Abstract: In the thesis, methods of the chiroptical spectroscopy (Raman optical activity, electronic and vibrational circular dichroism, circularly polarized luminescence) were utilized to obtain information on structure of chiral molecules. In four main projects, we focused on improving accuracy of quantum-chemical computations used for interpretation of experimental spectra by including anharmonic effects, solvent, molecular flexibility and dynamics. In the first project, the normal mode geometry optimization method was investigated and a suitable frequency limit providing realistic vibrational band broadening was found. Then the ability of harmonic and anharmonic computational approaches to describe the C-H stretching vibrations was explored for three terpene molecules and four spectroscopic methods. In the third project, we estimated the role of dispersion forces and different organic solvents for conformer equilibria and dynamics of cyclic dipeptides containing tryptophan. We focus on the extension of experimental feasability of vibrational optical activity (VOA). We have demonstrated a usability of intensity calibration in the field of Raman optical activity. Advantages for measurements on multiple instru- ments and/or using different configurations have been shown. A new instrumental setup has been developed for microsampling measurements of vibrational circular dichroism spectra with a spatial resolution of 1 mm. Using this technique, spatial inhomogeneities in a sample of protein fibrils have been observed. Epitaxní vrstvy oxidu ceru pro optoelektroniku / Epitaxial films of ceria for opto- electronics

This diploma thesis studies magneto-optical (MO) response of epitaxial thin films of Co-doped ceria. Thin films were characterized by XPS, LEED, STM, spectroscopic ellipsometry and measurement of MO activity. The work focuses on studying MO response of the films depending on film thickness, cobalt concentration, oxidation state of cerium and chemical state of cobalt. Spectra of MO response consist of low energy region where the MO activity is mediated by transitions from defect induced states to conduction band and high energy region where a peak of MO activity appears which we attribute to transitions from valence band to conduction band. In this work we qualitatively explain the effects of the physico- chemical states of the thin films on the structure of the obtained MO spectra, mainly on the appearance of the MO activity in the low energy region, and on the changes of the position of the MO peak. Compared to other preparation methods the epitaxial thin films allow achieving a shift of the MO peak in the direction of higher photon energy.

The terotical part deals with extension of LTC issues and follows up reserch from bachelor thesis. The teory is also focused on the issue of glass optical fibers. The experimental part deals with the influence of selected durability test on transparency of LTC. Transparency is determined by mesuring devise called black box. In the conclusion is summarized influences witch affects transparency and evaluation of whether the selected fibers are suitable for outdoor use.

Studium dohasínajícího dusíkového plazmatu pomocí titrace rtuťových par / Study of nitrogen post-discharge by mercury vapor titration

The aim of this master thesis is a study of nitrogen post-discharge by mercury vapours titration. The nitrogen post-discharge is investigated for many years theoretically as well as for a practical use. The object of this master thesis is a study of kinetic processes ongoing at titrations of mercury vapours during the nitrogen post-discharge at different pressures and applied powers. All experimental data were obtained from an optical emission spectroscopy of nitrogen post-discharge. DC discharge in flowing regime was chosen for measurements. The first part of experiments was carried out at the constant discharge current (100 mA), voltage (1300 V) and wall temperature (300 K). The total gas pressure was varied in range of 500-3000 Pa at nitrogen flow in range of 0.12-0.68 l/min. Nitrogen flow values were arranged to obtain constant nitrogen flow velocity for all gas pressures. The second set of experiments studied power dependencies. The current was varied in the range of 50-200 mA for constant voltage 1300 V. The total gas pressure in this case was 1000 Pa. Mercury vapours were introduced into the system by titration tube at different post-discharge time. The nitrogen pink afterglow effect was well visible at all experimental conditions. This effect corresponds to the maximum intensity of light emission, which expresses as considerable growth of characteristic pink radiation in the post-discharge time. Optical emission spectra of post-discharge were taken in the range of 320-780 nm. Besides three nitrogen spectral systems (first and second positive and first negative), the mercury line at 254 nm was recorded in the second order spectrum at 508 nm under these conditions if mercury was added. This spectral line is excited under post-discharge conditions by collisionally induced resonance energy transfer from nitrogen highly vibrationally excited ground state metastables and it opens an unique technique for their monitoring. The dependence of relative intensities on decay time for mercury spectral line and selected nitrogen spectral systems at different titration positions were measured. The relative intensities of nitrogen bands decrease with increasing of mercury line relative intensity for all total gas pressures. The pink afterglow phenomenon shifts to the later decay times with the increasing of total gas pressure. In the case of experiments at different power, it can be seen that with decreasing power mercury spectral line intensity decreases in post-discharge time. The first detailed tests of the unique detection for highly excited of nitrogen metastables were completed. However this master thesis is concentrated on the basic research which supports better indication of kinetic processes and reactions leading to transformation of excitation energy, this new knowledge should be applied in future also in technologies based on the long-lived metastable induced reactions.

