The theme of this diploma thesis is the manipulation in literature as a dialog between the I and the You or the I and the It. This I has a different position in each of the two types of manipulation. In the first type the I is the subject of manipulation - that is a manipulator. In the second one the I becomes the object of manipulation - thus a victim of a manipulator because the power over the I has been overtaken by other character's consciousness. The I and the You type is represented by an outside dialog (between characters), whereas the I and the It type is demonstrated in an inside dialog which does not occur between the hero and another character but between the hero and a voice of other character's consciousness. The opening part which deals with the manipulation in different contexts (philosophical, political and psychological) is followed by a part where I interpret the manipulation's representation in literature, especially in the works of Dostojevskij (The Double, The Village of Stepanchikovo, Notes from Underground, The Eternal Husband). With the characters of this author we can view the dark side of the human mind and also have a close look at various forms of arbitrariness, especially at the manipulation with the others.

The disertation thesis deals with the market abuse through the market manipulation. Its focuses, in particular, to the merits of the regulation itself and specific regimentation. Manipulation is one of two pillars which creates the subject of the area of law dealing with market abuse. The thesis is divided into five basic parts, which are further divided into seventeen chapters. We proceed from the general definition of basic concepts, categories, and economic context to clarify a particular concept of manipulation and its operation in practice to particular subject matters of manipulation. The conclusion is then added to the list of negative description and liability arising from manipulation. The first five chapters, which are contained in Part A deals with general introductory remarks (Chapter 1), including notes dealing with the concept and methodology of the thesis, its objectives and determination of verified hypotheses (Chapters 3, 4 and 5). Chapter 2 of this part is partially devoted to the issue of terminology used in the thesis. We note here the fact the existing regulation limiting ratio of the concepts and conclude that this thesis will use principally general terms, i.e the market (instead of the concept of capital or regulated market) and instrument (instead of the term financial...

Within my diploma thesis I have been trying to resolve the possibilities of optimization of artificial incubation of eggs and embryos in northern pike (Esox Lucius L.) under controlled conditions during three different experiments. In the first experiment was investigated the effect of three different semination solutions to the success of incubation of eggs and embryos in northern pike. I watched the survival of eggs and embryos, larvae hatching abnormalities and biometrics of larvae. It was found that the best overall results were achieved with semination solution composed of 15 ml of chemical crystalic urea per 1 liter of water, with an appropriate amount of abnormalities and highest larvae survival. In the second experiment were performed manipulation with developing eggs at given time intervals after fertilization of eggs. I watched the survival of eggs and embryos, abnormality and biometrics of hatched larvae. It was found that the lowest survival, abnormalities, and smallest TL and W were reached during manipulations within 24 hours after fertilization. In the third experiment was investigated the effect of water temperature on the length of the incubation of eggs and embryos in northern pike. With regard to the overall survival, larvae hatching, abnormalities and size of larvae achieved the best results temperatures 6 and 10 ° C. Conversely, it is entirely unsuitable for incubation temperature of 3 ° C (lowest survival, large amounts deformities). Different incubation temperatures have also confirmed the effect of water temperature on biometrics of hatched larvae.

miRNAs are small regulatory RNAs, which function as post-transcriptional mRNA regulators. They direct ribonucleoprotein complexes to cognate mRNA to repress them by translational inhibition and degradation. miRNAs regulate thousands of mRNAs in mammals and have been recognized as regulatory factors in most cellular and developmental processes. Dysregulation of the miRNA pathway can lead to severe defects and diseases. Interestingly, a unique situation exists in mouse oocytes, where all the miRNA pathway components are present, yet the pathway is dispensable and nonfunctional, the molecular foundation of this phenomenon and its significance still remain unclear. In spite of the pronounced effects of the miRNA pathway in gene regulation in somatic cells, study strategies of the pathway bare limitations. Current methods for studying the activity of the miRNA pathway employ corelative studies (such as NGS) or reporter assays, which have relatively low throughput and are prone to artifacts. Here, I present design and development of a new strategy for directly monitor global miRNA pathway activity and integrity in near physiological conditions in living cells, which could also be employed in vivo for studies of mouse oocytes. The main goal of this graduation thesis is to compose a set of special exercises with ball for sport game lessons at the primary school and to check whether it is more convenient and more interesting to use this special exercise than the classics gymnastics exercises. In my work, it is assumed that using this special warm-up exercise will conclude in the improvement in the fields of the children{\crq}s manipulation skills with the ball, their preferred sport game and that the lessons will be more attractive and entertaining for them.

Ověření dědičnosti barev okrasného kapra koi s využitím metod genomových manipulací / The assessment of colour inheritance in ornamental koi carp with the use of genome manipulation methodsŠTĚCH, Luděk January 2009 (has links)
The goal of this thesis was to assess colour inheritance in Showa tricolour variety of koi carp (black basis, white and red signs). Fish of the same colouration were pair-mated. Colouration of fingerlings was assessed in one day age. Colouration was observed in two forms, like melanin pigmentation and non-melanin pigmentation (black- pigmented and gold -pigmented fingerlings). In the tests, gynogenesis was employed, transmissing only mother`s genome to offspring. Gynogenesis was practiced in RIFCH USB in Vodnany. Gynogenetic offspring was further photographically documented to assess colouration of fingerlings and relationship between both variants of colouration. Fry from propagation was bred in ponds to the size stage of advanced fry (Kr; 90-day old, size 5- 6 cm). In this size, fingerlings were caught from breeding ponds, counted and further documented to check phenotype segregation.
