysis was a DCA analysis with the variables lakes, bottom types and emergent plants. ASPT index and Eq was also calculated and pH was measured during autumn for the lakes. The conclusion is that the there is no difference between the lakes but a significant effect when it comes to seasons according to the first ANOVA analysis. The second ANOVA analysis show that the lakes are not differing, that no factor is significant and there are no interactions. Deciphering Phosphorus and Aluminum Physiochemical Associations in Paleolake Sediments of Long Pond, GA

This study focuses on Long Pond, a groundwater-fed sinkhole lake in Georgia, which does not exhibit any evidence of eutrophication drivers despite high concentrations of sedimentary phosphorus (P) (> 3000 mg/kg P) in the recent sediment record. Chemical analyses have shown a strong correlation (r2>0.99) between P and aluminum (Al) throughout the core, suggesting Al plays a significant role in sequestering most of the P, and limiting its availability to phytoplankton, thereby inhibiting eutrophication. The purpose of this study is to decipher P and Al physicochemical associations in the sediments. After the samples were fractionated into amorphous and non-amorphous phases, the correlation was maintained in both phases. Evidence suggests two modes of Al-P associations: a sorption and/or co-precipitation occurring mostly in the amorphous phase and a mode whereby Al and P are being inputted to the lake bound together.

Warfare and fertility : a study of the Hor (Arbore) of Southern Ethiopia

This thesis is an ethnographic study of the Hor (Arbore) who live at the north-eastern end of the Limo river delta on Lake Stephanie in Southern Ethiopia. In the thesis the Hor belief in the link between warfare and fertility is described and analysed. The Hor do not go to war against all their neighbours. Instead they have categories of those whom they fight and whose shed blood is believed to be beneficial to the Hor and those whom they do not fight and whose shed blood is believed to be dangerous to the Hor. From the former they sometimes take wives and raid cattle while from the latter they neither take wives nor raid domestic animals. From a specific group in the first category known as Maale (and formerly from other groups) the Hor kill male victims whose genitals and bush knives they bring home as trophies. These outsider items are crucial in rituals for the reproduction of their society and culture and also for the reproduction of the societies and cultures of certain of their neighbours. The thesis discusses the link between fertility and various aspects of Hor life. Hor Qawots (ritual leaders) are empowered by the genitals brought from the exterior and it is mainly this empowerment that is believed to enable them to be effective in their political and religious roles in Hor country and among Hor neighbours. The study shows how strictly ranked senior and junior Qawots who are members of the braceletwearing clans, as well as metaphorically gendered age sets and ranked settlements, shape the br social and cultural world and the world of their neighbours. It also shows the crucial role of the outside both as a source of fertility and as a source of essential tools of production and ritual items.

Precipitation distribution in the Lake Pukaki Catchment, New Zealand

Mountain precipitation, as a major component of global ecology and culture, requires diverse observation-based distribution studies to improve process characterisation and so enhance environmental management and understanding. Analysis of data from an array of precipitation gauges within the nationally important, and internationally extreme, mountainous Lake Pukaki catchment in New Zealand has been undertaken in an effort to provide such a study, while also improving local hydrological understanding. An objective observation based undercatch-corrected 1971-2000 average annual precipitation distribution has been prepared for the mountainous Lake Pukaki catchment, New Zealand. Precipitation records from 58 gauges at 51 sites, augmented with 10 new gauges, were used in preparation of the distribution. The assessed undercatch correction of 17 % across the catchment indicates that mountain hydrological investigations in New Zealand that use precipitation data and yet do not consider undercatch will be in considerable error. The average annual distribution confirms the existence of high precipitation magnitudes and horizontal gradients in the catchment in comparison with other mountain regions around the world. The high magnitude is unusual when its position in the lee of the principal orographic divide is considered indicating rare precipitation distribution processes occur in the region. Consideration of river flows, glacial change and evaporation led to a confirmation of the gauge derived average catchment precipitation. Precipitation to wind direction relationships identified the predominant westerly wind to be the primary precipitation generating direction with large magnitude events biased towards the northerly direction. All directions from the eastern side of the mountain divide had the lowest frequency and daily precipitation magnitude. Derivation of wind-classed precipitation distributions identified a distinctive south east to north west precipitation gradient for all wind directions, most severe for the north west direction and least severe for the easterly direction. Precipitation extent was greatest for the northerly direction and least for the south south westerly. The wind-classed distributions enable the estimation of daily precipitation likelihood and magnitude at any location in the catchment based on knowledge of the synoptic wind flow direction and precipitation at just one reference site. Improved river flow and lake inflow estimates resulted from the use of wind classed daily precipitation estimates validating the quality of the wind classed distributions. From 1939 to 2000 there has been no statistically significant trend in precipitation magnitudes, frequencies, or extremes in the catchment. At Aoraki/Mt Cook village, in the upper catchment, there have been significant increases in magnitude, frequency and extremes associated with the phase change of the Interdecadal Pacific Oscillation (IPO) in 1978. This change can be explained by the increase in strength of westerly winds for the different IPO phases but not by a change in frequency of different wind directions. In the lower catchment the IPO relationship is of an opposite sense to that observed in the upper catchment, indicating that the areas operate under two different climate regimes with different precipitation controls. The significant relationship to the IPO phase indicates that it is more important than climate warming in terms of future precipitation distribution in the Lake Pukaki catchment, and by extension the Southern Alps. The distributions prepared provide a valuable tool for operational and academic hydrological applications in the region. In addition, they provide a valuable characterisation of the precipitation in a Southern Hemisphere mid-latitude lee to predominant westerlies glacierized mountain catchment. From this standpoint they highlight the contrast to Northern Hemisphere mountain precipitation distributions commonly used in model validation studies, thereby providing an extension of locations with which to refine orographic precipitation process understanding.

