Estimated dietary exposure to perfluorinated compounds in Canada

Perfluorinated carboxylates (PFCAs), sulfonates (PFSs) and perfluoroalkylsulfonamides (PFOSAs) have been detected in whole blood and serum of non-occupationally exposed humans, yet sources of exposure have not been fully elucidated. The objectives of this study were to estimate dietary exposure to perfluorooctane sulfonate (PFOS), PFCAs (C7--C 11) and fluorotelomer unsaturated carboxylates (FTUCAs) for the general Canadian and Inuit populations prior to the phase-out of perfluorooctyl-sulfonyl production by 3M and voluntary reductions in PFOA emissions under the PFOA stewardship program. PFCs were measured in 65 archived composite food samples prepared for the 1998 Canadian Total Diet Study (TDS) and 68 archived traditional foods from Nunavut using a newly developed methanol extraction combined with a solid phase extraction clean up. Dietary exposure was estimated using food intake data available from studies carried out between 1997 and 1998 in southern Canada and Nunavut. / PFCs were detected in eight composite food samples from the Canadian TDS and in 61 traditional food samples. Elevated concentrations of PFCs were found in caribou liver (6.2+/-5.5 ng/g), ringed seal liver (7.7, 10.2 ng/g), polar bear meat (7.0 ng/g), beluga meat (7.0, 5.8 ng/g), luncheon meats (5.02 ng/g), cookies (2.7 ng/g), processed cheese (2.1 ng/g) and peppers (1.8 ng/g). Low levels of total PFCs (<1.5 ng/g) were measured in 41 traditional foods including: meat (caribou, ptarmigan, snow goose, bearded seal, walrus, black duck), berries, and fish (lake trout, arctic char). PFCs were not detected in beverages, unprocessed meats, breads, cereals and fruits from the TDS composite samples analyzed. / The ranges of estimated daily exposure to PFCs were between 2 and 59 ng-person-1 and 210 to 610 ng-person-1 for average Canadians and Inuit in Nunavut respectively. There were no statistically significant differences in mean PFC exposure levels for different age and gender groups in the general Canadian population. Inuit men in the 41 to 60 year old age group had statistically significantly higher estimated daily exposure to PFCs (p<0.05) than younger men and women from the same age group. This higher exposure was associated with the consumption of beluga muktuk, caribou liver and bearded seal intestine. / Traditional foods contributed a higher percentage to PFC exposure than market foods in all age and gender groups for the Inuit population. In general, caribou meat, arctic char meat and cookies contributed most to dietary exposure for Inuit, with caribou flesh contributing 43 to 75 percent to daily PFC dietary exposure. Dietary exposure for the general Canadian population was associated with the consumption of cakes and cookies, processed cheese, and regular cheese. / Levels of dietary exposure to PFCs estimated in these studies do not pose any significant health risk to either population based on current toxicological information.

A Review of Perfluorooctanoic Acid Carcinogenicity and Application to Human Risk

Indiana University-Purdue University Indianapolis (IUPUI) / Perfluorooctanoic acid (PFOA) is a synthetic organic chemical that consists of an 8 carbon alkyl chain with a terminal carboxyl group in which the carbon-hydrogen bonds have been replaced with carbon-fluorine bonds except at the terminal carboxyl end. This perfluoralkyl carboxylate is a contemporary synthetic chemical that does not occur naturally in the environment and has only seen widespread use within the last 50 years. PFOA is environmentally persistent and is ubiquitously found in human serum. PFOA has been shown to induce a tumor triad consisting of liver adenomas, Leydig cell adenomas and pancreatic acinar cell tumors in male Spraque-Dawley rats. The ability of PFOA to produce tumors in rodents compounded by the fact that PFOA is accumulating not only in those occupationally exposed, but also in the general population, justifies concern about the carcinogenic potential of PFOA in humans. This paper reviews the data from current published research and reveals that some carcinogenic pathways identified in the tumors produced by PFOA in experimental animals may provide a plausible mode of action for human carcinogenesis.

<p>The toxicity of perfluorooctane sulfonic acid (PFOS) and perfluorooctanoic acid (PFOA) was measured. When Caco-2 cells from human adenocarcinoma are cultivated on a filter a monolayer is formed with properties similar to human duodenum epithelium. The Caco-2 cells grown on filter were exposed to the environmental contaminants PFOS and PFOA. The effects on the Caco-2 epithelium were examined by four different methods: trans-epithelial resistance (TEER), leakage of the intracellular protein lactate dehydrogenase (LDH), 14C-mannitol passage through the epithelium and protein content of the epithelium. TEER and C-mannitol passage show the Caco-2 cellmonolayer integrity, LDH leakage gives information of cytotoxicity and protein content of the epithelium shows cell adhension to the filter.</p><p>In the first study TEER decreased at the highest concentrations of PFOS and PFOA (1Mm). The 14C-mannitol passage increased at the highest PFOS concentration. No cytotoxicity was shown and protein-loss was not observed. The second study with PFOS doses of 0, 1, 10, 100 and 500µM and 1 and 10mM showed that the effect of PFOS on TEER was dose-dependent. The 14C-mannitol passage was very high at the highest PFOS-concentration (10mM) and a dose-response was indicated. No cytotoxicity was demonstrated and protein-quantity was not affected. In the third study it was demonstrated that the toxicity of PFOS did not depend on the different concentrations of the oil-emulsion used to dissolve PFOS and PFOA.</p>

