Role of DDE Exposure in Type 2 Diabetes Mellitus: Association with Biochemical Markers and Diabetes Prevalence

Type 2 diabetes mellitus (T2D) is a metabolic disease characterized by hyperglycemia, insulin resistance, dyslipidemia, and beta cell dysfunction. T2D prevalence has been increasing with not all clear causes identified, while the use of synthetic chemicals has risen. Because genetics can only explain a small percentage of disease development, more attention is being given to associations of environmental chemical exposure and disease. Epidemiological evidence suggests environmental exposure to organochlorine compounds (OC) including dichlorodiphenyltrichloroethane (DDT) and its bioaccumulative metabolite dichlorodiphenyldichloroethylene (DDE) is associated with T2D prevalence and is hypothesized to play a role in contributing to T2D. The purpose of this research was to perform anin vitro study of DDE exposure and its effect on liver hepatocyte and pancreatic beta cell functions that regulate biochemical markers implicated in T2D, and determine an association of DDE exposure with T2D from a population exhibiting a high prevalence of T2D living in an area once highly exposed to OC. Human blood samples from diabetics and non-diabetics were analyzed for any significant association of DDE levels with biochemical markers of T2D and T2D presence. The in vitro effect of DDE exposure on the regulation of hepatocyte lipid metabolism and secretion with respect to triglyceride (TG), apolipoprotein B (ApoB), sortilin-1 (Sort-1), and microsomal triglyceride transfer protein (MTP) levels implicated in dyslipidemia was investigated. Finally, the in vitro effect of DDE exposure on the regulation of beta cell insulin secretion with respect to insulin, oxidative stress (ROS), prohormone convertase (PC), and pancreatic-duodenal homeobox-1(PDX-1) levels implicated in beta cell dysfunction was investigated. Based on our results, DDE levels were not associated with an increased risk of T2D prevalence from this study population, although DDE levels were correlated with some biochemical markers of T2D. ApoB secretion, Sort-1 and MTP levels were increased after DDE exposure, while TG accumulation was decreased in hepatocytes. Insulin secretion and PC levels were increased after DDE exposure, while ROS and PDX-1 levels were increased but not significantly. Although no causative association of DDE exposure with T2D prevalence was found, a potential mechanism of DDE’s effect on regulating biochemical markers of T2D was identified.

Seleção de bactérias para biodegradação dos pesticidas organoclorados DDD, PCP e dieldrin / Selection of bacteria for biodegradation of organochlorine pesticides DDD, PCP and dieldrin

O objetivo deste trabalho foi a seleção de bactérias capazes de biodegradar os pesticidas organoclorados dieldrin, DDD e PCP. Inicialmente, foram realizados os ensaios de tolerância visando à seleção das bactérias degradadoras; posteriormente foram realizados os ensaios de biodegradação em meio liquido utilizando a bactéria selecionada. Dentre as 14 linhagens bacterianas isoladas testadas, selecionou-se a linhagem Pseudomonas aeruginosa L2-1 por apresentar maior tolerância a todos os pesticidas. Os ensaios de biodegradação foram realizados em diferentes meios de cultura, variando-se a concentração de glicose, a fonte de nitrogênio e a presença de ramnolipídeo. Os ensaios de biodegradação foram realizados determinando-se a concentração de pesticida, a concentração de glicose, o número de células viáveis, e o pH. O meio de cultura que mais favoreceu a biodegradação dos três pesticidas foi o meio com nitrato de sódio e 0,5% de glicose, obtendo-se biodegradação de aproximadamente 50% para cada pesticida após três dias de incubação. Na ausência de glicose, o meio com nitrato de amônio e 0,1% de ramnolipídeo, favoreceu a biodegradação, obtendo-se após 14 dias de incubação 36,8% de biodegradação de dieldrin; 33,7% de DDD e 42,8% de PCP. De uma forma geral, as taxas de biodegradação pela P. aeruginosa L2-1 foram maiores em menores concentrações de glicose e na presença de ramnolipídeo. Ao alterar a fonte de nitrogênio foram observados resultados diversos sobre a taxa de biodegradação: na ausência de glicose, o nitrato de sódio favoreceu a biodegradação de PCP, enquanto o nitrato de amônio favoreceu a biodegradação de dieldrin, na presença de glicose observou-se o inverso. A taxa de biodegradação do DDD não foi significativamente alterada ao variar a fonte de nitrogênio. A bactéria selecionada P. aeruginosa L2-1 apresentou potencial para biodegradação de pesticidas organoclorados, sendo que as condições nutricionais do meio influenciaram diretamente a biodegradação. / The objective of this work was the selection of bacteria capable of