Influence of Market Setting and Time of Purchase on Counts of Aerobic Bacteria, Escherichia Coli, and Coliform and Prevalence of Salmonella and Listeria in Beef, Pork, and Chicken in Vietnam

The objective of this study was to determine the influence of market type and sampling time on Salmonella and Listeria prevalence and microbiological quality of 540 beef, pork, and whole chicken samples collected in 6 supermarkets (SM), 6 indoor markets (IM), and 6 open markets (OM) at opening (T0) and 4 h after the opening (T4) in Vietnam. Salmonella and Listeria prevalence ranged from 30.4 to 71.0% and 56.6 to 99.9 %, respectively, in beef, pork, and chicken in Vietnam. Aerobic bacteria counts ranged from 10.5 to 11.6 log CFU/g, whereas, E. coli and coliform counts ranged from 7.2 to 11.4 log CFU/g in beef, pork, and chicken in Vietnam. E. coli counts were influenced by the interaction of market type and sampling time in beef and pork. Market characteristic data that were considered relevant to microbiological safety of fresh meat and poultry products were collected for individual samples.

Effect of irrigation water quality on the microbiological safety of fresh vegetables

Irrigation water is perhaps the leading pre-harvest source of contamination of fresh vegetables in the world. In this thesis, the effect of source water from the Olifants River and the Wilge River on the bacterial quality of water in the Loskop Canal that they feed and also the subsequent contribution to the bacterial contamination of fresh vegetables was determined for a period of twelve months. Also effect of attachment time on the survival of Listeria monocytogenes and the effect of chlorine on L. monocytogenes attached to vegetables were determined. Finally, a step-wise logistic regression analysis was made to determine whether various predictor variables could be used to predict the occurrence of L. monocytogenes, Salmonella spp and intestinal Enterococcus in irrigation water and vegetables (i.e., cauliflower and broccoli). COD and turbidity were higher in the Olifants River and the Wilge River than in the Loskop Canal that they feed, according to the water guidelines set by the World Health Organisation (WHO) and the Republic of South Africa (RSA). The level of the COD and turbidity were significantly different in terms of the two rivers in comparison with the canal. Levels of faecal coliforms and Escherichia coli were also higher than the WHO standard. Staphyloccocus aureus, intestinal Enterococcus, Salmonella, L. monocytogenes were recovered from the two rivers and the canal. Apart from L. monocytogenes that was not recovered from cauliflower, all bacterial pathogens recovered from the surface water were recovered from the vegetables. This study also indicated that L. monocytogenes could attach to both surface and subsurface structures of both tomatoes and spinach within 30 min, and that even after 72 h, it still remained viable. It also indicated that chlorine treatment is more effective against surface L. monocytogenes compared with subsurface inoculated L. monocytogenes. Finally, the logistic regression analysis of the sampled data showed that COD was statistically reliable to indicate a high probability of L. monocytogenes, turbidity reliable to indicate a high probability of intestinal Enterococcus and faecal coliforms and coliforms reliable to indicate a high probability of Salmonella in irrigation water. Étude des Mécanismes de Transfert des Nanoparticules au travers d'une Barrière de Confinement Dynamique / Study of transfer mechanism of nanoparticles across a dynamical air barrier

