A total of 30 goats, 15 were aged 8-9 months and 15 with 11-12 months of age. Of each goat were evaluated Longissimus dorsi (LD), Triceps brachii (TB), Infraspinatus (IS) and Biceps femoris (BF) to determine the proximate composition (moisture, protein, ash and lipids), cholesterol and fatty acids profile acids. The experimental design was completely randomized with a factorial 2 x 3 (ages two and three regions). There was significant interaction between age and the region of slaughter (muscle) studied. Goats slaughtered younger have higher concentrations of moisture and mineral matter on the back, shoulder and leg (P <0.05) compared to older animals. Since fat content were higher in goats slaughtered older (P <0.05). For cholesterol, only the leg of goats slaughtered younger levels were lower than other regions (P <0.05). There was no difference in the total cholesterol to goats slaughtered older (P> 0.05) in different regions of the carcass. Goats slaughtered older (11-12 months) had higher levels of C12:0, C14:0, C15:0, C16:0 and C18:0 (P <0.05) in muscle (LD). As for the muscles (TB) and (IS) there was no difference in C15:0 and C16:0. The same was observed for the muscle (BF) (P> 0.05). For the fatty acid C18:1 trans-9 and C18:1 trans-11, the goats slaughtered older had higher concentrations (P <0.05) in LD and BF muscles. While the muscles TB and IS only statistical difference (P <0.05) for C18: 1 trans-9, higher in older animals slaughtered. Regarding the content of polyunsaturated acids C18:2, C18:3, C20:3, C20:4, C20:5, C22:5 and C22:6 were higher in young kids (P <0.05) in the muscle LD. While TB and IS the C20:4 and C20:5 were higher in older kids. BF in the C18: 2 cis9, trans-11 (CLA) was higher in older kids (P <0.05), other muscles in this fatty acid did not differ (P> 0.05). With the progress of the slaughter of goats rises the amount of protein and fat, but cholesterol does not change. The regions of the substrate showed different chemical composition. There is direct relationship between age at slaughter increased with fat saturation and concentration of conjugated linoleic acid. The goat meat is a source of fat omega 3 and 6, showing a relationship beneficial to health. / O trabalho teve como objetivo avaliar o efeito da idade de abate sobre a composição química, colesterol e perfil de ácidos graxos da carne de cabritos criados na região do Alto Camaquã. Foram utilizados 30 cabritos, sendo 15 com idade de 8 9 meses e 15 com 11 12 meses de idade. De cada cabrito foram avaliados os músculos Longissimus dorsi (LD), Triceps brachii (TB), Infraspinatus (IS) e Biceps femoris (BF) para determinação da composição centesimal (umidade, proteína, matéria mineral e lipídios), colesterol e perfil de ácidos graxos. O delineamento utilizado foi o inteiramente casualizado, com arranjo fatorial 2 x 3 (duas idades e três regiões). Houve interação significativa entre a idade de abate e a região (músculo) estudada. Cabritos abatidos mais jovens apresentam maior concentração de umidade e matéria mineral no lombo, paleta e perna (P<0,05) em relação aos animais mais velhos. Já os teores de gordura foram superiores nos cabritos abatidos mais velhos (P<0,05). Para o colesterol, somente na perna de cabritos abatidos mais jovens os teores foram inferiores as demais regiões (P<0,05). Não houve diferença em relação ao colesterol total para cabritos abatidos mais velhos (P>0,05) nas diferentes regiões da carcaça. Cabritos abatidos mais velhos (11 12 meses) apresentam maiores teores de C12:0, C14:0, C15:0, C16:0 e C18:0 (P<0,05) no músculo (LD). Enquanto, para os músculos (TB) e (IS) não houve diferença para C15:0 e C16:0. O mesmo foi observado para o musculo (BF) (P>0,05). Para os ácidos graxos C18:1 trans-9 e C18:1 trans-11, os cabritos abatidos mais velhos apresentaram maiores concentrações (P<0,05) nos músculos LD e BF. Enquanto nos músculos TB e IS somente houve diferença estatística (P<0,05) para C18:1 trans-9, superior nos animais abatidos mais velhos. Em relação aos ácidos poliinsaturados os teores de C18:2, C18:3, C20:3, C20:4, C20:5, C22:5 e C22:6 foram superiores nos cabritos mais jovens (P<0,05) no músculo LD. Enquanto para TB e IS o C20:4 e C20:5 foram superiores nos cabritos mais velhos. No BF o C18:2 cis9, trans11 (CLA) foi superior nos cabritos mais velhos (P<0,05), nos demais músculos este ácido graxo não diferiu (P>0,05). Com o avanço da idade de abate dos cabritos eleva-se o teor de proteína e de gordura, porém o colesterol não se modifica. As regiões da carcaça apresentaram distinta composição química. Há relação direta entre a idade de abate com o aumento na saturação da gordura e concentração do ácido linoleico conjugado. A carne de cabrito é fonte de gordura ômega 3 e 6, apresentando uma relação benéfica a saúde.