biodegrading the organochloride pesticides dieldrin, DDD and PCP. Among the 14 bacterial isolated strains, Pseudomonas aeruginosa L2-1 was selected due to its greater tolerance to all pesticides. The biodegradation tests were conducted on different culture media, varying the concentrations of glucose, nitrogen source and presence of rhamnolipid. Biodegradation studies were performed by measuring the concentration of pesticide, the concentration of glucose, the number of viable cell and pH during time. The best medium for the biodegradation of all three pesticides was composed of sodium nitrate and 0.5% glucose, giving approximately 50% yield after three days of incubation. In the absence of glucose, the medium containing ammonium nitrate and 0.1% rhamnolipid improved biodegradation yielding, after 14 days of incubation, 36.8% biodegradation of dieldrin; 33.7% DDD and 42.8% of PCP. Obtenção de um efluente sintético do processo de branqueamento da indústria de papel e celulose / Obtainment of a synthetic effluent of the bleaching process in a pulp and paper mill

A necessidade de produção de papel por processo ambientalmente corretos é um grande desafio. A polpa celulósica branqueada gera um dos efluentes mais problemáticos e estudados atualmente. O tipo de efluente na planta de branqueamento é mais importante que a quantidade gerada. O efeito da DBO no corpo receptor desse resíduo não é o principal problema, o problema está na geração dos compostos organoclorados, que estão dissolvidos nesse efluente. Muitas substâncias desse grupo são tóxicas e também podem ter efeitos carcinogênicos e mutagênicos. O estudo desse tipo de efluente em sistemas de tratamento biológico é de extrema importância e é tema de diversos trabalhos científicos. Entretanto, há algumas dificuldades relacionadas com a viabilidade de composição, a obtenção, o manuseio e o armazenamento de amostras desse tipo de efluente em laboratórios de pesquisa. O objetivo desse trabalho foi, portanto, obter um efluente sintético em escala laboratorial, sob condições controladas, que pudesse simular o comportamento de um efluente gerado em uma planta de branqueamento, quando em um sistema de tratamento biológico. Para isso, partiu-se de uma amostra de polpa celulósica não-branqueada e realizou-se a etapa de branqueamento no laboratório sob condições semelhantes às industriais, obtendo-se assim, o efluente desejado, que foi chamado de Efluente Sintético. A comparação foi realizada através da caracterização físico-química deste efluente e de um efluente de uma planta industrial de branqueamento, chamado de Efluente Industrial. Reatores anaeróbios operando em batelada foram utilizados para avaliação do comportamento destes influentes de tratamento biológico. Observou-se que o comportamentos de ambos os efluentes pôde ser considerado semelhante. A avaliação foi feita através do acompanhamento da produção de gás metano e eficiências de remoção de DQO e AOX. / The necessity of producing paper by environmental friendly processes is a big challenge; bleached cellulose pulp produces one of the most problematic and studied kind of effluent nowadays. The kind of effluent produced in the bleaching mill is more important than the quantity of this effluent. The effect of the BOD of this effluent in the receiving water is not the main problem, the problem is on the generation of organochlorines compounds that are dissolved in this effluent. A lot of substances of this group are toxic and can show carcinogenic and mutagenic effects. The study of effluent in biologic systems of treatment is extremely important and is theme of a variety of scientific works. However, there are some difficulties related to the variability of composition, the obtainment, the handling and the storage of samples of this kind of effluent in a research laboratory. Therefore, the purpose of this work was to obtain a synthetic effluent in a laboratory scale, under controlled conditions, that could simulate the behavior of an effluent produced in a bleaching mill, when in a biological system of treatment. For this, it was used a sample of a unbleached cellulose pulp and it was done the stage of bleaching in the laboratory under similar conditions to the mill\'s, obtaining in this way the desired effluent, that was called Synthetic Effluent. It was done a comparison between this effluent and a bleaching mill effluent, that was called Industrial Effluent, by a physical and chemical characterization. It was used anaerobic batch reactors to make a evaluation of the behavior of these effluents in a biological system of treatment. It was observed that the behavior of both effluents can be considered similar. The evaluation was done by the observation of methane gas production and by the efficiencies of COD and AOX removal.