Les travaux de thèse ont permis de quantifier l'efficacité de confinement de deux dispositifs distincts (un poste de sécurité microbiologique et une sorbonne classique) lors de la production simultanée de nanoaérosols et d'un gaz traceur (SF6). Deux techniques de mesure différentes ont été exploitées : la première basée sur la mesure de la distribution granulométrique de l'aérosol s'échappant (SMPS-C), l'autre reposant sur la détection de fluorescence d'échantillons prélevés (fluorescéine sodée utilisée comme marqueur des nanoparticules). Les résultats ont permis d'établir une forte corrélation entre le comportement d'un nanoaérosol et celui d'un gaz traceur lorsqu'ils sont émis simultanément dans une enceinte ventilée. Plus encore, on a observé une rétrodiffusion gazeuse quasiment deux fois plus importante pour le gaz traceur que pour les nanoparticules testées dans différentes configurations. Le dépôt ainsi que l'agglomération présents dans le cas du transport d'un nuage de nanoparticules peuvent expliquer ces écarts dans le niveau global de confinement obtenu. Cependant, ce constat n'est pas un gage de protection suffisante dans la mesure où il n'existe pas de valeur spécifique de référence lors de l'exposition à des nanoparticules. Il est alors utile de respecter les règles de bonne conduite qui ont été définies dans de nombreux guides INRS ou au travers de multiples études de l'IRSN. En plus de ces études expérimentales, le banc d'essai développé à l'INRS a fait l'objet d'une simulation numérique permettant de valider un modèle eulérien de transport et de dépôt implémenté dans un code de CFD destiné à modéliser le comportement d'un nanoaérosol. Les résultats numériques/expérimentaux sont concordants ; les ordres de grandeur des niveaux de confinement atteints sont comparables / The thesis works have enabled us to quantify the containment efficiency of two devices (a microbiological safety cabinet and classical fume hood) during the simultaneous production of nanoaerosols and a tracer gas (SF6). Two different measurement techniques were used: the first based on the measurement of particle size distribution of the escaping aerosol (SMPS-C), the other based on the detection of fluorescence of samples (sodium fluorescein used as marker of nanoparticles). The results have established a strong correlation between the behavior of a nanoaerosols and the tracer gas when they are emitted simultaneously in a ventilated enclosure. More, we observed that tracer gas back diffusion was almost twice greater than for nanoparticles back diffusion in all the tested configurations. The deposit and the agglomeration present in the case of transport of a cloud of nanoparticles can explain these differences in the overall level of containment. However, this observation does not guarantee sufficient protection since there is no specific reference value for nanoparticle exposure. It is useful to observe the guidelines that have been defined in many INRS publications or through IRSN studies. In addition to these experimental studies, the test-rig developed at INRS has been numerically simulated to validate an eulerian transport and deposition model implemented in a CFD code for modeling the behavior of a nanoaerosol. Hurdles and potentials in value-added use of peanut and grape by-products as sources of phenolic compounds / Desafios e potencialidades na agregação de valor a subprodutos da agroindústria do amendoim e da uva como fonte de compostos fenólicos

