Nanoparticles (NP) are currently a progressive area of scientific research. The possibility of synthesizing them according to the required parameters opens up possibilities for their wide use also in biomedicine. One example is a nanoparticle that can detect cellular processes, such as pH. We already know that the pH of healthy and cancer cells differs by the opposite gradient on the intracellular and extracellular side of the membrane. In this context, this work deals with the study of fluorescent silicon nanoparticles (SiNP) tested on a human keratinocyte cell line from a healthy donor (HaCaT) and from skin cancer donor (A431). Once found that even the highest concentrations of SiNP used are not cytotoxic, they can be further studied by fluorescence, confocal and super-resolution microscopy. In order to assess the pH detection properties of these SiNPs, a method for measuring intracellular pH with a fluorescent raciometric probe SNARF-1 using fluorescence spectroscopy and flow cytometry was introduced. Since the pH values of the intracellular environment are closely related to cellular metabolism, the metabolism of A431 and HaCaT cells was characterized and compared. Příprava a charakterizace nanoliposomálních nosičů hydrofobních cytostatik s využitím nanofluidního směšování / Preparation and characterization of nanoliposomal carriers of hydrophobic cytostatics using nanofluidic mixing

This diploma thesis is focused on preparation of liposome by relatively new method called nanofluidisation. This method allows the controlled preparation of small unilamellar liposomes in one step. In my thesis I was dealing with optimalization of liposomes preparation which carry hydrophobic cytostatics using this method. Cytotoxic effect of liposomes carrying hydrophobic cytostatics in vitro on cell lines A549 and MCF-7 was determined. In cytotoxicyty test I compared the effect of hydrophobic cytostatics (paclitaxel and derivates of vitamin E specifically alfa-Tos, alfa-TEA) that were incorporated into liposomes prepared via nanofluidisation method and lipid film hydration method. Moreover, a technology of lyophilisation in the presence of cryoprotectants for preparation of liposomes using the method of nanofluidisation was developed.

Změna v občanské společnosti? Souvislost globalizace a sociokulturní štěpící linie s růstem populismu / Change in Civil Society? Connecting Globalisation and Sociocultural Cleavage with the Rise of Populism

This thesis employs the globalization and integration-demarcation cleavage theory formulated by Huttar [2014] and Kriesi [2012], conception of populism formulated by Mudde [2017] and draws on Gramscian conception of civil society and hegemony. Aim of this thesis is to build a model of causal influence of globalization on cleavage and on populism, as was suggested by Hutter [2014]. After building this model, the aim is to explore how this theoretical relationship hold's over the 30 years since 90's, when the connection between globalization and new sociocultural cleavage had been theoretically suggested. For this model I am using KOF Globalization Index, European Values Survey datasets and Authoritarian Populism Index constructed and published by Timbro in years 1990, 1999, 2008 and 2017. This model is built on a dataset containing 38 countries on European continent or being a candidate country for EU. I am elaborating Hutter's theoretical suggestion and framing it in Gramscian conception of civil society. This allows me to suggest that populists are using organic crisis in a society to attract people who feel disjointed from current hegemonical elite and to create counterhegemony. Kinase Src plays an essential role in signal transduction from activated surface receptors. Src is involved in signal pathways that participate in the control of cell proliferation, differentiation or motility. That is why Src activation undergoes strict and complex regulation. Inactive conformation is maintained by intramolecular inhibitory interactions. SH3 domain associates with a polyprolin helix in CD linker whereas SH2 domain binds phosphorylated C-terminal tyrosine 527. Both regulatory domains maintain contacts with the lobes of a kinase domain thereby stabilizing an inactive conformation of the catalytic domain. Transition to an active state is accompanied by a disruption of these inhibitory interactions. Conformation changes are substantially influenced by the phosphorylation status of key tyrosines 416 and 527. Phosphoproteomic analysis revealed new Src tyrosine residue, which can be phosphorylated in vivo. It has been found, that tyrosine works as an additional regulator of Src activity. This is Tyr 90, which forms one of the hydrophobic pockets in the binding surface of Src SH3 domain. Realizace programu uchování přeštického černostrakatého plemene prasat