Phytoremédiation des organochlorés. Etude mécanistique et fonctionnelle des capacités épuratrices du système plante-rhizosphère / Organochlorines phytoremediation. Mechanistic and functional studies of the system plant rhizosphere purifying capacitis.

Les organochlorés (OCs) -molécules utilisées dans des procédés industriels et en applications agricoles dont la plupart sont interdites d'utilisation de nos jours- sont considérés comme des polluants très persistants représentant une menace pour les eaux de surfaces et souterraines, le sol et l'atmosphère. Des méthodes alternatives de décontamination de sites pollués aux OCs sont développées utilisant des technologies in situ d'écoremédiation. L'objectif du projet de thèse est d'étudier les capacités de phytoremédiation des OCs par deux plantes modèles, Zea mays et Phragmites australis. Les OCs choisis dans cette étude sont, le lindane (γHCH), le monochlorobenzène (MCB), le 1,4-dichlorobenzène (DCB) et le 1,2,4-trichlorobenzène (TCB). Notre approche a été de considérer l'impact sur les fonctions physiologiques des plantes de chaque OC en mono-exposition, puis de comparer ces mêmes réponses physiologiques en multi-exposition. Ce travail a permis de mettre en évidence un effet synergique de l'action du mélange des OCs. Toutefois, Z. mays et P. australis tolérent des concentrations OCs largement supérieures aux concentrations environnementales mesurées in situ. La phytoextraction des OCs a été démontrée par nos expériences en conditions contrôlées et en utilisant des molécules 14C-OCs. Les OCs sont majoritairement bioconcentrés au niveau des organes souterrains des plantes. Enfin, cette étude montre une adaptation de la microflore bactérienne de la rhizosphère en présence d'un gradient d'exposition aux OCs (multi-exposition). Les approches d'isolement et d'études d'empreintes moléculaires (ARNr 16S, SSCP, pyroséquançage 454) ont permis de détecter des souches potentiellement capables d'assurer une rhizodégradation des OCs. / Organochlorines (OCs) -molecules used in industrial processes and in agricultural applications which are prohibited from use nowadays- are classified as very persistent pollutants and show an environmental risk for surface water and groundwater, soil and atmosphere. To restore polluted sites, alternative methods are developed using ecoremediation technologies in situ. The aim of this thesis is to study the OCs phytoremediation capacities using two model plants, Zea mays and Phragmites australis. OCs slected for this work are, lindane (γHCH), monochlorobenzene (MCB), 1,4-dichlorobenzene (DCB) and 1,2,4-trichlorobenzene (TCB). The work strategy was to compare the effects on plant physiological functions when plants were exposed to each OC (mono-exposure) and to a mixture of all OCs (multi-exposure). This study highlighted a synergistic effect of the OCs mixture. However, Z. mays and P. australis were able to tolerate OCs mixture concentrations much higher than the environmental concentrations measured in situ. The OCs phytoextraction was demonstrated by our experiments under controlled conditions and using 14C-OCs molecules. OCs were mostly bioconcentrated in the underground parts of the plants. This study also demonstrated an adaptation of the rhizospheric bacterial microflora under a gradient of OCs exposures (multi-exposure). The techniques used (16S rRNA, SSCP, 454 pyrosequencing) held to the detection of strains potentially able to provide OC-rhizodegradation.