Thesis (MTech (Chemistry))--Cape Peninsula University of Technology, 2016. / The release of wastewater to the aquatic environment is most likely to introduce some trace levels of organic contaminants, some of which may be toxic, carcinogenic, or endocrine disruptors, as well as, persistent in the environment. Additionally, increasing contamination of surface waters by wastewater effluents has made water treatment processes more challenging and expensive. The presence of these pollutants in the receiving water body may have negative effects on aquatic species and often pose potential human health risks through the reuse of treated wastewater for drinking purposes and other household use. In countries like South Africa, Namibia, USA, Singapore and Australia, water agencies are intensifying wastewater reclamation/wastewater reuse as part of their water resource agenda: in order to meet the demands of the growing populations. Nowadays, water reuse is generally considered as a viable method of water supply management. This study focused on the identification of the occurrence, quantification of emerging contaminants and evaluation of removal efficiency in wastewater treatment processes of three classes of emerging contaminants (ECs) in wastewater: 1) six types of perfluorinated compounds (PFCs), namely; perfluorooctanoic acid (PFOA), Perfluorooctane sulphonate (PFOS), perfluoroheptanoic acid (PFHpA), perfluorononanoic acid (PFNA), perfluorodecanoic acid (PFDA), and perfluoroundecanoic acid (PFUDA); 2) bisphenol A (BPA); and 3) Acetaminophen (ACP). These contaminants were identified and quantified in four wastewater treatment plants in the Western Cape. These treatment plants include three WWTPs in Cape Town, namely: Bellville WWTP, Scottsdene WWTP and Zandvliet WWTP and one WWTP in the central Karoo (Beaufort West wastewater reclamation plant).

The Cyclodextrin-Perfluorinated Surfactant Host-Guest Complex: Fundamental Studies for Potential Environmental Remediation and Therapeutic Applications

Perfluoroalkyl substances (PFASs) are contaminants of emerging concern, and have been detected in drinking water, wildlife, humans, and the environment. Cyclodextrins (CDs), cyclic sugars composed of glucose monomers, are proposed as a potential remediation strategy. CDs can form host-guest complexes with hydrophobic molecules; this complexation could be capitalized on for PFAS removal and sequestration. These dissertation projects aim to study the fundamental host-guest interactions between a variety of PFASs and CDs for eventual applications in environmental and biological remediation. 1D and 2D Nuclear magnetic resonance (NMR) spectroscopic methods were employed to determine the strength, dynamics, and structure of the CD:PFAS host-guest complexes. Legacy and emerging PFASs were studied with the three native CDs (α-, β-, and γ-CDs) as well as β-CD derivatives. β-CD and its derivatives exhibit the strongest complexation with all studied PFASs, with association constants of 102-105 M-1, depending on PFAS chain length, functional groups, and branching. The host-guest complex was not significantly disturbed under different environmental conditions, such as changing pH, ionic strength, and in the presence of humic acid. A competition study between perfluorooctanoic acid (PFOA), β-CD, and human serum albumin (HSA), the most abundant protein in blood serum, was then conducted using NMR, circular dichroism, and fluorescence spectroscopies. Excess β-CD was able to totally reverse all PFOA binding to HSA. Finally, the host-guest complex was studied within a biological organism to test its viability as a remediation strategy. The attenuation of the toxicity of PFOA in zebrafish embryos, a model organism for toxicology studies, was tested with β-CD. Excess β-CD increased the LC50 (lethal concentration for 50 % of the population) of PFOA compared to PFOA in the absence of β-CD (p < 0.0001). These dissertation projects suggest that the encapsulation of PFASs by CDs has potential in PFAS remediation strategies.

Thesis (DTech (Environmental Health))--Cape Peninsula University of Technology, 2017. / This study represents the first monitoring campaign to assess the seasonal trend of nine perfluorinated compounds (PFCs) in surface water and sediment from the Plankenburg and Diep Rivers in the Western Cape, South Africa. An analytical protocol was developed and validated for qualitative and quantitative routine determination of nine perfluorinated compounds (PFCs), in water and sediment samples using Ultra performance liquid chromatography-mass spectrometry quadrupole time of flight (UPLC-QTOF-MS). This method was applied to determine levels of PFOA and PFOS in environmental samples. Samples were collected along the Diep (Milnerton) and Plankenburg (Stellenbosch) Rivers respectively. Samples were pre-treated, cleaned-up and extracted using solid-phase extraction (SPE) procedures with hydrophilic-lipophilic balance (HLB) C-18 cartridges. Seasonal variation and distribution of PFCs in surface water and sediment was also investigated. Levels of PFCs were monitored in four seasons (summer, autumn, winter and spring) to establish their trend in the environment. The removal of PFOA and PFOS from aqueous solutions using agro-waste biomass of Vitis vinifera (grape) leaf litter was also studied. Activated carbons were produced from the biomass and chemical activation achieved with phosphoric acid (H3PO4) and potassium hydroxide (KOH) for the modification of the carbons’ (AC-H3PO4 and AC-KOH respectively). Activated carbons were characterized using Fourier Transform infrared Spectroscopy (FTIR), Scanning Electron Microscopy (SEM), and Brunauer- Emmett-Teller (BET) in order to understand the removal mechanisms of the contaminants by activated carbons. The effects of solution concentration, pH, adsorbent dosage, contact time, and the temperature were optimized for evaluation of the removal efficiency of the activated carbons. Adsorption isotherm models were used to analyze the equilibrium data obtained and kinetic models were applied to study sorption mechanisms. A fixed bed column study was conducted using: AC-H3PO4 adsorbent. Experimental parameters such as initial concentration of the solution, column bed height, flow rate and initial concentrations of the influent were optimized to establish the best adsorption efficiency parameters of the column system. Breakthrough curve and exhaustion time were predicted using Adam-Bohart, Yoon-Nelson, and Thomas models for the fixed bed column under varying experimental conditions.
