Aim of diploma work is research of plasma influence on seed germination in some plant species (wheat and oat). The complex work includes parts both physical-technological and biological. The work discusses a selection of useful seeds for the plasma treatment and describes parameters of plasma processing. It discusses results of germination testing and early growth of seedlings. Parts of the work are electron microscopy output, data processing result, and appendix.

The system of mixed function oxygenase (MFO system) participates in significant roles in the metabolism of endogenous compounds and xenobiotics. This system contains cytochrome P450, NADPH:cytochrome P450 reductase, and also there are assigned NADH: cytochrome b5 reductase and cytochrome b5. It was proved that cytochrome b5 can stimulate or inhibit cytochrome P450 (CYP)-dependent reactions and even change the ratio of resulting metabolites. The mechanism of cytochrome b5 action has not been fully elucidated yet. Elucidation of protein-protein interactions in MFO system and determination of topology of this system could explain the mechanism of cyt b5 action. The covalent cross- linking technique is suitable method for identifying protein-protein interactions within the membrane. Cytochrome b5 contains 3 methionines and in 2 cases the methionines are localized in a short hydrophobic C-terminal membrane anchor. Interactions with cytochrome P450 in the membrane environment can be identified by substitution of two methionine for photoactivatable analogue of methionine (photo-methionine) and subsequent photoactivation. The diploma thesis Body Modifications in Cyberpunk Literature deals with the representation and role of body modifications in selected works of cyberpunk literature. The issue analysis is carried out by means of combination of quantitative and qualitative approaches. The first part of the thesis covers both areas theoretically and provides essential insight into the issue. In addition, the author develops her own definition of body modification and provides its genesis. In the research part, the author first explores a body of selected literary cyberpunk works obtained through a pilot research, namely novels Neuromancer, Snow Crash, Schismatrix and Blade Runner and short stories collections Mirrorshades and Burning Chrome. Furthermore, the author quantitatively evaluates 503 mentions of body modifications in a given sample, using 25 variables. The qualitative analysis, structured in 7 thematic areas, amplifies the quantitative findings and among other things deals with the role of body modifications in the construction of cyberpunk literary texts and their overlapping with reality. First chapter is devoted to modifications of the stochastic process to the space of continuous functions, modifications of submartingale to the set of right-continuous with finite left-hand limits functions and separable modifications of stochastic process. In the second chapter is the attention on the regularization of random probability measure in Markov kernel focused. In particular, we work with random probability measures on the Borel subset of the Polish space, or Radon separable topological space.