Studium vlivu vlhkosti na celkový sterilizační účinek dielektrického bariérového výboje / Influence of humidity on total sterilisation effect of dielectric barrier discharge

The main subject of this diploma thesis is the study of the effect of humidity on the total sterilization effect of the dielectric barrier discharge. Sterilization is a process which can eliminate all forms of life. The plasma sterilization is one of the methods that are suitable for sterilization of temperature and chemical sensitive materials. This sterilization method was proved to be effective on the wide spectrum of procaryotic and eucaryotic microorganisms. Basically, the main inactivation factors for cells exposed to plasma are heat, UV radiation and various reactive species. Dielectric barrier discharge (DBD) operating at atmospheric pressure was used for the sterilization of the samples. The discharge was generated in dry air and in humid air. The plasma power densities were 2 160 mW.cm-3, 2 279 mW.cm-3 and 2 760 mW.cm-3 (dry air) or 2 326 mW.cm-3 and 2 850 mW.cm-3 (humid air). Humidity of air was achieved using a wash bottle filled with water through which air flowed into the DBD reactor. Fungi spores of Aspergillus niger were used as model microorganisms. Whatman paper No. 1 was used as the carrying medium. When comparing sterilization efficiency of humid and dry air operating at the same conditions, the higher sterilization effect was observed in humid air. The sterilization effect of the DBD generated in air was compared with results obtained during plasma generation in argon and nitrogen. At the same conditions, the highest sterilization effect was observed in argon, followed by humid air, nitrogen and dry air. It was found out that in our experimental setup the active species are probably the main inactivation mechanism. The influence of temperature on the inactivation of microorganisms was completely negligible. The discharge parameters were studied by means of the optical emission spectroscopy (OES). Plasma treated samples were analyzed employing scanning electron microscopy (SEM). Damage of the microorganisms due to the effect of plasma as well as plasma effect on the structure of the carrying medium was evaluated.

Studium utváření elastohydrodynamických mazacích filmů u hypoidních převodů / Study of Elastohydrodynamic Film Formation in Hypoid Gears

This PhD thesis deals with elastohydrodynamic lubrication (EHL) under the conditions that occur between mated hypoid gear teeth. The aim is to describe experimentally the behaviour of lubricating film and mechanisms of its formation. The focus is mainly placed on clarifying the influence of direction and magnitude of sliding velocity in circular and elliptical contacts. Generally, it is still assumed that the direction of sliding velocity has no effect on the resulting lubricant film. In this study optical interferometry is used to determine distribution of lubricant film thickness. This work includes new and original results which clarify the studied problem. It was found that the direction of sliding velocity affects shape of the film at high speeds. This relates to the effect on heat flow through the contact. The results have an impact on the development of general EHL theory and provide knowledge applicable in film thickness prediction in the design of real tribological nodes.

Genetic and Hormonal Regulation of Children's Growth MUDr. Jan Vosáhlo Abstract Growth in childhood is a complex process of changing the body, which can be disrupted by various illnesses including endocrine disorders, particularly growth hormone deficiency. Tumors or other processes affecting hypothalamic-pituitary area can be a postnatal cause of GHD; prenatal causes include 1) developmental disorders of the pituitary as part of complex syndromes, 2) developmental disorders of the pituitary due to defects in regulatory genes and 3) defects in genes involved in the synthesis and secretion of GH. The first topic of the thesis was septo-optic dysplasia - a complex syndrome involving optic nerve hypoplasia, structural brain abnormalities and pituitary dysfunctions. We extensively described phenotype in 11 Czech patients; we observed both complete SOD and incomplete forms variously combining two of the three main components of the syndrome. The cohort then became a part of an international study of 68 patients, in which we studied the phenotype in dependence on the brain morphology. We found correlation between the severity of clinical symptoms and the degree of septum pellucidum abnormities and also a correlation between hippocampus and falx abnormities and neurological symptoms.