Recent studies have demonstrated that peanut and grape processing by-products may be richer sources of bioactive compounds as compared to their original raw material and feedstock; however, before their application as a source of nutraceuticals or in the prevention of lipid oxidation in food systems, certain technological challenges have to be addressed. This study discusses recent advances in the application of plant food processing by-products as sources of phenolic compounds with special emphasis on the profiling and screening of phenolics using high-performance liquid chromatography-mass spectrometry, their potential health benefits, and microbiologial safety. The major findings are summarized in chapters 2, 3, and 4. The first chapter deals with phenolics from grape by-products. In general, insoluble-bound phenolics were more effective in inhibiting copper-induced human LDL-cholesterol oxidation in vitro than free and esterified phenolics. Phenolic extracts from all fractions inhibited peroxyl radical-induced DNA strand breakage. The third chapter brings about the effects of gamma-irradiation on the microbial growth, phenolic composition, and antioxidant properties of peanut skin. Gamma-irradiation at 5.0 kGy decreased the microbiological count of the product. Total phenolic and proanthocyanidin contents, ABTS radical cation, DPPH radical, hydrogen peroxide, and hydroxyl radical scavenging capacities as well as the reducing power of the sample were increased upon gamma-irradiation in both the free and insoluble-bound phenolic fractions. The bioactivity of the free phenolics against in vitro human LDL-cholesterol oxidation and copper induced DNA strand breakage was improved upon gamma-irradiation. Phenolic compounds were positively or tentatively identified and their distribution was in the decreasing order of free > esterified > insoluble-bound forms. Procyanidin dimer A was increased in all phenolic fractions, whereas procyanidin dimer B decreased. Gamma-irradiation induced changes may be explained by molecular conversion, depolymerization, and cross-linking. In the fourth chapter, the ability of selected enzymes in improving the extraction of insoluble-bound phenolics from the starting material (experiment I) or the residues containing insoluble-bound phenolics (experiment II) were evaluated. Pronase and Viscozyme improved the extraction of insoluble-bound phenolics. Viscozyme released higher amounts of gallic acid, catechin, and prodelphinidin dimer A compared to Pronase treatment. Furthermore, p-coumaric and caffeic acids, as well as procyanidin dimer B, were extracted with Viscozyme but not with Pronase treatment. Solubility plays an important role in the bioavailability of phenolic compounds, hence this study may assist in better exploitation of phenolics from winemaking by-products as functional food ingredients or supplements. / Estudos recentes têm demonstrado que subprodutos da indústria processadora de amendoim e uva podem ser mais ricos em compostos bioativos em comparação às suas matérias-primas. No entanto, alguns desafios tecnológicos precisam ser enfrentados antes da sua aplicação como fonte de compostos nutracêuticos ou na prevenção da oxidação lipídica em sistemas alimentares. Este estudo discute os recentes avanços na aplicação de subprodutos da indústria processadora de amendoim e uva como fontes de compostos fenólicos. Especial ênfase foi dada a sua caracterização por cromatografia líquida acoplada à espectrometria de massas, aos potenciais benefícios à saúde e à segurança microbiológica. As principais conclusões estão apresentadas nos capítulos 2, 3 e 4. O primeiro capítulo trata de compostos bioativos de subprodutos da indústria de suco de uva e da produção vinícola. A fração da qual foram extraídos os compostos fenólicos ligados à parede celular foi predominante. Em geral, esta fração também foi a mais eficaz na inibição da oxidação do LDL - colesterol in vitro quando comparada à fração que continha os fenólicos livres e os esterificados. Os compostos fenólicos de todas as frações inibiram o dano oxidativo ao DNA induzido por radicais peroxila. O terceiro capítulo fala sobre os efeitos da irradiação gama sobre a carga microbiana, a composição fenólica e as propriedades antioxidantes da película de amendoim. A irradiação gama (5,0 kGy) diminuiu a contagem microbiana do produto. Os compostos fenólicos totais, o teor de proantocianidinas e a capacidade dos extratos em neutralizar radicais como o ABTS, DPPH e espécies reativas de oxigênio como o peróxido de hidrogênio e radicais hidroxila, assim como o poder redutor da amostra, aumentaram devido à irradiação gama em ambas as frações (contendo fenólicos livres e ligados à parede celular). A bioatividade dos compostos fenólicos livres contra a oxidação do LDL-colesterol in vitro e contra os danos oxidativos ao DNA aumentou com a irradiação gama. Os compostos fenólicos foram positivamente ou tentativamente identificados, distribuindo-se entre: fenólicos livres > esterificados > ligados. Houve aumento na concentração de dímeros de procianidina A em todas as frações, enquanto a concentração de dímeros de procianidina B diminuiu. Essas alterações podem ser explicadas pela conversão molecular, despolimerização e formação de ligações cruzadas. No quarto e último capítulo, enzimas selecionadas foram aplicadas à matéria-prima inicial (experimento I) ou nos resíduos contendo apenas compostos fenólicos insolúveis (experimento II). Pronase e Viscozyme aumentaram a extração de compostos fenólicos insolúveis (ligados à parede celular). Viscozyme liberou maiores quantidades de ácido gálico, catequina e dímero de prodelfinidina A em comparação ao tratamento com Pronase. Além disso, os ácidos p-cumárico e ácido caféico, bem como o dímero de procianidina B, foram extraídos com Viscozyme, mas não com Pronase. A solubilidade desempenha um papel importante na biodisponibilidade de compostos fenólicos. Desta forma, o terceiro estudo oferece uma alternativa para a exploração de compostos fenólicos de subprodutos da indústria vinícola como ingredientes alimentares com propriedades funcionais ou suplementos alimentares.