Os compostos organoclorados causam grande impacto na natureza devido a três características básicas: persistência ambiental, bioacumulação e alta toxicidade. Os mamíferos marinhos estão entre os organismos mais vulneráveis à toxicidade crônica desses contaminantes porque, além de concentrá-los em grande quantidade, a fêmea transfere parte de sua carga ao filhote durante a gestação e a lactação. Assim, o presente trabalho teve como objetivos otimizar uma metodologia para determinação de organoclorados (pesticidas e PCBs) em matrizes gordurosas e verificar a ocorrência dos mesmos na gordura subcutânea de mamíferos marinhos amostrados na costa de São Paulo (Brasil) e na Ilha Rei George (Antártica). No protocolo metodológico otimizado, a extração foi realizada em extrator Soxhlet (8 h) com uma mistura de n-hexano e diclorometano. A etapa de purificação foi feita através de tratamento ácido e o extrato final analisado em cromatógrafo a gás equipado com detector de captura de elétrons (GC-ECD). A performance do método foi avaliada com material de referência certificado, enquadrando-se dentro de padrões internacionais de controle de qualidade. O limite de detecção do método foi em média 2 ng g-1. As análises apontaram DDTs e PCBs como os grupos que mais causam impacto nos cetáceos da costa de São Paulo. Isso refletiu o histórico de ambos no Brasil, tanto em indústria como em agricultura e saúde pública. Em contrapartida, HCHs e HCB não apresentaram concentrações elevadas, fato que pode ser atribuído à volatilidade dos mesmos em regiões de clima tropical. Da mesma maneira, a-clordano, g-clordano e mirex não foram detectados em níveis significativos. A foca de Weddell (Leptonychotes weddelli), habitante do continente antártico, evidenciou as menores cargas de contaminante entre os animais estudados. As toninhas (Pontoporia blainvillei) e o golfinho-nariz-de-garrafa (Tursiops truncatus) também apresentaram baixos níveis de organoclorados. Os botos-cinza (Sotalia fluviatilis) revelaram concentrações de DDT iguais ou superiores a cetáceos da Índia, país onde esse pesticida ainda não está proibido. Já o golfinho-de-dentes-rugosos (Steno bredanensis) mostrou a maior contaminação entre os animais analisados, comparável a espécies estudadas em águas costeiras de países desenvolvidos (onde os organoclorados foram muito utilizados). / Organochlorine compounds cause strong impact on the nature, as a consequence of three basic characteristics: environmental persistence, bioaccumulation, and high toxicity. The marine mammals are one of the most vulnerable organisms to the chronic toxicity of these contaminants. Besides the high concentration in the body, the female transfers part of her load to the offspring during gestation and lactation. The aim of this study was (1) the optimization of a methodology for determining chlorinated hydrocarbons (pesticides and PCBs) in fatty biological matrices and (2) the analysis of organochlorines in marine mammals blubber sampled along São Paulo coast (Brazil) and King George Island (Antarctica). According to the optimized methodology, the extraction was carried out in Soxhlet apparatus (8 h) with a mixture of n-hexane and dichloromethane. The clean-up was carried out with acid treatment and the resulting extract injected into gas chromatography coupled to electron capture detector (GC-ECD). The method performance was evaluated with certified reference material and fitted for international standards of control quality. The mean method detection limit was 2 ng g-1. DDTs and PCBs were the most concentrated organochlorines in the cetaceans from São Paulo coast. These findings reflected their past usage in Brazil by industry, agriculture, and public health. On the other hand, both HCHs and HCB were not found in high concentration likely due to their volatility in tropical climate areas. Mirex, a-chlordane and g-chlordane were not detected in elevated levels. The Weddell seal (Leptonychotes weddelli), from Antarctic continent, presented the smallest load among the studied animals. As the same way, the franciscanas (Pontoporia blainvillei) and the bottlenose dolphin (Tursiops truncatus) presented low organochlorine levels. The marine tucuxi (Sotalia fluviatilis) showed equal or higher DDT concentration than Indian cetaceans where that pesticide is still in use. The rough-toothed dolphin (Steno bredanensis) revealed the greatest contamination among the analyzed animals, comparable to species studied in the coastal waters of developed countries (where organochlorines were extensively used).

