Analýza chovu plemene Shagya-arab v České republice

The aim of this thesis was the first part of their work to process data on the number of foals born, import and export, trends in breeds of horses Shagya Arabian (Sh-a) and data horse's active Sh-a in the Czech Republic for the year 2014. The data were processed in Microsoft Office Excel 2007 and have created graphs. However, the number of foals born, it was found that most of them were born in 2009 and 2002. Import greatly outweighed exports, but in recent years also began to flourish and export. States Shagya-Arabians in the Czech Republic since 1994, continues to grow. In 2014, the Czech Republic 283 broodmares, 176 geldings, 142 mares, 133 stallions and 31 sire. In the second part of their work was the aim statistically evaluate data mares entered in the Stud book. This program was used Unistat 5.5. First, the statistical effect was studied by GLM and positive results have been used multiple comparisons by Tukey-B. It was found that on the basic colour has a highly significant influence line and family. Were examined influences on 4 basic body measurements (withers high tape, withers high rod, chest circumference, circumference of the cannon bone), where it was found that withers high tape, withers high rod, and chest circumference have a statistically significant effect of a family and line. The overall gain points is statistically highly significant influence lines and the number of ox 4th generation.

Antioxidační, anti-proliferační a imunomodulační účinky ovoce, zeleniny a hub in vitro / Antioxidant, anti-proliferative and immunomodulatory effect of fruits, vegetables and mushrooms in vitro

Recently there has been an increasing interest in discovering of new species of plants and mushrooms which have antioxidant or anti proliferative activity. The interest is caused by the fact that these species have medicinal and food utilization. These properties of plants and mushrooms can be used when dealing with many diseases which may be connected with oxidative stress (inflammatory bowel disease, cardiovascular disease, hypertension, and tumour disease, etc.). The thesis propounds characteristics of in vitro antioxidant and anti proliferative activity of 19 types of juices and 28 methanol extracts of fruits and vegetables, which are common parts of a diet. In the thesis there are also characteristics of 13 ethanol extracts of edible mushrooms of the order of Polypore (Popyporales). Antioxidant activity was quantifie by 2, 2 diphenyl 1 picrylhydrazyl (DPPH), oxygen radical absorbance capacity (ORAC), and inhibition of nitric oxide (NO) production. Cytotoxicity was measured by MTT (3 (4,5 dimethylthiazol 2 yl) 2,5 diphenyltetrazolium bromide. Immunomodulatory activity was determined by an assay of phagocytic activity of human neutrophil granulocytes. Further a total phenolic content (TPC) and total beta glucan content were investigated; these are considered to be the potentially active constituents participating in mentioned activities. From the obtained results it is evident that juices (TPC = 1603.2 mg GAE/L; ORAC = 438.5 umol TE/g) and a bilberry (Vaccinium myrtillus) fruit extract (ORAC 836.6 umol TE/g; DPPH = 404.6 umol TE/g) showed the highest antioxidant activity of all tested samples of fruits and vegetables. Capsicum (Capsicum L.) juices (TPC = 642.1 mg GAE/L; ORAC = 127.9 umol TE/g) and a radish (Raphanus sativus L.) extract (ORAC 724.5 umol TE/g; DPPH = 52.2 umol TE/g) also proved to have the high values of antioxidant activity. From the tested fruits and vegetables the following inhibited to produce a nitric oxide: onion (Allium cepa L) juices (lowered the NO production by 57%), tangerine juices (Citrus reticulata Blanco)(by 52%), broccoli (Brassica oleracea var. botrytis italic) extract (by 21%), and orange (Citrus sinensis Pers.) extract (by 10%). Concerning edible mushrooms, Lentinus tigrinus (Bull.) Fr. (TPC = 216.2 umol GAE/g of extract), Ganoderma lucidum (Curtis) P. Kras (TPC = 257.9 umol GAE/g of extract), and Royoporus badius (Pers.) A.B. De (TPC = 257.8 umol GAE/g of extract) presented the highest phenolic content. Sparassis crispa (Wulfen) Fr. (117.4 mg/g of extract) had the highest content of beta glucan. Substantial effect on phagocytic activity of granulocytes was noticed in connection with Neolentinus lepideus (Fr.) Redhead & Ginns, Polyporus squamosus (Huds.) Fr., and S. crispa. In connection with the latter of mentioned mushrooms, a moderate inhibitory effect towards HT 29 human colorectal adenocarcinoma cell line (IC50 = 107 ug/ml of extract) was noticed. The results indicate that the tested plants and mushrooms can be beneficial when reducing the negative effects of oxidative stress. The oxidative stress has been recognized as a contributing factor to a whole range of diseases and the reduction of the oxidative stress may lead to the decrease in the possibility of the diseases´ progression. The results show the possible favourable effects on the human health. When evaluating the results, the following fact should be taken into consideration; the in vitro tests and screenings are considered to be the first phase of systematic research of the effects and serve for the choice of respondents for further detailed studies.