Vitiligo is a skin disease with 2 % prevalence in a worldwide population. It is characterised by loss or decrease in activity of epidermal melanocytes, which lead to skin and hair depigmentation. It has negative impact on psyche, social relationships of patients and reduces the protection of the organism against UV radiation. One of the treatment methods is autologous transplantation of melanocytes or suspension of melanocytes with keratinocytes. Use of the biocompatible membrane, which allows the cultivation of these cells with resulting transplantation on the depigmented lesion, could improve treatment and make it more efficient. The main goal of this work was to create the biocompatible membrane from nanofiber layers of polyvinylalcohol (PVA) which can stand as a carrier for cell transplants in vitiligo therapy. PVA scaffolds were prepared by electrostatic spinning and later on modified by cold methane plasma (CH4) for lowering their hydrofility. Samples of modified nanofiber carriers were analysed according to their physical and chemical characteristics (visualization fiber morphology by SEM, XPS and surface Zeta potential analysis and contact angle). 4 Abstract Phosphodiesterases (PDEs), enzymes that hydrolyze cyclic nucleotides, are important components of signal transduction pathways in eukaryotic cells. Second messenger 3'-5'- cyclic adenosine monophosphate (cAMP) is hydrolyzed by specific PDEs. By controlling concentration levels of cAMP in cell, PDEs preserve favorable environment for successful transmission of the cAMP signal. Moreover, PDEs are activated by protein kinase A (PKA) in response to elevated cAMP concentration, which is a feature crucial for signal termination. PDE8A1 is a high-affinity cAMP-specific IBMX insensitive phosphodiesterase, an enzyme important for cAMP signaling. However, mostly due to a lack of specific inhibitor, its role has not been assessed in detail. This thesis reports cloning of PDE8A1, identification of its posttranslational modifications and subcellular localization, as well as an alternative approach to address PDE biology by the use of cyclase toxin from Bordetella pertussis. The diploma thesis analyses written academic text. Academic prose is frequently characterized as a highly complex style which is structurally elaborated, contains a large number of subordinate clauses and expresses meaning relations explicitly. However, new research (e.g. Biber & Gray, 2010) shows that complexity of academic writing occurs on the level of noun phrases which often contain extensive premodification and/or postmodification. The thesis studies noun phrase structure in research articles from two disciplines: medicine and sociology. Two articles from each discipline were selected, each yielding 50 complex noun phrases. These 200 examples were analysed with respect to their modification, its form and levels of embedding. The results were compared for both disciplines. The aim of the thesis was to describe complex noun phrase structure and identify its relation to the type of academic discipline. Electrospinning is widely used technique to produce nanoscale constructs for tissue engineering. This technique can be used to spin wide range of polymers. One of them is polyvinyl alcohol (PVA), which has very good properties for use in this field. PVA is nontoxic, has good mechanical strength and it's degradable and biocompatible. Electrospun PVA nanofibers have limitations because of their -OH side groups. These groups cause solubility of PVA in water. The solubility can be adjusted with crosslinking techniques, but PVA still remains very hydrophilic, which is causing low adhesion of cells. In recent research we decided to reduce the hydrophilicity of PVA using plasma modification. Polymer modification with cold plasma is an economic and quite simple process to change the surface chemistry without side effects that come with conventional chemical treatment. With radical, formed by discharge, we have deposited hydrocarbons on the PVA surface and we rapidly increased hydrophobicity of the polymer surface. The change of surface chemistry has only a little effect on the fiber morphology. The increase of hydrophobicity allowed better adhesion of mesenchymal stem cells on plasma modified PVA as compared to non-modified PVA and a huge change in cell morphology was observed. Alternative splicing (AS) is involved in expansion of transcriptome and proteome during cell growth, cell death, pluripotency, cell differentiation and development. There is increasing evidence to suggest that splicing decisions are made when the nascent RNA is still associated with chromatin. Here, I studied regulation of AS via chromatin modification with main focus on histone acetylation. First, we demonstrate that activity of histone deacetylases (HDACs) influences splice site selection in 700 genes. We provided evidence that HDAC inhibition induces histone H4 acetylation and increases RNA Polymerase II (RNA Pol II) processivity along an alternatively spliced element. In addition, HDAC inhibition reduces co-transcriptional association of the splicing regulator SRp40 with the target fibronectin exon. Further we showed that histone acetylation reader, Brd2 protein, affect transcription of 1450 genes. Besides, almost 290 genes change their AS pattern upon Brd2 depletion. We study distribution of Brd2 along the target and control genes and find that Brd2 is specifically localized at promoters of target genes only. Surprisingly, Brd2 interaction with chromatin cannot be explained solely by histone acetylation, which suggests that other protein-domains (in addition to bromodomains) are important for...

Title: Analysis of the game performance in relation to different types of the match mini-basketball players. Objectives: The objective of this study is analysis and comparison of the selected performance indicators of the player in relation to the current competiton system in the Czech minibasketball (5x5 vs 3x3 with one basket). Methods: As a research method, the method of observation of the video recordings. Using the basic statistic and arithmetic methods, the measured data were processed statistically and consequently, graphically also. Results: It was discovered that frequency of selected performance indicators is higher , for the whole team as well as individual players in form of modified minibasketball 3x3, in most cases.
