Determinação e quantificação das vitaminas C e E associadas em produtos cosméticos / Determination and quantification of vitamins C and E associated in cosmetic products

A vitamina C e a vitamina E são antioxidantes naturais encontrados em diversas frutas e verduras frescas, em óleos vegetais e germem de trigo. Produtos cosméticos, contendo essas vitaminas em forma de ésteres, têm sido desenvolvidos e disponibilizados comercialmente, ressaltando-se que o aspecto natural dessas induz seu uso em cremes e loções. Por isso, torna-se necessário a validação de metodologias analíticas para identificar essas vitaminas em preparações cosméticas, tanto para o controle da qualidade do produto como para a segurança do consumidor. O presente trabalho tem como objetivo a validação das metodologias analíticas para determinação e quantificação do derivado do ácido ascórbico, tetraisopalmitato de ascorbila e o princípio ativo acetato de tocoferila que estão associados nas formulações cosméticas. A técnica de espectrofotometria no ultravioleta (UV) e a análise térmica foram empregadas na caracterização desses princípios ativos. Para a quantificação dos ativos foram desenvolvidas metodologias analíticas empregando as seguintes técnicas: espectrofotometria derivada no UV; eletroforese capilar e cromatografia líquida de alta eficiência (CLAE). A metodologia desenvolvida por espectrofotometria derivada no UV não demonstrou ser aplicável devido à interferência dos excipientes presentes na formulação. Para determinação dos ativos tetraisopalmitato de ascorbila e acetato de tocoferila, foi validado um método por CLAE em fase reversa, empregando-se a coluna Synergi Hidro® C18, fase móvel metanol:2-propanol (25:75), vazão 1,0 mL/min e detecçao UV à 222 nm. As amostras foram extraídas sem grande complexidade e os ativos puderam ser determinados em 3 min e 6 min, para o acetato de tocoferila e o tetraisopalmitato de ascorbila respectivamente. O desenvolvimento da metodologia por eletroforese capilar utilizando-se a técnica de MEEKC (Cromatografia Eletrocinética em Microemulsão) usando a microemulsão óleo-aquosa não demonstrou ser adequada para determinação dos princípios ativos em estudo, sendo que o trabalho terá como perspectivas a tentativa da MEEKC usando a microemulsão aquosa-oleosa. / Vitamin C and vitamin E are natural antioxidants found in many fruits and fresh vegetables, vegetable oils and seed wheat. Cosmetics containing these vitamins in the form of esters have been developed and made available commercially, highlighting the fact that the \"natural\" induces its use in these creams and lotions. Than, it is necessary the validation of analytical methodologies to identify these vitamins in cosmetic preparations, both for the control of product quality and for the safety of consumers. The present work aims the validation of analytical methodologies for determining and quantifying the derivative of ascorbic acid, ascorbyl tetraisopalmitate and the active tocopheryl acetate that are associated in cosmetic formulations. The techniques of UV spectrophotometry and the thermal analysis were used in the characterization of these active ingredients. For the quantification of the actives, analytical methodologies were developed using the following techniques: the UV first-derivative spectrophotometry (UVD), high performance liquid chromatography (HPLC) and capillary electrophoresis (CE). The methodology developed by UV first-derivative spectrophotometry do not showed to be applicable due to the interference of excipients in the formulation. To determine the actives ascorbyl tetraisopalmitate and tocopheryl acetate, a method has been validated by reverse-phase HPLC, using the column Synergi Hidro ® C18, mobile phase methanol :2-propanol (25:75), flow rate 1.0 mL / min and UV detection 222 nm. The samples were extracted without great complexity and the actives could be determined in 3 min and 6 min, for the tocopheryl acetate and ascorbyl tetraisopalmitate respectively. The development of methodology for capillary electrophoresis using the technique of MEEKC (Microemulsion Electrokinetic Chromatography) using the oil-in-water (o/w) microemulsion do not showed to be suitable for determination of active ingredients in the study, and that the work will attempt the prospects of using MEEKC the water-in-oil (w/o) microemulsion.