O estudo objetivou desenvolver e validar um método para identificar e quantificar poluentes orgânicos persistentes, conhecidos como POPs, em solo de regiões industriais dos municípios de Caieiras e Franco da Rocha SP, via cromatografia a gás acoplada a Espectrometria de Massas (GC-MS) e Detector por Captura de Elétrons (ECD). Em observância ao Tratado de Estocolmo, patrocinado pela Organização das Nações Unidas ONU, que prevê o banimento de pelo menos doze dos POPs, e no qual o Brasil é um dos 113 países signatários, o trabalho visou colaborar positivamente com essa questão ambiental tão importante. Estes compostos são tóxicos e altamente estáveis no ambiente e em organismos vivos, dentre os quais, são abordados nesse trabalho o clordano cis/trans (C10H6Cl8), o heptacloro (C10H5Cl7), o heptacloro epóxido cis/trans (C10H5Cl7) e os isômeros α-, β-, γ- e δ-BHC (C6Cl6). Para garantir a confiabilidade das análises realizadas, foram realizados ensaios de Validação da metodologia, com base nas diretrizes do INMETRO. A técnica de extração utilizada foi o QuEChERS, obtendo resultados de recuperação na faixa de 70 a 120% para a maioria dos compostos estudados, considerados aceitáveis para matrizes complexas. Os limites de detecção e quantificação do método compreenderam a faixa de 0,0002 e 0,01 μg.g1, respectivamente. As amostras analisadas apresentaram contaminação pelos compostos hexaclorobenzeno α-, β-, γ- e δ-, muitas das quais estão acima dos limites máximos permissíveis, de acordo com as legislações nacionais e internacionais vigentes. / The study aimed to develop and validate a method to identify and quantify persistent organic pollutants, known as POPs in soil of industrial regions Caieiras and Franco da Rocha municipalities in São Paulo, via gas chromatography coupled with Mass Spectrometry (GC-MS) and Electron Capture Detector (ECD). In compliance with the Treaty of Stockholm, sponsored by the United Nations - UN, which provides for a ban of at least twelve POPs, in which Brazil is one of the 113 signatory countries, the work aims to contribute positively to the environmental issue as important . These compounds are highly toxic and stable in the environment and living organisms, among which are address in this work chlordane cis / trans (C10H6Cl8), heptachlor (C10H5Cl7), heptachlor epoxide the cis / trans (C10H5Cl7) and the isomers α-, β-, γ- and δ-BHC (C6Cl6). To ensure reliability of the analysis carried out, tests were carried out validation method, based on the guidelines of INMETRO. The extraction technique was used QuEChERS, achieving recovery in the range 70 to 120% for the most of compounds, acceptable for complex matrices. The limits of detection and quantification of the method comprises the range of 0.0002 and 0.01 μg.g-1, respectively. O estudo objetivou desenvolver e validar um método para identificar e quantificar poluentes orgânicos persistentes, conhecidos como POPs, em solo de regiões industriais dos municípios de Caieiras e Franco da Rocha SP, via cromatografia a gás acoplada a Espectrometria de Massas (GC-MS) e Detector por Captura de Elétrons (ECD). Em observância ao Tratado de Estocolmo, patrocinado pela Organização das Nações Unidas ONU, que prevê o banimento de pelo menos doze dos POPs, e no qual o Brasil é um dos 113 países signatários, o trabalho visou colaborar positivamente com essa questão ambiental tão importante. Estes compostos são tóxicos e altamente estáveis no ambiente e em organismos vivos, dentre os quais, são abordados nesse trabalho o clordano cis/trans (C10H6Cl8), o heptacloro (C10H5Cl7), o heptacloro epóxido cis/trans (C10H5Cl7) e os isômeros α-, β-, γ- e δ-BHC (C6Cl6). Para garantir a confiabilidade das análises realizadas, foram realizados ensaios de Validação da metodologia, com base nas diretrizes do INMETRO. A técnica de extração utilizada foi o QuEChERS, obtendo resultados de recuperação na faixa de 70 a 120% para a maioria dos compostos estudados, considerados aceitáveis para matrizes complexas. Os limites de detecção e quantificação do método compreenderam a faixa de 0,0002 e 0,01 μg.g1, respectivamente. As amostras analisadas apresentaram contaminação