The feeding was based on the ratio of 60% forage (alfalfa hay), and 40% commercial concentrate with two levels of lipids (3% and 5%). The variables were: average milk production, behavior of the lactation curve, milk chemical composition and total time of lactation against the profile of saturated, monounsaturated and polyunsaturated. The increase in the percentage of ether extract oil-based rice did not affect milk production, which presented as mean ± 2658 379 mL milk / goat / day, between 11 weeks and the chemical composition of milk, which characterized by having 3.78 ± 0.59% fat, 3.10 ± 0.18% crude protein, 4.46 ± 0.20% lactose and 12.19 ± 0.72% of total solids. However significant (P <0.05) fatty acid profile of milk, reducing the concentrations of saturated fatty acids, palmitic acid C16: 0, myristic C14: 0, capric C10: 0 and lauric C12: 0, which were higher in 3% treatment (26.87, 8.60, 5.90 and 3.06% respectively) and increasing concentrations of unsaturated C16: 1, C17: 1 and C20: 1 (0.43, 0.12 and 0.10%, respectively). The inclusion of rice bran oil at levels of 3% and 5% of dry matter in the diet of goats does not affect milk production and the chemical composition until the 11th week of lactation, however, is able to alter the fatty acid profile of significantly. / Testou-se o efeito de dois níveis de extrato etéreo a base de óleo de arroz no concentrado de 30 cabras Saanen durante a fase inicial de lactação. A alimentação baseou-se na relação 60% de volumoso (feno de alfafa), e 40% de concentrado comercial com dois níveis de extrato etéreo (3% e 5%). As variáveis estudadas foram: Produção, composição química, perfil de ácidos graxos saturados, mono e polinsaturados no leite. O aumento no percentual de extrato etéreo na ração não influenciou a produção de leite, que apresentou como média 2658±379 mL de leite/cabra/dia, no período de 11 semanas e nem a composição química do leite, que se caracterizou por ter 3,78 de gordura; 3,10 de proteína bruta; 4,46 de lactose e 12,19 de sólidos totais. O perfil de ácidos graxos do leite foi influenciado pelo acréscimo de extrato etéreo na ração. No tratamento de maior proporção houve um aumento das concentrações de ácidos graxos insaturados C18:1n 9c (Oléico) e C18:3n3, (25,79 e 0,80%, respectivamente) e no Ácido Linoléico Conjugada (CLA) que aumentou de 0,37 para 0,72% com o acréscimo de óleo; e uma redução dos ácidos graxos saturados: palmítico C16:0, mirístico C14:0, cáprico C10:0 e láurico C12:0, que foram maiores no tratamento 3% (26,87; 8,60; 5,90 e 3,06%, respectivamente).

