Entre 1890 et 1914, les pionniers de l'aviation entretiennent des liens étroits par le biais des correspondances. A travers elles, ils échangent sur les quatre grandes étapes des débuts de l'aviation : la théorisation, l’expérimentation, la pratique sportive et l'industrialisation de l'aéroplane. Des frères Wright à Roland Garros en passant par les lettres de Clément Ader et d'Otto Lilienthal, les pionniers ont, pour la plupart, produit des écrits intimes dans lesquels ils expriment leurs doutes, leurs ambitions ou encore leurs peurs les plus profondes. En s'intéressant à la sociabilité épistolaire des acteurs de l'aéronautique, l'étude ambitionne de mettre en lumière les constructions imaginaires, les quêtes de reconnaissance, les stratégies de communication, les positionnements sociaux ou encore la guerre des brevets. Il s'agit également de démontrer l'existence des solidarités et des tensions au sein d'un groupe en pleine évolution puisque entre 1890 et 1914, des générations se succèdent entraînant avec elles des quêtes identitaires différentes. Dans cette étude, les représentations socio culturelles de l'aviation sont examinées à la lumière des lettres et des mémoires laissant ainsi au lecteur les moyens de découvrir sous un angle nouveau les débuts palpitants de l'aviation. / Between 1890 and 1914, aviation pioneers maintained close ties through correspondence. Through them, they exchange on the four major stages of the beginnings of aviation: theorization, experimentation, sporting practice and industrialization of the airplane. From the Wright brothers to Roland Garros, through the letters of Clement Ader and Otto Lilienthal, the pioneers have for the most part produced intimate writings in which they express their doubts, their ambitions or their deepest fears. By studying the epistolary sociability of aeronautics actors, the study aspires to highlight imaginary constructions, recognition quests, communication strategies, social positioning and patent warfare. It is also necessary to demonstrate the existence of solidarity and tensions within a group in full evolution, since between 1890 and 1914, generations succeed each other, bringing with them different identity quests. In this study, the socio-cultural representations of aviation are examined in the light of letters and memoirs, leaving the reader the means of discovering from a new angle the thrilling beginnings of aviation.

The TALE Factors and Nuclear Factor Y Cooperate to Drive Transcription at Zygotic Genome Activation

The TALE factors, comprising the pbx and prep/meis gene families, are transcription factors (TFs) vital to the proper formation of anterior anatomical structures during embryonic development. Although best understood as essential cofactors for tissue-specific TFs such as the hox genes during segmentation, the TALE factors also form complexes with nuclear factor Y (NFY) in the early zygote. In zebrafish, Pbx4, Prep1, and NFY are maternally deposited and can access their DNA binding sites in compact chromatin. Our results suggest that TALE/NFY complexes have a unique role in early embryonic development which is distinct from each factor’s independent functions at later stages. To characterize these TALE/NFY complexes, we employed high-throughput transcriptomic and genomic techniques in zebrafish embryos. Using dominant negatives to disrupt the function of each factor, we find that they display similar, but not identical, loss-of-function phenotypes and co-regulate genes involved in transcription regulation and embryonic development. Independently, the TALE factors regulate homeobox genes and NFY governs cilia-related genes. ChIP-seq analysis at zygotic genome activation reveals that the TALE factors occupy DECA sites adjacent to CCAAT boxes near genes expressed early in development and involved with transcription regulation. Finally, DNA elements containing TALE and NFY binding sites drive reporter gene expression in transgenic zebrafish, and disruption of TALE/NFY binding via mutation or dominant negatives eliminates this expression. Taken together, this data suggests that the TALE factors and NFY cooperate to regulate a set of development and transcription control genes in early zygotic development but also have independent roles after gastrulation.