pelos compostos hexaclorobenzeno α-, β-, γ- e δ-, muitas das quais estão acima dos limites máximos permissíveis, de acordo com as legislações nacionais e internacionais vigentes. / The study aimed to develop and validate a method to identify and quantify persistent organic pollutants, known as POPs in soil of industrial regions Caieiras and Franco da Rocha municipalities in São Paulo, via gas chromatography coupled with Mass Spectrometry (GC-MS) and Electron Capture Detector (ECD). In compliance with the Treaty of Stockholm, sponsored by the United Nations - UN, which provides for a ban of at least twelve POPs, in which Brazil is one of the 113 signatory countries, the work aims to contribute positively to the environmental issue as important . These compounds are highly toxic and stable in the environment and living organisms, among which are address in this work chlordane cis / trans (C10H6Cl8), heptachlor (C10H5Cl7), heptachlor epoxide the cis / trans (C10H5Cl7) and the isomers α-, β-, γ- and δ-BHC (C6Cl6). To ensure reliability of the analysis carried out, tests were carried out validation method, based on the guidelines of INMETRO. The extraction technique was used QuEChERS, achieving recovery in the range 70 to 120% for the most of compounds, acceptable for complex matrices. The limits of detection and quantification of the method comprises the range of 0.0002 and 0.01 μg.g-1, respectively. Thus, this research aimed to determine the concentrations of organochlorine and organophosphorus pesticides present in the sediments of an urban lake located in the Southern region of Brazil, as well as the potential ecological risk and possible sources of contamination according to the use and occupation of the soil through time. Samples of sediment on the surface of the lake were collected every three months, from April 2010 to June 2013, covering the four hydrological stations, in five different collection points, accounting for 65 samples. The methodology used in the extraction, identification and quantification of pesticides was the QuEChERS method. The reading was performed in Gas Chromatograph attached to a mass spectrum. The analyses indicated that organochlorine pesticide contamination dates back to past agricultural activity. Concentrations of organophosphorus pesticides indicate recent and continuous occurrence from urban activities. In general, concentrations of organochlorines rarely indicate a potential ecological risk to organisms in the benthic environment. However, for organophosphates, especially Disulfoton, the risk factor indicates a significant potential adverse effect on organisms in the environment. / O reconhecimento de concentrações residuais de pesticidas no ambiente é importante para compreender as ameaças destes compostos ao meio ambiente e à saúde humana, sobretudo, quando recursos naturais são utilizados para consumo humano. Desta forma, esta pesquisa buscou determinar as concentrações de pesticidas organoclorados e organofosforados presentes nos sedimentos do lago urbano situado na região Sul do Brasil, bem como o potencial risco ecológico e as possíveis fontes de contaminação de acordo com o uso e a ocupação do solo ao longo do tempo. As amostras de sedimentos superficiais do lago foram coletadas a cada três meses, no período de abril de 2010 a junho de 2013, abrangendo as quatro estações hidrológicas, em cinco diferentes pontos de coleta, totalizando 65 amostras. A metodologia utilizada na extração, identificação e quantificação dos pesticidas foi o método QuEChERS. A leitura foi realizada em Cromatógrafo Gasoso acoplado a espectro de massa. As análises indicaram que a contaminação por pesticidas organoclorados se refere às atividades agrícolas no passado. As concentrações dos pesticidas organofosforados indicam ocorrência recente e contínua, proveniente das atividades urbanas. De modo geral, as concentrações dos organoclorados indicam raro potencial risco ecológico aos organismos em ambiente bentônico. Entretanto, para os organofosforados, especialmente o Disulfoton, o coeficiente de risco indica potencial significativo de efeito adverso aos organismos no ambiente.