Sarapatel caprino: valor nutricional e qualidade microbiológica / Goat Sarapatel: nutritional value and microbiological quality

It represents an economical alternative to the use of the by goat slaughter. This product, typical of northeastern Brazil, presents no standardization in its formulation, which can lead to a wide variation in its composition and nutritional quality. The objective of this research was, therefore, to evaluate the microbiological and nutritional of the quality goat sarapatel which is commercialized at different places points and under different storage conditions in the city of João Pessoa, Paraíba, Brazil. For this experiment work, a total of 48 samples of goat sarapatel were analyzed; there were obtained from a variety of places, such as street fairs, butcher shops and supermarkets. As a result of being acquired from different places the condition of each samples varied: some were frozen, others were refrigerated and there were a few under room temperature. The physico-chemical quality of goat sarapatel presented an remarkable nutritional value (each 100g of goat sarapatel presented, on average, 72g of moisture, 2g of ash, 18g of protein, 9g of fat and 2g of carbs).Its fatty acid profile showed a high amount of unsaturated fatty acids, especially oleic, linoleic and linolenic acids. In the amino acid profile analysis, 18 amino acids were identified, especially the amino acids lysine, histidine, leucine, phenylalanine and valine. There amino acids are considered essential, which makes the sarapatel a protein source with a high nutritional value. The samples also presented a high level of cholesterol, with an average concentration of 282mg/100g. The results of the microbiological analyses of the samples revealed the absence of Salmonella spp. and L monocytogenes. However, high levels of total coliform, thermotolerant and Clostridium sulphite reducer were found, ranging from 1,5x104 to >1,1x105 NMP/g; from 2,4x104 to >1,1x105 NMP/g; and from 3,2x103 to 9,8x105 UFC/g, respectively. Positive Staphylococcus coagulase was present in 31.25% of the samples, with level counts ranging from 4,3x104 to 1,2x105 UFC/g. These results show that the samples of goat sarapatel sold in the city of João Pessoa/PB present acceptable nutritional value; however, their microbiological quality is unsatisfactory, which may represent a risk to consumer health. The involvement of regulatory agencies in the production process is therefore necessary, in order to promote health and safety measures and make sure this products is safe for consumption. / O sarapatel caprino é produzido a partir de vísceras e sangue, representando uma alternativa econômica para aproveitamento dos subprodutos do abate caprino. Este produto, típico do Nordeste do Brasil, não apresenta padronização em sua formulação, o que pode gerar grande variação na sua composição e qualidade nutricional. Objetivou-se, assim, avaliar a qualidade microbiológica e nutricional do sarapatel caprino comercializados em diferentes pontos de vendas e sob diferentes condições de armazenamento, na cidade de João Pessoa, Paraíba, Brasil. Para tanto foram analisadas 48 amostras de sarapatel caprino provenientes de feiras-livre, açougues e supermercados, mantidas sob refrigeração, congelamento e temperatura ambiente. A qualidade físico-química do sarapatel caprino mostrou destacado valor nutricional (cada 100g do sarapatel caprino apresentou, em média, 72 g de umidade, 2 g de cinzas, 18 g de proteínas, 9 g de lipídeos e 2 g de carboidratos), além de perfil de ácidos graxos rico em ácidos graxos insaturados, principalmente oléico, linoléico e linolênico. Na análise de perfil de aminoácidos foram identificados 18 aminoácidos, destacando-se lisina, histidina, leucina, fenilalanina e valina, considerados essenciais, sendo, portanto uma fonte proteica de alto valor biológico. As amostras apresentaram ainda, teor de colesterol com concentração média de 282 mg/100 g. Os resultados das análises microbiológicas revelaram ausência de Salmonella spp. e L. monocytogenes nas amostras avaliadas. No entanto foram encontrados elevados valores de Coliformes totais, termotolerantes e Clostridium sulfito redutor, variando entre 1,5x104 a >1,1x105 NMP/g; 2,4x104 a >1,1x105 NMP/g e 3,2x103 a 9,8x105 UFC/g, respectivamente; além da ocorrência de Staphylococcus coagulase positiva em 31,25 % das amostras analisadas, com contagens variando entre 4,3x104 e 1,2x105 UFC/g. Estes resultados mostram que as amostras de sarapatel caprino, comercializadas na cidade de João pessoa/PB, apresentam bom percentual proteico e perfil de ácidos graxos e aminoácidos, embora com elevado teor de colesterol e qualidade microbiológica insatisfatória, podendo representar risco à saúde do consumidor, necessitando de envolvimento de órgãos fiscalizadores com ações que promovam segurança na cadeia de processamento deste produto o caprino derivado.
