Využití českého teplokrevníka u jízdních složek státní policie České republiky / Using of the Czech Warmblood for the Czech Republic Mounted Police

The horse evolution started with Hyracotherium, which was a small animal inhabiting forests of the Eocene. Since then, it evolved into more or less successful forms in which the conditions of the surroundings of its occurrence were reflected. When it comes to the initial usage of horses, they were used as a source of meat. Later, in the period before 4,600 years ago, evidence of the domestication of these animals emerged. Due to the improved system of breeding, horse as a draught animal and a mean of transport gradually became riding horse, which contributed not only in spreading human population, but also in influencing history of many civilizations. Nowadays, horse is represented through a number of breeds, which are used in equestrian sport, work with cattle, horse races, in forestry labours, or as a good companion. In addition, horse gained its irreplaceable role in the service of the Mounted Police of Czech Republic where it is daily deployed in diverse actions; for example in city and peripheral guarding, in searching for missing persons or in restoration of disturbed public peace. The most useful breed for these purposes is Czech Warmblood, where the most preferred geldings are in brown or chestnut color. High demands are placed on physiological characteristics; however, the most important criterion is the excellent character of this breed. Quality of these characteristics consists in the length of duty and in the reasons for their release from duty. Some of the most frequent reasons for the release from duty health problems and age. The release due to age is associated with strong and resistant horses. Regarding the lineage of the police horses on duty, the best evaluated were the descendants of stallions 1154 Quick Lauro Z, Mykonos and 1028 Manillon Rouge. However, there is a correlation between these descendants and their number on active duty. After categorization of all traced stallions into relevant lineages, the lineage 1100 Przedswit became the most numerous lineages (7 stallions). Nevertheless, this lineage is not positively evaluated because of its descendants, which did not perform well on duty. Another numerous lineages were 5500 Pythagoras, 4700 Amour du Bois, Orange Peel xx-Almé Z, 67 Dark Ronald or 4800 Ladykiller XX. The central purchase system of duty horses should help to unify these horses as well as to prolong the length of duty, via universally established criteria. Results demonstrate the unification of typological characteristics of these horses. However, the average withers height of measured horses fails to meet the requirements. Regarding the length of duty, the results were not conclusive; therefore, the hypothesis was confirmed just partially.

Vývoj místní samosprávy v obci Kamenný Újezd / The Development of Local Government in the Municipality of Kamenný Újezd

This dissertation focuses on current issues of Kamenný Újezd´s local council. Kamenný Újezd is a municipality with over two thousand inhabitants. This assessment looks into the local political process in the municipality. This assessment is divided into two parts - theoretical and practical. Theoretical part focuses on the local councils, general establishments and elections into the local authorities in the Czech Republic. It also focuses on local political partisanship, coalition and the theory of political cleavage. The practical part of the dissertation describes some demographic and socioeconomics indicators in the municipality. Majority of the practical part is devoted to the local elections from 1994 - 2014. The results from these elections are put into graphs and charts. The collected data is then compared. In the last part of the dissertation are interviews with local politicians. Main public and political problems in the municipality are discussed. Also public relations and main political cleavages between individual politicians are discussed in these interviews.

Přežívání bifidobakterií v trávicím traktu dospělých dobrovolníků / Survival of bifidobacteria in digestive tract of adult volunteers

Probiotics are living microorganisms, which contribute to health improvement. The bacteria of the genus Bifidobacterium are one of the most commonly used probiotics. These bacteria are naturally occurring especially in the digestive tract of mammals. They affect the digestive process by helping to optimize the composition of the intestinal microbiota. The probiotic bacteria are expected to survive in the digestive tract and to be able to colonize the intestinal mucosa. The aim of this diploma thesis was to test bifidobacteria of different origin for certain functional properties. Three strains of bifidobacteria were selected: Bifidobacterium animalis ssp. lactis DAN (isolated from Danone yoghurt), Bifidobacterium bifidum JKM (isolated from infant feces) and Bifidobacterium longum ssp. suis 5/9 (isolated from feces of a calf); and tested in vitro a in vivo. The ability to survive in simulated conditions of digestive tract was tested. The bifidobacteria were exposed to low pH and bile salts. All the strains survived the conditions in unaltered amount till the end of the test. Autoaggregation was observed as well. The strain Bifidobacterium animalis ssp. lactis DAN showed average autoaggregation only 7,69 %. The other two strains had average autoaggregation 16,10 % and 79,05 %. The autoaggregation correlated with adherence. Adherence was tested on intestinal cell lines HT-29, Caco-2 and HT29-MTX, which differed in their ability to produce mucin. The strain Bifidobacterium bifidum JKM showed the best ability to adhere to intestinal cells among all the tested bifidobacteria. They best adhered to Caco-2 cells. Further, it was necessary to determine if the in vitro study is in correlation with in vivo testing and the bifidobacteria strain with the best ability to adhere will survive in the digestive tract of adult volunteers for the longest time. Rifampicin resistant variants of tested bacteria were given to adult volunteers to distinguish the introduced bifidobacteria and the bifidobacteria naturally occurring in intestine. All the tested bacteria survived the passage through the digestive tract in large numbers (106 a 107 KTJ/g), the bacteria were not detected 12th day after the introduction. None of the strains is able to permanently colonize the colon of the volunteers. The hypothesis was not confirmed that the most adherent strain in the digestive tract will survive the longest.