It participates in the inflammatory reaction in the distinct compartments of the living organism. As a transcription factor, it controls the gene expression of many genes, especially cytokines (tumor necrosis factor alfa, interleukins: IL-1β, IL-2, IL-6, IL-12; chemokines etc.). NF-κB is also a key factor in the activation of monocytes and macrophages In this diploma thesis I focused on the role of NF-κB in the monocyte cell line THP-1. This line is an important model of human macrophages in which the THP-1 line can be differentiated. Using available literature, I summarized all the available knowledge on this issue. At the same time, I conducted several experiments on NF-κB activation in the THP- 1 line as a potential model in the research and development of therapeutic intervention in NF-κB signaling to suppress inflammation.

The goal of this thesis is the analysis of the milk yield on a particular organic farm breeding goats. The milk yield was monitored and evaluated in the control range 2015-2017. The data was obtained from the yield checks carried out by SCHOK (Association of Goat and Sheep Breeders) and from the farm's own records. The testing was run on 526 White Shorthaired goats yielding 760,08 kg of milk for standard lactation with a fat content of 3.4% in 2017. There has been a trend of rising milk yields on the farm since 2015 when the yield reached 627.77 kg of milk with a fat content of 3.02%. This trend was statistically highly potent (P<0.01), as well as the protein content (%) in milk which has dropped from 2.97% to 2.73% during the the same period of time. In addition, 184 goats on the 1st lactation and 177 goats on the 2nd lactation belonging to 16 father lines were evaluated. These lines were compared to each other by individual milk yield indicators. In the lactose content (%) the Krasus line dominated (4.83%) which showed statistically significant difference (P<0.05) compared to the Molch and Emil lines whose lactose content was the lowest (4.51% and 4.53% respectively). The 2nd lactation testing evaluated the number of somatic cells in the milk, with an average value of the group monitored being 1543.12 thousand of somatic cells per one mililiter. The highest number of the SC was measured in the Bera line (4437.17 thousand SC/ml) with statistical conclusiveness (P<0.05) compared to Bernd (758.5 thousand SC/ml), Curt (858.42 thousand SC/ml), Zub (444.24 thousand SC/ml), and Zuzek (696.77 thousand SC/ml) lines which reached the lowest values. In other observed milk yield parameters, the lines on both lactations showed marked differences in the values achieved but they were not statistically proven.

Title of diploma thesis: Determination of organ toxicity of BRAF inhibitors in vitro. Malignant melanoma is one of the most serious skin diseases today. Therapy of advanced melanoma is difficult and often ineffective. BRAF inhibitors (dabrafenib and vemurafenib) have dramatically changed the results of melanoma treatment in the last few years. BRAF inhibitors are one of the most effective drugs against melanoma, but their clinical application is largely limited by drug resistance. Available clinical studies have shown an adverse nephrotoxic effect of BRAF inhibitors, but information on its mechanism is limited. Published studies further suggest that the toxic effect of BRAF inhibitors is primarily directed to podocytes located in the glomerular membrane. Thus, the aim of our study was to assess the cytotoxic effect of BRAF inhibitors on selected model renal cells in vitro in order to confirm the renal target toxicity. The main objective of the study was to analyse whether the nephrotoxic effect of BRAF inhibitors is specifically limited to podocytes or whether it can damage other renal cells. They work on the principle of inhibition of certain types of protein kinases and turned out to be very efficient for the treatment of melanoma. One of their disadvantages is relatively early onset of resistance; thus, it is important to look for new combinations of drugs that are already in use or work on the development of new structures with similar inhibition efficacy on melanoma cells. Encorafenib and its combination with binimetinib have been shown to be very promising drugs from the group of BRAF inhibitors, however, potential renal toxicity may be a therapeutic limitation. This thesis was focused on the determination of in vitro cytotoxicity of encorafenib on different types of renal cells in three time intervals and on its comparison with two drug standards - amphotericin B and paracetamol. Three types of morphologically and functionally different kidney cells (PODO / TERT256, HK-2 and HEK293) were used for this purpose.