Atualmente no Brasil mais da metade da população adulta tem excesso de peso e 21% estão obesos. A obesidade é uma doença que se encontra em evidente crescimento, sendo considerada a epidemia do século XXI. Como alternativa de tratamento e prevenção, o uso de ácidos graxos que possuem habilidade de induzir a expressão de genes com importante papel em modulações metabólicas e mitocondriais têm sido estudados. O ácido linoleico conjugado (CLA, 18:2) é da família ômega-6, descrito por sua propriedade antiobesidade relacionada à diminuição da adiposidade e ao aumento do metabolismo corporal. O óleo de peixe (OP) é uma mistura de ácidos graxos poli-insaturados eicosapentaenóico (EPA, 20:5) e docosahexaenóico (DHA, 22:6) da família ômega-3, conhecido por aumentar a sensibilidade à insulina, o colesterol-HDL, pelas suas propriedades antiinflamatórias e sua ação protetora sobre o sistema nervoso. O objetivo deste estudo foi verificar os efeitos da suplementação da dieta de camundongos C57BL6 com CLA em conjunto com OP durante 60 dias sobre aspectos bioquímicos, moleculares e fisiológicos do metabolismo mitocondrial e corporal. Verificamos que a suplementação da dieta com CLA e OP in vivo: aumenta o metabolismo corporal, efeito atribuído à ambos os óleos; prejudica o metabolismo da glicose circulante, proporcionado exclusivamente pelo CLA; reduz o nível de movimentação, proporcionado exclusivamente pelo OP. No fígado: aumenta a expressão de UCP2, a atividade de proteínas desacopladoras e a ?- oxidação, efeito atribuído à ambos os óleos; aumenta o número de mitocôndrias, proporcionado exclusivamente pelo OP. CLA aumenta a produção de espécies reativas de O2 (EROs) a qual é revertida pelo OP em conjunto. No músculo sóleo: aumenta a biogênese mitocondrial via PGC- 1? e a expressão de UCP2, proporcionados pelo OP. Por fim, no hipocampo: aumenta o número de mitocôndrias, estimulo dado por ambos os óleos; aumenta a atividade desacopladora e reduz a produção de EROs, proporcionados exclusivamente pelo CLA. Concluímos que na suplementação conjunta os efeitos do OP são predominantes nos metabolismos corporal, hepático e muscular, e na movimentação corporal, enquanto que o efeito predominante do CLA é a diminuição na sensibilidade à insulina. Já no cérebro, o OP potencializa os efeitos do CLA. / Currently in Brazil more than a half of adult population has overweight, and 21% are obese. Some fatty acids have been considered an alternative treatment and prevention strategy for obesity due to their ability to stimulate gene expression with important role in cellular and mitochondrial metabolisms. Conjugated linoleic acid (CLA, 18:2) from omega-6 family, with anti-obesity properties related to diminution of adiposity and increments in body metabolism. The fish oil (FO) is a mixture of the poli-unsaturated fatty acids eicosapentaenoic (EPA, 20:5) and docosahexaenoic (DHA, 22:6) from omega-3 family, known for improving insulin sensibility and HDL-cholesterol, anti-inflammatory properties and protective action over the central nervous system. The objective of this study was to examine the effects of dietary supplementation of CLA in conjunction with FO during 60 days over biochemical, molecular and physiological aspects of mitochondrial and body metabolism in C57BL6 mice. Diet supplementation with CLA and FO in vivo: raise body metabolism, an effect attributed to both oils; affect glucose metabolism, exclusively proportionate by CLA; diminish the level of mice movement, exclusively proportionate by FO. In liver: increase UCP2 expression, uncoupling proteins activity and ?-oxidation, stimulated by both oils; increase mitochondrial density, exclusively proportionate by FO. CLA also raises the reactive oxygen species (ROS) production, which is reversed by FO in conjunction. In soleus muscle: increase mitochondrial biogenesis through PGC-1? and the UCP2 expression, exclusively proportionate by FO. Lastly, in hippocampus: increase mitochondrial density, stimulated by both oils; stimulate uncoupling activity and diminish ROS production, exclusively proportionate by CLA. In conclusion, in the dietary supplementation with CLA and FO in conjunction the FO effects are prevalent in metabolisms of body, liver and muscle, and in body movement, while the CLA effects are prevalent in decreasing insulin sensitivity. However in the brain, the FO potentiates the effects of CLA.