Analýza zainteresovaných stran v projektech ICT / Stakeholder management in ICT projects

This work deals with the questions of the analysis and management of the parts engaged in information and communication technology projects. The stakeholder (the term "stakeholder" is used in Czech as well) is a subject, i.e. an individual or a corporate body, that is concerned about the result of the project and has the possibility to influence the project. The main goal of the thesis is to map the contemporary approaches toward the stakeholders analysis and management in the ICT projects, to analyze the problems of the influence of individual parts from both technocracy and human (psychological) point of view and to subsequently suggest the methodology for an effective analysis and management of the engaged parts, which could be used at the management of the projects in the ICT field. It is convenient to introduce the used terms and notions , their history and the way of use in the first part. There are dissect the known methodologies and tools used for the analysis and the management of the engaged parts. Subsequently, the stakeholders are systematically divide, it is accomplished their analysis from different perspectives and discussed mainly the aspects which can be crucial in the project management. The most common interests and motivations of the particular engaged parts are presented, including the influence and interest groups and the ways how to procure these parts for the aims of the projects. In the second part a research is described, which is carried out among managers of ICT companies and participants of ICT projects. There are elaborated some hypotheses on grounds of practical experiences from ICT projects and these are verified by this examination. The third part is focused on the presentation of the methodology for the analysis and the management of stakeholders in the ICT projects, which should serve the project managers for the stakeholders analysis and management in the ICT projects. Practical use of this methodology in the project of the SAP system implementation is described in the last, fourth part. This thesis completes relevant sources with the practical experiences at work on the projects in the ICT field. The effected research attested the original notion that this issue is relatively little known in the Czech Republic. The outcomes of this work could be of use for the project management in the ICT field.

The goal of this thesis is to implement a web portal for administration and classification of data from distributed sources. To achieve the goal, it is necessary to test different methods and find the most appropriate one for web articles classification. From the results obtained, there will be developed an automated application for downloading and classification of data from different sources, which would ultimately be able to substitute a user, who would process all the tasks manually.

[ES] La interoperabilidad semántica de la información sanitaria es un requisito imprescindible para la sostenibilidad de la atención sanitaria, y es fundamental para afrontar los nuevos retos sanitarios de un mundo globalizado. Esta tesis aporta nuevas metodologías para abordar algunos de los aspectos fundamentales de la interoperabilidad semántica, específicamente aquellos relacionados con la definición y gobernanza de modelos de información clínica expresados en forma de arquetipo. Las aportaciones de la tesis son: - Estudio de las metodologías de modelado existentes de componentes de interoperabilidad semántica que influirán en la definición de una metodología de modelado de arquetipos. - Análisis comparativo de los sistemas e iniciativas existentes para la gobernanza de modelos de información clínica. - Una propuesta de Metodología de Modelado de Arquetipos unificada que formalice las fases de desarrollo del arquetipo, los participantes requeridos y las buenas prácticas a seguir. - Identificación y definición de principios y características de gobernanza de arquetipos. - Diseño y desarrollo de herramientas que brinden soporte al modelado y la gobernanza de arquetipos. Las aportaciones de esta tesis se han puesto en práctica en múltiples proyectos y experiencias de desarrollo. The contributions of the thesis are: - Study of existing modeling methodologies of semantic interoperability components that will influence in the definition of an archetype modeling methodology. - Comparative analysis of existing clinical information model governance systems and initiatives. - A proposal of a unified Archetype Modeling Methodology that formalizes the phases of archetype development, the required participants, and the good practices to be followed. - Identification and definition of archetype governance principles and characteristics. - Design and development of tools that provide support to archetype modeling and governance. The contributions of this thesis have been put into practice in multiple projects and development experiences. Katechetická práce s dětmi v Královehradecké diecézi CČSH / Katechetical Work with Children in the Hradec Králové Diocese of The Czechoslovak Hussite Church

This thesis is devoted to the topic catechetical work with children in the Czechoslovak Hussite Church. After presenting into context depicts the subject from two point sof wiew. The first part is devoted to theoretical catechetics, which describes how the catechesis of children theoretically takes place. The sekond part shows you how it really takes place in Hradec Kralove diocese of the Czechoslovak Hussite Church. These two parts are equally divided into three sections. By comparing these sections it is concluded that in practice the fundamental katechetice coincides with the theory. In a formal katechetics, basic points also coincide, but practice may inspire theory rather than vice verse. The material part is of the biggest contradictions, especially in materials that Czechoslovak Hussite Church does (not) offer. Mismatch in this material part of catechetics is essential, and unless a consensus is reached again, the problems that the practice faces, cannot be solved.
