Summary Natural phytochemicals are currently used in the treatment of many diseases. Cancers are just ones of them and they are ranked among the most common and the most serious. Phytochemicals in the form of cytostatics are used in chemotherapeutic treatment of cancer. In future there could be included among cytostatics also some alkaloids from the family of Amaryllidaceae, whose testing for a selective cytostatic effect on tumor cell lines of colorectal carcinoma Caco-2 and HT-29 and on normal cell lines of human intestinal epithelial FHs 74 Int is a subject of this thesis. There were tested 17 alkaloids isolated from plants of Chlidanthusfragrans, Zephyranthes robusta and Nerine bowdenii. Particularly alkaloids from plant Zephyranthes robusta namely haemanthamine with this values: IC50 = 0.99 plus/minus 0.14 microM for tumor cells, Caco-2, 0.59 plus/minus 0.01 microM for tumor cells HT-29 and 19.47 plus/minus 8.86 microM for normal cells FHs 74 Int, Lycorine with values IC50 = 0.99 plus/minus 0.08 microM for tumor cells Caco-2, 1.2 plus/minus 0.01 microM for tumor cells HT-29 and 22.68 plus/minus 0.09 microM for normal cells FHs 74 Int and Haemanthidin with values IC50 = 3.29 plus/minus 0.91 microM to tumor cells Caco-2, 1.72 plus/minus 0.11 microM to tumor cells HT-29, and 11.63 plus/minus 0.86 microM for normal cells FHs 74 Int proved a significant anti-proliferative activity. From these results there is evident the selectivity against colorectal cancer cell lines. For this reason, those tested alkaloids are suitable for further testing and for study of their biological activity against tumor cells in the terms of in vitro and in vivo.

Zhodnocení reprodukce a vlastní užitkovosti v nukleovém chovu přeštického černostrakatého plemene

The main objective of the thesis was to evaluate the reproductive utility and the achieved parameters of its own utility of the nucleus herd reared on the farm in Mladotice. The detected results have been compared with the average of the Prestice black-pied breed population. The attention has been paid to the lines of the boar representation. The number of all born piglets, the alive born ones and the weaned ones has been regarded during reproductive qualities assessment. The average daily growth, the muscle proportion and the back fat height have been evaluated while assessing the own utility. The origin of the Prestice black-pied breed with the following regeneration process with the assistance of Mirgorod, Liven, Cornwall, Berkshire, Wessex and German saddleback breeds, which contributed to the resistance of the Prestice black-pied breed, has been described in the literature review. The improvement process followed after the regeneration, especially by using Pietrain, Hampshire and Landrase breeds. The characteristics of the Prestice breed with reference to its tilted ear and typical black-pied coloring without body parts defining have been elaborated. Furthermore, the utility, the population development in the last five years and the preservation of the breed as a genetic resource have been described there. In the thesis the own utility of the Prestice black-pied breed of the nucleus herd reared on the Mladotice farm has been evaluated that does not reach breeding goal. The Akoga line is the best line considering its own utility. The reproductive utility has been evaluated as excellent. The Wiskont line has the best results. Statistically significant differences have been found among the lines considering both its own utility and reproduction.

The thesis dealt with the issue of planting, economy and integration of alleys into the landscape. The aim of the study was to evaluate the economy of new plantings alleys, success rate of planting, integration into the landscape, aesthetics and health status in comparison with the assumptions presented in the literature. It was observed six model alleys, which were selected especially with respect to their age structure in order to verify the basic assumptions of this work. Alleyways were examined with regard to health, aesthetics, integration into the landscape and influence of these characteristics by human activities. Establishment of new alleys was considered in detail from economy point of view.