Monitoring a natural autoxidation process of methyl linoleate by using GC-MS

The methyl ester of an unsaturated fatty acid, methyllinoleate was reacted with oxygen in a pressurized system at a controlled temperature. A natural autoxidation of methyl linoleate was observed without the addition of an initiating reagent. This autoxidation process could be used to mimic the course of lipid peroxidation, which is the major cause ofradical damage to living cells. The technology of GC-MS was employed to monitor the autoxidation of methyllinoleate. Eight of the autoxidation products separated by GC column were identified by interpreting the corresponding EI ion mass spectra. The products from 9-alkoxy methyl linoleate radical were methyl octanate, 2,4-decadienal, nonanoic acid, 9-oxo-methyl ester, and its further oxidation product, nonanedioic acid, monomethyl ester. All of them formed through a pathway of beta-cleavage. The products from 13-alkoxy methyl linoleate radical were tridecanoic acid, 9, 11-diene-13-oxo-methyl ester, hex anal, and its further oxidation product, hexanoic acid. They were also formed through a mechanism of beta-cleavage. The fourth product from 13-alkoxy methyllinoleate radical was 13-keto-9, 11-octadecadienoic acid, methyl ester, which was obtained through a pathway of keto formation. Observation of their concentrations in the samples at different autoxidation periods revealed the time-course of formation of these products.

Modifying Fatty Acid Composition of Bovine Milk by Abomasal Infusion or Dietary Supplementation of Seed Oils or Fish Oil

The potential for enhancing oleic acid (cis-18:1) and linoleic acid (18:2) content and lowering medium chain fatty acid (MCFA) content of bovine milk was investigated by abomasal infusion or dietary supplementation of oils. In experiment 1, olive oil, sesame oil, sunflower oil, or fish oil was abomasally infused (155 to 219 g/d) into Jersey cows during the last 6 d of each of four 14-d periods. In experiment 2, canola oil, olive oil, high-oleic sunflower oil, or distilled water (control) was abomasally infused (342 to 371 g/d) into three Holsteins and three Jerseys during the last 5 d of each of four 10-d periods. The intestinal digestibility and concentration of cis-18:1 and 18:2 in milk were proportional to flow of these fatty acids to the duodenum. Also, greater concentration of cis-18:1 in milk was associated with lowered yield of MCFA. During olive oil or sesame oil infusion in experiment 1, for each 100 g of cis-18:1 infused into the abomasum, milk cis-18:1 yield was increased by an average of 47 g, and MCFA yield was reduced by 42 g. The yield of 18:2 in milk was increased by approximately 46 g for each 100 g of infused 18:2 during olive oil or sesame oil infusion. Milk produced during sesame oil infusion, however, had an off-flavor when evaluated by a taste panel. In experiment 2, each 100 g of cis-18:1 infused daily increased milk cis-18:1 yield in Holsteins and Jerseys by 41 and 39 g/d, respectively, whereas recovery of infused 18:2 was 34 g/d for Jerseys and 42 g/d for Holsteins. In experiment 3, 22 Jersey cows were fed a basal diet, or the basal diet supplemented with 3.5% high-oleic canola oil, 3.5% soybean oil, or 1.75% high-oleic canola oil plus 1.75% soybean oil for 5 wk. Dietary canola oil supplementation increased conjugated linoleic acid (CLA) percentage in milk to a moderate level without raising trans-18:1 percentage, whereas feeding either supplement containing soybean oil raised both CLA and trans-18:1 percentages. Concentrations of trans-18:1 and CLA in milk apparently reflected the extent of unsaturated fatty acid biohydrogenation in the rumen. Dietary supplementation with canola oil increased yield of cis-18:1 in milk by 21 g for each 100 g of supplemental cis-18:1 intake. Yield of 18:2 in milk was raised by 3 g for each 100 g of supplemental 18:2 intake by cows fed soybean oil. Using abomasal infusion as an indicator