Spatial and Temporal Coordination of oskar mRNA Localization and Translation During Drosophila Oogenesis

In the fruit fly, Drosophila melanogaster, accumulation of osk mRNA at the posterior pole of the oocyte and local translation initiate assembly of the pole plasm, which is required for germ cell formation and posterior patterning of the embryo. I have used fluorescence in situ hybridization (FISH) in combination with immunofluorescence and laser scanning confocal microscopy to examine the spatial and temporal control of osk transcript localization and translation. Drosophila oocytes develop within cysts of 16 interconnected cells. One cell in each cyst differentiates to form the oocyte while the remaining cells form nurse cells that produce RNAs and proteins that are transported to the oocyte. osk mRNA is produced by the nurse cells and accumulates in the oocyte throughout oogenesis, but is only specifically localized to the posterior pole and translated during mid to late oogenesis. My studies help define distinct steps in the osk mRNA localization process. An early step in posterior localization is removal of osk mRNA from most of the cortex, leading to accumulation in the oocyte interior. This process requires microtubules, the microtubule motor protein Kinesin I, the actin binding protein Tropomyosin, and the RNA binding protein Staufen. Transcript then moves from the oocyte interior to the posterior pole through a microtubule independent process. The genes cappuccino, chickadee, spire, armitage, maelstrom, par-1 and gurken are all required for this next step in osk mRNA localization. The final capturing or tethering osk mRNA at the cortex requires an intact actin filament system, but additional components of this anchoring system remain to be identified. I also find that osk mRNA first begins to accumulate at the posterior pole during oogenesis stage 8, but protein is not detectable until stage 9. In addition, grk and par-1 mutations that block osk mRNA localization to the posterior pole and lead to transcript accumulation in the interior do not prevent translation; again, Osk protein production is only observed during stage 9 and later. These observations indicate that posterior localization is neither sufficient nor necessary to trigger osk mRNA translation, which appears to be under tight temporal control.

The religious experience is one among these many existing demands. It is undeniable that Brazil is going through a process of effervescence in this area. And this regards both religious and non-religious people. Due to that scenario, it is necessary to accurately investigate it, expanding and contributing academically with the area of psychology of religion. The Protestant pastor Oskar Pfister is known as an important scholar in the fields of psychology and religion. The aim of this dissertation is the understanding of the relevance and contribution of Oscar Pfister to the field of psychology of religion. In order to achieve that, an intense investigation was taken on the referred author bibliography, as well as on other correlative authors. In the first chapter, it was presented the biography of Oskar Pfister and his main epistemological influences. Its goal is to find out how Pfister builds his thought until his contact with the Freudian psychology. In the second chapter, the contribution of Pfister to the psychology of religion is investigated. Its aim is to suggest some pfisterian approaches to the religious context, as well as his legacy in the field of psychology of religion. In the third chapter, the dialogue between the pfisterian thought and the religious phenomenon is presented. Its purpose is to approximate psychological and religious subjects to each other, and to show how joining them together is crucial to the understanding of the relationship between these two areas. In the final thoughts, the author aims to reclaim the discussion on the so far established relationship, keeping alive the fruitful legacy of Oskar Pfister. / A contemporaneidade traz variadas demandas ao homem. Diante deste cenário se tem a necessidade da investigação criteriosa a partir de referenciais que buscam o balizamento equilibrado e assim expandir os estudos no campo da psicologia da religião. O pastor protestante Oskar Pfister foi um árduo pesquisador no campo, tanto da religião como da psicologia. O objetivo do presente trabalho busca compreender a relevância de Oskar Pfister e sua contribuição ao campo da psicologia da religião. Para tanto, se fez uso de uma extensa investigação bibliográfica do autor e obras correspondentes de outros autores, que serviram de base para aproximações do debate psicologia e religião, isto em três capítulos. No primeiro capítulo, se realizou uma biografia de Oskar Pfister e as principais influências epistemológicas. Tem como objetivo investigar como Oskar Pfister constrói seu pensamento até o encontro com a psicologia freudiana. No segundo capítulo, se pesquisa a contribuição de Oskar Pfister a psicologia da religião. Tem como objetivo, levantar e apontar as aproximações pfisterianas ao contexto religioso, bem como, o legado deixado no campo da psicologia da religião. Die künstlerische Entwicklung des Dresdner Malers Oskar Zwintscher (1870–1916) ist eng mit seinem frei gewählten ,Exil‘ in Meißen verknüpft. Ausgebildet an der Dresdner Kunstakademie zog sich der junge Maler 1892 nach seinem Studium in die nahegelegene kleine Stadt zurück, wo er bis zu seiner Berufung an die Akademie im Jahr 1903 lebte. Trotz dieses Rückzugs hat Zwintscher aber, anders als in der älteren Literatur angenommen, nicht abgewandt von den Entwicklungen seiner Zeit gearbeitet, sondern sich mit modernen Fragestellungen und Maltechniken auseinandergesetzt. Seine Schaffensphase in Meißen war eine äußerst produktive Zeit. Neben Porträts und ersten vielfigurigen Kompositionen entstanden in den 1890er Jahren auch eine Reihe von Landschaften. Die charakteristische Architektur der Stadt taucht in verschiedenen seiner Gemälde auf, wie in den Werken Rote Dächer, Der alte Turm, Mondnacht, Familie und Selbstbildnis mit Tod.

This master thesis presents the character of Job and his reception in German-speaking literature on the basis of three works of the twentieth century, written by Joseph Roth, Oskar Kokoschka and Günter Kunert. The starting point for the literary reception is the biblical Old Testament book Job, whose story, form and circumstances of origin are mentioned in the first part of the work. Following are the performances of individual authors and the interpretation of their works. The core of the work is a comparison of literary reception of the figure of Job in the texts with a biblical original. The subject of the study was, in particular, the question of how the selected authors deal with the biblical substance, how they use Job for their works and which meaning have the comic elements in their texts.