Návrh a testování způsobů zvýšení účinnosti transfekce živočišných buněk

Master’s thesis deals with the design and testing of methods of increasing the transfection efficiency in mammalian cells. In this work a polyethylenimine (PEI) transfection method was used. The transfection efficiency was tested using MDA-MB-231 breast cancer cell line and pcDNA3.1-GFP-hMT-TOPO vector. We focused on optimization of PEI (1-6 µg) and plasmid DNA (1-3 µg) concentration, transfection time (0-48 h), number of cells per 9 cm2 (100,000-2 million) and medium composition. A complex containing linear penetration peptides (CPPs) KALA and GALA was assembled. Concentration of CPPs (7,5-45 µg) was optimized and a zeta potential and a size of the complexes were investigated. Due to the formation of large aggregates (over 1200 nm), modification with polyethyleneglycol (PEGylation) of PEI was performed. With the resulting complexes (~15 nm), transfection was performed. Transfection and apoptosis induction rates were compared between PEI and PEGylated PEI and as the best was chosen conjugate of PEGylated PEI (3 µg) with 45 µg of GALA and 3 µg pDNA with efficiency of 75%.

Vyhodnocení úrovně zabřezávání skotu na farmě využívající řízené reprodukce stáda

The aim of this thesis was to evaluate the level of mating on the farm using controlled herd reproduction. The evaluation of the level of pregnancy was carried out in dependence on the individual month of observation, the monitored parameters of milk from the performance control of individual lactations and the lines of fathers of cows. Performance data for January 2017 - April 2018 were used for the assessment. Based on the result, it can be concluded that the seasonal percentage is affected by the season. It has been shown that the percentage of pregnancy has decreased in the summer month. The significant decrease in the rate of mating was occurred with the increasing age of cows. The percentage of lactation, regardless of the order of lactation and the line of bulls, is negatively influenced by the milk yield and by the positive effect of the number of lactation days when embedded. To confirm the negative effect of milking yield on the breeding, the NEB breeders showed the highest milk yield and the lowest percentage of pregnancy. Significant fluctuation were occurred in value when evaluating individual indicator depending on the reference month. In the middle of the reporting period, the amount of urea in the milk had a positive effect on pregnancy. A similar result was obtained for the T / B ratio.

Analýza liniové skladby populace slezského norika

The aim of the diploma thesis is to analyze linear composition population of silesian noric breed, characteristic of SN lines in terms of exterior and performance, database processing and statistical evaluation of individual lines SN by scoring exterior, perfor-mance and monitored effects withers height tape, withers height stick, chest circumfe-rence, circuit shin. We developer the database based on manual entry of the point data evaluation and observed effects. We used method GLM in softawre program Unistat 6.5. Multiple comparisons, we have found that the effect of the reference point evaluati-on for exterior and performance stallions are on average better assessment than mares and the differencies between the lines was proved. The differencies between lines and also sex were proved also at withers height stick and withers height tape. Stallions reach bigger hight. Further was proved that mares have bigger chest circumference, then stalli-ons and at circuit shin the stallions have bigger value compare to the mares.

Dactylography - History, Present and Future This thesis introduces one of the most important identification methods - dactylography (or dactyloscopy), e.g. the method based on analysis and classification of patterns of friction ridges (especially fingerprints). This thesis consists of three main chapters considering its name. The first chapter describes interesting history of this method from first discoveries, through various breakthroughs, to reaching the status of forensic evidence. This chapter mainly focuses on individual pioneers on the field of dactylography, but tells as well the story of Czechoslovak dactylography and reminds cases, that were very close to prove this method wrong at its fragile beginning. The second chapter focuses on the present of dactylography. This chapter starts with three basic laws of dactylography, but the essence of it lies in the term "dactyloscopic evidence". The methods of revelation, analysis and preservation of dactyloscopic evidence are described here as well as their comparison. If it comes to analysis, I am trying to compare two approaches used today (holistic and numerical) and offer my opinion which one is better. FINGERPRINTING - PAST, PRESENT AND FUTURE The objective of this thesis is to present the forensic fingerprinting as a scientific method which, although in comparison with other forensic sciences being one of the oldest, is not out-dated at all, quite to the contrary - it presents an ever-changing and continuously developing field of science. The objective is approached through three distinctive and yet intertwined chapters corresponding with the title of the thesis - past, present and future of the fingerprinting. They are also designed, at least in part, to capture this constant forward motion. Accordingly, the first chapter aims to describe briefly scientific roots of the method, the way it drew in its beginnings from various scientific discoveries, inspired by the rise of Darwinism. Particular emphasis is put on the description of the formulation and empirical confirmation of three physiological laws of fingerprinting that even today serve as its grounds. Concept and nature of dactyloscopy The aim of the thesis is to provide a comprehensive view on the issue of dactyloscopy. Especially, to introduce readers about dactyloscopy and institutes related to this method. The thesis is systematically divided into several chapters. The thesis is complemented by visual and textual appendices for the better and easier understanding of the various institutes. The introduction contains basic information about dactyloscopy and the outline of what the thesis will contain. Chapter One deals with the subject, concept and importance of the dactyloscopy. Chapter Two is divided into two parts. The first subchapter concerns the historical development of forensic dactyloscopy in the world.