The biotic and abiotic interactions influencing organochlorine contaminants in temporal trends (1992-2003) of three Yukon lakes: focus on Lake Laberge

Periodic monitoring of contaminant levels in fish from the Yukon Territory indicated that organochlorine (OC) contaminants had rapidly declined since the early 1990s. This study examined OC concentrations, including chlordane (sigma-CHL), sigma-DDT, hexachlorocyclohexane (sigma-HCH), toxaphene (sigma-CHB), sigma-PCB and chlorinated benzenes (sigma-CBz) in sentinel fish (species of consistent annual observation and collection) from two Yukon lakes (Kusawa, Quiet), and from the aquatic food web of a focus lake (Lake Laberge) across several temporal points between 1993 and 2003. OC analysis and phytoplankton counts from dated sediment cores as well as climate data were also collected. Population, morphological (length, weight, age), biochemical (lipid content, delta-13C, delta-15N) and OC contaminant data for fish and invertebrates (zooplankton, snails, clams) were reviewed to elucidate the primary causes for these OC declines. Although some spatial differences in contaminant levels exist between the Yukon lakes, OC concentrations were declining for lake trout in all three lakes, with declines also noted for burbot from Lake Laberge. Several other fish species as well as zooplankton from Lake Laberge exhibited decreases in contaminant levels except northern pike, which registered consistently higher levels from 1993 to 2001. There was no evidence to support the hypotheses of changes in fish trophic levels or food sources with the exception of burbot, which marginally decreased, and northern pike, which climbed a half trophic level. Through OC flux analysis in dated sediments, the hypothesis that declines in abiotic deposition affected the contaminant levels in the food web was also negated. The closure of the Lake Laberge commercial fishery resulted in faster fish growth and larger fish populations, which are contributing to biomass dilution of OC concentrations, higher OC biomagnification factors for some species and likely changes in predator-prey interactions as resource competition increases. The large ratio of OC decreases in the lower vs. higher trophic levels of Lake Laberge have increased food web magnification factors (FWMF) for all six OC groups. It is also suspected that above-average temperatures and below-average precipitation in the lower Yukon region over the 1990s may have contributed towards an increase in lake primary production resulting in biomass dilution of contaminants in zooplankton for all three study lakes. Concurrently, shifts in the Lake Laberge zooplankton community, from climate fluctuations or increased fish predation, have gone from an abundance of Cyclops scutifer in 1993 to dominance by Diaptomus pribilofensis in 2001, although sample sites were limited. Characteristics specific to each species (e.g. body size, composition and metabolism) likely play a role in the significant OC declines measured in zooplankton. Fluctuations in population dynamics, species characteristics and OC contaminant concentrations in the Lake Laberge ecosystem may continue for several years to come. Sentinel species such as lake trout, burbot, whitefish, cisco and plankton should continue to be monitored in all three Yukon lakes for future temporal correlations with contaminants or climate change. / May 2006