Pathway Pioneer: A Web-Based Graphical Tool for the Organization and Flux Analysis of Metabolic Networks

As stoichiometric metabolic models increase in complexity and delity, design and reconstruction tools are urgently needed to increase the productivity of this time-consuming process. Engineers require software for the exploration, evaluation, and rapid analysis of model alternatives within an intuitive visualization and data management framework. This thesis introduces such a tool: Pathway Pioneer (www.pathwaypioneer.org), a web based system built as a front-end graphical user interface to the ux balance analysis tool COBRA. Pathway Pioneer adds additional functionality for customized network layout and model-engineering collaboration through shared models and model version control. Pathway Pioneer is a dynamic, clickable, browser-based visualization system for metabolic network models retrieved from databases such as BiGG or developed in-house as SBML or XLS compliant les. The user can customize the network layout to visually organize the metabolites and reactions into functional modules. The tool supports zooming and panning, level-of-detail control, ux visualization, keyword searching, and hierarchical subsystem organization. A reaction may be knocked out, set as an objective, looked up in a database or many other operations by a single click on the visualized network. Following each operation the visualization is refreshed with the new ux values. The system supports model revision control to manage alternative network congurations and supports sharing of models and layouts to the broader community. By moving the computationally intensive model analysis from the user computer to remote servers, Pathway Pioneer enables the application of high performance cloud-based resources for greater eciency and scalability. I demonstrate the utility of Pathway Pioneer through application in model reconstruction and analysis of many standard models and also two new models under development: Eukaryotic multicompartment Chinese Hamster Ovary (Cho) cells and in a large-scale Escherichia coli system for bio-manufacturing.

The term pioneer simply means one who pushes ahead to remove obstacles and to prepare the way for others who are to follow. Nevertheless, when we speak of the pioneers in American history, we are really speaking of three different types of men who have only a little in common. All pioneers leave behind them settled communities and highly civilized life and push out into an unknown and. unsettled country;y. Also, all pioneers are seekers, but they do not all seek the same things, Some are seeking adventure. These are the explorers, the pioneers seized with the wanderlust, those who move on and on from one place to another, and, if they live long enough, finally come back; to their friends telling wonderful tales of what they have seen and done. The second group includes those who are seeking for gold, some literally and some figuratively. The miners, the trappers, the hunters might all be classed here. The third group includes those who seek an opportunity to build new homes. These are the pioneers whose work is permanent and most worthwhile. In this sense the Puritans and other early settlers were pioneers of the truest type. They were willing to leave home and friends in order to found a new and better home for themselves and their children. The explorer and gold seeker were necessary for the complete settlement of .the United States. but after all, it was the home seeker who finally conquered the forests and the plains. The home-seeker, to be a good pioneer, must have within him. however, the courage and impetuous desire of the explorer to push forward if he is to be successful. He must be willing to endure any number of hardships and even defeat if he is to carry out his purpose. It is this last type of pioneer whom the modern novelist has sought to portray and interpret, and in this discussion only those novels which deal with such pioneers will be discussed.

This study is a history of a group of Latter-day Saints who travelled from Kirtland, Ohio to Adam-ondi-Ahman, Caldwell County, Missouri during the late summer and early autumn, 1838. The group numbered between five and six hundred when they left Kirtland and had diminished to approximately half that many at the end of their journey.The official name, "Kirtland Camp," was adopted by the group which was originally founded and governed by seven men, i.e. the Seventies Presidency in Kirtland.The camp was organized to assist Church members who otherwise would have been unable to escape persecutions in Ohio. Camp membership consisted of those who were elderly, infirm or lacking necessary finances to flee by themselves. Although sickness, death and dissension plagued the camp members during their exodus, faith, determination and able leadership contributed to the successful completion of the trek.The formulation of Kirtland Camp and the unique nature of its Camp Constitution became the pattern for the organization of later Mormon pioneer exiles under Brigham Young.

But in addition to this, the effort has been made to picture one part of a phase in general Utah history, a phase which followed the original settlement, a phase in which an effort was made to tap the vast mineral and timber resources of the Wasatch Mountains and valleys.