1 Abstract Fingerprinting - Past, Present and Future The objective of the presented diploma thesis is to provide a comprehensive overview of findings in fingerprints area. Fingerprinting is a unique method enabling quick, inexpensive and reliable personal identification. To provide a comprehensive understanding of the presented issue, fingerprint reputation as ever-changing and continuously developing method is put into contrast with its contradictions and current problems. The first part of the thesis describes roots and genesis of the method from its beginnings in ancient realms up to date. The process of shaping of the knowledge in the field of fingerprints into a science is presented through the works of the most prominent fingerprints personalities. Despite the difficult beginnings, fingerprints finally reached the position of trustworthy and respectful area of forensic science. Due to that; the historical part follows the journey of fingerprints evidence into the police investigation and court rooms. Final point brings an attention to the establishment of fingerprinting in Czech criminal investigation. The main part of the thesis is dedicated to the practical aspect of fingerprints expertise. The tested substances have been provided by the Department of Organic and Bioorganic Chemistry at the Faculty of Pharmacy in Hradec Králové, Charles University, as potential antifungal pharmaceuticals and medicine effective against Methicillin-resistant Staphylococcus aureus (MK-NO2-1, MK-NO2-2, DAB-5-K, PABA-Me-5, PABA-Et-5, MK-F-1, PABAN- 3, PABAN-5, MK-F-2, MK-CF3-1, MK-CF3- 2). In order to determine the toxicity we have implemented two methods. The first method is based upon measuring metabolic activity of cells by means of reducing tetrazolium to a colored product. The second method detects the amount of LDH released as a marker of toxicity. The human hepatoma cell line HepG2was used as a model. . The IC50 and EC50 parameters were used to assess the degree of viability and cytotoxicity. Segmentace žeber v hrudních CT skenech / Segmentation of ribs in thoracic CT scans

This thesis deals with design and implementation of an algorithm for segmentation of ribs from thoracic CT data. For the segmentation method of rib centerlines detection is chosen. The first step of this approach is to extract the centerlines of all the bones located in the scan. These centerlines are divided into short primitives, which are subsequently classified into couple of categories, depending on whether they represent the centerline of the rib. Subsequently, the centrelines of ribs are used as the seed points of the region growing algorithm in three-dimensional space, which realizes the final segmentation of the ribs. Within the work, a database of 10 CT scans was manually annotated, which was subsequently used to validate a performance of the proposed segmentation approach. The achieved success rate of primitive classification is 96,7 %, the success rate of rib segmentation (Dice coefficient) is 86,8 %.

The application of nanoparticles in the field of theranostics requires knowledge of the specific interaction of nanoparticles with the immune system. One of the first cells with which nanoparticles interact when given to the body are cells of the mononuclear phagocytic system. The aim of this diploma thesis is to prepare an in vitro study that describes the effect of two types of gold and three types of silicon ultra-small nanoparticles on immune cells. Immune cells are presented in the form of primary PBMCs isolated from whole blood , and cells of monocytic cell line THP-1 in the form of monocytes and differentiated macrophages. During the experiments with primary cells, emphasis is placed on maintaining the concept of personalized protein corona. After characterization of the immune cells used, cells are subsequently stimulated with ultra-small nanoparticles and the influence of these nanoparticles on cell metabolism, viability, degree of differentiation and secretion of pro- and antiinflammatory cytokines is monitored. Peptide CART (cocaine- and amphetamine- regulated transcript) is a neuropeptide acting in the hypothalamus to reduce food intake (anorexigenic peptide). Despite all efforts the receptor and the mechanism of action is still unknown. This peptide has two biologically active forms, CART(55-102) and CART(61-102). Peptide CART is able to bind to pheochromocytoma cells PC12. PC12 cells differentiated in neuronal phenotype with NGF (nerve growth factor) showed a higher number of binding sites (11250 ± 2520 binding sites/cell) compared to undifferentiated cells (3600 ± 570 binding sites/cell). PC12 cells differentiated by dexamethasone to chromaffin cells showed high non-specific binding. Peptide CART contains three disulfide bridges. To clarify the importance of each disulfide bridge to maintain biological activity, analogues with one (analogue 3, 4 and 5) or two (2, 6, 7 and 8) disulfide bridges and a peptide analogue of CART (61-102), which has methionin at position 67 replaced with norleucine were synthesized. We showed that biological activity was unchanged at analogue 1 and analogue 7 containing disulfide bridges in positions 74-94 and 88-101. Auxin is one of the most important plant hormones, which provides development of a plant. PIN1 and PIN7 proteins belong to the PIN family of transporters which is among the most important auxin efflux carriers. This thesis deals with the of AtPIN1 and AtPIN7 auxin efflux carriers (from Arabidopsis thaliana) in human embryonic kidney 293 cell line. Biological activity of these proteins was tested by using radiolabeled auxins accumulation. Further inhibitors of auxin transport have been tested - NPA, CHPAA and BFA.
