The main goal of this thesis is to study and compare electron microscopy images of Nicotiana Occidentalis chloroplasts, obtained from two types of transmission electron microscopes,which work with different accelerating voltage of 80kV and 5kV. The two instruments, TEM JEOL 1010 and low voltage electron microscope LVEM5 are employed for experiments. In the first theoretical part, principle of electron microscopy and chloroplast morphology is described. In experimental part, electron microscopy images of chloroplast under different conditions of sample preparation are shown and discussed.

The thesis is focused on the medium dynamics simulation in the environmental scanning electron microscope. Specifically, it researches (examines) the conical aperture location effect in the differential pumped chamber and the width of the pumping channel in this chamber. The theoretical part deals with the environmental scanning microscope generally and other tools used in this thesis, e.g. ANSYS Fluen software, fluid turbulence, medium free path of molecules and electrons scattering. In the practical part, the work is focused on data processing from the ANSYS Fluent program and on their evaluation.

This diploma thesis deals with the problems of analysis and the subsequent design of a precision manipulator used as a microscope table. The first part od the final thesis deals with the problems associated with the movement of the test sample within the electron microscope. Subsequently, possiblle design variants were proposed, further developing the next steps fot the correctness and functionality of manipulator. The individual force loads of the components were calculated. The result of the work was constructed as a CAD model with subsequent transfer to the assembly drawing.

Kryogenní cela pro studium vodního ledu v mikroskopu ESEM / Cryogenic cell for study of water ice in ESEM microscope

The presented thesis focuses on designing a cryogenic cell for the study of water ice in an environmental scanning electron microscope (ESEM). This cell allows the study ice in the temperature range 80 K – 300 K in a nitrogen gas environment with a pressure of up to 400 Pa. The cell is cooled by a flow cooling system, where liquid or gaseous nitrogen is used as a refrigerant. The cell consists of a double-walled vessel with vacuum insulation, a flow-through heat exchanger, a sample well, and a cooled cell lid. The heat exchanger was designed to be able to dissipate the heat load at the level of 1 W. The exchanger is equipped with an electric resistance heater with an output of approximately 60 W, enabling heating of the sample at speeds of up to 100 K·min-1. The design also includes an LN2 gateway located on the door of the vacuum chamber of the microscope, to which the capillaries of the heat exchanger for the intake and outlet of refrigerant are connected. During the experimental verification of the cryogenic cell in the test vacuum chamber with a pressure of GN2 ~ 400 Pa, the limit temperature of 77.5 K on the sample well was reached.

Návrh automatizovaného procesu elektrolytického leštění vzorků pro elektronový mikroskop / Design of automated electro-polishing apparatus for electron microscopy specimens preparation

This diploma thesis deals with the automation of the electropolishing process, which is per-former as the last step in the preparation of metallographic samples intended for observation in an electron microscope. A complete hardware design of a single-purpose machine has been developed, which provides the automatic preperation of up to six samples per insertion. There was the design of a manipulator for sample handling together with chemically re-sistant sample holder suitable for automatic operation as a part of solution. The design of the whole machine was developed with regard to the safety of the operator. The thesis includes detailed 3D model of the device and the desing of an application for measurement in the LabVIEW. It describes the future working process of the machine, including a description of a software for controlling the machine and sending process data of each sample to the to the database in accordance with the principles of industry 4.0. In the conclusion, the achieved results and the proposal of further steps necessary for the realization of the machine are for-mulated.

PACMan : An automated Chlorophyll-a fluorescence acquisition platform for single cell microalgae analysis

In this thesis a robust Python based software for controlling a Chlorophyll-a Pulse-Amplitude-Modulated (PAM) fluorescence microscope and analysing subsequent data has been developed and validated. The automation software, called PACMan (PAM Automation Control Manager) was made for the purpose of increasing the amount of single cell data generated per experiment. PACMan includes an autofocus algorithm and the ability to vary experimental parameters during experiments. The analysis software, called PAMalysis, processes and facilitates interpretation of PAM experimental data, printing both text files and creating graphical output. PACMan was used on two different phytoplankton species of the Symbiodiniacae family to characterize them under thermal stressors while immobilized on a microfluidic device. The heterogeneity of the phytoplanktons response to increasing thermal stress was evaluated and the best performers under heat stress have been removed using Laser Capture Microdissection for downstream cultivation. PACMan was also used to compare the response of 4 Symbiodiniacae species to increasing relaxation time between light pulses and to image the heterogeneity of response of the common eukaryotic model organism C. reinhartii to a chemical gradient of the common herbicide DCMU (3-(3,4-dichlorophenyl)-1,1-dimethylurea).

Celulózová nanovlákna a materiály v aerosolu / Cellulose Nanofibers and Aerogel Materials

Tato diplomová práce se zabývá přípravou nových kompozitů na bázi mikrofibrilované celulozy a multi-walls carbon nanotubes (MFC/MWCNTs). Tři různé metody byly použity pro modifikaci MWCNTs. Byl zkoumán vliv použitých modifikací na kvalitu MWCNTs suspense. Dále byly připraveny kompozity obsahující MFC a MWCNTs a vliv MWCNTs na mechanické a elektrické vlastnosti a morfologii byl zkoumán. MWCNTs suspense byly charakterizovány za použití UV-VIS spektrofotometrie, Rastrovacího elektronového mikroskopu (SEM) a Termogravimetrické analýzy (TGA). Elektrické vlastnosti byly měřeny pomocí Keithly Electrometer/High Resistivity Meter. Mechanické vlastnosti v tahu byly měřeny za použití Miniature Materials Tester a pro studium morfologie byl použit SEM. Žádný významný vliv MWCNTs na mechanické vlastnosti nebyl pozorován. Hodnoty získané z měření rezistivity vykazují typický perkolační trend a výrazného snížení rezistivity bylo dosaženo přidáním 1 – 2 wt.% MWCNTs. Nicméně hodnoty se velice liší pro horní a spodní stranu téhož vzorku. Ze získaných výsledků vyplývá, že klíčovým krokem ke zdokonalení vlastností materiálu jsou dobrá suspense MWCNTs a také dosažení vzájemné kompatibility obou složek.