The aim of the thesis was to capture trends in population of Prestice Black Pied breed. In the literature section there was described formation and regeneration of breed characteristics and typical features of Prestice Black Pied breed, breed performance, the structure of the breeding base and the expected utilization and marketing. In own work there were described changes in the breeding base, which were in western Bohemia and moved to other places in the country. The largest companies that breed Prestice Black Pied breed are the ZD Mladotice and Žihelský statek SpA, located in the Plzeň region and Zemet Ltd. Tečovice, based in Zlin region and Chovservis Inc. Radostov breeding, based in Hradec Kralove region. In 2013 it was bred 348 sows, in 2014 404 sows, 2015 sows and 413 in 2016, 369 sows. Furthermore, the age structure was evaluated in basic herd of sows. The farmers had the most sows in the first and second litter, compared to that of the fifth litter sows had a few breeders. The requirement for longevity therefore not been complied with. The results of performance tests were evaluated reproductive performance of sows of basic herd in collections (the number of all born piglets, the number of live born piglets, the number of surviving piglets and milkiness). The average value of a breeding population did not meet the target. Some companies, however, had excellent results, especially ZD Mladotice and Zemet Ltd. Tečovice. In field tests were evaluated gain, the percentage of lean meat and backfat thickness. Breeding goal was achieved with values backfat thickness and percentage of lean meat. Gilts had lower gain than required by breeding goal. The gain for young boars has been in 2013 and 2016 higher than the required target and breeding in 2014 and 2015 below the required target. There were also evaluated linear track sires in breeding. It was found that, of the 10 extant lines has long represented the biggest line Wiskont and least represented of the Sudet line. The entire work was focused on monitoring results of individual breeders and their comparison with the breeding goal for Prestice Black Pied breed. Furthermore, the evaluation of the quality of semen of boars Prestice Black Pied breed, operating in the artificial insemination center in Kout Na Šumavě. All boars met the minimum parameters.

Planning is a very important manager work which is necessary for governing and functioning every society. The aim of this diploma paper is to analyse the system of financial plans, define planning of society, whether it is effective and reliable, describe setting short run and long run plans. For these case study was chosen a leasing company which plans on the base of accountancy of international standards, the medium {--} term plan and at that time also the contemporary plan is checked. The medium {--} term plan relates with expenses and profits. The plan is effected by the market share, environment and the market competition. The leasing company planes for three forefronts {--} cars, consumer goods and direct loans. Significant influence on the product, if it is profitable or not, has delinquency, which is the highest in the first year of the repayment. The company creates provisions for delinquent contracts. In the first phase of ratification insurance some products is necessary to be a part of the products. The Planning department and Department of Control are in charge of following forefronts of individual product. Conclude that the leasing company is in its planning and implementation successful. Profitable products of all product forefronts are the result. Cars, with their highest position in the market, are the star of the company. Nonprofitable products are cancelled and there are substituted by other products. Liquidity is not followed much because the company is owned by a multinational company.

The master thesis "Comparison of the development of party systems in the Czech Republic and in the Slovak Republic in 2010-2020" is focused on the analysis of the development of the Czech and Slovak party system in the years 2010-2020 and their mutual comparison. So the milestone for the analysis was the years 2010-2020. During this period there were fundamental changes in the Czech and Slovak party systems, which was the main reason for choosing this topic. The individual periods of development of both party systems will be defined by parliamentary elections to the Chamber of Deputies of the Parliament of the Czech Republic and to the National Council of the Slovak Republic. The analysis is based on the methods mentioned in the theoretical part of the work and on the classical typologies of party systems of M. Duverger, J. Blondel and G. Sartori. In the analysis itself, electoral statistics, surveys of electoral preferences and media sources are largely used. Among the main criteria for the analysis, the number and size of political parties and their relevance were determined. The relevance means the determination of coalition or blackmail potential. RNA interference (RNAi) is a process mediated by small RNAs (sRNA), which is significantly involved in the regulation of gene expression in plants. Diverse RNAi pathways can be divided into two basic mechanisms, which are post-transcriptional and transcriptional gene silencing (PTGS and TGS). Production of sRNAs is dependent on the presence of a double-stranded RNA molecule (dsRNA), which is cleaved by one of DCL proteins to produce sRNAs usually of 21-24 nt in length. One strand of the sRNA is subsequently loaded onto AGO protein. During PTGS, the AGO-sRNA complex interacts with the target RNA based on its sequence complementarity to the sRNA and cleaves it or blocks its translation. In the case of TGS, AGO interacts with plant-specific RNA Pol V and its transcripts, which are again complementary to the sRNA. This interaction allows assembling of a protein complex facilitating DNA and histone methylation inhibiting RNA Pol II transcription. There are numerous ways the dsRNA can arise. A significant part of dsRNA cell production is dependent on synthesising the complementary strand of the dsRNA by RDR6 (RNA-dependent RNA polymerase 6).