of the maximum potential for apparent recovery of cis-18:1 in milk (39 to 49%), cis-18:1 recovery in response to supplemental cis-18:1 in the diet was approximately half of the potential response due to partial biohydrogenation in the rumen. The apparent recovery of dietary 18:2 in milk was reduced to only one-tenth of the potential yield (31 to 47%) indicated by abomasal infusion of seed oils. Results indicated that the fatty acid profile of bovine milk was altered in a manner that would be beneficial to human health when cows were fed supplemental oleic acid, but further research should focus on safe and economical methods to protect dietary unsaturated fatty acids from biohydrogenation. / Ph. Akrylbaserat bencement är den gyllene standarden vid ryggförstärkningsförfaranden. Modifiering av akrylbencement med linolsyra har resulterat i attraktiva egenskaper vilka möjliggör lättare hantering för kirurger och minskar efterföljande komplikationer såsom närliggande ryggradsfrakturer. Även om de attraktiva egenskaperna hos linolsyramodifierat bencement är kända är ännu förståelsen för hur linolsyra påverkar egenskaperna outforskade. Som en komponent i bencement måste linolsyra steriliseras innan den används i bencement. Det finns emellertid oro för att autoklavsterilisering av linolsyra orsakar nedbrytning. Dessutom är det oklart vad som händer med linolsyra i härdat bencement över tid. I detta examensarbete utvärderades steriliserad och osteriliserad linolsyra. Linolsyra blandades med olika komponenter som finns i bencementet såsom aktivator, initiator, monomer och inhibitor, i närvaro och frånvaro av lösningsmedel. De efterföljande förändringarna studerades genom 1H NMR och UV-VIS. Resultaten visade att linolsyra bryts ned av sterilisering och oxidation. Oxidationen av linolsyra berodde på sterilisering, temperatur, lösningsmedel och mängden syrexponering. Det bekräftades genom 1H NMR och UV-VIS att linolsyra reagerade (steriliserad och osteriliserad) med aktivatorn, initiatorn och monomeren. Dessa reaktioner kan minska tillgängligheten av komponenterna för in situ-polymerisation av monomeren, och därmed förändra bencementens egenskaper, vilket i sin tur bidrar till lättare hantering under förstärkningsförfarandet och reducerar intilliggande ryggradsfrakturer efter operation. / Acrylic bone cement is the gold standard in vertebral augmentation procedures. Modification of acrylic bone cement using linoleic acid has resulted in attractive properties that enable convenient handling by surgeons and reduce follow-up complications such as adjacent vertebral fractures. Even though the attractive properties of linoleic acid-modified bone cement are acknowledged, the understanding of how it imparts those properties remain unexplored. As a component in bone cement, linoleic acid needs to be sterilized before its use in bone cement. However, there are apprehensions whether autoclave sterilization of linoleic acid causes degradation. In addition, it is unclear what happens with linoleic acid in hardened bone cement over time.  In this thesis, sterilized and unsterilized linoleic acid were evaluated alone and treated with different components of the bone cement such as activator, initiator, monomer, and inhibitor, in the presence and absence of solvent, and the ensuing changes were monitored through 1H NMR and UV-VIS. The results showed that linoleic acid degraded due to sterilization and oxidation. The oxidation of linoleic acid depended on sterilization, temperature, solvent, and the amount of oxygen exposure. As confirmed through 1H NMR and UV-VIS, linoleic acid (sterilized and unsterilized) reacted with the activator, initiator, and monomer. These reactions could reduce the availability of the components for the in-situ polymerization of the monomer, thus altering the properties of the bone cement including convenient handling during the operation and reduced adjacent vertebral fractures post the operation.