An investigation into the impact of environmental change upon the vegetation of Widdybank Fell, Upper Teesdale

Widdybank Fell is located in the Upper Teesdale National Nature Reserve in the Northern Pennines. It is internationally renowned for its unique late-glacial relict assemblage of plant species, particularly those on the "sugar limestone” outcrops. Cow Green Reservoir was constructed in the early 1970s and flooded the lower western slopes of Widdybank Fell. At the time there was concern amongst ecologists that the reservoir might alter the local climate and have adverse effects on the remaining vegetation. A comprehensive vegetation survey of Widdybank Fell was carried out by Jones (1973). Aspects of this survey were repeated as part of the present study. There have been significant changes in the composition of some of the plant communities since the 1970s. These changes include a considerable loss of bryophyte diversity and lichen abundance. Overall there has been a decline in "stress tolerant" species, and a loss of calcicole species on the calcareous grasslands. Few of the nationally rare plant species have changed in abundance. Using data from the meteorological stations at Widdybank Fell and from nearby Moor House, it has been demonstrated that the presence of Cow Green Reservoir has resulted in significant changes in the local climate. These changes are consistent with those expected by a classic "lake effect" and include the all year round moderation of minima (e.g. resulting in a reduction in the number of ground frosts). The reservoir has also produced cooler mean air temperatures in spring and warmer mean temperatures in autumn. Despite the observed local climate impact of Cow Green Reservoir it seems most likely that other factors have been responsible for the observed vegetation changes. Acid deposition has probably caused the loss of calcicoles on the calcareous grasslands, and atmospheric nitrogen deposition could explain the loss of bryophyte diversity and lichen abundance.

Phytoplankton Production In Lake Victoria, East Africa

This thesis develops, validates and applies an empirical model that provides the first spatially explicit estimates of gross and net phytoplankton production in Lake Victoria. Gross and net phytoplankton production are in turn used to estimate the maximum sustainable yield (MSY) of Lake Victoria's fishery following an empirical formula and the carbon efficiency transfer method. Chapter 2 presents results from three inshore areas where diurnal and sub-seasonal gross and net phytoplankton production was derived using an adapted version of the phytoplankton production model developed by Fee (1990). Spatial and temporal trends of chlorophyll (chl), PI parameters, the vertical attenuation of PAR (kPAR), Secchi depths (SD) and respiration rates are identified. kPAR and SD are highly correlated to chl within the euphotic zone, as well as to each other. Furthermore, the two PI parameters, PBM and aB, exhibit a strong linear relationship and both decline along an increasing chl gradient, presumably due to increased light-limitation, a taxonomic shift from diatoms to cyanobacteria with increasing chl as well as an increased need for biologically fixed nitrogen. These hypotheses are supported by observed synchronous changes in the PSII:PSI ratio of phytoplankton and changes in the chl-specific attenuation of PAR (kchl). Relationships are also derived between biomass-specific respiration rates (RB) with chl and PBM; similar to PI parameters RB decreases with increasing chl. Owing to these correlative trends, only one parameter is required to estimate gross phytoplankton production through the empirical model developed in this thesis. The empirical model predicts that gross phytoplankton production increases in a near linear fashion between chl of 0 to 10 mg. m-3, begins to flatten out as chl approaches 20 mg. m-3 and then slightly decreases when chl exceeds 40 mg. m-3 where the maximum PPG of 13. 1 g O&#8322;. m-2. day-1 is reached and is in close agreement with a theoretical argument proposed by Talling (1965). Areal respiration and consequently net phytoplankton production are sensitive to chl within the mixed layer as well as mixed layer depths. Overall, the lakewide averages of gross and net phytoplankton production are 9. 68 and 2. 2 g O&#8322;. m-2. day-1 respectively. Significant temporal variability was observed on sub-seasonal scales within the inshore of Lake Victoria, and changes in limnological parameters coincided with changes in water column temperatures in each of the three bays. In Fielding Bay, the availability of meteorological data revealed that strong nocturnal wind events decreased both the water column temperature and chl, while both parameters generally increased in the absence of any such wind event. Lateral exchange of water with deeper areas through strong wind events essentially flushes Fielding Bay causing the observed decreases in both the water column temperature and chl; this hydrodynamic event also influences other limnological parameters according to their respective correlative regression equations with chl. Spatial trends were also observed between inshore areas. The deepest area, Napoleon Gulf, has the lowest values of chl while the shallowest area, Inner Murchison Bay, has the highest chl as the mean depth of a bay sets an approximate upper limit on chl. With respect to diurnal variability, PI parameters decline through the day, kPAR increases over the day and no statistically valid trends were ascertained for chl and RB. Chapter three examined spatial and seasonal patterns of chlorophyll fluorescence, temperature, dissolved oxygen and water transparency from four lakewide cruises. Significant spatial variability of each parameter confirmed that lakewide data is required to generate spatially explicit estimates of phytoplankton production. Complex patterns in the thermal structure during each cruise illustrated that physical processes in Lake Victoria are at times more complex that a previously stated unidirectional hypothesis of warm water in the north and cool water in the south (Spigel and Coulter 1996), and these patterns influence spatial patterns in dissolved oxygen and Secchi depths. Similar to Chapter 2, estimates of chl within the mixed layer were highly correlated to mixed depths, while lakewide averages of chl are lower than previously reported offshore values (Mugidde 1993, 2001).