Stearoyl-COA Desaturase Gene Transcription, mRNA, And Activity In Response To Trans-Vaccenic Acid And Conjugated Linoleic Acid Isomers

Studies were conducted to investigate: 1) desaturation of dietary trans-vaccenic acid (TVA, trans11-18:1) to the cis9,trans11-18:2 isomer of conjugated linoleic acid (9/11CLA), 2) effects of two conjugated linoleic acid isomers [9/11CLA or trans10,cis12-18:2 (10/12CLA)] and TVA on enzyme activities and mRNA abundance for lipogenic enzymes, and 3) regulation of stearoyl-CoA desaturase (SCD) gene transcription. In the first study, lactating mice were fed 3% linoleic acid (LA), or 2% LA plus 1% stearic acid (SA), 1% TVA, or 1% CLA mixture. Dietary TVA enriched the 9/11CLA content of carcass, liver, and mammary tissue of lactating mice. A similar enhancement of 9/11CLA also was observed in liver, but not carcass, of suckling pups nursing TVA-fed dams. The CLA mixture decreased mammary acetyl-CoA carboxylase (ACC) activity compared with other treatments. However, total fatty acid content of mammary tissue was reduced only when compared with TVA. In the second experiment, lactating mice were fed 3% canola oil (OA), or 2% OA plus 1% SA, 1% TVA, 1% 9/11CLA, or 1% 10/12CLA. Dietary TVA, 9/11CLA, and 10/12CLA decreased mRNA abundance for ACC and fatty acid synthase (FAS) in mammary tissue, suggesting each had the potential to reduce de novo fatty acid synthesis. However, only the CLA isomers decreased ACC activity in mammary tissue and concentration of medium-chain fatty acids (MCFA = 12:0+14:0+16:0) in milk fat. The 10/12CLA isomer caused greater reductions in MCFA and milk fat percentage than the 9/11CLA, indicating that 10/12CLA is the primary CLA isomer affecting lipid metabolism in the mammary gland. Dietary TVA, 9/11CLA, or 10/12CLA decreased SCD enzyme activity and mRNA abundance in mammary tissue. In study 3, mouse (COMMA-D/MME) and bovine (Mac-T) mammary epithelial cells were transfected with the putative promoter (600 bp) of SCD gene. The 9/11CLA reduced SCD gene transcription in mouse cells, but not bovine cells. Transcription, however, was reduced in both cell lines by 10/12CLA, linoleic acid, and linolenic acid. Thus, reduced SCD transcription in response to the CLA isomers in mouse mammary cells in vitro may provide an explanation for reduced SCD enzyme activity and mRNA abundance in mammary tissue when lactating mice were fed either of the CLA isomers. In contrast, stearic acid, oleic acid, and TVA did not affect SCD transcription. Although TVA did not reduce SCD transcription in mouse mammary cells in vitro, it did reduce SCD enzyme activity and mRNA abundance in mammary tissue when fed to lactating mice. The results suggested TVA may influence SCD mRNA processing or stability in the nucleus after transcription. Despite the reduction in SCD mRNA and enzyme activity, however, substantial quantities of TVA were desaturated to the 9/11CLA isomer when TVA was fed to lactating mice in the first two studies. Thus, dietary TVA provides an alternate supply of the anticarcinogenic 9/11CLA isomer in tissues. / Ph. D.

Qualitative and Quantitative Assessment of Fatty Acids of Buddleja asiatica by GC-MS

No / To analyze the fatty acid contents of Buddleja asiatica Lour, both the non-volatile oil and fat obtained from the n-hexane soluble sub-fraction were subjected to GC/MS using BSTFA (N,O-bis(trimethylsilyl) trifloroacetamide) derivatization. The oil showed the presence of six fatty acids including palmitic acid (46.75 %), linoleic acid (37.80 %), stearic acid (10.98 %), arachidic acid, margaric acid and lignoceric acid (< 3 %). Analysis of the fat revealed nine fatty acids including lignoceric acid (43.12 %), behenic acid (26.39 %), arachidic acid (9.29 %) and stearic acid (5.3 %). Cerotic acid, montanic acid, melissic acid and palmitic acid were found in low amounts (< 5 %) while trycosylic acid (4.83 %) was the only fatty acid with odd number of carbon atoms.
